Аннотация: The biodegradability of oriented fibers made of polyhydroxybutyrate (P(3HB)) and its co-polymer with ?-hydroxyvalerate (P(3HB-co-3HV)) was investigated in buffer solutions and in biological media in vitro and in vivo. The fibers of both polymer types demonstrated resistance to hydrolytic degradation in buffer solutions at 38В°C and pH from 4.5 to 7.0 (for up to 180 days). It has been found that the biodegradation of the fibers in vitro in blood and serum and in vivo is accompanied by weight losses and minor changes in the microstructure with no significant losses in the tensile strength over a long time (up to 180 days). The biodegradation rate of the less crystalline co-polymer P(3HB-co-3HV) fibers was 1.4-2.0-times higher than that of the homopolymer P(3HB). It has also been shown that the degradation of the fibers in vivo is influenced both by tissue fluid enzymes and cells (macrophages and foreign-body giant cells). The fibers were eroded on the surface only with no gross defects and no dramatic effects on their mechanical performance. В© VSP 2005.

Аннотация: Polyhydroxyalkanoate (PHA) sutures were implanted to test animals intramuscularly, and tissue reaction was investigated and compared with the reaction to silk and catgut. Tested monofilament sutures made of PHAs of two types polyhydroxybutyrate (PHB) and a copolymer of hydroxybutyrate and hydroxyvalerate (PHV) featured the strength necessary for the healing of muscle-fascial wounds. The reaction of tissues to polymeric implants was similar to their reaction to silk and was less pronounced than the reaction to catgut; it was expressed in a transient post-traumatic inflammation (up to four weeks) and the formation of a fibrous capsule less than 200 ?m thick, which became as thin as 4060 ?m after 16 weeks, in the course of reverse development. Macrophages and foreign-body giant cells with a high activity of acid phosphatase were actively involved in this process. PHB and PHB/PHV sutures implanted intramuscularly for an extended period (up to one year) did not cause any acute vascular reaction at the site of implantation or any adverse events, such as suppurative inflammation, necrosis, calcification of the fibrous capsule or malignant tumor formation. No statistically significant differences were revealed in the tissue response to polymer sutures of the two types. Capsules around silk and catgut sutures did not become significantly thinner.

