Аннотация: The morphotypes of human macrophages (MPh) were studied in the culture on nano-structured biopolymer substrates, made from polyhydroxyalcanoates (PHAs) of five various monomer compositions, followed by the solvent evaporation. Its surface relief, which was further in direct contact with human cells in vitro, was analyzed by atomic force microscopy (AFM) and scanning electron microscopy (SEM). It was shown, that the features of the micro/nano relief depend on the monomeric composition of the polymer substrates. Monocytes (MN) of patients with atherosclerosis and cardiac ischemia, undergoing stenting and conventional anti-atherosclerotic therapy, were harvested prior and after stenting. MN were isolated and cultured, with the transformation into MPh in direct contact with biopolymer culture substrates with different monomer composition and nano-reliefs, and transformed into MPh, in comparison with the same process on standard culture plastic. Sub-populations of cells with characteristic morphology in each phenotypic class were described, and their quantitative ratios for each sample of polymers were counted as an intermediate result in the development of “smart” material for cardiovascular devices. The results obtained allow us to assume, that the processes of MPh differentiation and polarization in vitro depend not only on the features of the micro/nano relief of biopolymer substrates, but also on the initial state of MN in vivo and general response of patients. © 2018 The Authors

Аннотация: In this study, polymer-based microparticles are used to improve the therapeutic properties of ceftriaxone (CEF) and render them safer. Poly-3-hydroxybutyrate (P3HB) and poly-3-hydroxybutyrate/polyethylene glycol (P3HB-PEG)-based microparticles were prepared by two methods: a double emulsification technique and spray-drying. The microparticles were characterized in terms of size and zeta potential, morphology, total drug loading and drug release. The microparticles had spherical shapes with diameters of a size range from 0.74 to 1.55 µm (emulsification technique) and from 3.84 to 6.51 µm (spray-drying); CEF encapsulation efficiency was around 63% and 49% for these methods respectively. The CEF release from microparticles obtained by spray-drying reached 100% after 150 h, while for microparticles obtained by emulsification technique the total release of CEF did not exceed 34% after 312 h. The release profiles could be best explained by Zero order kinetics model, Higuchi and Korsmeyer-Peppas models, as the plots showed high linearity. Antibacterial activity of the microparticles was evaluated against gram-positive and gram-negative bacterial strains. In general, CEF encapsulation in polymeric microparticles preserves the therapeutic efficacy of the CEF and provides its prolonged effect. © 2018, © 2018 Taylor & Francis.

Аннотация: We describe a low-cost three-dimensional underwater particle tracking velocimetry system to directly measure particle settling rate and flux in low-turbulence aquatic environments. The system consists of two waterproof cameras that acquire stereoscopic videos of sinking particles at 48 frames s?1 over a tunable sampling volume of about 45 ? 25 ? 24 cm. A dedicated software package has been developed to allow evaluation of particle velocities, concentration and flux, but also of morphometric parameters such as particle area, sinking angle, shape irregularity, and density. Our method offers several advantages over traditional approaches, like sediment trap or expensive in situ camera systems: (1) it does not require beforehand particle collection and handling; (2) it is not subjected to sediment trap biases from turbulence, horizontal advection, or presence of swimmers, that may alter particulate load and flux; (3) the camera system enables faster data processing and flux computation at higher spatial resolution; (4) apart from the particle settling rates, the particle size distribution, and morphology is determined. We tested the camera system in Lake Stechlin (Germany) in low turbulence and mean flow, and analyzed the morphological properties and settling rates of particles to determine their sinking behavior. The particle flux assessed from conventional sediment trap measurements agreed well with that determined by our system. By this, the low-cost approach demonstrated its reliability in low turbulence environments and a strong potential to provide new insights into particulate carbon transport in aquatic systems. Extension of the method to more turbulent and advective conditions is also discussed. © 2019 The Authors. Аннотация: Four PHA types were synthesized in the culture of Cupriavidus eutrophus B-10646 under special conditions, poly(3?hydroxybutyrate) [P(3HB)] and of copolymers, which contained 3HB monomers and 4?hydroxybutyrate (4HB), 3?hydroxyvalerate (3HV), or 3?hydroxyhexanoate (3HHx). All copolymers had the Mw of about 550–670 kDa, and the homopolymer P(3HB) had a significantly higher Mw – 920 kDa. P(3HB?co?4HB) and P(3HB?co?3HHx) had the lowest Cx (42 and 49%) while P(3HB?co?3HV) and P(3HB) exhibited higher Cx values (76%). Polymer films were prepared from different PHAs. Electron microscopy showed differences in the surface microstructure of the films. Films prepared from the P(3HB) were more hydrophobic and the arithmetic mean surface roughness of 71–75 nm, than the copolymer films, which were hydrophilic (57–60°) and had considerably higher roughness (158–177 nm). Blood parameters (hemoglobin and hemolysis) and response of the cells (erythrocytes, platelets, and monocytes) were studied in experiments with blood directly contacting the surface of the films of PHAs with different compositions. Cultivation of blood cells on polymer films did not cause any adverse effects on adhesion and morphology of all cell types. Results of studying blood cell response suggested that the films made from low-crystallinity copolymers containing 4?hydroxybutyrate and 3?hydroxyhexanoate were the best for contact with blood. © 2019 Elsevier B.V.

