Аннотация: Polyhydroxyalkanoate (PHA) sutures were implanted to test animals intramuscularly, and tissue reaction was investigated and compared with the reaction to silk and catgut. Tested monofilament sutures made of PHAs of two types polyhydroxybutyrate (PHB) and a copolymer of hydroxybutyrate and hydroxyvalerate (PHV) featured the strength necessary for the healing of muscle-fascial wounds. The reaction of tissues to polymeric implants was similar to their reaction to silk and was less pronounced than the reaction to catgut; it was expressed in a transient post-traumatic inflammation (up to four weeks) and the formation of a fibrous capsule less than 200 ?m thick, which became as thin as 4060 ?m after 16 weeks, in the course of reverse development. Macrophages and foreign-body giant cells with a high activity of acid phosphatase were actively involved in this process. PHB and PHB/PHV sutures implanted intramuscularly for an extended period (up to one year) did not cause any acute vascular reaction at the site of implantation or any adverse events, such as suppurative inflammation, necrosis, calcification of the fibrous capsule or malignant tumor formation. No statistically significant differences were revealed in the tissue response to polymer sutures of the two types. Capsules around silk and catgut sutures did not become significantly thinner.

Аннотация: Polyhydroxyalkanoate (PHA) sutures were implanted to test animals intramuscularly, and tissue reaction was investigated and compared with the reaction to silk and catgut. Tested monofilament sutures made of PHAs of two types-polyhydroxybutyrate (PHB) and a copolymer of hydroxybutyrate and hydroxyvalerate (PHV)-featured the strength necessary for the healing of muscle-fascial wounds. The reaction of tissues to polymeric implants was similar to their reaction to silk and was less pronounced than the reaction to catgut; it was expressed in a transient post-traumatic inflammation (up to four weeks) and the formation of a fibrous capsule less than 200 mum thick, which became as thin as 40-60 mum after 16 weeks, in the course of reverse development. Macrophages and foreign-body giant cells with a high activity of acid phosphatase were actively involved in this process. Results of the study of tissue response to biodegradable polyoxyalkanoate suture threads in the postoperative period are analyzed. These suture threads have been shown to have the required strength and meet a number of other requirements for currently available materials.

Аннотация: Tissue reaction to implantation of polymeric microparticles from resorbable polymer (polyhydroxybutyrate) is characterized by slight inflammatory reaction and pronounced progressive macrophage infiltration with the presence of mono-and multinuclear foreign body giant cells resorbing the polymeric matrix. No fibrous capsules were formed around the polymeric microparticles; neither necrosis nor other adverse morphological changes and tissue transformation in response to implantation of the PHB microparticles were recorded. The results indicate good prospects of using polyhydroxybutyrate for the construction of long-acting dosage forms as microparticles for intramuscular injection. Аннотация: The research on cellulose fiber-reinforced nanocomposites has increased by an unprecedented magnitude over the past few years due to its wide application range and low production cost. However, the incompatibility between cellulose and most thermoplastics has raised significant challenges in composite fabrication. This paper addresses the behavior of plasma-modified polyethylene (PE) reinforced with cellulose nanofibers extracted from isora plants (i.e., isora nanofibrils (INFs)). The crystallization kinetics of PE–INF composites were explained using the Avrami model. The effect of cellulose nanofillers on tuning the physiochemical properties of the nanocomposite was also explored in this work. The increase in mechanical properties was due to the uniform dispersion of fillers in the PE. The investigation on viscoelastic properties confirmed good filler–matrix interactions, facilitating the stress transfer. © 2021 by the authors. Licensee MDPI, Basel, Switzerland.

Аннотация: Biodegradable poly-3-hydroxybutyrate [P(3HB)] and natural fillers - clay, peat, and birch wood flour – were used to prepare powdered composites to form pellets and granules. Pellets were produced by cold pressing of polymer and filler powder whereas granules were produced from the powders wetted with ethanol. Characterization techniques like IR spectroscopy, differential scanning calorimetry, X-ray analysis, mechanical analysis and electron microscopy were employed to study the properties of the initial P(3HB) and fillers and the composites. Analysis of the IR spectra of the composites showed the absence of chemical bonds between the components, i.e. the composites were physical mixtures. Young's moduli of the pellets prepared from initial materials varied considerably, and the highest value was obtained for P(3HB) pellets (350 MPa). Studies of biodegradation of composite pellets and granules in the soil for 35 days showed that the residual mass of the pellets had decreased to 68% for P(3HB); 56.4% for P(3HB)/peat; 67% for P(3HB)/wood flour, and 64% for P(3HB)/clay; granules exhibited a similar mass loss, residual mass of the granules of P(3HB) was 68.4%, P(3HB)/peat 46.4%; P(3HB)/wood flour 77%, and P(3HB)/clay 74%. This shows the significance of the material as an eco-friendly composite without sacrificing its mechanical properties. © 2018

