Аннотация: The circadian rhythm of grazing of microalgae by a laboratory culture of Ceriodaphnia quadrangula under continuous illumination was studied by continuous registration of chlorophyll fluorescence at the outlet of a flow-through cultivator. A culture of green alga Chlorella vulgaris was used as a feed. The data obtained were treated by the statistical spectral analysis. It was found that animals preliminarily grown under a 12 h light : 12 h dark regime and transferred to constant light showed two maxima in the circadian rhythm of grazing with periods of 0.7 and 1.1 h. Animals preliminarily grown under constant light showed no circadian rhythm of grazing. It was concluded that the circadian rhythm of grazing of C quadrangula has endogenous nature and can change according to light conditions.

Аннотация: The relationship between the induction of the luminescent operon of lux-genes fused with the naphthalene and salicylate degradation genes and the inhibition of light emission caused by these compounds was studied. The quantitative correlations between these processes manifest themselves in the fact that light intensity linearly increased in a narrow concentration range of the inductor and then decreased due to the inhibition of the luminescence reaction itself, which is not related to the regulation of expression of lux-genes.

Аннотация: We used a Lake Shira numerical model to estimate the response of the ecosystem of a saline meromictic lake to variations in weather parameters during the growing season. The sensitivity analysis of the model suggests that compared to other external (nutrient inflows) and internal (spring biomasses of food-web components) factors, weather parameters are among the most influential for both mixolimnetic (phyto- and zooplankton) and monimolimnetic (purple sulfur bacteria, sulfur reducing bacteria and hydrogen sulfide) food-web components. Calculations with different weather scenarios shows how changes in the water temperature and mixing depth affect mixolimnetic and monimolimnetic food-web components and the depth of the oxic-anoxic interface in a meromictic lake. When weather forcing stimulates an increase in the biomass of food-web components in the mixolimnion, it produces cascading effects that lead to three results: 1) a higher content of detritus in the water column; 2) a higher content of hydrogen sulfide in the monimolimnion; 3) raising of the oxic-anoxic interface closer to the water-air surface. This cascading effect is complicated by the negative correlation between two light dependent primary producers located at different depths—phytoplankton in the mixolimnion and purple sulfur bacteria at the oxic-anoxic interface. Thus, weather conditions that stimulate higher phytoplankton biomass are associated with a higher detritus content and lower biomass of purple sulfur bacteria, a higher content of hydrogen sulfide and a shallower oxic-anoxic interface. The same weather conditions (higher wind, lower cloud cover, and lower air temperature) promote a scenario of less stable thermal stratification. Thus, our calculations suggest that weather parameters during the summer season strongly control the mixing depth, water temperature and the mixolimnetic food web. An effect of biogeochemical and physical interactions on the depth of the oxicanoxic interface is also detectable. However, intra- and interannual climate and weather effects will be more important for the control of meromixis stability. © 2018, Chinese Society for Oceanology and Limnology, Science Press and Springer-Verlag GmbH Germany, part of Springer Nature.

Аннотация: Upcoming professional sports authorities seek rapid noninvasive biosensing tools for regular monitoring of athletes' physiological states. The analysis of saliva through luminescence-based biosensors has been perceived as a suitable candidate for such purposes. The present study reports a qualitative bioluminescence assay based on a coupled enzyme system that consists of bacterial luciferase (BLuc) and nicotinamide adenine dinucleotide (NADH):flavin mononucleotide (FMN) oxidoreductase (Red), BLuc-Red, for the express diagnostics of athletes' stress levels before and after physical exertion. The volunteers who participated in the study were grouped as freestyle wrestlers and students who adapted to different levels of physical activities. Under physical exertion modelling conditions, the influence of participant saliva on BLuc-Red catalyzed light emission was investigated. Results showed a significant increase in residual luminescence (I-exp, mean maximum bioluminescence intensity of the experimental measurement (I-exp); I-c, luminescence intensity in control; I-exp/I-c, %) values for participants in the wrestler group while a decrease in the student group (P < 0.05). Such contrasting residual luminescence values in both groups were found to be dependent on the catalase activity of saliva. The proposed bioluminescence assay can be utilized as a potential nonspecific biosensing tool for determining the physical state of athletes under high loads.