Аннотация: We have analyzed the partial sequences of the mitochondrial COI gene along with the amino acid sequences of cytochrome oxidase subunit I, encoded by this gene region, in representatives of 11 genera of salmonoid fish. For amino acid sequences, two alternative networks are constructed with outgroups represented by either Esocoidei or Osmeroidei as the supposed ancestral groups. This way, Osmeroidei appear to be closer to the salmonoid fish than Esocoidei, and their presence in the network as an outgroup explains the available data on the morphology and karyology of salmonoids much better. A number of the results of this study are fundamentally new. In particular, the slowing down of the molecular evolution of the grayling (Thymallidae) is shown. We conclude that the charr (Salvelinus) is one of the modern genera of salmonoids closest to their ancestor. The hypothesis of the phylogenetic proximity of the genera Brachymystax, Hucho, and Salmo has been confirmed. We also discuss the possibility that it is namely the changes in the amino acid sequence of cytochrome oxidase subunit I that lead to postzygotic reproductive isolation between taxa.

Аннотация: The results of a study of the specific features of morphological and genetic polymorphism of the population of the anadromous White Sea common whitefish (Coregonus lavaretus L.) from the Keret' River are presented. As assessed by its morphological features, the whitefish population of the Keret' is homogenous. A comparison of the Keret' whitefish with whitefish populations of both Europe and Siberia revealed clear differences. These differences relate first and foremost to the fish locomotion and orientation in space: body depth, length and positions of fins, and interorbital width. The characteristics of skull features (craniological analysis) of the White Sea whitefish are presented for the first time. The results of an analysis of the polymorphism of the ND1 fragment of the mitochondrial DNA confirmed an earlier suggestion about the mixed origin of the Keret' River whitefish: the descendants of three different phylogenetic lineages were revealed in the population.

Аннотация: The study investigates toxic effects of the fungicide tebuconazole (TEB) on Fusarium-infected wheat (Triticum aestivum) plants based on the morphological characteristics of root apices and changes in the integrated parameters of redox homeostasis, including the contents of free proline and products of peroxidation of proteins (carbonylated proteins, CP) and lipids (malondialdehyde, MDA) in roots. In two-day-old wheat sprouts infected by Fusarium graminearum, the levels of proline, CP, and border cells of root apices are higher than in roots of uninfected sprouts by a factor of 1.4, 8.0, and 3, respectively. The triazole fungicide tebuconazole (TEB) at the concentrations of 0.01, 0.10, and 1.00 ?g ml?1 of medium causes a dose-dependent decrease in the number of border cells. The study of the effects of TEB and fusarium infection on wheat plants in a 30-day experiment shows that the effect of the fungicide TEB on redox homeostasis in wheat roots varies depending on the plant growth stage and is significantly different in ecosystems with soil and plants infected by Fusarium phytopathogens. The study of the morphology of root apices shows that the toxic effects of TEB and fusarium infection are manifested in the destructive changes in root apices and the degradation of the root tip mantle. © 2018 Elsevier Masson SAS

