Аннотация: Obelin from the hydroid Obelia longissima and aequorin are members of a subfamily of Ca2+-regulated photoproteins that is a part of the larger EF-hand calcium binding protein family. On the addition of Ca2+, obelin generates a blue bioluminescence emission (lambda(max) = 485 nm) as the result of the oxidative decarboxylation of the bound substrate, coelenterazine. The W92F obelin mutant is noteworthy because of the unusually high speed with which it responds to sudden changes of [Ca2+] and because it emits violet light rather than blue due to a prominent band with lambda(max) = 405 nm. Increase of pH in the range from 5.5 to 8.5 and using D2O both diminish the contribution of the 405 nm band, indicating that excited state proton transfer is involved. Fluorescence model studies have suggested the origin of the 485 nm emission as the excited state of an anion of coelenteramide, the bioluminescence reaction product, and 405 nm from the excited neutral state. Assuming that the dimensions of the substrate binding cavity do not change during the excited state formation, a His22 residue within hydrogen bonding distance to the 6-(p-hydroxy)-phenyl group of the excited coelenteramide is a likely candidate for accepting the phenol proton to produce an ion-pair excited state, in support of recent suggestions for the bioluminescence emitting state. The proton transfer could be impeded by removal of the Trp92 H-bond, resulting in strong enhancement of a 405 nm band giving the violet color of bioluminescence. Comparative analysis of 3D structures of the wild-type (WT) and W92F obelins reveals that there are structural displacements of certain key Ca2+-ligating residues in the loops of the two C-terminal EF hands as well as clear differences in hydrogen bond networks in W92F. For instance, the hydrogen bond between the side-chain oxygen atom of Asp 169 and the main-chain nitrogen of Arg112 binds together the incoming alpha-helix of loop III and the exiting cc-helix of loop IV in WT, providing probably concerted changes in these EF hands on calcium binding. But this linkage is not found in W92F obelin. These differences apparently do not change the overall affinity to calcium of W92F obelin but may account for the kinetic differences between the WT and mutant obelins. From analysis of the hydrogen bond network in the coelenterazine binding cavity, it is proposed that the trigger for bioluminescence reaction in these Ca2+-regulated photoproteins may be a shift of the hydrogen bond donor-acceptor separations around the coelenterazine-2-hydroperoxy substrate, initiated by small spatial adjustment of the exiting a-helix of loop IV.

Аннотация: Upon binding their metal ion cofactors, Ca2+-regulated photoproteins display a rapid increase of light signal, which reaches its peak within milliseconds. In the present study, we investigate bioluminescence kinetics of the entire photoprotein family. All five recombinant hydromedusan Ca2+-regulated photoproteinsaequorin from Aequorea victoria, clytin from Clytia gregaria, mitrocomin from Mitrocoma cellularia and obelins from Obelia longissima and Obelia geniculatademonstrate the same bioluminescent kinetics pattern. Based on these findings, for the first time we propose a unanimous kinetic model describing the bioluminescence mechanism of Ca2+-regulated photoproteins.

Аннотация: Luciferase of copepod Metridia longa (MLuc) is a naturally secreted enzyme catalyzing the oxidative decarboxylation of coelenterazine with the emission of light. To date, three nonallelic isoforms of different lengths (17-24 kDa) for M. longa luciferase have been cloned. All the isoforms are single-chain proteins consisting of a 17-residue signal peptide for secretion, variable N-terminal part and conservative C-terminus responsible for luciferase activity. In contrast to other bioluminescent proteins containing a lot of aromatic residues which are frequently involved in light emission reaction, the C-terminal part of MLuc contains only four Phe, two Tyr, one Trp and two His residues. To figure out whether Tyr residues influence bioluminescence, we constructed the mutants with substitution of Tyr to Phe (Y72F and Y80F). Tyrosine substitutions do not eliminate the ability of luciferase to bioluminescence albeit significantly reduce relative specific activity and change bioluminescence kinetics. In addition, the Tyr replacements have no effect on bioluminescence spectrum, thereby indicating that tyrosines are not involved in the emitter formation. However, as it was found that the intrinsic fluorescence caused by Tyr residues is quenched by a reaction substrate, coelenterazine, in concentration-dependent manner, we infer that both tyrosine residues are located in the luciferase substrate-binding cavity.

