Аннотация: Using mathematical modeling methods, the work investigates possible existence of a phytoflagellate population in the ecosystem of a stratified lake (Lake Shira, Khakasia, Russia). Until this study was carried out, no phytoflagellate population had been detected in the lake, although there had been indirect evidence of the possible presence of a cryptomonad population in this water body. To answer the question if this population may be present in this water body, as the first step of investigation, a mathematical model was developed describing the microorganism biomass dynamics and phytoflagellate relationship with the lake food web. The calculations showed that cryptophytic algae were abundant in the water column layers above the lake chemocline at the beginning of summer but showed low biomass concentrations during other periods of time. In the summer following the construction of the model, samples were collected from Lake Shira, as the next step of the study. A cryptophytic algae population was found in the water column, and the pattern of its distribution in the ecosystem was similar to that predicted by the model. In addition to that, we studied theoretically possible reasons for the early summer development of the population in the deep water layers and found that it was the result of the mixotrophic feeding strategy of cryptomonads and trophic interactions in the food web. The study shows that preliminary modeling of the locational and temporal distribution of populations whose presence in the ecosystem is only suggested by indirect evidence may be useful as a tool for searching for the species in natural ecosystems and as an approach enabling a more accurate description of the food web structure of a water body. Аннотация: Two-section enclosures were designed for the investigation of the effect of various physicochemical and biological factors on vertical distribution of zooplankton in situ. The framework of the enclosure was a cylindrical polyethylene column without any partitions inside, in which the isolation of animals in different sections after in situ exposure was achieved by pinching the flexible central part of the column. Enclosures were tested at the brackish stratified meromictic Lake Shira (Russia, Khakasia). The absence of fish and carnivorous zooplankton in the lake suggests that the vertical distribution of zooplankton is mainly determined by physicochemical gradients in the water column. Experiments and field observations demonstrated that all age and size groups of Arctodiaptomus salinus and Brachionus plicatilis strongly avoided surface layers during the daylight. The escape of zooplankton from the anoxic hypolimnion was less active. Statistically significant avoidance was observed only for copepodites C4-C5 and females of A. salinus. The relatively simple construction of the columns and easy handling during the experiment were the factors that favoured the use of this device to perform in situ basic tests of the effect of different factors on the vertical distribution of zooplankton.

Аннотация: Three levels of chemical communications involved plankton Crustacea are considered: 1) Influence of zooplankton excretion on phytoplankton; 2) Influence of zooplankton excretion on the individuals of the same or other species of the same trophic level; 3) Influence of chemical cues released by predatory zooplankton and fish on herbivorous zooplankton. The data on the influence of excreted cues on some physiological (growth, reproduction, feeding, etc.) and behavioural (vertical and horizontal migrations) characters of planktonic crustaceans are presented. Ecological role and chemistry cues responsible for the interactions of different trophic levels can be different. It is considered that chemical communications in aquatic ecosystems can be provided with: 1) Species-specific cues that strictly influence particular biological functions (communication system of feromone type); 2) Non-specific cues that strictly influence particular functions (system of regulator, that act at the whole ecosystem as the hormonal system of an organism). 3) Non-specific substances with broad (nonspecific) influence-toxic substances of "biocondition substances" according to classification of Novikov and Kharlamova (2000).

