Аннотация: Spontaneous assembling of a simplest bacterial cell even if all necessary molecules are present in a solution seems to be extremely rare event and from the scientific standpoint has to be considered as impossible. Therefore, a predecessor of a living cell has to be very simple for providing its self-assembling and at the same time it should be able of progressive increase in complexity. Now phase-separated particles, first of all micelles, are put forward as possible predecessors of living cell. According to the offered working concept only phase-separated particles possessing autocatalytic properties can be considered as predecessors of living cells. The first stage of evolution of these phase-separated autocatalytic systems is the appearance of pre-biotic metabolism providing synthesis of amphiphiles for formation of capsules of these systems. This synthesis is maintained by the energy of a base reaction being a component of a planet-chemical cycle. Catalytic system providing functioning of pre-biotic metabolism is based on multivariate oligomeric autocatalyst, which reproduces itself from monomers, penetrating the particles from the outside. Since the autocatalyst realizes random polymerization then a collection of other oligomers possessing different catalytic functions is produced. Аннотация: This review examines the general principles of bioluminescent enzymatic toxicity bioassays and describes the applications of these methods and the implementation in commercial biosensors. Bioluminescent enzyme system technology (BEST) has been proposed in the bacterial coupled enzyme system, wherein NADH: FMN-oxidoreductase-luciferase substitutes for living organisms. BEST was introduced to facilitate and accelerate the development of cost-competitive enzymatic systems for use in biosensors for medical, environmental, and industrial applications. For widespread use of BEST, the multicomponent reagent "Enzymolum'' has been developed, which contains the bacterial luciferase, NADH: FMN-oxidoreductase, and their substrates, co-immobilized in starch or gelatin gel. Enzymolum is the central part of Portable Laboratory for Toxicity Detection (PLTD), which consists of a biodetector module, a sampling module, a sample preparation module, and a reagent module. PLTD instantly signals chemical-biological hazards and allows us to detect a wide range of toxic substances. Enzymolum can be integrated as a biological module into the portable biodetector-biosensor originally constructed for personal use. Based on the example of Enzymolum and the algorithm for creating new enzyme biotests with tailored characteristics, a new approach was demonstrated in biotechnological design and construction. The examples of biotechnological design of various bioluminescent methods for ecological monitoring were provided. Possible applications of enzyme bioassays are seen in the examples for medical diagnostics, assessment of the effect of physical load on sportsmen, analysis of food additives, and in practical courses for higher educational institutions and schools. The advantages of enzymatic assays are their rapidity (the period of time required does not exceed 3-5 min), high sensitivity, simplicity and safety of procedure, and possibility of automation of ecological monitoring; the required luminometer is easily available.

Аннотация: A "producer-consumer" (Chlorella vulgaris-Paramecium caudatum) closed aquatic system has been investigated experimentally and theoretically. It has been found that there is a direct relationship between the growth of the paramecia population and their release of ammonia nitrogen, which is the best form of nitrogen for Chlorella growth. The theoretical study of a model of a "producer-consumer" aquatic biotic cycle with spatially separated compartments has confirmed the contribution of paramecia to nitrogen cycling. It has been shown that an increase in the concentration of nitrogen released as metabolites of paramecia is accompanied by an increase in the productivity of microalgae. (c) 2005 COSPAR. Аннотация: A model consisting of two blocks (equations) was proposed for the analytical, study of the biosphere-climate system over great periods of time. The first equation describes the balance of carbon dioxide in the Atmosphere and re presents the biological block of the model. The second equation is the equation of the energy balance or the physical block of the system. The model is based on the most general conceptions of living matter and the evolution process. A possible interpretation of some events and phenomena in the earth history in terms of the model is given.

Аннотация: The bioluminescent bacterial enzyme system NAD(P)H:FMN-oxidoreductase-luciferase has been used as a test system for ecological monitoring. One of the modes to quench bioluminescence is the interaction of xenobiotics with the enzymes, which inhibit their activity. The use of endogenous flavin fluorescence for investigation of the interactions of non-fluorescent compounds with the bacterial luciferase from Photobacterium leiognathi and NAD(P)H:FMN-oxidoreductase from Vibrio fischeri has been proposed. Fluorescence spectroscopy methods have been used to study characteristics of endogenous flavin fluorescence (fluorophore lifetime, the rotational correlation time). The fluorescence anisotropy behaviour of FMN has been analysed and compared to that of the enzyme-bound flavin. The fluorescence characteristics of endogenous flavin of luciferase and NAD(P)H:FMN-oxidoreductase have been shown to be applicable in studying enzymes' interactions with non-fluorescent compounds. Аннотация: The biomodule of bioluminescent biosensor based on a coupled enzyme system NADH:FMN-oxidoreductase and luciferase, co-immobilized with substrates in dried starch or gelatin gels, has been developed. We studied the impact of several stabilizers - dithiothreitol (DTT), bovine serum albumin (BSA) and mercaptoethanol (ME) on the biomodule's activity, storage stability and sensitivity to toxic substances. The inclusion of stabilizers increases the activity of the biological module by more than 150%. To achieve the combination of high activity, prolonged storage time and acute sensitivity to toxic substances within maximum permissible concentration we used starch gel as a carrier adding 100 ?M DTT to the immobilized preparation. The gelatin-based biological module had greater storage stability than the starch-based one but demonstrated less sensitivity to toxic substances. © 2015 Elsevier B.V. All rights reserved.

