Stephanodiscus hantzschii Grun. -- chlorophyll a -- efficiency in using photosynthetically active radiation
Аннотация: In the periods of summer and autumn bloom of the Stephanodiscus hantzschii Crun. in recreational water bodies, studies on the vertical distribution of chlorophyll a, its contents per unit biomass, efficiency in using photosynthetically active radiation (E-PhAR), and assimilative activity of microalgae were performed. The results confirmed the existence of two ecophysiological forms of St. hantzschii and provided evidence that both forms are typically autotrophic and can efficiently use low-intensity PhAR for photosynthesis.

Аннотация: The circadian rhythm of grazing of microalgae by a laboratory culture of Ceriodaphnia quadrangula under continuous illumination was studied by continuous registration of chlorophyll fluorescence at the outlet of a flow-through cultivator. A culture of green alga Chlorella vulgaris was used as a feed. The data obtained were treated by the statistical spectral analysis. It was found that animals preliminarily grown under a 12 h light : 12 h dark regime and transferred to constant light showed two maxima in the circadian rhythm of grazing with periods of 0.7 and 1.1 h. Animals preliminarily grown under constant light showed no circadian rhythm of grazing. It was concluded that the circadian rhythm of grazing of C quadrangula has endogenous nature and can change according to light conditions.

Аннотация: The work analyzes functioning of a "producer-consumer" closed aquatic system with spatially separated links, where each component consisted of two species. Producers in the system were the microalgae of Chlorella vulgaris and Scenedesmus sp., consumers - Paramecium caudatum infusoria and Brachionus sp. rotifers. In the experiment the competing predators were consuming on a mixed culture of algae, and the competition of algae was studied under nitrogen limitation. Under these conditions competitiveness of Scenedesmus was higher than that of Chlorella vulgaris. Metabolism products of Scenedesmus algae have been found to have negative effect on reproduction of Paramecium caudatum protozoa. Predator population dynamics in the "consumer" link demonstrated that the rotifers that consume two algal species are more competitive compared to protozoa feeding on chlorella only. (C) 2001 COSPAR. Аннотация: 1. We studied variation in the composition of fatty acids in the seston of a small freshwater reservoir with changes in phytoplankton composition during four growth seasons. We focused on the dynamics of the omega3 fatty acids because of their potential importance for zooplankton nutrition. 2. Total diatoms were related to the 20:5omega3 fatty acid (eicosapentaenoic, EPA) content in seston. Among two dominant diatom genera, Cyclotella was not associated with EPA content. In contrast, there was a significant correlation between Stephanodiscus and the percentage contribution and content of EPA throughout the study. Hence, freshwater diatoms can differ strongly in content of the essential EPA. 3. We considered abundant cyanobacteria as a potential source of 18:3omega3 fatty acid (linolenic, ALA) to aquatic food webs. Among four dominant cyanobacteria species, two (Anabaena flos-aquae and Planktothrix agardhii) showed significant correlation with the ALA content of the seston, while the other two (Aphanizomenon flos-aquae and Microcystis aeruginosa) did not. 4. Dinophyta had a relatively high level of 22:6omega3 (docosahexaenoic, DHA) for freshwater species and can be also a source of EPA to aquatic food webs. 5. Our results show that various species of diatoms as well as cyanobacteria can be of contrasting nutritional value for zooplankton because of their different content of the essential PUFAs. Diatoms, which are low in EPA, could not be considered as a valuable food, while some field populations of cyanobacteria might be valuable sources of essential ALA.

Аннотация: 1. We studied the composition of fatty acids (FAs) in the seston from two small freshwater reservoirs (Bugach and Lesnoi) with distinct periodicity of domination by cyanobacteria and eukaryotic algae during the growth season. 2. The diatoms in the both reservoirs were characterised by a high content of 14:0 and C16 unsaturated acids, whereas that of the essential FA 20:5omega3 [eicosapentanoic acid (EPA)] was low. The correlation between this polyunsaturated FA (PUFA) and diatom biomass was not significant in either reservoir. The percentage of 20:5omega3 in seston significantly correlated with the biomass of euglenophyta in Bugach and dinophyta in Lesnoi. Hence the diatoms, usually referred as a valuable food for zooplankton, were not an important source of the essential PUFA in these systems. 3. The dominant cyanobacteria in Bugach, and the green algae in Lesnoi, both contained the same marker acids: 18:3omega3 and 18:2omega6. Hence, a discrimination between these two phytoplanktonic groups on the basis of FA biomarkers may be difficult in some cases. 4. We found no significant correlation between the content of 20:5omega3 in seston and the biomass of the dominant daphniids in either reservoir. This is contrary to expectations, based on the literature, that EPA is generally important. Rather, the biomass of the two dominant Daphnia species in Bugach correlated strongly with the content of 18:3omega3 in the seston. The cyanobacteria were a probable source of this omega3 FA for Daphnia . We conclude that EPA is not always important for Daphnia populations although, in such cases, some other PUFA (e.g. 18:3omega3) might be related to their growth.