Аннотация: Stability of LSS based on biological regeneration of water, air and food subject to damaging factors is largely dependent on the behavior of the photosynthesizing component represented, mainly, by higher plants. The purpose of this study is to evaluate the tolerance of uneven-aged wheat and radish cenoses to temperature effects different in time and value. Estimation of thermal tolerance of plants demonstrated that exposure for 20 h to the temperature increasing to 45degreesC brought about irreversible damage both in photosynthetic processes (up to 80% of initial value) and the processes of growth and development. Kinetics of visible photosynthesis during exposure to elevated temperatures can be used to evaluate critical exposure time within the range of which the damage of metabolic processes is reversible. With varying light intensity and air temperature it is possible to find a time period admissible for the plants to stay under adverse conditions without considerable damage of metabolic processes. Аннотация: The research on cellulose fiber-reinforced nanocomposites has increased by an unprecedented magnitude over the past few years due to its wide application range and low production cost. However, the incompatibility between cellulose and most thermoplastics has raised significant challenges in composite fabrication. This paper addresses the behavior of plasma-modified polyethylene (PE) reinforced with cellulose nanofibers extracted from isora plants (i.e., isora nanofibrils (INFs)). The crystallization kinetics of PE-INF composites were explained using the Avrami model. The effect of cellulose nanofillers on tuning the physiochemical properties of the nanocomposite was also explored in this work. The increase in mechanical properties was due to the uniform dispersion of fillers in the PE. The investigation on viscoelastic properties confirmed good filler-matrix interactions, facilitating the stress transfer.

Аннотация: The present study demonstrates a simple approach to enhancing thermal stability of butyrylcholinesterase (BChE) by using natural polymers. Analysis of thermal inactivation of the tetrameric BChE in starch and gelatin gels at 50-64 degrees C showed that thermal inactivation followed second-order kinetics and involved two alternating processes of BChE inactivation, which occurred at different rates (fast and slow processes). The activation enthalpy Delta H-# and the activation entropy Delta S-# for BChE in starch and gelatin gels were evaluated. The values of Delta H-# for the fast and the slow thermal inactivation of BChE in starch gel were 61 +/- 3, and 22 +/- 2 kcal/mol, respectively, and the values of Delta S-# were 136 +/- 12 and -2.03 +/- 0.05 cal center dot K-1 center dot mol(-1), respectively. Likewise, the values of Delta H-# for BChE in gelatin gel were 58 +/- 6 and 109 +/- 11 kcal/mol, and the values of Delta S-# were 149 +/- 16 and 262 +/- 21 cal center dot K-1 center dot mol(-1), respectively. The values of the activation parameters obtained in this study suggest that starch gel produced a stronger stabilizing effect on BChE exposed to elevated temperatures over long periods compared with gelatin gel.

Аннотация: We examined the dynamics of the vertical stratification of physical, chemical and biological factors in a brackish meromictic lake with a deep-water chlorophyll maximum (Shira Lake, Russia, Khakasia) during the growing season and estimated how the vertical distribution of these factors influences the vertical distribution of the zooplankton community. The vertical distribution of zooplankton was restricted by the anoxic hypolimnion. Nauplii and younger copepodides (C1-C3) of the copepod, Arctodiaptomus salinus, and the rotifer, Brachionus plicatilis, were found in the upper warm waters. During summer stratification the maximum of A. salinus biomass, which consisted mainly of older copepodides (C4-C5) and females, was associated with the deep-water maximum of biomass of green algae, which are the preferred diet for this species. The vertical distribution of the rotifer Hexarthra oxiuris was bimodal with numerical peaks in the epi- and hypolimnion. Reproduction peaks of dominant species were separated in time. The reproduction peak of A. salinus was at the beginning of summer when A. salinus constituted up to 99% of total zooplankton biomass. The development of rotifers was detected after the reproduction peak of A. salinus when the biomass of rotifers reached 50% of total zooplankton biomass. В© 2007 Springer Science+Business Media B.V.

