Аннотация: This study investigates kinetic and production parameters of a glucose-utilizing bacterial strain, C. eutrophus B-10646, and its ability to synthesize PHA terpolymers. Optimization of a number of parameters of bacterial culture (cell concentration in the inoculum, physiological activity of the inoculum, determined by the initial intracellular polymer content, and glucose concentration in the culture medium during cultivation) provided cell concentrations and PHA yields reaching 110 g/L and 80%, respectively, under two-stage batch culture conditions. Addition of precursor substrates (valerate, hexanoate, propionate, c-butyrolactone) to the culture medium enabled synthesis of PHA terpolymers, P(3HB/3HV/4HB) and P(3HB/3HV/3HHx), with different composition and different molar fractions of 3HB, 3HV, 4HB, and 3HHx. Different types of PHA terpolymers synthesized by C. eutrophus B-10646 were used to prepare films, whose physicochemical and physicalmechanical properties were investigated. The properties of PHA terpolymers were significantly different from those of the P3HB homopolymer: they had much lower degrees of crystallinity and lower melting points and thermal decomposition temperatures, with the difference between these temperatures remaining practically unchanged. Films prepared from all PHA terpolymers had higher mechanical strength and elasticity than P3HB films. In spite of dissimilar surface structures, all films prepared from PHA terpolymers facilitated attachment and proliferation of mouse fibroblast NIH 3T3 cells more effectively than polystyrene and the highly crystalline P3HB.

Аннотация: The recombinant Ca(2+)-activated photoprotein obelin was used as a reporter protein in a solid-phase bioluminescent hybridization DNA assay. Oligonucleotide probes were immobilized on the surface of polymer methacrylate beads or microbiological plates of different types. A 30-mer oligonucleotide or its derivative with the biotin residue on the 3'-terminus, as well as a denatured double-stranded PCR fragment of the hepatitis C virus with the sequence of the 30-mer oligonucleotide was used as a DNA template. The probe in the hybridization complex was labeled by the elongation of the chain using a Taq DNA polymerase in the presence of biotinylated deoxyuridine triphosphate. The results of the bioluminescent assay were compared with the results of colorimetric analysis obtained with alkaline phosphatase as a reporter protein. It was shown that the use of the bioluminescent obelin label substantially accelerates the DNA detection procedure, provides a high sensitivity of the assay (no less than 10(-15) mol of DNA template), and ensures a quantitative determination of the amount of DNA template in the tested sample.

Аннотация: Samples of a hybrid composite of polyhydroxybutyrate (PHB), a biodegradable polyester, and hydroxyapatite (HA), with different PHB/HA ratios, have been prepared using mechanical-physical method. Electron microscopy, X-ray structure analysis and differential thermal analysis have been used to investigate the structure and physicochemical properties of the composite, depending on the PHB/HA ratio. The properties of the surface of the HA-loaded composite are significantly different from those of the pure polymer. As the HA percentage in the composite increases, free interface energy, the cohesive force, i.e., the strength of the adhesive bond between the composite surface and the water phase, and surface wettability increase. The HA percentage of the composite does not influence its melting temperature, but affects the temperature for the onset of decomposition: as the HA content increases from 0 to 10% (w/w), Td decreases from 260В°C to 225В°C. The degree of crystallinity of PHB/HA increases from 77% to 89% with an increase in the HA fraction from 10% to 50%. Functional properties of the composites have been investigated in vitro and in vivo. The best parameters of growth and differentiation of murine marrow osteoblasts are registered on PHB/HA samples containing 10% and 20% HA. In ectopic bone formation assay it has been proven that the hybrid PHB/HA composites can function as scaffolds and that bone tissue develops on their surface and in pores. В© VSP 2006.