Аннотация: The effect of ambient temperature on the composition of intracellular fatty acids and the release of free fatty acids (FFA) into a medium by cyanobacterium Spirulina platensis and eukaryotic microalgae, Chlorella vulgaris and Botryococcus braunii, was studied using their batch cultures. It was found that all the species studied, regardless of their taxonomic status, responded to the temperature regime by similar changes in their intracellular fatty acid composition: the relative content of more unsaturated fatty acids decreased with the elevation of temperature. At the same time, in the prokaryote, this temperature shift blocked, first of all, the elongation of 16:0 to 18:0 and then their further desaturation. In eukaryotes, the change in the desaturation of dienoic to trienoic fatty acids was the most pronounced process. The ratio of dienoic to trienoic fatty acids remained almost unchanged in S. platensis. The relative content of extracellular unsaturated FFA increased in the prokaryotic organism S. platensis at a higher temperature. But no significant changes in the composition of extracellular unsaturated FFA were detected in eukaryotic algae upon temperature elevation.

Аннотация: The use of relative variable fluorescence (RVF) of chlorophyll, as measured in the presence of Diuron, an inhibitor of electron transfer, for the estimation of the photosynthetic activity of plankton microalgae was analyzed under a wide range of light intensities in the PAR region. Oxygen evolution rates (estimated by the method of light and dark bottles and the amperometric method), RVF, and chlorophyll a concentration were measured in parallel in natural algal cenoses and microecosystems. When the previously used regression equation, in the form A = b(DeltaF/F-d)CchlI, where A is O-2 evolution rate (g/(m(3) h), DeltaF/F-d is RVF (relative units), C-chl is chlorophyll a concentration (mg/m(3)), and I is light intensity (W/m(2)), was verified in the PAR region, we observed a nonlinear dependence of the correction coefficient b on I, which can be described by the formula b = 6.227 x 10(3)rootI. This result agrees with the hypothesis that chlorophyll a fluorescence quenching comprises photochemical (qQ) and energy (qE) components. On the basis of the energy model, we determined the upper limit b(max) = 0.003 for light intensity range I < 4.4 W/m(2) and the lower limit b(min) = 0.0003 for I = 400 W/m(2).

Аннотация: We have reviewed foreign and domestic literature devoted to the study of the amino acid (AA) composition of aquatic organisms representing major groups of producers (green microalgae and diatoms, and cyanobacteria) and primary consumers (zooplankton). Based on published data, we estimate the composition of essential and nonessential AAs of microalgae, cyanobacteria, and zooplankton and determine their differences. It is concluded that the AA composition of major groups of plankton is heterogeneous. The role of AAs as a limiting factor for the development of herbivorous zooplankton is discussed. The prospects and the need for further study of AA composition in order to develop a complete theory of functioning of aquatic ecosystems have been demonstrated.

Аннотация: Conservation and sustainable management of aquatic ecosystems is a priority in environmental programs worldwide. However, these aims are highly dependent on the efficiency, accuracy and cost of existent methods for the detection of keystone species and monitoring of biological communities. Rapid advances in eDNA, barcoding and metabarcoding promoted by high-throughput sequencing technologies are generating millions of sequences in a fast way, with a promising cost reduction, and overcoming some difficulties of the traditional taxonomic approaches. This paper provides an updated broad perspective of the current developments in this dynamic field presented in the special session (SS) "The use of molecular tools in ecological and biodiversity assessment of aquatic ecosystems" of the XIX Congress of the Iberian Association of Limnology (AIL2018), held in Coimbra, Portugal. Developments presented are mainly focused on the Iberian Peninsula (Portugal and Spain, including Atlantic Macaronesian islands) but include studies in France, Germany, Finland, Russia (Siberia) and South America. The networks within which these researchers are involved are yet even broader, profiting from existing molecular facilities, and traditional taxonomic expertise, which can be viewed as a characteristic of this new research area. It was evident in the SS that the use of molecular tools is widespread, being used to study a diversity of aquatic systems, from rivers' headwaters to estuaries and coastal lagoons, and volcanic, mountain and frozen lakes to hot springs. The organisms targeted are likewise varied and include fish, macroinvertebrates, meiofauna, microalgae such as diatoms and dinoflagellates, other protists, fungi, and bacteria (cyanobacteria and other). Some studies address the whole biodiversity (i.e., all species present independently of the taxonomic group) from environmental samples of water, biofilms and preservative solution from field samples (e.g., ethanol from macroinvertebrate samples). Great advances were acknowledged in the special session, namely in the use of metabarcoding for detecting hidden biodiversity, juvenile stages, low-abundance species, non-indigenous species and toxicity potential, and ultimately for ecological monitoring of diatoms and invertebrates. Yet, several drawbacks were highlighted and need further work, which include: taxonomic gaps in the reference databases (including gaps at species level and on intraspecific variability) or absence of public databases (e.g. for meiofauna), still high sequencing costs, the need of a substantial bioinformatics effort, difficulties in establishing the amount of environmental sample necessary for a good DNA extraction and the need for testing different genetic markers to obtain accurate results.