Аннотация: A set of polyhydroxyalkanoates are synthesized, and a comparative study of their physicochemical properties is performed. The molecular masses and polydispersities of polyhydroxyalkanoates are found to be independent of their chemical structures. It is shown that the temperature characteristics and degrees of crystallinity of polyhydroxyalkanoates are affected by the chemical compositions of the monomers and their quantitative contents in the polymers. The incorporation of 4-hydroxybutyrate, 3-hydroxyvalerate, and 3-hydroxyhexanoate units into the chain of poly(3-hydroxybutyrate) decreases its melting point and thermal degradation temperature relative to these parameters of a homogeneous poly(3-hydroxybutyrate) sample (175 В± 5 C and 275 В± 5 C, respectively). The higher the content of the second monomer units in the poly(3-hydroxybutirate) chain, the greater the changes. The degrees of crystallinity of polyhydroxyalkanoate copolymers are generally lower than that of poly(3-hydroxybutyrate) (75 В± 5%). The effect on the ratio of the amorphous and crystalline phases of the copolymer samples becomes more pronounced in the series 3-hydroxy-valerate-3-hydroxyhexanoate-4- hydroxybutyrate. The addition of free pesticides adversely affected the abundance of soil microorganisms; the decrease varied from 1.4 to 56.0 times for different types of pesticides. The slow release pesticide formulations, in contrast to the free pesticides, exerted a much weaker effect on soil microorganisms, no significant inhibition in the abundance of saprotrophic bacteria was observed, partly due to the positive effects of the composite matrix (polymer/natural material), which was a supplementary substrate for microorganisms. The slow release fungicide formulations, like the free fungicides, reduced the total abundance of fungi and inhibited the development of the phytopathogens Fusarium and Alternaria. Thus, slow release formulations of pesticides preserve the bioremediation potential of soil microorganisms, which are the main factor of removing xenobiotics from the biosphere. © 2020, Springer-Verlag GmbH Germany, part of Springer Nature.

Electron microscopy showed differences in the surface microstructure of the films. Films prepared from the P(3HB) were more hydrophobic and the arithmetic mean surface roughness of 71–75 nm, than the copolymer films, which were hydrophilic (57–60°) and had considerably higher roughness (158–177 nm). Blood parameters (hemoglobin and hemolysis) and response of the cells (erythrocytes, platelets, and monocytes) were studied in experiments with blood directly contacting the surface of the films of PHAs with different compositions. Cultivation of blood cells on polymer films did not cause any adverse effects on adhesion and morphology of all cell types. Results of studying blood cell response suggested that the films made from low-crystallinity copolymers containing 4?hydroxybutyrate and 3?hydroxyhexanoate were the best for contact with blood. © 2019 Elsevier B.V.

Аннотация: The ability of marine luminescent bacteria to synthesize polyesters of hydroxycarboxylic acids (polyhydroxyalkanoates, PHA) as reserve macromolecules was studied. Twenty strains from the collection of the luminescent bacteria CCIBSO (WDCM839) of the Institute of Biophysics, Siberian Branch, Russian Academy of Sciences, assigned to different taxa (Photobacterium leiognathi, Ph. phosphoreum, Vibrio harveyi, and V. fischeri) were analyzed. The most productive strains were identified, and the conditions ensuring high polymer yields in batch culture (40-70% of the cell dry mass weight) were determined. The capacity for synthesizing two-and three-component polymers containing hydroxybutyric acid as the main monomer and hydroxyvaleric and hydroxyhexanoic acids was revealed in Ph. leiognathi and V. harveyi strains. The results allow luminescent microorganisms to be regarded as new producers of multicomponent polyhydroxyalkanoates. В© 2008 MAIK Nauka.