Аннотация: The structure elucidation and synthesis of the luciferin from the recently discovered luminous earthworm Fridericia heliota is reported. This luciferin is a key component of a novel ATP-dependent bioluminescence system. UV, fluorescence, NMR, and HRMS spectroscopy studies were performed on 0.005 mg of the isolated substance and revealed four isomeric structures that conform to spectral data. These isomers were chemically synthesized and one of them was found to produce light when reacted with a protein extract from F. heliota. The novel luciferin was found to have an unusual extensively modified peptidic nature, thus implying an unprecedented mechanism of action.

Аннотация: The study addresses reflection and spectral-polarization characteristics of forest stands and farm crops obtained under field conditions. The study of the reflection properties of farm crops shows that during the summer plant growing season, the major factors influencing the plant canopy reflectance are morpho-physiological parameters, plant architectonics, solar elevation h0, and viewing angle. The crop reflectance minimum was recorded at viewing angles 25-30В° with respect to the nadir. Coniferous and broadleaf forest stands had similar reflectance spectra of polarized light. The polarized component was smaller for all coniferous stands than for broadleaf ones. For broad-leaved farm crops (wheat and corn), the polarized component of the spectral brightness coefficients had a greater influence on the plant reflectance in the red and near-infrared spectral regions, ?>720nm. Аннотация: In this work we experimentally estimated the capacities of the key components of the Yenisei River (Russia): particulate suspended matter (seston), diatom microalgae, and submerged macrophytes for accumulating Am-241 from water. In our experiments large particles of seston (>8 mu m), comparable in size with diatoms, took up most of americium from water. The accumulation of americium by isolated diatom algae (Asterionella formosa and Diatoma vulgare) was lower than by total seston. The concentration factors (CFs) of Am-241 for seston of the Yenisei River in our experiments were (2.8-6.9).10(5); for diatoms -(1.5-4.2)-10(4). The Us for aquatic plant Elodea canadensis were within the same order of magnitude as those for diatoms. Activity concentration and CFs of Am-241 were nearly the same in experiments under dark and light conditions. This is indicative of an energy independent mechanism of americium uptake from the water by diatoms and submerged macrophytes. (C) 2009 Elsevier Ltd. All rights reserved.

Аннотация: Light-sensitive photoprotein berovin accounts for a bright bioluminescence of ctenophore Beroe abyssicola. Berovin is functionally identical to the well-studied Ca2+-regulated photoproteins of jellyfish, however in contrast to those it is extremely sensitive to the visible light. Berovin contains three EF-hand Ca2+-binding sites and consequently belongs to a large family of the EF-hand Ca2+-binding proteins. Here we report the spatial structure of apo-berovin with bound Mg2+ determined at 1.75 A. The magnesium ion is found in each functional EF-hand loop of a photoprotein and coordinated by oxygen atoms donated by the side-chain groups of aspartate, carbonyl groups of the peptide backbone, or hydroxyl group of serine with characteristic oxygen-Mg2+ distances. As oxygen supplied by the side-chain of the twelfth residue of all Ca2+-binding loops participates in the magnesium ion coordination, it was suggested that Ca2+-binding loops of berovin belong to the mixed Ca2+/Mg2+ rather than Ca2+-specific type. In addition, we report an effect of physiological concentration of Mg2+ on bioluminescence of berovin (sensitivity to Ca2+, rapid-mixed kinetics, light-sensitivity, thermostability, and apo-berovin conversion into active protein). The different impact of physiological concentration of Mg2+ on berovin bioluminescence as compared to hydromedusan photoproteins was attributed to different affinities of the Ca2 +-binding sites of these photoproteins to Mg2+. © 2015 Elsevier B.V. Аннотация: Two-section enclosures were designed for the investigation of the effect of various physicochemical and biological factors on vertical distribution of zooplankton in situ. The framework of the enclosure was a cylindrical polyethylene column without any partitions inside, in which the isolation of animals in different sections after in situ exposure was achieved by pinching the flexible central part of the column. Enclosures were tested at the brackish stratified meromictic Lake Shira (Russia, Khakasia). The absence of fish and carnivorous zooplankton in the lake suggests that the vertical distribution of zooplankton is mainly determined by physicochemical gradients in the water column. Experiments and field observations demonstrated that all age and size groups of Arctodiaptomus salinus and Brachionus plicatilis strongly avoided surface layers during the daylight. The escape of zooplankton from the anoxic hypolimnion was less active. Statistically significant avoidance was observed only for copepodites C4-C5 and females of A. salinus. The relatively simple construction of the columns and easy handling during the experiment were the factors that favoured the use of this device to perform in situ basic tests of the effect of different factors on the vertical distribution of zooplankton.