Аннотация: The paper presents the results of biomedical investigations of sutures made of polyhydroxyalkanoates (PHAs) of two types (poly-3-hydroxybutyrate (PHB) and poly-3-hydroxybutyrate-co-poly-3-hydroxyvalerate (PHB/PHV)), synthesized by the bacteria Ralstonia eutropha B 5786 in the experiment on test animals in vivo, in comparison with silk and catgut sutures. PHB and PHB/PHV (PHBV) implants produced no adverse effect on physiological, biochemical and functional parameters of the animals during the post-surgery period. The tested PHA sutures featured the necessary strength throughout the healing period of the muscle-fascial cuts. The reaction of tissues to the implantation of PHB and PHB/PHV fibers fitted into the usual scheme characteristic of the wound process and of the reaction to a foreign-body invasion. This reaction and the reaction of tissues to silk had the similar nature and period of inflammation, but it was much less pronounced than the reaction to catgut. The tissue response to the implantation of PHAs consisted in a short-duration (up to 2 weeks) post-traumatic inflammation and the formation of a fibrous capsule less than 200 ?m thick during weeks 4-8, which in 4-6 months was reduced to 40-60 ?m in the course of reverse development. There were no adverse changes, such as suppurative inflammation, necrosis, calcification, and malignization of the cicatrice, at the site of implantation of PHA filaments, unlike in the cases with silk and catgut. In the case of PHA implantation there was a typical prolonged (throughout the post-surgery monitoring period) pronounced macrophagal stage with a large number of macrophages present. The macrophages were of the phagocytic type and multinucleate giant foreign cells, with a high activity of acid phosphomonoesterase that correlated with the activity of the enzyme in blood. Throughout the period of monitoring no differences in the tissue response to the implantation of the polymer filaments of two PHA types were recorded. В© 2003 Elsevier Science B.V. Аннотация: Abstract: Fatty acid composition and content of 2 forms of sockeye salmon Oncorhynchus nerka from lakes in Kamchatka Peninsula (Russia) were compared. One form of sockeye salmon was anadromous ("marine"), that is, adult fish migrated in ocean to feed and grow and than return in the lake to breed. Fish of another form, kokanee, never migrate in the ocean. Per cent levels of the main indicators of nutritive value, eicosapentaenoic acid (EPA, 20:5n-3), and docosahexaenoic acid (DHA, 22:6n-3), were significantly higher in the landlocked O. nerka. However, concentrations of EPA and DHA per wet weight of filets were higher in the marine form, because of the relatively higher content of sum of fatty acids in their muscle tissue. As concluded, fish fed in marine environment had higher contents of long-chain n-3 fatty acids per wet weight than fish of the same species, fed in fresh waters. In general, both the anadromous sockeye salmon and the landlocked kokanee salmon can be recommended for human diet as a valuable product concerning contents of EPA and DHA. Understanding the trophic relationships of consumers is central to ecology, but constructing meaningful food webs is often difficult because of a lack of detailed information on consumption versus assimilation and high degrees of omnivory.2. We used fatty acid analyses to examine the trophic relationships of three common larval anurans (Pseudacris crucifer, Lithobates catesbeianus and Lithobates clamitans) that are often classified as grazers or detritivores. Tadpoles and potential food sources were sampled in four ponds in southern Illinois and analysed for fatty acid composition. Single linkage cluster analysis was then used to compare fatty acid profiles among tadpole gut contents, tadpole muscle tissues and available food resources.3. Diets varied among species and within species among ponds, but organic sediments consistently contributed most to the fatty acid composition of the gut contents of all species. Fatty acid profiles also indicated that larval insects and phytoplankton were consumed by both L. catesbeianus and L. clamitans in one pond, while L. clamitans and P. crucifer consumed mainly periphyton along with sediments in another pond, and these diet differences appeared linked to physical differences among ponds, with periphyton and/or phytoplankton contributing more to tadpole diets in less shaded ponds.4. The fatty acid composition of muscle tissues of L. clamitans, the dominant tadpole in these systems, indicated that plant detritus and bacteria, which were the dominant components of organic sediments in the ponds, were common components of the assimilatory diet.5. Results demonstrate the utility of fatty acid analyses for assessing both consumption and assimilation. The tadpole assemblages we examined derive much of their energy from heterotrophic and allochthonous sources and exhibit high dietary plasticity. This information will allow for more accurate and comprehensive assessments of trophic interactions in freshwater habitats, as well as aid in amphibian conservation, management and captive propagation efforts. Аннотация: A formula was derived for quantification of benefit-risk ratio (hazard quotient) for the intake of a product containing essential polyunsaturated fatty acids vs. heavy metals. The quotient was used in a three year case study of the contents of essential fatty acids and heavy metals in Siberian grayling, the main food fish from the middle section of the Yenisei River. As found, in general the fish intake was potentially very beneficial for human health, except on a few occasions, when the risk overweighed the benefit because of high contents of chromium in the fish muscle tissue. The data demonstrated the necessity for regular monitoring of the hazard quotients for food fish in wild conditions, based at least on monthly sampling frequency. В© 2008 Elsevier Ltd. Аннотация: Frozen samples of common fish species, sea trout (Salmo trutta), from Norway and Siberia, herring (Clupea harengus pallasi), rock sole (Lepidopsetta bilineata) and cod (Gadus morhua maris-albi), collected from a wholesale market in Krasnoyarsk city (Siberia, Russia) were analyzed. Special attention was paid to long-chain essential polyunsaturated fatty acids: eicosapentaenoic, 20:5?3 (EPA) and docosahexaenoic, 22:6?3 (DHA). Heat-treatment (cooking and frying) did not in general significantly decrease the contents of EPA and DHA compared to raw fish species, except for a modest reduction in Norwegian trout during frying. Boiled trout appeared to be a more valuable fish dish for obtaining the officially recommended appropriate daily intake of EPA + DHA for humans. Аннотация: Seasonal variations of fatty acid contents in muscle tissue of one of the main food fish species in Siberian rivers, grayling, Thymallus arcticus, were studied over 3 years. Under a comparatively low range of water temperature variations, spawning appeared to be the main cause of seasonal changes in contents of quantitatively prominent and essential fatty acids in fish filets. In general, fish accumulated essential polyunsaturated fatty acids (PUFA), such as eicosapentaenoic (EPA) and docosahexaenoic (DHA), in their muscle tissue before reproductive seasons and then the PUFA seemed to be transferred into gonads during their formation. Hard roe of Siberian grayling had 3-4 times higher PUFA contents, than had that of the muscle tissue. The fish species, T. arcticus, was found to be a valuable source of the essential PUFA, including EPA and DHA, for human diet. В© 2007 Elsevier Ltd. All rights reserved.