Аннотация: The synthesis of polyhydroxyalkanoates (PHAs) was scaled up to pilot production in a 150-L fermenter on sugars (fructose and glucose) and purified and crude glycerol in a culture of the wild-type strain Cupriavidus necator B-10646. Over 60 h of cultivation, a cell concentration of 150-160 g/L was obtained on purified glycerol and glucose; cultivation on fructose and crude glycerol resulted in a cell concentration of 130 ± 10 g/L. Polymer content and yield coefficients for the biomass were similar on all substrates (80-85 wt % and 0.29-0.33 kg biomass/kg carbon substrate, respectively). Copolymers poly(3-hydroxybutyrate-co-3-hydroxyvalerate) and poly(3-hydroxybutyrate-co-4-hydroxybutyrate) and terpolymers poly(3-hydroxybutyrate-co-3-hydroxyvalerate-co-4-hydroxybutyrate) having a decreased degree of crystallinity (36-46%) were first synthesized in the scaled-up process using C. necator B-10646 cultivated on glycerol. These results will provide the basis for scaling-up PHA synthesis in an organotrophic C. necator B-10646 culture. © 2019 American Chemical Society.

Аннотация: The present study addresses growth parameters and physiological and biochemical characteristics of the aerobic CO-oxidizing carboxydobacterium Seliberia carboxydohydrogena Z-1062. Poly(3-hydroxybutyrate) yields were investigated in experiments with limiting concentrations of mineral nutrients (nitrogen or sulfur or nitrogen and sulfur) in batch culture of S. carboxydohydrogena Z-1062 grown on gas mixtures consisting of CO2, O-2, H-2, and CO. CO concentrations of 10, 20, and 30 % v/v did not affect polymer synthesis, whose content after 56-h cultivation under limiting concentrations of nitrogen and sulfur was 52.6-62.8 % of biomass weight at a productivity of 0.13-0.22 g/L h. The inhibitory effect of CO on cell concentration was revealed at CO concentration of 30 % v/v. That also caused a decrease in substrate (H-2 and O-2) use efficiency. Thus, this carboxydobacterium can be regarded as a potential producer of polyhydroxyalkanoates from industrial hydrogenous sources.

Аннотация: BACKGROUND: P(3HB-co-3HHx) copolymers are very promising biomaterials. The main challenge in the production of these polymers is to simultaneously achieve high cell biomass; high P(3HB-co-3HHx) content; and high molar fraction of 3HHx in P(3HB-co-3HHx). The most common approach to production of these copolymers is the use of recombinant bacterial strains. The purpose of this study was to optimize the process of production of P(3HB-co-3HHx) copolymers containing high molar fractions of 3HHx by using the wild-type strain Cupriavidus eutrophus B10646. RESULTS: Kinetic properties of C. eutrophus B10646 were studied during cultivation of the cells on substrates necessary for P(3HB-co-3HHx) synthesis: glucose, nitrogen, sodium hexanoate, and sodium acrylate. The physiological ranges of their effects were determined experimentally, and C. eutrophus B10646 was grown in culture media with different dosages of these substrates. P(3HB-co-3HHx) copolymers with different molar fractions of 3HHx, including high ones (12 to 68%), were synthesized, and their physicochemical and mechanical properties were investigated. CONCLUSION: For the first time, cultivation conditions of Cupriavidus eutrophus B10646 enabled production of high biomass yields (5-6gL-1) and high content of the polymer (60-75%) that contained high 3HHx molar fraction. By varying the 3HB/3HHx ratio, one can change physicochemical and mechanical properties of P(3HB-co-3HHx) copolymers. © 2014 Society of Chemical Industry © 2016 Society of Chemical Industry.

Аннотация: Synthesis of 3- and 4-hydroxybutyrate copolymer (3HB-co-4HB), the most promising member of the biodegradable polyhydroxyalkanoate (PHA) family, has been studied. Cultivation conditions of naturally occurring strains of hydrogen-oxidizing bacteria Ralstonia eutropha B5786 and Cupriavidus eutrophus B10646 have been optimized to ensure efficient synthesis of the 3HB-co-4HB copolymer. A set of highly pure samples of the 3HB-co-4HB copolymer with 4HB content varying from 8.7 to 24.3 mol% has been obtained. Incorporation of 4HB into the copolymer was shown to cause a more pronounced decrease in polymer crystallinity than the incorporation of 3-hydroxyvalerate or 3-hydroxyhexanoate; samples with a degree of crystallinity below 30% have been obtained. The weight average molecular mass of the 3HB-co-4HB copolymers was shown to be independent on the monomer ratio and to vary broadly (from 540 to 1110 kDa). В© 2010 Pleiades Publishing, Ltd.