Аннотация: The current lithospheric geodynamics and tectonophysics in the Baikal rift are discussed in terms of a nonlinear oscillator with dissipation. The nonlinear oscillator model is applicable to the area because stress change shows up as quasi-periodic inharmonic oscillations at rifting attractor structures (RAS). The model is consistent with the space-time patterns of regional seismicity in which coupled large earthquakes, proximal in time but distant in space, may be a response to bifurcations in nonlinear resonance hysteresis in a system of three oscillators corresponding to the rifting attractors. The space-time distribution of coupled MLHВ >В 5.5 events has been stable for the period of instrumental seismicity, with the largest events occurring in pairs, one shortly after another, on two ends of the rift system and with couples of smaller events in the central part of the rift. The event couples appear as peaks of earthquake 'migration' rate with an approximately decadal periodicity. Thus the energy accumulated at RAS is released in coupled large events by the mechanism of nonlinear oscillators with dissipation. The new knowledge, with special focus on space-time rifting attractors and bifurcations in a system of nonlinear resonance hysteresis, may be of theoretical and practical value for earthquake prediction issues. Extrapolation of the results into the nearest future indicates the probability of such a bifurcation in the region, i.e., there is growing risk of a pending MВ ?В 7 coupled event to happen within a few years. Аннотация: The vertical and seasonal distributions of the phytoflagellate Cryptomonas spp., and its most common, the planktonic ciliate predators (Oligotrichida, Scuticociliatida, Hypotrichida and Prostomatida) were investigated in chemocline region of small saline, meromictic lake Shunet (Siberia, Russia) during 2003 and 2005. The lake has a pronounced chemocline, with abundance of purple and green sulphur bacteria. Vertical distribution of the Cryptomonas populations near the oxic/anoxic boundary layer was studied at close intervals in water sampled using a hydraulically operated thin-layer sampler. In both summer and winter, Cryptomonas peaked in water stratum 5-10 cm above anoxic zone or in the anoxic zone water column in the chemocline (about 5 m). Ciliate densities and biomass were also much higher in chemocline than in mixolimnion. The range of diurnal migration of Cryptomonas population was not very wide, and it was restricted to layers with high light intensity. Аннотация: Methods of atomic absorption, flame photometry, and emission spectral analysis were used to study the concentrations of metals in water and major ecosystem components of the Yenisei River upstream of Krasnoyarsk City (conventionally background area). The mean bulk concentrations of Al and Cu in water exceeded the MAC for water bodies used for fishery. Cu concentration in freshwater shrimp was found to be reliably higher than that in the link of primary producers (periphyton), and Cd concentration in caddis fly larvae was found to exceed that in water moss. The maximal concentrations of metals among the examined aquatic organisms were recorded in periphyton. Cr concentration in the muscles of Arctic grayling was found to exceed some international standards. В© 2009 Pleiades Publishing, Ltd.

Аннотация: Populations of brown trout in the Mzymta River and its tributaries include anadromous (mainly female) and resident (mainly males) fish. Some resident males in the basin of the Mzymty River attain sexual maturity at the age 1+, and resident females mature at the age 2+ or 3+. The maximum age of resident fish is 4+ in the samples studied. Migrations of anadromous brown trout to the sea occur at the ages 1+, 2+, or 3+. Future spawners spend from 1 to 4 years at feeding grounds in the sea. Smolts of the population are characterized by performing not only spring but also autumn migrations to the sea. One smolt specimen has been detected upstream from the dam in the river where spawners of anadromous brown trout do not migrate; this means that the capability for sea migrations persists long in the population represented only by resident specimens of brown trout. The diversity of life cycles and ecological forms in populations of brown trout is not lower than in populations of brown trout in Northern and Western Europe. The comparison of the data obtained with published data makes it possible to come to the conclusion about the high plasticity of ontogenesis of Black Sea brown trout.

Аннотация: Based on experimental and field data, the character of daily variations in the production and distribution of the phytoplankton biomass under various hydrodynamic and thermodynamic conditions is analyzed. The effects of density stratification, wind and light conditions, vertical water flows, and daily migration of the Zooplankton on the life activity of the phytoplankton are simulated. It is shown that the vertical distribution of the maximum concentrations of the phytoplankton depends on daily variations in a number of abiotic factors. Аннотация: The effect of humic acid on absorption-release processes in the bottom sediments-Yenisei river water system was studied by dual-column ion chromatography and ?-ray spectrometry. With the use of ion chromatography, it was found that processes related to the absorption of SO 42- and Cl- anions by a solid phase with the release of NO 3- , PO 43- , and F- to a liquid phase competed in the test systems as the concentration of water-soluble organic carbon (WSOC) was increased. Only the test anions were released in the systems without the introduction of an additional amount of WSOC as humic acid. With the use of ?-ray spectrometry, it was found that the release of 60Co, 152Eu, and 241Am radionuclides to the liquid phase in the systems with added humic acid began much earlier than in the system without the addition of humic acid. In this case, the amount of released radionuclides was greater than the amount of radioisotopes released in the system without the addition of humic acid: ?25% 241Am, ?3% 152Eu, and ?0.8% 60Co in the system with added humic acid or 0.8% 152Eu and <0.1% 60Co in the system without the addition of humic acid. The 241Am radionuclide was not determined in the system without the addition of humic acid. An increase in the concentration of WSOC in the experimental system of bottom sediments-Yenisei river water initiated the release of 60Co, 152Eu, and 241Am anthropogenic radionuclides from bottom sediments because of the formation of soluble complexes capable of migration. An increase in the concentration of WSOC had almost no effect on the release of 40K and 137Cs radionuclides. В© Pleiades Publishing, Inc., 2006.

