Аннотация: The life support systems (LSS) for long-term missions are to use cycling-recycling systems, including biological recycling. Higher plants are the traditional regenerator of air and producer of food. They should be used in many successive generations of their reproduction in LSS. Studies of influence of microbial associations on germination of wheat seeds and on growth of seedlings under impact of heavy metals are necessary because of migration of heavy metals in LSS. Microbial associations are able to stimulate growth of plants, to protect them from pathogenic organisms and from toxicity of heavy metal salts. The goal of this work was to investigate effect of microbial associations on the germination of wheat seeds and on the growth of seedlings under impact of different concentrations of ZnSO4. The results of investigations showed that:(1)Zinc salt had an adverse effect on germination of wheat seeds, beginning with concentrations of 8 MPC (Maximum Permissible Concentration) and higher.(2)Microbial associations (concentrations -104 to 107 cells/ml) were able to decrease (partly or completely) the adverse effect of ZnSO4 on germination of wheat seeds.(3)Concentrations (104-107 cells/ml) of microbial associations were able to decrease partly the adverse effect of zinc salts (intervals: from 1 to 32 MPS{cyrillic}) on the growth and development of wheat plantlets during heterotrophic phase.(4)The root system of plants was more sensitive to the adverse effect of ZnSO4 than shoots of plants. Аннотация: The speciation of artificial radionuclides 60Co, 137Cs, 152Eu, and 241Am in surface layers of bottom sediments of the Yenisei river, collected within the near zone of impact of the Mining and Chemical Combine (Rosatom), was studied. In these samples the radionuclides 60Co, 152Eu, and 241Am are mainly associated with fractions of humic and fulvic acids. The fraction of 152Eu associated with nonsilicate iron is approximately 92%. The migration capability of radionuclides collected near the Atamanovo settlement decreases in the order 152Eu > 241Am > 60Co > 137Cs. For samples collected near Bol'shoi Balchug settlement, this order is as follows: 152Eu ? 241Am > 60Co > 137Cs. The presence of radionuclide-bearing micro-particles in bottom sediments considerably complicates the distribution of radionuclides, in particular, of 241Am and 137Cs, among migration forms. Аннотация: The seasonal formation of annual rings on the scales of Baikal grayling from the middle reaches of the Yenisei River has been studied to find out the reasons for the high growth rate of the grayling under altered temperature regime downstream of the dam of the Krasnoyarsk Hydroelectric Plant. The number of circuli outside the last identified annuli and in the second, third, and fourth completed annuli was estimated on 569 fish caught during the annual cycle. The von Bertalanffy growth function was used to describe the increment in the number of circuli over a year. The calculation showed that a new annual ring was produced in July. In November, there was no statistically significant difference between the circulus number in scale increment and the circulus number in the corresponding completed annuli of older fish. Thus, despite the increased duration of the period with optimal water temperatures downstream of the dam of the Krasnoyarsk Hydroelectric Plant, fish growth was observed in a limited period of the year, from July to November. The probable reason for the high growth rate of grayling in the study area is that the fish do not have to waste energy on seasonal migration to the tributaries. Taking into account that grayling biomass production in the middle Yenisei occurs from July to November, we can recommend shifting the dates of commercial fishing for grayling to the end of this period-November-December.

Аннотация: Two species "prey-predator model" where the individuals purposefully migrate between two habitats is considered. A migration is regarded as transference from one habitat to another yielding in maximization of average (over two habitats) coefficient of reproduction. Targeted migration results in the increase in average abundance of population comparing with discrete analogies of classic equations of Volterra and also to the enlargement of the area of the admissible meanings of parameters. A great variety of dynamic regimes has been observed, some of them could be interpreted as outbreak explosion.

Аннотация: The new model was proposed to study the dynamic of spatially distributed populations with nonrandom migrating individuals. All migrations are resulted in maximization of the average net reproduction rate taking into account the cost of migration. The model belongs to a box type: individuals migrate between two stations and have complete information about living conditions both in the stations of residence and migration. The information is determined by dependence of reproduction rate on the abundance of individuals in the station. Numerical experiments showed that taking into account the goal-directed migrations caused the following effects: significant extension of the value of external conditions leading to the stable existence of population; increase in the average abundance of population; broadening of the observed set of dynamic regimes. The limitation of the model application are discussed.