Аннотация: Contents of fatty acids in filets of unfrozen (control), boiled, fried, roasted and boiled in a small amount of water humpback salmon, collected from a wholesale market in Krasnoyarsk city (Siberia, Russia) were analyzed. Special attention was paid to essential polyunsaturated fatty acids of ?3 family: eicosapentaenoic, 20:5?3 (EPA) and docosahexaenoic, 22:6?3 (DHA). Heat treatment in general did not decrease content of EPA and DHA in humpback, except a modest reduction during frying. Cooked humpback appeared to be the valuable source of essential ?3 PUFAs, namely EPA and DHA. It was hypothesized that the absence of significant reduction of PUFAs' contents in red flesh of fishes of Salmonidae family during heat treatment may be due to a high level of natural antioxidants which formed in the course of evolution as adaptation to their ecological niche. Аннотация: Fatty acid content of raw and cooked zander (Sander lucioperca) was studied. Special attention was paid to long-chain polyunsaturated fatty acids: eicosapentaenoic, 20:5 n-3 (EPA) and docosahexaenoic, 22:6 n-3 (DHA), and also to the n-6/n-3 ratio, which are regarded as indicators of nutritive value. As found, the heat treatments, boiling, stewing and frying, including those in a convection steam oven (CSO), did not significantly decrease the content of EPA and DHA in the products. Boiling and stewing appeared to give products of a higher nutritive value, regarding the above indicators, than frying and cake preparation. Frying of zander in CSO was found to be more beneficial for nutrition compared to pan-frying. The cooked zander had higher EPA and DHA contents than many other popular food fish species, and also had a high nutritive value due to the low n-6/n-3 ratio when boiled and stewed.

Аннотация: The paper presents the results of biomedical investigations of sutures made of polyhydroxyalkanoates (PHAs) of two types (poly-3-hydroxybutyrate (PHB) and poly-3-hydroxybutyrate-co-poly-3-hydroxyvalerate (PHB/PHV)), synthesized by the bacteria Ralstonia eutropha B 5786 in the experiment on test animals in vivo, in comparison with silk and catgut sutures. PHB and PHB/PHV (PHBV) implants produced no adverse effect on physiological, biochemical and functional parameters of the animals during the post-surgery period. The tested PHA sutures featured the necessary strength throughout the healing period of the muscle-fascial cuts. The reaction of tissues to the implantation of PHB and PHB/PHV fibers fitted into the usual scheme characteristic of the wound process and of the reaction to a foreign-body invasion. This reaction and the reaction of tissues to silk had the similar nature and period of inflammation, but it was much less pronounced than the reaction to catgut. The tissue response to the implantation of PHAs consisted in a short-duration (up to 2 weeks) post-traumatic inflammation and the formation of a fibrous capsule less than 200 mum thick during weeks 4-8, which in 4-6 months was reduced to 40-60 mum in the course of reverse development. There were no adverse changes, such as suppurative inflammation, necrosis, calcification, and malignization of the cicatrice, at the site of implantation of PHA filaments, unlike in the cases with silk and catgut. In the case of PHA implantation there was a typical prolonged (throughout the post-surgery monitoring period) pronounced macrophagal stage with a large number of macrophages present. The macrophages were of the phagocytic type and multinucleate giant foreign cells, with a high activity of acid phosphomonoesterase that correlated with the activity of the enzyme in blood. Throughout the period of monitoring no differences in the tissue response to the implantation of the polymer filaments of two PHA types were recorded. (C) 2003 Elsevier Science B.V. Аннотация: Polyhydroxyalkanoate (PHA) sutures were implanted to test animals intramuscularly, and tissue reaction was investigated and compared with the reaction to silk and catgut. Tested monofilament sutures made of PHAs of two types-polyhydroxybutyrate (PHB) and a copolymer of hydroxybutyrate and hydroxyvalerate (PHV)-featured the strength necessary for the healing of muscle-fascial wounds. The reaction of tissues to polymeric implants was similar to their reaction to silk and was less pronounced than the reaction to catgut; it was expressed in a transient post-traumatic inflammation (up to four weeks) and the formation of a fibrous capsule less than 200 mum thick, which became as thin as 40-60 mum after 16 weeks, in the course of reverse development. Macrophages and foreign-body giant cells with a high activity of acid phosphatase were actively involved in this process. PHB and PHB/PHV sutures implanted intramuscularly for an extended period (up to one year) did not cause any acute vascular reaction at the site of implantation or any adverse events, such as suppurative inflammation, necrosis, calcification of the fibrous capsule or malignant tumor formation. No statistically significant differences were revealed in the tissue response to polymer sutures of the two types. Capsules around silk and catgut sutures did not become significantly thinner. Аннотация: Long-chain polyunsaturated n-3 fatty acids (LC-PUFA), including eicosapentaenoic (EPA), and docosahexaenoic (DHA) are an essential part of human diets, but their main source, the wild fish catch, is limited. Terrestrial production animals, such as cattle, pigs, and chicken, can synthesize these compounds from the plant-derived precursor -linolenic acid, and the synthesis occurs mainly in liver. We estimated the putative contribution of liver products to the LC-PUFA supply in human nutrition, as an alternative to fish. We measured EPA and DHA contents in raw and cooked livers of the animals. Calculations of global production of EPA and DHA in the livers of these animals were done. Mean contents of LC-PUFA in raw livers of chicken, pigs, and cattle were comparable with those in some fish. Culinary treatments of the liver did not result in a decrease of EPA and DHA contents. Global production of EPA and DHA in animals' livers was estimated as approximate to 410(6)kg/year, while the global EPA+DHA supply through the wild fish catch is known to be 18010(6)kg/year. Thus, liver of production animals is an additional source of LC-PUFA for human nutrition, rather than an alternative to fish. Practical applications: Consumption of food with a high PUFA content and a low n-6/n-3 ratio is recommended for humans to prevent cardiovascular disease and psychiatric disorders. We found that cooked pork and beef liver is a valuable source of n-3 LC-PUFA in human nutrition. In contrast, cooked chicken liver had too high of a n-6/n-3 ratio and, thereby, had a lower nutritive value concerning n-3 LC-PUFA. An estimate has been made of the putative contribution of liver products of terrestrial animals to the LC-PUFA supply as alternative to fish in human nutrition.