The Am-241 radionuclide was not determined in the system without the addition of humic acid. An increase in the concentration of WSOC in the experimental system of bottom sediments-Yenisei river water initiated the release of Co-60, Eu-152, and Am-241 anthropogenic radionuclides from bottom sediments because of the formation of soluble complexes capable of migration. An increase in the concentration of WSOC had almost no effect on the release of K-40 and Cs-137 radionuclides.

Аннотация: An experimental approach for investigation of genetically modified microorganisms (GMMO) introduced into model ecosystems to evaluate potential risk of propagation of recombinant plasmids in surrounding medium has been developed. The object of modeling was Escherichia coli Z905 strain with a recombinant plasmid with bacterial luminescence genes, which was introduced into water microcosms of different structure. The approach involves comprehensive investigation of GMMO at four hierarchical levels: molecular (retaining the structure of the plasmid and expression of cloned genes); cellular (variation of metabolic activity); population (competitive power and metabolic interactions of GMMO with indigenous microflora, migration of recombinant and natural plasmids); ecosystem (effect of GMMO and cloned genes on ecosystem parameters). The experimental evidence and theoretical estimates are intended to form grounds to develop a basic version of an ecological certificate for different GMMO variants.

Аннотация: The present study reports construction of wound dressing materials from degradable natural polymers such as hydroxy derivatives of carboxylic acids (PHAs) and 3-hydroxybutyrate/4-hydroxybutyrate [P(3HB/4HB)] as copolymer. The developed polymer films and electrospun membranes were evaluated for its wound healing properties with Grafts—elastic nonwoven membranes carrying fibroblast cells derived from adipose tissue multipotent mesenchymal stem cells. The efficacy of nonwoven membranes of P(3HB/4HB) carrying the culture of allogenic fibroblasts was assessed against model skin defects in Wistar rats. The morphological, histological and molecular studies revealed the presence of fibroblasts on dressing materials which facilitated wound healing, vascularization and regeneration. Further it was also observed that cells secreted extracellular matrix proteins which formed a layer on the surface of membranes and promoted the migration of epidermal cells from the neighboring tissues surrounding the wound. The wounds under the P(3HB/4HB) membrane carrying cells healed 1.4 times faster than the wounds under the cell-free membrane and 3.5 times faster than the wounds healing under the eschar (control).The complete wound healing process was achieved at Day 14. Thus the study highlights the importance of nonwoven membranes developed from degradable P(3HB/4HB) polymers in reducing inflammation, enhancing angiogenic properties of skin and facilitating better wound healing process. © 2016, Springer Science+Business Media New York.

Аннотация: MicroRNAs epigenetically regulate physiological and pathological processes. Previously, we found that miR-204-5p is expressed at low levels in melanoma cells, and an increase in its level leads to a change in proliferation, migration, and invasion of these cancer cells. Now, using bioinformatics analysis, it has been shown that the target of miR-204-5p is FOXC1 transcription factor, which is implicated in carcinogenesis. Using the luciferase reporter assay, it was found that miR-204-5p suppresses expression of the FOXC1 gene by binding to its 3' non-coding region. Transfection of small interfering RNA (siRNA) targeting FOXC1 into melanoma cells caused a decrease in miR-204-5p levels, which is consistent with the generally accepted concept of feedback regulation of miRNA expression by target genes. According to the results of the MTT test and fluorescence microscopy, the proliferation level of melanoma cells under the influence of siRNA to FOXC1 decreased 72 h after transfection. Changes in the ratio of cells by cell cycle phase were analyzed using flow cytometry. Regulatory relationships between FOXC1 and miR-204-5p, and an inhibitory effect of FOXC1 knockdown on melanoma cell proliferation were revealed. Based on the results, it can be assumed that miR-204-5p regulates proliferation of melanoma cells by affecting FOXC1 expression.

Аннотация: The specific composition, vertical distribution and daily migration of plankton in a meromictic salt lake, Lake Shira, have been investigated. The main structural components of the ecosystem are bacteria, algae and crustaceans. Since the late 1970s, blue-green algae are reported as dominant. Nowadays the phytoplankton is dominated by Lyngbya contorta, Microcystis ichtyoblabe (Cyanophyta), Cyclotella tuberculata (Bacillariophyta) and Dictyosphaerium tetrachotomum (Chlorophyta). The zooplankton of Lake Shira comprises Arctodiaptomus salinus (Copepoda), Brachionus plicatilis and Hexarthra oxiuris (Rotatoria). Investigation revealed a stratified vertical distribution of all dominant species except Cyclotella with a maximum at a depth of 12-15 m during the summer growth period. Zooplanktons differ in the pattern of distribution and daily migration, but most zooplankton tend to concentrate near the surface. The possible reasons for the vertical distribution of the plankton are discussed.