Аннотация: A procedure has been developed to fabricate polymer tubes using solution casting inside template tubes. Polyester solution placed inside the vertically fixed template evaporated forming a hollow tube on the inner walls of the template. Silicone tubes used as the templates were permeable to solvent vapors and had relatively low adhesion to materials of the fabricated tubes, enabling their effortless removal. Parameters of the polymer tubes were determined by a type of the polymer, its concentration in the solution, and the inner diameter of the template. In the first method, a clamp was placed on the lower end of the template tube. A thickness of the new tube walls gradually increased from the open to the clamped end of the template tube. In another method viscous polymer solution without clamping was used; in this case, a polymeric bulkhead was formed in the middle of the tube. Two microbial polyesters, poly-3-hydroxybutyrate and poly-3-hydroxybutyrate-co-3-hydroxyvalerate, and also synthetic poly-L-lactide were used as model polymers. (C) 2020 Elsevier B.V. In the present study, the metribuzin (MET), tribenuron-methyl (TBM), and fenoxaprop-P-ethyl (FPE) herbicides were encapsulated in microparticles of degradable microbial polymers – polyhydroxyalkanoates (PHAs) – of two types – poly-3-hydroxybutyrate [P(3HB)] and poly(3-hydroxybutyrate-co-3-hydroxyvalerate [P(3HB/3HV)]. The use of P(3HB) resulted in higher yields of microparticles (63% to 79%) and larger sizes of the particles, whose average diameter was 0.60 ± 0.06–0.75 ± 0.11 ?m, while the average diameter of copolymer particles varied between 0.43 ± 0.12 and 0.55 ± 0.05 ?m. Encapsulation efficiency was rather determined by the type of herbicide and its solubility, varying from 24.7% to 48.2%. In vitro herbicide release from microparticles to water was affected by herbicide solubility and PHA chemical composition. The readily soluble MET showed the highest release rate, and over 30 days, 64% and 78% of the encapsulated amounts were released from P(3HB) and P(3HB/3HV) microparticles, respectively. High herbicidal activity of microparticles loaded with metribuzin and tribenuron-methyl was demonstrated in the laboratory stands of the Elsholtzia ciliata weed plant. © 2021 Wiley Periodicals LLC.

The use of P(3HB) resulted in higher yields of microparticles (63% to 79%) and larger sizes of the particles, whose average diameter was 0.60 +/- 0.06-0.75 +/- 0.11 mu m, while the average diameter of copolymer particles varied between 0.43 +/- 0.12 and 0.55 +/- 0.05 mu m. Encapsulation efficiency was rather determined by the type of herbicide and its solubility, varying from 24.7% to 48.2%. In vitro herbicide release from microparticles to water was affected by herbicide solubility and PHA chemical composition. The readily soluble MET showed the highest release rate, and over 30 days, 64% and 78% of the encapsulated amounts were released from P(3HB) and P(3HB/3HV) microparticles, respectively. High herbicidal activity of microparticles loaded with metribuzin and tribenuron-methyl was demonstrated in the laboratory stands of the Elsholtzia ciliata weed plant.

Аннотация: The synthesis of polyhydroxybutyrate by the carboxydobacterium Seliberia carboxydohydrogena under various growth conditions was studied. The bacterium was able to accumulate poly-beta-hydroxybutyric acid and a heteropolymer that was a product of copolymerization of hydroxybutyric and hydroxyvaleric acids. Nitrogen and sulfur exerted the strongest effect on polymer accumulation in carboxydobacteria: The maximum polymer concentrations in cells reached 28%. Specimens of polyhydroxybutyrate films were obtained, and their degradability by soil microflora was shown.

Аннотация: A series of 3D implants and filling materials prepared from powdered biodegradable polymers, polyhydroxyalkanoates (PHAs), have been designed for the purposes of reparative osteogenesis. The 3D implants are made of resorbable polymer of hydroxybutyric acid (poly-3-hydroxybutyrate, P3HB) and a composite of this polymer with hydroxyapatite (HA) (P3HB/HA). The properties of the implants were studied in vivo in a model of segmental osteotomy and compared with commercial material Bio-Oss®. All implants containing P3HB as the main component facilitate reconstructive osteogenesis. P3HB and P3HB/HA show pronounced osteoplastic properties; their in vivo degradation is slow and corresponds to the growth of a new bone tissue, facilitating normal reparative osteogenesis. Also, powdered P3HB and P3HB/tienam can be used as filling materials for osteoplasty of bone cavities infected by Staphylococcus aureus. Biodegradable 3D implants and P3HB-based filling materials show pronounced osteoplastic properties and degrade in vivo at a slow rate, enabling normal reparative osteogenesis.

Аннотация: The study addresses consequences of subcutaneous implantation of film matrices prepared from different PHAs to laboratory animals. No negative effects of subcutaneous implantation of PHA matrices on physiological and biochemical characteristics of the animals were determined. Independently of the matrices composition and duration of the contact with the internal environment of the organism we did not observe any deviations in the behavior of animals, their growth and development, as well as blood functions. Response of the tissues to PHA matrices was comparable with the response to polylactide, but substantially less expressed at the earlier time periods after implantation. Tissues response to implantation of PHA of all types is characterized by short-term (up to 2 weeks) post-traumatic inflammation with formation of fibrous capsules by 30th-60th days with the thickness less than 100 microns, which get thinner down to 40-60 microns by 180th day as the result of involution. No differences in response of tissues and the whole organism were observed for the matrices produced from the homopolymer of 3-hydroxybutyric acid (P3HB), copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids (P3HB/4HB), 3-hydroxybutyric acid and 3-hydroxyvalerianic acids (P3HB/3HV), 3-hydroxybutyric and 3-hydroxyhexanoate acids (P3HB/3HH). Macrophages and foreign-body giant cells actively participate in the response of the tissues to PHAs. In the studied conditions matrices from the copolymers containing 3-hydroxyhexanoate and 4 hydroxybutyrate were determined as more actively degraded PHA. The next less degraded matrices were matrices from the copolymer of P3HB/3HV and the most resistant were P3HB matrices. The slower degradation of PHA matrices was accompanied by delayed development of giantcells response. The studied PHA matrices can be placed in the following range by their degradation: P3HB/3HH - P3HB/4HB - P3HB/HV - P3HB.