Аннотация: Changes in the intracellular concentration of free ionized calcium ([Ca2+]i) control a host of cellular processes as varied as vision, muscle contraction, neuronal signal transmission, proliferation, apoptosis etc. The disturbance in Ca2+-signaling causes many severe diseases. To understand the mechanisms underlying the control by calcium and how disorder of this regulation relates to pathological conditions, it is necessary to measure [Ca2+]i. The Ca2+-regulated photoproteins which are responsible for bioluminescence of marine coelenterates have been successfully used for this purpose over the years. Here we report the results on comparative characterization of bioluminescence properties of aequorin from Aequorea Victoria, obelin from Obelia longissima, and clytin from Clytia gregaria charged by native coelenterazine and coelenterazine analogues f, i, and hcp. The comparison of specific bioluminescence activity, stability, emission spectra, stopped-flow kinetics, sensitivity to calcium, and effect of physiological concentrations of Mg2+ establishes obelin-hcp as an excellent semisynthetic photoprotein to keep track of fast intracellular Ca2+ transients. The rate of rise of its light signal on a sudden change of [Ca2+] is almost 3- and 11-fold higher than those of obelin and aequorin with native coelenterazine, respectively, and 20 times higher than that of the corresponding aequorin-hcp. In addition, obelin-hcp preserves a high specific bioluminescence activity and displays higher Ca2+-sensitivity as compared to obelin charged by native coelenterazine and sensitivity to Ca2+ comparable with those of aequorin-f and aequorin-hcp. © The Royal Society of Chemistry and Owner Societies 2015.

Аннотация: Triploid fish has become an important item of commercial aquaculture, but data on its fatty acid (FA) composition are still controversial, especially regarding essential polyunsaturated fatty acids, eicosapentaenoic acid (20:5n-3, EPA) and docosahexaenoic acid (22:6n-3, DHA). We studied FA composition and content of diploid and triploid pink salmon Oncorhynchus gorbuscha, reared in aquaculture in a bay of the White Sea (Russia). FA composition, measured as percentages of total FA of triploids and immature diploid females significantly differed from that of mature diploid fish. Specifically, mature diploids had higher percentage of EPA and DHA in their muscle tissue (filets) compared to that of triploids and immature diploid females. Nevertheless, the contents of EPA and DHA per mass of the filets in diploid and triploid specimens were similar. Thus, no special efforts are needed to improve EPA and DHA contents in filets of triploids. © 2016 Elsevier Ltd