Аннотация: In studies of fungal bioluminescence, not only fruiting bodies and spores of the fungus, but also samples of luminescent wood, leaf litter or soil may need to be used to derive pure mycelial culture. This study describes an approach to isolating the culture of luminescent fungal mycelium from samples of light-emitting wood found on Borneo Island in November-December 2013. A GelDoc XR Imaging System (Bio-Rad Laboratories, Inc., U.S.) was used for the first time to monitor luminescence and select luminous samples. This study shows that for successful isolation of the culture of luminescent mycelium out of the luminescent wood found in the forest, it is imperative to keep the samples moist (mycelium alive until there is water), while immediate and aseptic delivery of the samples to the laboratory is not a crucial condition (inner layers of wood is "sterile"). Investigation of the growth features of the isolated mycelium in various growing conditions revealed some peculiar properties of its luminescence in comparison with the known luminescent cultures of basidiomycetes. When grown on solid nutrient media, mycelium exhibits low growth rates, long-lasting luminescence (140 days or longer), and emergence and disappearance of local zones with high levels of light emission. Mycelium produced in submerged culture does not emit light, and this effect must be caused by the absence or a very low level of the luminescent reaction substrate in the biomass. The luminescence system isolated from mycelial biomass did not induce luminescent reaction in vitro upon the addition of NADPH (recording intensity is 60 100 URL/sec). We found that enzymes of the luminescence systems isolated from mycelium pellets retained their activity and catalyzed luminescent reaction when a hot extract of the luminous fungus Armillaria sp. (IBSO 2360) was added (near 1900 URL/sec). The same effect was obtained after addition of hot extracts from the fruiting bodies of nonluminous higher fungi Pholiota squarrosa, Cortinarius sp., Hypholoma capnoides and Chroogomphus rutilus (near 3500 URL/sec). The pure culture of luminescent mycelium has been registered in the Culture Collection of IBP SB RAS as IBSO 2371; now it can be used for various in vivo and in vitro studies, including identification of the fungus.

Аннотация: The paper addresses the possibility of controlling the microbial community composition through metabolites produced by microbes. The comparative analysis of experimental data has shown that volatile metabolites make a much greater contribution to the microbial interactions than nonvolatile ones. It has been found that interaction of microorganisms via the volatiles they release occurs frequently and is typical of a number of microorganisms. Volatile metabolites released by microorganisms produce an inhibitory, sometimes bactericidal, effect on the vital functions of bacteria. The stimulating action occurs 6-8 times less frequently. The range of action on the growth of the test cultures and the range of sensitivity to the effect of volatile metabolites of the study microbes have been found to be individual. Comparative cluster analysis of the ranges of action of 100 study cultures has shown that in the investigated set of microorganism species interaction was performed via a set of volatiles of an inhibiting action (82) and of a stimulating action (52). It has been found that release of volatile metabolites by the studied microorganisms depends upon the culture age, concentrations of components of the nutrient medium, and volatile by-products released by other microorganisms. This production can be increased or decreased by the action of volatile metabolites of other microbes. This is related to strain features and culture age. The prospects of using these regulating metabolites depend on the "range", specificity and safety for other members of the microbial community in insufficient concentrations. Volatiles produced by plants and microorganisms as well as by other components of the system - humans and processing equipment installed inside the closed ecosystem - could influence the formation not only of the microbial community but also of the gas composition of the system's atmosphere, through which they could affect the state of the plants. Specially performed experiments have shown that volatile metabolites of microorganisms can accumulate in their habitat, dissolve in the atmospheric water, and maintain their biological activity for days. It has been determined that volatile metabolites of some microorganisms are toxic for plants. Growth of seedling roots is inhibited more than growth of stems. В© 2006 COSPAR.