Аннотация: The present study describes production of bacterial cellulose composites with silver nanoparticles and antibiotics and compares their properties. Bacterial cellulose (BC) composites synthesized in the culture of the strain of acetic acid bacterium Komagataeibacter xylinus VKPM B-12068 with silver nanoparticles, BC/AgNps, were produced hydrothermally, under different AgNO3 concentrations (0.0001, 0.001, and 0.01 M) in the reaction medium. The presence of silver in the BC/AgNp composites was confirmed by elemental analysis conducted using scanning electron microscopy with a system of X-ray spectral analysis. Analysis showed that the average atomic number of silver particles in composite samples depended on the concentration of AgNO3: as AgNO3 concentration in the reaction solution was increased, silver content in the composites increased from 0.044 to 0.37 mg/cm2. BC composites with amikacin and ceftriaxone were prepared by immersing dry BC films in solutions containing different concentrations of the antibiotics. The surface structure and properties and physicochemical and mechanical characteristics of composites were investigated using SEM, DSC, X-ray analysis, the system for measuring water contact angles, and electromechanical tensile testing machine. The disk-diffusion method and the shake-flask culture method used in this study showed that all experimental composites had pronounced antibacterial activity against E. coli, Ps. eruginosa, K. pneumoniae, and St. aureus, and the BC/antibiotic composites were more active than BC/AgNp ones; S. aureus was the most susceptible to the effect of BC composites. No potential cytotoxicity was detected in any of the BC/AgNp composites in the NIH 3T3 mouse fibroblast cell culture, in contrast to the BC/antibiotic composites. These results suggest that BC composites constructed in the present study hold promise as dressings for managing wounds, including contaminated ones. © 2017 Elsevier Ltd

Аннотация: In this study, tebuconazole (TEB)-loaded poly-3-hydroxybutyrate (P3HB)-based microparticles were developed and comprehensively characterized. TEB-loaded microparticles with the initial loading amounts of the fungicide of 10, 25, and 50% of the polymer mass (TEB 10, TEB 25, and TEB 50%) were prepared using emulsion technique. Encapsulation efficiency of TEB varied from 59 to 86%. As the loading amount was increased, the average diameter of microparticles increased too, from 41.3 to 71.7 µm, while zeta potential was not influenced by TEB loading, varying between –32.6 and –35.7 mV. TEB was gradually released from the microparticles to the model medium, and after 60 d, from 25 to 43% of TEB was released depending on the content of the encapsulated fungicide. The data obtained from in vitro TEB release were fitted to different mathematical models. It was shown that the release profiles of TEB could be best explained by the Zero-order, Higuchi, and Hixson–Crowell models. The antifungal activity of the P3HB/TEB microparticles against phytopathogenic fungi Fusarium moniliforme and Fusarium solani was demonstrated by in vitro tests conducted in Petri dishes. Thus, hydrophobic agrochemicals (TEB) can be effectively encapsulated into P3HB microparticles to construct slow-release formulations. © 2019, © 2019 Informa UK Limited, trading as Taylor & Francis Group.

Аннотация: The efficacy of slow-release formulations of tribenuron-methyl (TBM) embedded in the matrix of degradable poly(3-hydroxybutyrate) blended with birch wood flour [polymer/wood flour/herbicide 50/30/20 wt.%] was compared with the efficacy of TBM as the active ingredient of the Mortira commercial formulation, which was applied as post-emergence spray to treat spring wheat cv. Novosibirskaya 15. The study was conducted in Central Siberia (in the environs of the city of Krasnoyarsk, Russia) from May to August 2020. The biological efficacy of the embedded TBM was 92.3%, which was considerably higher than the biological efficacy of the Mortira formulation used as the post-emergence spray (15.4%). The embedding of TBM into degradable blended matrix enabled long-duration functioning of this unstable herbicide in soil. The sensitivity of weed plants to TBM differed depending on the species. TBM was more effective against A. retroflexus and A. blitoides, which were killed at an earlier stage, than against C. album and G. aparine, whose percentage increased in the earlier stage and which were controlled by the herbicide less effectively and at later stages. On the plot treated with the embedded herbicide, the parameters of the wheat yield structure were the best, and the total yield was the highest: 3360 +/- 40 kg/ha versus 3250 +/- 50 kg/ha in the group of plants sprayed with the Mortira formulation. The grain produced in all groups was of high quality and was classified as Grade 1 food grain. The highest quality parameters (grain hectoliter mass, gluten, and protein contents) were obtained in the group of plants treated with the embedded herbicide. The study of the embedded TBM confirmed the high efficacy of the experimental formulation.