Аннотация: In several Russian northern lakes and rivers, Arctic cisco Coregonus autumnalis, least cisco C. sardinella, peled C. peled, tugun C. tugun, broad whitefish C. nasus, whitefish C. lavaretus and vendace C. albula were sampled in periods of officially permitted commercial fishery. Special attention was paid to contents (mg g(-1) of wet weight) of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in muscle tissues (filets), which are essential for human nutrition. The highest values of EPA + DHA content in semi-anadromous fish and freshwater fish were recorded for C. autumnalis from the Yenisei River, 17.60 mg g(-1) wet weight, and for C. lavaretus from the Sobachye Lake, 16.61 mg g(-1) wet weight, respectively. Intra-genus variations of EPA + DHA contents of Coregonus species were from 1.87 to 17.60 mg g(-1) wet weight. Since the congeneric species were genetically close to each other, the variations in EPA and DHA contents were thought to be caused primarily by ecological factors: migrational capability, type of feeding and trophic status of aquatic ecosystems. In general, the majority of studied species appeared to be of a high nutritive value for humans, although unfavorable environmental conditions could considerably diminish this value.

Аннотация: Fish are recognized as the main source of physiologically important omega-3 long-chain polyunsaturated fatty acids, namely, eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), for human nutrition. However, muscle tissue contents of these fatty acids in diverse fish species, i.e., their nutritive value for humans, varied within two orders of magnitude. We reviewed contents of EPA and DHA, measured by similar methods using an internal standard during chromatography as mg per g of wet mass in 172 fish species belonging to 16 orders, to evaluate probable variations in phylogenetic and ecological drivers. EPA + DHA content varied from 25.6 mg g(-1) of wet mass (Sardinops sagax) to 0.12 mg g(-1) (Gymnura spp.). Multidimensional redundancy analysis revealed that among phylogenetic, ecomorphological and abiotic environmental factors, the highest proportion of variation contribution belonged to the shared contribution of sets of phylogenetic and ecomorphological factors. Specifically, the highest values of EPA + DHA content were characteristic of fish belonging to the orders Clupeiformes or Salmoniformes, were pelagic fast swimmers, ate zooplankton and inhabited marine waters or migrated from fresh to marine waters (anadromous migrations). High EPA and DHA content in muscle tissues of the above species appeared to be a metabolic adaptation for fast continuous swimming. In contrast to common beliefs, our meta-analysis did not support the significant influence of higher trophic levels (piscivory) and cold environments (homeoviscous adaptation) on EPA and DHA content in fish. However, many causes of high and low levels of physiologically important fatty acids in certain fish species remained unexplained and require evaluation in future studies.

Аннотация: Two fish species reared in aquaculture (pink salmon Oncorhynchus gorbuscha and whitefish Coregonus lavaretus) and ten fish species from natural habitats (whitefish C. lavaretus, tugun Coregonus tugun, broad whitefish Coregonus nasus, least cisco Coregonus sardinella, vendace Coregonus albula, boganid charr Salvelinus boganidae, charr Salvelinus alpinus complex, northern pike Esox lucius, sharp-snouted lenok Brachymystax lenok, and taimen Hucho taimen) have been studied. The content of two long-chain polyunsaturated omega-3 fatty acids (PUFAs), eicosapentaenoic acid (20:5n-3, EPA) and docosahexaenoic acid (22:6n-3, DHA), in the muscle tissue of the fish and in their food (intestine contents) are compared. In the aquacultures of whitefish and pink salmon, the total content of EPA and DHA is significantly higher in feed than in the muscle tissue of the fish, which indicates losses of PUFA in the two-link food chain of the aquaculture during their transfer to the upper trophic level. EPA and DHA losses in aquaculture, which are confirmed by numerous literature data, mean an inefficient usage of the available sources of PUFAs and the aggravation of the global deficit of these biochemicals in the human diet. A study of natural fish populations reveals the accumulation of EPA and DHA in their biomass compared to food in many cases, although opposite phenomena are also observed. An assumption on the presence of an optimal, physiologically adequate species-specific level of PUFA in the fish muscle tissue has been made based on our data and literature data. If the level of PUFAs in the muscles is lower than optimal, their accumulation (bioaccumulation) from food and/or de novo synthesis are observed. When the optimal level is exceeded, the content of EPA and DHA in biomass approaches maximum species-specific values; however, part of these PUFAs entering from food is not digested or is catabolized. According to the obtained data, the species of the order Salmoniformes have an optimal level of 2 to 6 mg/g of wet weight. It has been found that in aquaculture approaching to maximum values of EPA + DHA content was accompanied by their losses (scattering) in the food chains, while in natural ecosystems the maximum values of PUFA content in the fish biomass are achieved by their accumulation from the lower trophic level. Boganid charr S. boganidae had the highest content of EPA + DHA in the muscle tissue among all known fish species (32.78 mg/g of wet weight).