Аннотация: Sorption of radiotracers 241Am and 242Pu and weighable amounts of uranium and stable Eu on bottom sediments in the simulated system bottom sediments-Yenisei river water was studied. At a contact time of 40 min these metals are completely sorbed on the bottom sediments, presumably via association with organomineral complexes and hydrated gels. Along with sorption, these metals can form soluble complexes and stable colloids (pseudocolloids). The distribution factors of 241Am and 242Pu, and stable Eu in the simulated system between bottom sediments and liquid phases were calculated from the results of sorption experiments. The introduced 241Am and 152Eu initially present in radioactively contaminated bottom sediments of the Yenisei river show similar pattern of distribution over differently mobile fractions. The distribution of weighable amounts of stable Eu significantly differs from that of initially present 152Eu and tracer 241Am due to decelerated sorption of stable Eu and its considerably higher concentration. A significant part of 242Pu, 241Am, and 152Eu is associated with mobile fractions of bottom sediments, which, under certain environmental conditions, can pass into river water as migrating species. Аннотация: Data of two field studies were used to analyze the concentrations of Na, K, Ca, Mg, Fe, Mn, Zn, Cu, Al, Cr, Ni, Cd, and Pb in the water, bottom deposits, zoobenthos, fish, and macrophytes of the Pond Bugach and in the soils near the pond. It was established that the majority of metals in the soils and bottom deposits correlate and their concentrations are governed by the universal geochemical factors of the region. The heavy metals were recognized that originate from anthropogenic sources and their concentrations were found to exceed the maximum admissible values for different components of the ecosystem. Five types of heavy metals migration were recognized in the soil-bottom deposits-zoobenthos-fish chain and in macrophytes. Statistically significant difference was found to exist between the concentrations of some heavy metals in the muscles of fish species with different food types (crucian carp and perch) as well as between the correlations of metals. The recorded concentrations were compared with the concentrations of metals measured in the last decade in other limnetic ecosystems in Siberia, Europe, North America, and China.

Аннотация: The specific composition, vertical distribution and daily migration of plankton in a meromictic salt lake, Lake Shira, have been investigated. The main structural components of the ecosystem are bacteria, algae and crustaceans. Since the late 1970s, blue-green algae are reported as dominant. Nowadays the phytoplankton is dominated by Lyngbya contorta, Microcystis ichtyoblabe (Cyanophyta), Cyclotella tuberculata (Bacillariophyta) and Dictyosphaerium tetrachotomum (Chlorophyta). The zooplankton of Lake Shira comprises Arctodiaptomus salinus (Copepoda), Brachionus plicatilis and Hexarthra oxiuris (Rotatoria). Investigation revealed a stratified vertical distribution of all dominant species except Cyclotella with a maximum at a depth of 12-15 m during the summer growth period. Zooplanktons differ in the pattern of distribution and daily migration, but most zooplankton tend to concentrate near the surface. The possible reasons for the vertical distribution of the plankton are discussed.

Аннотация: MicroRNAs epigenetically regulate physiological and pathological processes. Previously, we found that miR-204-5p is expressed at low levels in melanoma cells, and an increase in its level leads to a change in proliferation, migration, and invasion of these cancer cells. Now, using bioinformatics analysis, it has been shown that the target of miR-204-5p is FOXC1 transcription factor, which is implicated in carcinogenesis. Using the luciferase reporter assay, it was found that miR-204-5p suppresses expression of the FOXC1 gene by binding to its 3' non-coding region. Transfection of small interfering RNA (siRNA) targeting FOXC1 into melanoma cells caused a decrease in miR-204-5p levels, which is consistent with the generally accepted concept of feedback regulation of miRNA expression by target genes. According to the results of the MTT test and fluorescence microscopy, the proliferation level of melanoma cells under the influence of siRNA to FOXC1 decreased 72 h after transfection. Changes in the ratio of cells by cell cycle phase were analyzed using flow cytometry. Regulatory relationships between FOXC1 and miR-204-5p, and an inhibitory effect of FOXC1 knockdown on melanoma cell proliferation were revealed. Based on the results, it can be assumed that miR-204-5p regulates proliferation of melanoma cells by affecting FOXC1 expression.