Аннотация: Coelenterazine-dependent copepod luciferases containing natural signal peptide for secretion are a very convenient analytical tool as they enable monitoring of intracellular events with high sensitivity, without destroying cells or tissues. This property is well suited for application in biomedical research and development of cell-based assays for high throughput screening. We report the cloning of cDNA gene encoding a novel secreted non-allelic 16.5-kDa isoform (MLuc7) of Metridia longa luciferase, which, in fact, is the smallest natural luciferase of known for today. Despite the small size, isoform contains 10 conservative Cys residues suggesting the presence of up to 5 S-S bonds. This hampers the efficient production of functionally active recombinant luciferase in bacterial expression systems. With the use of the baculovirus expression system, we produced substantial amounts of the proper folded MLuc7 luciferase with a yield of similar to 3 mg/L of a high purity protein. We demonstrate that MLuc7 produced in insect cells is highly active and extremely thermostable, and is well suited as a secreted reporter when expressed in mammalian cells ensuring higher sensitivity of detection as compared to another Metridia luciferase isoform (MLuc164) which is widely employed in real-time imaging. Аннотация: This work considers antioxidant properties of natural detoxifying agents-humic substances (HS) in solutions of model inorganic and organic compounds of oxidative nature-complex salt K-3[Fe(CN)(6)] and 1,4-benzoquinone. Bioluminescent system of coupled enzymatic reactions catalyzed by NAD(P) H:FMN-oxidoreductase and bacterial luciferase was used as a bioassay in vitro to monitor toxicity of the oxidizer solutions. Toxicities of general and oxidative types were evaluated using bioluminescent kinetic parameters-bioluminescence intensity and induction period, respectively. Antioxidant activity of HS was attributed to their ability to decrease both general and oxidative toxicities; the HS antioxidant efficiency was characterized with detoxification coefficients D-GT and D-OxT, respectively. Dependencies of D-GT and D-OxT on HS concentration and time of preliminary incubation of the oxidizers with HS were demonstrated. The optimal conditions for detoxification of the oxidizers were >20-min incubation time and 0.5x10(-4) to 2x10(-4) M of HS concentration. The present study promotes application of the enzymatic luminescent bioassay to monitor toxicity of pollutants of oxidative nature in environmental and waste waters in remediation procedures.

Аннотация: For the identification of ecologically significant dominant groups of phytoplanktonic algae a polychromatic DCMU-induced fluorescence method is recommended. A special fluorometer equipped with a system of replaceable filters is used to differentiate three regions of the spectrum (410 +/- 20, 510 +/- 20 and 540 +/- 10 nm) that can excite the basic light-harvesting pigments. Total and differential (for every algal taxon studied) chlorophyll a calculated from the fluorescence signals is in good agreement with biomass estimates from direct cell counts for several different trophic types of aquatic systems. This is made possible by the vizualization of the ratios of fluorescence signal values in their own coordinates: first, to decide whether it is necessary to correct linear equations in order to eliminate negative solutions; second, to determine the possibility of nulling the negative solution if a point is situated close to a side of the triangle; and third, to reduce the number of linear algebraic equations to two if the points are situated along one of the triangle sides or to one if the points are gathered at the apex. The polychromatic DCMU-induced fluorescence method can be used for monitoring natural phytoplankton populations to detect changes in their taxonomic structure.