Аннотация: We synthesized large-area single-crystal graphene sheets to use them in biosensors based on field-effect transistors (FET) for quantitative analysis of interaction kinetics and affinity between the imatinib drug and its target protein kinase Abl1. The G-FET biosensor showed an excellent performance and recognized imatinib at as low as 15.5 fM. The biosensor also showed a linear response to the logarithm of imatinib concentration in the 0.1 pM-10 mu M range. This graphene-based FET biosensor (G-FET) was also applied toquantify Abl1 Y253 F mutation and Abl1 dependency on Mg2+ to bind to imatinib in real-time. Results demonstrated in this work clearly showed that the novel G-FET biosensors are very promising to analyze interactions between proteins and low molecular weight drugs.

Аннотация: The paper presents results and analysis of a study on polarized characteristics of the reflectance factor of different plant canopies under field conditions, using optical remote sensing techniques. Polarization characteristics were recorded from the elevated work platform at heights of 10-18 m in June and July. Measurements were performed using a double-beam spectrophotometer with a polarized light filter attachment, within the spectral range from 400 to 820 nm. The viewing zenith angle was below 20 degree. Birch (Betila pubescens), pine (Pinus sylvestris L.), wheat (Triticum acstivum) [L.] crops, corn (Zea mays L ssp. mays) crops, and various grass canopies were used in this study. The following polarization characteristics were studied: the reflectance factor of the canopy with the polarizer adjusted to transmit the maximum and minimum amounts of light (R-max and R-min), polarized component of the reflectance factor (R-q), and the degree of polarization (P). Wheat, corn, and grass canopies have higher R-max and R-min values than forest plants. The R-q and P values are higher for the birch than for the pine within the wavelength range between 430 and 740 nm. The study shows that polarization characteristics of plant canopies may be used as an effective means of decoding remote sensing data. (C) 2014 Elsevier Ltd. All rights reserved.

Аннотация: This study was performed to investigate water and gas exchange in the experimental model of a closed ecological system (CES) intended for an estimated portion of a human in the long-duration (several-month) experiment. The diversity of the vegetable conveyor in the system was increased. Human wastes were involved in mass exchange processes, and human respiratory function was periodically connected to the experimental model of a CES. The experimental model of a CES was used to quantify regeneration of the gaseous atmosphere with oxygen and carbon dioxide loops by linking the photosynthesizing compartment with the heterotrophic compartment (soil-like substrate) and by the periodic connection of the human respiratory function. Under the preset light and temperature conditions, atmospheric CO2 concentration in the CES model intended for a portion of a human was maintained at a level that neither limited photosynthetic processes nor was harmful to humans (800–2000 ppm) during the 154-day experiment. At the same time, O2 concentration did not either drop below 20.8% or rise above 22.6%. The amount of the evapotranspiration water collected in the system could satisfy 50% of the daily water requirement of a human (with all the water used and excreted by the human being processed and used to irrigate plants). The evapotranspiration water did not need to be additionally purified before being used by humans. Thus, in the experimental model of the closed ecological system, human oxygen and food requirements (per 0.05 portion of a human) were matched to the function of the heterotrophic compartment and the photosynthesizing activity of the multispecies uneven-aged higher plant community. © 2018 IAA

Аннотация: Bioluminescence of isolated enzymes is a perspective phenomenon for biosensors development due to simplicity of registration of a physiological parameter - light intensity. Enzyme-based bioluminescent assay is widely used to evaluate a decrease in biochemical toxicities. Also the enzyme-based assay is used for the direct biochemical monitoring of oxidative toxicity. This work considers antioxidant properties of fullerenols, water-soluble polyhydroxylated derivatives of fullerenes and perspective pharmaceutical agents, in solutions of model inorganic and organic toxicants of oxidative type K-3[Fe(CN)(6)] and 1,4-benzoquinone. Two fullerenol preparations were used: C60O2-4(OH)(20-24) and mixture of two types of fullerenols C60O2-4(OH)(20-24)+C70O2-4(OH)(20-24). The enzyme-based assays showed the peculiarities of the detoxification processes: ultralow concentrations of fullerenols were active (ca 10(-17)-10(-5)g/L); no monotonic dependence of detoxification efficiency on fullerenol concentrations was observed, and detoxification of organic oxidizer solutions was more effective than that of the inorganic oxidizer. The antioxidant effects of highly diluted fullerenol solutions were attributed to hormesis phenomenon; the detoxification was concerned with stimulation of adaptive cellular response under low-dose exposures.