Аннотация: The small synanthropic and peregrine earthworm Microscolex phosphoreus (Duges, 1837) is reported for the first time from Siberia. Morphological and DNA barcode (COI) analyses of this and widely separate samples worldwide demonstrate that, as currently identified, M. phosphoreus is a heterogeneous taxon, with divergent lineages occurring often in the same locality and hardly providing geographically structured genetic signals. The combined morphological and genetic evidence suggests that at least four of the found clades should be reclassified as separate species, both morphologically and genetically distinct from each other. However, as the specimen number was limited and only the COI gene was studied for the genetic work, we hesitate in formally describing new species. There would also be the problem of assigning the available names to specific lineages. Our findings encourage careful external and anatomical examination and using reliable characters such as the interchaetal distances and spermathecal morphology for correct identification and for deeper evaluation of cryptic diversity in this interesting bioluminescent worm.

Аннотация: Single-crystal diamonds in the form of micrometer-scale pyramids were produced using a combination of hot-filament (HF) chemical vapor deposition (CVD) and thermal oxidation processes. The diamond pyramids were compared here with similar ones that were manufactured using plasma-enhanced (PE) CVD. The similarities revealed in the morphology, Raman, and photoluminescent characteristics of the needles obtained using the hot-filament and plasma-enhanced CVD are discussed in connection with the diamond film growth mechanism. This work demonstrated that the HF CVD method has convincing potential for the fabrication of single-crystal diamond needles in the form of regularly shaped pyramids on a large surface area, even on non-conducting substrates. The experimental results demonstrated the ability for the mass production of the single-crystal needle-like diamonds, which is important for their practical application.

Аннотация: Morphological analysis of muksun Coregonus muksun of the Khatanga River basin revealed that along with its typical high-density-rakered form, individuals of the low-density-rakered form also live in this region. Analysis of the genetic polymorphism of three marker regions of mitochondrial (ND1 and COI fragments) and nuclear (ITS1) DNA indicates the polyphyletic origin of these forms. The revealed low level of genetic differentiation of muksun and whitefish C. lavaretus, along with the available data on the absence of clear diagnostic species characteristics, suggest that they belong to the same biological species C. lavaretus.

Аннотация: In this work, delivery systems in the form of microparticles and films containing 1,2,4-trioxolane (ozonide, OZ) based on polyhydroxyalkanoates (PHAs) were developed. Main systems' characteristics were investigated: the particle yield, average diameter, zeta potential, surface morphology, loading capacity, and drug release profile of microparticles, as well as surface morphology and release profiles of OZ-containing films. PHA-based OZ-loaded microparticles have been found to have satisfactory size, zeta potential, and ozonide loading-release behavior. It was noted that OZ content influenced the surface morphology of obtained systems.

Аннотация: A series of films was obtained from blends of poly-3-hydroxybutyrate (PHB) and poly-s-caprolactone (PCL). The structure, physical, mechanical and biological properties of the films were studied, depending on the ratio of the components. The microscopic structure of mixed films was determined by the main component of the blend: the films were porous with a predominance of PHB and relatively smooth with a predominance of PCL. The highest values of elongation at break (229-269%), as well as the local maximum values of tensile strength (14-15 MPa) were noted for blends with 5-25% PHB content. At PHB content of 25% and 75%, local maxima of Young's modulus were observed (507 and 655 MPa, respectively). When the ratio of the components in the blend was equal (50:50 by weight), the films were heterogeneous and had the lowest values of mechanical strength (ultimate strength and elongation at break were 3 MPa and 15.5%, respectively). A study of the biocompatibility of mixed films in a culture of NIH 3T3 mouse fibroblasts based on determination of the number of adhered cells in the MTT test showed the absence of cytotoxic action, as well as high adhesion and cell proliferation, in some cases exceeding the data for films from pure PHB and PCL. This demonstrates the suitability of PHB-PCL blends for the production of cell carriers for tissue engineering and other reconstructive technologies. © 2021 Author(s).

