Аннотация: The effects of elevated temperature (35 and 45В°C) on photosynthesis, respiration, and both the qualitative and quantitative compositions of volatile emissions (VE) of wheat (Triticum aestuvum L. cultivar 232) cenoses at light intensities of 70, 150, or 240 W/m2 of photosynthetically available radiation (PAR) were studied. At a PAR of 240 W/m2, the thermal stabilities of photosynthesis and respiration increased at 35В°C and decreased at 45В°C. Elevated temperatures nonuniformly changed the rates and direction of VE syntheses. In this process, the highest increase in VE evolution was observed at 70 W/m2 and 35В°C; the lowest, at 240 W/m2. In addition, the concentrations and composition of VE during the repair period differed from the initial values.

Аннотация: The peroxidase and catalase activities in the mycelium of luminous basidiomycetes Armillaria borealis and Neonothopanus nambi in normal conditions and under stress were compared. An increase in the luminescence level was observed under stress, as well as an increase in peroxidase and catalase activities. Moreover, the peroxidase activity in extracts of A. borealis mycelium was found to be almost one and a half orders of magnitude lower, and the catalase activity more than two orders of magnitude higher in comparison with the N. nambi mycelium. It can be suggested that the difference between the brightly luminescent and dimly luminescent mycelium of N. nambi is due to the content of (HO2)-O-2 or other peroxide compounds.

?-ketothiolase -- Controlled synthesis -- Poly(hydroxybutyrate-co-hydroxyvalerate) -- Ralstonia eutropha -- Bacteria (microorganisms) -- Ralstonia -- Wautersia eutropha
Аннотация: The paper deals with the study of the synthesis of 3-hydroxybutyrate (3HB) and 3-hydroxyvalerate (3HV) copolymers by the bacterium Ralstonia eutropha B-5786 grown under different carbon nutrition conditions (growth on carbon dioxide, fructose, and CO2-valerate and fructose-valerate mixtures). The parameters to be analyzed included the yield of biomass; the yield, synthesis rate, and composition of copolymers; the activity of the key enzymes of polyhydroxyalkanoate (PHA) synthesis ?-ketothiolase, acetoacetyl-CoA reductase, and PHA synthase); the maximum tolerable concentration of valerate to the bacterium; and the conditions that govern the incorporation of hydroxyvalerate into copolymers. This allowed the relationship between cultivation conditions and the proportion of monomers in the copolymers to be deduced. We were able to synthesize a range of 3HB/3HV copolymers and found that the thermal characteristics and the degree of crystallinity of such copolymers depend on the molar fraction of 3HV. Аннотация: The paper addresses the possibility of controlling the microbial community composition through metabolites produced by microbes. The comparative analysis of experimental data has shown that volatile metabolites make a much greater contribution to the microbial interactions than nonvolatile ones. It has been found that interaction of microorganisms via the volatiles they release occurs frequently and is typical of a number of microorganisms. Volatile metabolites released by microorganisms produce an inhibitory, sometimes bactericidal, effect on the vital functions of bacteria. The stimulating action occurs 6-8 times less frequently. The range of action on the growth of the test cultures and the range of sensitivity to the effect of volatile metabolites of the study microbes have been found to be individual. Comparative cluster analysis of the ranges of action of 100 study cultures has shown that in the investigated set of microorganism species interaction was performed via a set of volatiles of an inhibiting action (82) and of a stimulating action (52). It has been found that release of volatile metabolites by the studied microorganisms depends upon the culture age, concentrations of components of the nutrient medium, and volatile by-products released by other microorganisms. This production can be increased or decreased by the action of volatile metabolites of other microbes. This is related to strain features and culture age. The prospects of using these regulating metabolites depend on the "range", specificity and safety for other members of the microbial community in insufficient concentrations. Volatiles produced by plants and microorganisms as well as by other components of the system - humans and processing equipment installed inside the closed ecosystem - could influence the formation not only of the microbial community but also of the gas composition of the system's atmosphere, through which they could affect the state of the plants. Specially performed experiments have shown that volatile metabolites of microorganisms can accumulate in their habitat, dissolve in the atmospheric water, and maintain their biological activity for days. It has been determined that volatile metabolites of some microorganisms are toxic for plants. Growth of seedling roots is inhibited more than growth of stems. В© 2006 COSPAR.

Аннотация: The study of heterotrophic bacteria isolated from the brackish waters of Lake Shira has shown that some of them contain plasmid pSH1 of approximately 2.7 kb in size. The number of plasmid copies in plasmid-containing strains cultivated at a minimal concentration of sodium chloride is found to be low, whereas the sub-culturing of these strains at high salt concentrations increases the plasmid number. The role of natural pSH1 plasmid in the osmotolerance of host bacteria is discussed.

Аннотация: The lipid composition of the green alga Botryococcus was studied at three different cultivation temperatures: suboptimal (18В°C), optimal (25В°C), and supraoptimal (32В°C). Cultivation at the supraoptimal temperature was found to considerably inhibit the synthesis of nearly all intracellular lipids, except for triacylglycerides, and to influence their fatty acid composition. In particular, the content of trienoic fatty acids was significantly lower at the supraoptimal than at the optimal cultivation temperature. At the same time, the fatty acid composition of the extracellular lipids of the alga virtually did not depend on cultivation temperature. В© 2002 MAIK "Nauka/ Interperiodica".