Аннотация: Abstract: We have reviewed foreign and domestic literature devoted to the study of the amino acid (AA) composition of aquatic organisms representing major groups of producers (green microalgae and diatoms, and cyanobacteria) and primary consumers (zooplankton). © 2019, Pleiades Publishing, Ltd.

    Comparison of rapid methods used to determine the concentration, size structure and species composition of algae / E. S. Zadereev, A. V. Drobotov, T. S. Lopatina [и др.] // J. Sib. Fed. Univ. - Biol. - 2021. - Vol. 14, Is. 1. - С. 5-27, DOI 10.17516/1997-1389-0338 . - ISSN 1997-1389

Flow cytometry -- Fluorescence -- Microscopy -- Particle counter -- Phytoplankton -- Size distribution
Аннотация: Traditionally, the abundance, cell size distribution and species identification of algae are determined by microscopic counts. In recent years, various rapid methods have been developed for routine algal studies. However, each of these methods has its drawbacks. It is important for aquatic ecologists to understand the advantages, disadvantages, and limitations of these methods. We compared the sensitivity of three rapid methods (multichannel fluorimeter FluoroProbe, imaging flow cytometer FlowCam, and CASY particle counter) to changes in cell abundance of three algae species (Chlorella vulgaris Beyerinck, Arthrospira platensis Gomont, and Nostoc sp.). We also assessed the ability of rapid methods to estimate the cell abundance of different species in the mixed samples. All instruments showed high sensitivity to changes in the cell abundance of different algae species and a mixture of these species. Any one of these methods, once calibrated, can be reliably used to estimate the abundance of a single-species/laboratory culture of microalgae. At the same time, FlowCam, without preliminary calibration, recorded the cell abundance closest to microscopic counts. When analysing a mixture of three microalgae differing in their cell sizes and spectral characteristics, FluoroProbe showed the highest accuracy in assessing the proportions of species in the mixture and FlowCam - in assessing their abundance. To study mixtures of algae and/or natural phytoplankton communities, it is advisable to use jointly a flow cytometer and a multichannel fluorimeter. The images of algae saved by the flow cytometer, if necessary, can be used to identify them, with a certain accuracy, to the species. Information on cells size and spectral characteristics obtained by two methods will be detailed enough to perform such common tasks as studying trophic interactions between phyto- and zooplankton or creating warning systems to inform of unwanted blooms of phytoplankton and their individual groups (for example, cyanobacteria). © Siberian Federal University. Аннотация: Long-chain alkenones (LCAs)-lipides produced by some microalgae of the Haptophyta group in seas and continental water bodies-are promising paleomarkers of climate changes because they are well-preserved in bottom sediments. The hydrocarbon chain and the degree of unsaturation of double bonds of this class of lipides can vary depending on habitat conditions of their producers. For the first time, we have discovered LCAs in the bottom sediments of two saline meromictic lakes Shira and Uchum located in arid steppes of southern Siberia (the North Minusinsk Depression) and estimated the distribution of their total content, length, and the degree of unsaturation. The highly abundant population of producers of LCAs-haptophyte algae of the genus Isochrysis (Haptophyta)-is revealed in the water column of Lake Uchum. It is shown that the species composition of the haptophyte algae and composition of the LCAs in the lakes under study are similar to those in saline stratified lakes of North America (Saskatchewan, Canada) with a similar climate and salt composition. The abundance of the C37:4 alkenone in the bottom sediments in the last 100 years reflected the variations in lake salinity caused by the documented changes in the water level. In this way, it is shown that the LCAs may be used as a paleomarker of climate-driven changes in the water level of the saline lakes in southern Siberia.

Аннотация: A "producer-consumer" (Chlorella vulgaris-Paramecium caudatum) closed aquatic system has been investigated experimentally and theoretically. It has been found that there is a direct relationship between the growth of the paramecia population and their release of ammonia nitrogen, which is the best form of nitrogen for Chlorella growth. The theoretical study of a model of a "producer-consumer" aquatic biotic cycle with spatially separated compartments has confirmed the contribution of paramecia to nitrogen cycling. It has been shown that an increase in the concentration of nitrogen released as metabolites of paramecia is accompanied by an increase in the productivity of microalgae. (c) 2005 COSPAR. Аннотация: The possibility of symbiotic, mutualistic (++) interrelations between epibiontic microalgae (Characidiopsis ellipsoidea, Colacium vesiculosum) and cyclopoids (Cyclops vicinus, Mesocyclops leukarti) in the Bugach recreational reservoir with a bloom of cyanobacteria is demonstrated. Overgrown cyclopoids, compared to those free from microalgae, have advantages in oxygen supply, which may be insufficient in the period of intensive algal bloom. Calculations have shown that oxygen requirements of cyclopoids may be fully satisfied due to C. vesiculosum photosynthetic activity in the dysphotic zone if the density of overgrowth exceeds 1000 algal cells per individual. There is no significant correlation between the natural mortality of crustaceans and the indices of their overgrowth by microalgae.