Аннотация: The variability of craniological characters has been studied for anadromous and lacustrine forms of whitefish Coregonus lavaretus (Linnaeus, 1758) from waterbodies of the Karelian coast of the White Sea. The significant phenotypic plasticity of craniological characters of the whitefish depends on the environmental conditions. Moreover, there is a correlation between indexes of the craniological traits (except for the index of the snout length) and the body and/or head length (allometric effect). These results suggest that it is unreasonable to separate anadromous and lacustrine forms of C. lavaretus as distinct taxonomical units and argues for the invalidity of C. pidschian and C. oxyrinchus species.

Аннотация: The anoxygenic phototrophic bacterial community of the brackish meromictic Lake Shira (Khakassia) was investigated in August 2001, July 2002, and February-March 2003. In all the periods of investigation, the prevailing microorganisms were purple sulfur bacteria similar to Lamprocystis purpurea in morphology and pigment composition. Their highest number (3 ? 10 5 cells/ml) was recorded in July 2002 at the depth of 15 m. According to 16S rRNA gene analysis, the strain of purple sulfur bacteria isolated in 2001 and designated ShAm01 exhibited 98.6% similarity to the type strain of Thiocapsa roseopersicina and 97.1-94.4% similarity to the type strains of Tca. pendens, Tca. litoralis, and Tca. rosea. The minor microorganisms of the anoxygenic phototrophic bacterial community within the period of investigation were nonsulfur purple bacteria phylogenetically close to Rhodovulum strictum (98.3% similarity, strain ShRb01), Ahrensia kielensis (of 93.9% similarity, strain ShRb02), Rhodomicrobium vannieli (of 99.7% similarity, strain ShRmc01), and green sulfur bacteria, phylogenetically close to Chlorobium limicola (of 98.7% similarity, strain ShCl03). Аннотация: We studied how chemicals obtained as filtrates from algal monocultures (algal chemicals) and from rotifer cultures with or without algae (rotifer chemicals) affected feeding rates of the rotifer Brachionus calyciflorus on its food algae, both directly and indirectly (through chemical-induced changes in algal morphology). Algal chemicals had a strong stimulating effect on the feeding rate of B. calyciflorus, but these effects were counteracted by rotifer chemicals. In functional response experiments, rotifer chemicals lowered maximum ingestion rates and had strong effects on assimilation rates and assimilation efficiencies of B. calyciflorus, probably due to the release of unspecific (auto)toxic metabolites. Furthermore, rotifer chemicals induced colony formation in the food alga Scenedesmus obliquus. Above the optimum particle size for ingestion by B. calyciflorus, larger algal colony sizes increased the food-handling time, thus lowering ingestion and assimilation rates. Through their effects on trophic interactions, infochemicals may play a role in structuring and the functioning of aquatic food webs. Аннотация: The paper describes the production and investigation of flexible films made of high-purity polyhydroxyalkanoates (PHAs) - polyhydroxybutyrate [poly-(3HB)] and poly-3-hydroxybutyrate-co-poly-3-hydroxyvalerate [poly(3Hl3-co-3HV)], containing 4-30 mol % hydroxyvalerate. Poly(3HB-co-3HV) films have a more porous structure than poly-(3HB) films, which are more compact, but their surface properties, such as wettability and surface and interface energies, are the same. Sterilisation of the PHA films by conventional methods (heat treatment and gamma-irradiation) did not impair their strength. Cells cultured on PHA films exhibited high levels of cell adhesion. Cell morphology, protein synthesis and DNA synthesis were estimated by extent of H-3-thymidine incorporation into the animal cell cultures of various origins (fibroblasts, endothelium cells, and isolated hepatocytes) in direct contact with PHAs. The investigation showed that this material can be used to make matrices for in vitro proliferous cells. The investigated properties of poly-(3HB) and poly(3HB-co-3HV) films proved to be fundamentally similar. (C) 2004 Kluwer Academic Publishers.