Аннотация: The CO oxidase (COX) activity of carboxydobacteria Seliberia carboxydohydrogena cells and the rate of CO oxidation by this culture were studied under various modes of fermentation. The CO oxidase activity of cells was maximal (60-80 U) during limitation of bacterial growth by the source of energy or carbon. The rate of CO oxidation by the culture was 1.4-1.71/g. The adaptive type of dependence of the CO oxidase activity of cells on the concentration of CO in the medium within the range of 0-20% was established.

Аннотация: The CO-resistant strain B5786 of the hydrogen-oxidizing bacterium Alcaligenes eutrophus was found to be able to synthesize polyhydroxyalkanoates (PHAs) under the conditions of growth limitation by nitrogen deficiency (the factor that promotes PHA synthesis) and growth inhibition by carbon monoxide. The gas mixtures that contained from 5 to 20 vol % CO did not inhibit the key enzymes of PHA synthesis-?-ketothiolase, acetoacetyl-CoA reductase, hydroxybutyrate dehydrogenase, and PHA synthase. In the presence of CO, cells accumulated up to 70-75 wt % PHA (with respect to the dry biomass) without any noticeable increase in the consumption of the gas substrate. Chromatographic-mass spectrometric analysis showed that the PHA synthesized by A. eutrophus is a copolymer containing more than 99 mol % ?-hydroxybutyrate and trace amounts of ?-hydroxyvalerate. The PHA synthesized under the conditions described did not differ from that synthesized by A. eutrophus cells from electrolytic hydrogen. В© 2001 MAIK "Nauka/Interperiodica".

Аннотация: Colonies grown from vegetative Bacillus subtilis 2335 cells had a standard structure, with bacillar cells occupying the whole colony volume. At the same time, the colonies of this bacterium grown from germinated spores had an abnormal structure characterized by the location of cells in a surface layer 100-200 mum thick at the colony boundary with the air. The glycocalyx of the colonies grown from spores was characterized by a wetting angle theta(e) of 120degrees-160degrees, whereas that of the colonies grown from vegetative cells had an angle theta(c) as low as 5degrees-30degrees. It is suggested that spores and vegetative cells follow different strategies of substrate colonization and that the architectonics of bacterial colonies is determined by the physicochemical properties of the glycocalyx.

Аннотация: The ability of marine luminescent bacteria to synthesize polyesters of hydroxycarboxylic acids (polyhydroxyalkanoates, PHA) as reserve macromolecules was studied. Twenty strains from the collection of the luminescent bacteria CCIBSO (WDCM839) of the Institute of Biophysics, Siberian Branch, Russian Academy of Sciences, assigned to different taxa (Photobacterium leiognathi, Ph. phosphoreum, Vibrio harveyi, and V. fischeri) were analyzed. The most productive strains were identified, and the conditions ensuring high polymer yields in batch culture (40-70% of the cell dry mass weight) were determined. The capacity for synthesizing two-and three-component polymers containing hydroxybutyric acid as the main monomer and hydroxyvaleric and hydroxyhexanoic acids was revealed in Ph. leiognathi and V. harveyi strains. The results allow luminescent microorganisms to be regarded as new producers of multicomponent polyhydroxyalkanoates. В© 2008 MAIK Nauka.

Аннотация: The present study addresses growth parameters and physiological and biochemical characteristics of the aerobic CO-oxidizing carboxydobacterium Seliberia carboxydohydrogena Z-1062. Poly(3-hydroxybutyrate) yields were investigated in experiments with limiting concentrations of mineral nutrients (nitrogen or sulfur or nitrogen and sulfur) in batch culture of S. carboxydohydrogena Z-1062 grown on gas mixtures consisting of CO2, O-2, H-2, and CO. CO concentrations of 10, 20, and 30 % v/v did not affect polymer synthesis, whose content after 56-h cultivation under limiting concentrations of nitrogen and sulfur was 52.6-62.8 % of biomass weight at a productivity of 0.13-0.22 g/L h. The inhibitory effect of CO on cell concentration was revealed at CO concentration of 30 % v/v. That also caused a decrease in substrate (H-2 and O-2) use efficiency. Thus, this carboxydobacterium can be regarded as a potential producer of polyhydroxyalkanoates from industrial hydrogenous sources.

Аннотация: The paper considers problems of biosynthesis of higher plants' biomass and "biological incineration" of plant wastes in a working physical model of biological LSS. The plant wastes are "biologically incinerated" in a special heterotrophic block involving Califomian worms, mushrooms and straw. The block processes plant wastes (straw, haulms) to produce soil-like substrate (SLS) on which plants (wheat, radish) are grown. Gas exchange in such a system consists of respiratory gas exchange of SLS and photosynthesis and respiration of plants. Specifics of gas exchange dynamics of high plants - SLS complex has been considered. Relationship between such a gas exchange and PAR irradiance and age of plants has been established. Nitrogen and iron were found to the first to limit plants' growth on SLS when process conditions are deranged. The SLS microflora has been found to have different kinds of ammonifying and denitrifying bacteria which is indicative of intensive transformation of nitrogen-containing compounds. The number of physiological groups of microorganisms in SLS was, on the whole, steady. As a result, organic substances - products of exchange of plants and microorganisms were not accumulated in the medium, but mineralized and assimilated by the biocenosis. Experiments showed that the developed model of a man-made ecosystem realized complete utilization of plant wastes and involved them into the intrasystem turnover.