Аннотация: It has been found that the carbon monoxide (CO)-resistant strain of the hydrogen bacteria Ralstonia eutropha B5786 is able to synthesise polyhydroxyalkanoates (PHAs) in the presence of CO under autotrophic conditions. This strain, grown on model gas mixtures containing 5-25% CO v/v), accumulates up to 70-75% (of absolutely dry matter) PHA, without significant variation in the yield coefficient on hydrogen. No suppression of the activities of the key enzymes of PHA synthesis (beta-ketothiolase, acetoacetyl-CoA-reductase, butyrate dehydrogenase and poly-3-hydroxybutyrate synthase) was recorded. The PHA synthesised is a copolymer containing mostly beta-hydroxybutyrate (more than 99 mol%) with trace amounts of beta-hydroxyvalerate. The investigated properties of the polymer (molecular weight, crystallinity, temperature characteristics) do not differ from those of the polymer synthesised on electrolytic hydrogen.

During the particle preparation process, P(3-HB) molecular weight decreased by 15-20%. Sterile microparticles (mean diameter 2.4 ?m) were injected into the tail veins of Wistar rats (5 mg/rat). The effects of the particles administered to rats were studied based on the general response of animals and local response of internal organ tissues and blood morphology; no adverse effects on growth and development of the animals or unfavorable changes in the structure of the tissues of internal organs were observed. Measurements of radioactivity in tissues showed that 14C concentrations are different in different organs, changing during the course of the experiment. The main targets for 14C-labeled microparticles were tissues of the liver, spleen, and kidneys. Comparison of radioactivity levels and residual contents of high-molecular-weight matrix in tissues suggested that the most rapid metabolism and degradation of P(3-HB) occurred in the liver and spleen. Gel-permeation chromatography showed that at 3 h after the microparticles were injected into the bloodstream, polymer degradation started in all examined organs, except the lungs; at 12 weeks, the M w of the polymer matrix was as low as 20-30% of its initial value. The presence of high-molecular-weight (undegraded) polymer in the tissues at 12 weeks after administration of the particles suggests that P(3-HB) is degraded in tissues of internal organs slowly and, hence, P(3-HB)-based microparticles can be used as sustained-release drug-delivery systems. Аннотация: Microspheres have been prepared from the resorbable linear polyester of ?-hydroxybutyric acid (polyhydroxybutyrate, PHB) by the solvent evaporation technique and investigated in vitro and in vivo. Biocompatibility of the microspheres has been proved in tests in the culture of mouse fibroblast cell line NIH 3T3 and in experiments on intramuscular implantation of the microspheres to Wistar rats for 3 months. Tissue response to the implantation of polymeric microspheres has been found to consist in a mild inflammatory reaction, pronounced macrophage infiltration that increases over time, involving mono- and poly-nuclear foreign body giant cells that resorb the polymeric matrix. No fibrous capsules were formed around polymeric microparticles; neither necrosis nor any other adverse morphological changes and tissue transformation in response to the implantation of the PHB microparticles were recorded. The results of the study suggest that polyhydroxybutyrate is a good candidate for fabricating prolonged-action drugs in the form of microparticles intended for intramuscular injection. Аннотация: BACKGROUND: Increasing use of mineral fertilisers can lead to accumulation of fertilisers in soil, water and foodstuffs. One of the approaches to preventing these problems is to develop controlled release forms of fertilisers. RESULTS: Experimental formulations of the nitrogen fertiliser urea loaded in a degradable matrix of the natural polymer poly-3-hydroxybutyrate (P3HB) in the form of films, pellets and coated granules were constructed and investigated. Nitrogen release into soil occurred as the polymer was degraded, and it was dependent on the geometry of the carrier and the amount of nitrogen loaded in it, showing that nitrogen release can last for 30 days or longer and that release rates can be controlled by varying the fabrication technique employed. P3HB/urea formulations have a favourable effect on the soil microbial community. The use of embedded urea has a beneficial influence on the growth of creeping bentgrass (Agrostis stolonifera) and lettuce (Latuca sativa) and reduces removal of nitrogen with drain water. CONCLUSION: The slow-release nitrogen formulations developed in this study can be buried in soil together with seeds preventing nitrogen deficiency. The use of such slow-release formulations can decrease the amounts of chemicals in the environment and prevent their adverse effects on the biosphere. © 2016 Society of Chemical Industry. © 2016 Society of Chemical Industry