Аннотация: Kinetics of the triple bioluminescent enzyme system: alcohol dehydrogenase - NADH:FMN-oxidoreductase - luciferase in the presence of quinones and phenols has been studied. The correspondence between the bioluminescent kinetic parameters, redox potentials and concentrations of the quinones and phenols has been estimated. The substances have been shown to change bioluminescent kinetics through moving off the NAD(+)/NADH balance in the enzyme processes. This system is proposed to be used as enzymatic biotest in ecological monitoring. (C) 1998 Elsevier Science Ltd. All rights reserved.

Аннотация: The effects of viscous media with glycerol and sucrose (10–40%) on the kinetics of the bacterial bioluminescent reaction have been investigated by stopped-flow technique. Increment of quantum yield in media with 10% of both osmolytes was shown. Higher concentrations of glycerol, up to 30–40%, were found to reduce the efficiency of the reaction, while this effect was not observed in the media with sucrose. The molecular dynamics simulation was used to study the structure of bacterial luciferase surrounded by either water molecules solely or by mixture of water with various numbers of glycerol/sucrose molecules. It was found that both cosolvents at studied concentrations did not cause a significant change in conformation of bacterial luciferase. The calculated root-mean-square fluctuation for C?-atoms of bacterial luciferase ?-subunit indicated that the higher flexibility of the enzyme mobile loop could be responsible for increment of quantum yield in the presence of 10% of both osmolytes. The active site of bacterial luciferase was found to be accessible for glycerol molecules while sucrose did not enter catalytic gorge. Moreover, at 30 and 40% concentration the glycerol molecules were found to locate in the active site of bacterial luciferase throughout the whole simulation time. © 2020, Pleiades Publishing, Inc.

Аннотация: Using experimental microecosystems the kinetics of phenol disappearance in small forest pond waters (Siberia, Russia) in the summer of 1995-96 were investigated. Despite of high variability of components of the ecosystem (plankton biomass and species composition) and two pronounced 'blooms' of green algae Volvox aureus the same kinetics of the disappearance took place over the investigated period. Half-lives of the pollutant depended on water temperature only. A comparison of the self-purification of the pond with that of the Krasnoyarsk reservoir, 'blooming' with blue-greens was carried out. Half-lives in the pond were significantly lower than that in the reservoir. During the periods of 'blooms' of the green algae in the pond the concentrations of inorganic nutrients were comparatively high and the phenol-degrading bacteria likely were not limited by these nutrients, in contrast to the periods of 'bloom' of the blue-green algae in the reservoir.