Аннотация: The present study reports construction of wound dressing materials from degradable natural polymers such as hydroxy derivatives of carboxylic acids (PHAs) and 3-hydroxybutyrate/4-hydroxybutyrate [P(3HB/4HB)] as copolymer. The developed polymer films and electrospun membranes were evaluated for its wound healing properties with Grafts—elastic nonwoven membranes carrying fibroblast cells derived from adipose tissue multipotent mesenchymal stem cells. The efficacy of nonwoven membranes of P(3HB/4HB) carrying the culture of allogenic fibroblasts was assessed against model skin defects in Wistar rats. The morphological, histological and molecular studies revealed the presence of fibroblasts on dressing materials which facilitated wound healing, vascularization and regeneration. Further it was also observed that cells secreted extracellular matrix proteins which formed a layer on the surface of membranes and promoted the migration of epidermal cells from the neighboring tissues surrounding the wound. The wounds under the P(3HB/4HB) membrane carrying cells healed 1.4 times faster than the wounds under the cell-free membrane and 3.5 times faster than the wounds healing under the eschar (control).The complete wound healing process was achieved at Day 14. Thus the study highlights the importance of nonwoven membranes developed from degradable P(3HB/4HB) polymers in reducing inflammation, enhancing angiogenic properties of skin and facilitating better wound healing process. © 2016, Springer Science+Business Media New York.

Аннотация: An experimental approach for investigation of genetically modified microorganisms (GMMO) introduced into model ecosystems to evaluate potential risk of propagation of recombinant plasmids in surrounding medium has been developed. The object of modeling was Escherichia coli Z905 strain with a recombinant plasmid with bacterial luminescence genes, which was introduced into water microcosms of different structure. The approach involves comprehensive investigation of GMMO at four hierarchical levels: molecular (retaining the structure of the plasmid and expression of cloned genes); cellular (variation of metabolic activity); population (competitive power and metabolic interactions of GMMO with indigenous microflora, migration of recombinant and natural plasmids); ecosystem (effect of GMMO and cloned genes on ecosystem parameters). The experimental evidence and theoretical estimates are intended to form grounds to develop a basic version of an ecological certificate for different GMMO variants.

Аннотация: The effect of humic acid on absorption-release processes in the bottom sediments-Yenisei river water system was studied by dual-column ion chromatography and gamma-ray spectrometry. With the use of ion chromatography, it was found that processes related to the absorption of SO42- and Cl- anions by a solid phase with the release of NO3-, PO43-, and F- to a liquid phase competed in the test systems as the concentration of water-soluble organic carbon (WSOC) was increased. Only the test anions were released in the systems without the introduction of an additional amount of WSOC as humic acid. With the use of gamma-ray spectrometry, it was found that the release of Co-60, Eu-152, and Am-241 radionuclides to the liquid phase in the systems with added humic acid began much earlier than in the system without the addition of humic acid. In this case, the amount of released radionuclides was greater than the amount of radioisotopes released in the system without the addition of humic acid: similar to 25% Am-241, similar to 3% Eu-152, and similar to 0.8% Co-60 in the system with added humic acid or 0.8% Eu-152 and 0.1% Co-60 in the system without the addition of humic acid. The Am-241 radionuclide was not determined in the system without the addition of humic acid. An increase in the concentration of WSOC in the experimental system of bottom sediments-Yenisei river water initiated the release of Co-60, Eu-152, and Am-241 anthropogenic radionuclides from bottom sediments because of the formation of soluble complexes capable of migration. An increase in the concentration of WSOC had almost no effect on the release of K-40 and Cs-137 radionuclides.