Аннотация: The anthropogenic impact on the Earth's ecosysterns are leading to dramatic changes in ecosystem functioning and even to destruction of them. System analysis and the use of heuristic modeling can be an effective means to determine the main biological interactions and key factors that are of high importance for understanding the development of ecosysterns. Cycling of limiting substances, induced by the external free energy flux, and trophic links interaction is the basis of the mathematical modeling studies presented in this paper. Mathematical models describe the dynamics of simplified ecosysterns having different characteristics:1) different degrees of biotic turnover closure (from open to completely closed); 2) different numbers of trophic links (including both "top-down", "bottom-up" regulation types); 3) different intensities of input - output flows of the limiting nutrient and its total amount in the system. Adaptive values of the changes of lower hierarchical levels (populational, trophic chain level) are to be estimated by integrity indices for total system functioning (e.g. NPP, total photosynthesis). The approach developed can be used for evaluating the contributions of lower hierarchical levels to the functioning of the higher hierarchical levels of the system. Аннотация: The term Closed Ecological System (CES) is in wide use. However there is no generally accepted measure of the closure of ecological systems. In order to obtain reproducibility of experiments with natural and man-made CES (with respect to degree of closure) some universal estimate needs to be developed. Understanding ecological systems as a network and closure as the degree of matter recycling allows the use of matrix graphs. Graphs are very natural forms for the presentation of the network of matter flows in ecosystems. An estimate equal to the sum of products of weights of oriented edges that constitute contour is suggested as a measure of the degree of closure in ecosystems. It is shown that this estimate can be uniformly applied to ecosystems of arbitrary size and configuration of flows. (C) 2003 COSPAR. Published by Elsevier Science Ltd. All rights reserved.

Аннотация: Sustainability is one of the most important criteria in the creation and evaluation of human life support systems intended for use during long space flights. The common feature of biological and physicochemical life support systems is that basically they are both catalytic. But there are two fundamental properties distinguishing biological systems: 1) they are auto-catalytic: their catalysts - enzymes of protein nature - are continuously reproduced when the system functions; 2) the program of every process performed by enzymes and the program of their reproduction are inherent in the biological system itself - in the totality of genomes of the species involved in the functioning of the ecosystem. Actually, one cell with the genome capable of the phenotypic realization is enough for the self-restoration of the function performed by the cells of this species in the ecosystem. The continuous microalgal culture of Chlorella vulgaris was taken to investigate quantitatively the process of self-restoration in unicellular algae population. Based on the data obtained, we proposed a mathematical model of the restoration process in a cell population that has suffered an acute radiation damage. (C) 2003 COSPAR. Аннотация: The levels of free and bound forms of IAA, ABA, cytokinins (CK), and gibberellins, as well as growth characteristics and productivity were investigated in two wheat lines. The plants were grown under controlled conditions in an artificial ecosystem that allowed the irradiance, CO2 concentration, and rhizosphere temperature to be changed. The main difference in the hormonal status of leaves of tall spring wheat, line 232, and dwarf wheat, line 95-3, was the absence of GA(9) gibberellins in the latter. It was found that the light intensity and temperature of rhizosphere insignificantly affected the balance of endogenous phytohormones and HI in wheat. The elevation of CO2 concentration resulted in a considerable increase in the content of free IAA, an appearance of free GA(9), and a rise in the productivity of wheat, line 232. The concentration of CO2 was shown to be a major parameter that determined HI in the experimental ecological system.

Аннотация: Samples of olyhydroxyalkanoates (PHAs), polyhydroxybutyrate (PHB) and copolymers poly(hydroxybutyrate-co-hydroxyvalerate) (PHBV) with 4 and 18 mol% hydroxyvalerate, synthesized by the bacteria Ralstonia eutropha B5786, were investigated. PHA films in contact with blood did not activate the hemostasis system at the level of cell response, but they did activate the coagulation system and the complement reaction. To detect biologically-active components in the PHAs, a detailed analysis of the composition of the polymers was conducted. Gas chromatography-mass spectrometry revealed long-chain fatty acids (FAs) in the tested PHAs. Their total concentration in the polymer ranged from tenths of mol% to 2-3 mol%, depending on the purification method. C-16:0 constituted the largest proportion, up to 70%. Of the long-chain hydroxy acids, only beta-OH-C-14:0 was detected and it did not exceed 0.06 mol%. The analysis of the hemocompatibility properties of the PHAs purified by a specialized procedure, including the quantitative and morphological estimation of platelets adherent to the surface of polymer films, the plasma recalcification time and complement activation studies, indicated that PHB and PHBV can be used in contact with blood. It has been found out that the lipopolysaccharides of bacteria producing PHAs, which contain mostly long-chain hydroxy acids, can be the factor activating the hemostasis systems. Thus, the technology of PHA purification must satisfy rather stringent specific requirements.