Аннотация: We studied soil-like substrate (SLS) as a potential candidate for plant cultivation in bioregenerative life support systems (BLSS). The SLS was obtained by successive conversion of wheat straw by oyster mushrooms and worms. Mature SLS contained 9.5% humic acids and 4.9% fulvic acids. First, it was shown that wheat, bean and cucumber yields as well as radish yields when cultivated on mature SLS were comparable to yields obtained on a neutral substrate (expanded clay aggregate) under hydroponics. Second, the possibility of increasing wheat and radish yields on the SLS was assessed at three levels of light intensity: 690, 920 and 1150 ?mol m-2 s-1 of photosynthetically active radiation (PAR). The highest wheat yield was obtained at 920 ?mol m-2 s-1, while radish yield increased steadily with increasing light intensity. Third, long-term SLS fertility was tested in a BLSS model with mineral and organic matter recycling. Eight cycles of wheat and 13 cycles of radish cultivation were carried out on the SLS in the experimental system. Correlation coefficients between SLS nitrogen content and total wheat biomass and grain yield were 0.92 and 0.97, respectively, and correlation coefficients between nitrogen content and total radish biomass and edible root yield were 0.88 and 0.87, respectively. Changes in hormone content (auxins, gibberellins, cytokinins and abscisic acid) in the SLS during matter recycling did not reduce plant productivity. Quantitative and species compositions of the SLS and irrigation water microflora were also investigated. Microbial community analysis of the SLS showed bacteria from Bacillus, Pseudomonas, Proteus, Nocardia, Mycobacterium, Arthrobacter and Enterobacter genera, and fungi from Trichoderma, Penicillium, Fusarium, Aspergillus, Mucor, Botrytis, and Cladosporium genera. Аннотация: BACKGROUND: P(3HB-co-3HHx) copolymers are very promising biomaterials. The main challenge in the production of these polymers is to simultaneously achieve high cell biomass; high P(3HB-co-3HHx) content; and high molar fraction of 3HHx in P(3HB-co-3HHx). The most common approach to production of these copolymers is the use of recombinant bacterial strains. The purpose of this study was to optimize the process of production of P(3HB-co-3HHx) copolymers containing high molar fractions of 3HHx by using the wild-type strain Cupriavidus eutrophus B10646. RESULTS: Kinetic properties of C. eutrophus B10646 were studied during cultivation of the cells on substrates necessary for P(3HB-co-3HHx) synthesis: glucose, nitrogen, sodium hexanoate, and sodium acrylate. The physiological ranges of their effects were determined experimentally, and C. eutrophus B10646 was grown in culture media with different dosages of these substrates. P(3HB-co-3HHx) copolymers with different molar fractions of 3HHx, including high ones (12 to 68%), were synthesized, and their physicochemical and mechanical properties were investigated. CONCLUSION: For the first time, cultivation conditions of Cupriavidus eutrophus B10646 enabled production of high biomass yields (5-6gL-1) and high content of the polymer (60-75%) that contained high 3HHx molar fraction. By varying the 3HB/3HHx ratio, one can change physicochemical and mechanical properties of P(3HB-co-3HHx) copolymers. © 2014 Society of Chemical Industry © 2016 Society of Chemical Industry.

Аннотация: Ca2+-regulated photoproteins, which are responsible for light emission in a variety of marine coelenterates, are a highly valuable tool for measuring Ca2+ inside living cells. All of the photoproteins are a single-chain polypeptide to which a 2-hydroperoxycoelenterazine molecule is tightly but noncovalently bound. Bioluminescence results from the oxidative decarboxylation of 2-hydroperoxycoelenterazine, generating protein-bound coelenteramide in an excited state. Here, the crystal structures of the Y138F obelin mutant before and after bioluminescence are reported at 1.72 and 1.30 angstrom resolution, respectively. The comparison of the spatial structures of the conformational states of Y138F obelin with those of wild-type obelin gives clear evidence that the substitution of Tyr by Phe does not affect the overall structure of both Y138F obelin and its product following Ca2+ discharge compared with the corresponding conformational states of wild-type obelin. Despite the similarity of the overall structures and internal cavities of Y138F and wild-type obelins, there is a substantial difference: in the cavity of Y138F obelin a water molecule corresponding to W2 in wild-type obelin is not found. However, in Ca2+-discharged Y138F obelin this water molecule now appears in the same location. This finding, together with the observed much slower kinetics of Y138F obelin, clearly supports the hypothesis that the function of a water molecule in this location is to catalyze the 2-hydroperoxycoelenterazine decarboxylation reaction by protonation of a dioxetanone anion before its decomposition into the excited-state product. Although obelin differs from other hydromedusan Ca2+-regulated photoproteins in some of its properties, they are believed to share a common mechanism.

Аннотация: Motivation: Bacterial luciferases are heterodimeric enzymes that catalyze a chemical reaction, so called bioluminescence, which causes light emission in bacteria. Bioluminescence is vastly used as a reporter system in research tools and commercial developments. However, the details of the mechanisms that stabilize and transform the reaction intermediates as well as differences in the enzymatic kinetics amongst different bacterial luciferases remain to be elucidated. Results: Amino acid sequences alignments for 21 bacterial luciferases (both alpha- and beta-subunits) were analyzed. For alpha-subunit, containing the enzyme active center, 48 polymorphic amino acid positions were identified. According to them, the sequences fell into two distinct groups known as slow and fast based on the decay rate of the bioluminescence reaction. The differences in the enzyme active site induced by structural polymorphism are analyzed.