Аннотация: This paper presents a mathematical model of interactions between two herbivorous invertebrates (ciliate Paramecium caudatum and rotifer Brachionus plicatilis) and two planktonic algae (Chlorella vulgaris and Scenedesmus quadricauda) spatially segregated in two compartments of a chemostat - type experimental microcosm system. The model mimics a "producer-consumer" aquatic biotic cycle, describing the dynamics of the mixed culture of ciliates and rotifers, as "consumer" compartment, feeding on the mixed algal culture, as "producer" compartment, under N-limiting conditions. We experimentally found that metabolites of the alga Scenedesmus produce an adverse effect on the reproduction of ciliate Paramecium. Taking this effect into account improved the behavior of the model, the results of which came into qualitative agreement with the experimental results. Both our experimental and modeling approaches demonstrated that, even in conditions of a spatially - segregated "producer-consumer" biotic cycle, species coexistence is impossible either in the mixed algal culture or in the mixed invertebrate culture. Scenedesmus excluded Chlorella, whereas Brachionus excluded Paramecium.

Аннотация: The question on the essence of life as phenomenon is the key one for astrobiology, since the answer to this question determines "breadth of our outlook". Taking Earth's version of life as the pattern extremely under-estimates our estimation of the probability of life origin and respectively expected probability of extraterrestrial life discovery. In the paper the hypothetical key attribute of life in general is selected on the base of comparative analyses and deductive inference. Simulation conducted on the base of neural network model shows that the same function could be realized by means of great variety of structures, which originated in the course of an evolutionary process. So multiplicity of evolutionary outcomes essentially increases the probability of final result - realization of an integrated function providing fitness to environment. Life as the integrated function can be realized via great variety of development ways and structures. A logical consequence of definitions for life as phenomenon is suggested. Final one is "Life is specific organization of informational and energetic processes coupling, enabling choice-making, and displayed as anomalies of different kinds". Anomalies of visible form, mechanical movement, chemical composition and noticeable response are considered. Presented in the paper sweeping generalization is not rigorously proven, however it can play heuristic role in increasing the level of specificity of searching for extraterrestrial life. Аннотация: We recently introduced a new electrophoretic method, nonequilibrium, capillary electrophoresis of equilibrium mixtures (NECEEM). NECEEM provides a unique way of finding kinetic and equilibrium parameters of the formation of intermolecular complexes from a single electropherogram and allows for the use of weak affinity probes in protein quantitation. In this work, we study theoretical bases of NECEEM by developing a mathematical model for the new method. By solving a system of partial differential equations with diffusion in linear approximation, we found the analytical solution for concentrations of components involved in complex formation as functions of time from the beginning of separation and position in the capillary. The nonnumerical nature of the solution makes it a powerful tool in studying the theoretical foundations of the NECEEM method and modeling experimental results. We demonstrate the use of the model for finding binding parameters of complex formation by nonlinear regression of NECEEM electropherograms obtained experimentally.

Аннотация: An experimental model of a biological life support system was used to evaluate qualitative and quantitative parameters of the internal mass exchange. The photosynthesizing unit included the higher plant component (wheat and radish), and the heterotrophic unit consisted of a soil-like substrate, California worms, mushrooms and microbial microflora. The gas mass exchange involved evolution of oxygen by the photosynthesizing component and its uptake by the heterotroph component along with the formation and maintaining of the SLS structure, growth of mushrooms and California worms, human respiration, and some other processes. Human presence in the system in the form of "virtual human" that at regular intervals took part in the respirative gas exchange during the experiment. Experimental data demonstrated good oxygen/carbon dioxide balance, and the closure of the cycles of these gases was almost complete. The water cycle was nearly 100% closed. The main components in the water mass exchange were transpiration water and the watering solution with mineral elements. Human consumption of the edible plant biomass (grains and roots) was simulated by processing these products by a unique physicochemical method of oxidizing them to inorganic mineral compounds, which were then returned into the system and fully assimilated by the plants. The oxidation was achieved by "wet combustion" of organic biomass, using hydrogen peroxide following a special procedure, which does not require high temperature and pressure. Hydrogen peroxide is produced from the water inside the system. The closure of the cycle was estimated for individual elements and compounds. Stoichiometric proportions are given for the main components included in the experimental model of the system. Approaches to the mathematical modeling of the cycling processes are discussed, using the data of the experimental model. Nitrogen, as a representative of biogenic elements, shows an almost 100% closure of the cycle inside the system, The proposed experimental model of a biological system is discussed as a candidate for potential application in the investigations aimed at creating ecosystems with largely closed cycles of the internal mass exchange. The formation and maintenance of sustainable cycling of vitally important chemical elements and compounds in biological life support systems (BLSS) is an extremely pressing problem. To attain the stable functioning of biological life support systems (BLSS) and to maintain a high degree of closure of material cycles in them, it is essential to understand the character of mass exchange processes and stoichiometric proportions of the initial and synthesized components of the system.
