Аннотация: In this work, delivery systems in the form of microparticles and films containing 1,2,4-trioxolane (ozonide, OZ) based on polyhydroxyalkanoates (PHAs) were developed. Main systems' characteristics were investigated: the particle yield, average diameter, zeta potential, surface morphology, loading capacity, and drug release profile of microparticles, as well as surface morphology and release profiles of OZ-containing films. PHA-based OZ-loaded microparticles have been found to have satisfactory size, zeta potential, and ozonide loading-release behavior. It was noted that OZ content influenced the surface morphology of obtained systems.

Аннотация: A key component of the recently described bioluminescent system of higher fungi is luciferase, a new class of proteins. The properties of fungal luciferase and their relationship with its structure are interesting both for improving autoluminescent systems already created on its basis and for creating new ones. Therefore, it is extremely important to understand the spatial structure of this protein. We have performed heterologous expression and purification of Neonothopanus nambi luciferase, obtained a protein suitable for subsequent crystallization, and also determined some biochemical properties of the recombinant luciferase.

Аннотация: Traditionally, the abundance, cell size distribution and species identification of algae are determined by microscopic counts. In recent years, various rapid methods have been developed for routine algal studies. However, each of these methods has its drawbacks. It is important for aquatic ecologists to understand the advantages, disadvantages, and limitations of these methods. We compared the sensitivity of three rapid methods (multichannel fluorimeter FluoroProbe, imaging flow cytometer FlowCam, and CASY particle counter) to changes in cell abundance of three algae species (Chlorella vulgaris Beyerinck, Arthrospira platensis Gomont, and Nostoc sp.). We also assessed the ability of rapid methods to estimate the cell abundance of different species in the mixed samples. All instruments showed high sensitivity to changes in the cell abundance of different algae species and a mixture of these species. Any one of these methods, once calibrated, can be reliably used to estimate the abundance of a single-species/laboratory culture of microalgae. At the same time, FlowCam, without preliminary calibration, recorded the cell abundance closest to microscopic counts. When analysing a mixture of three microalgae differing in their cell sizes and spectral characteristics, FluoroProbe showed the highest accuracy in assessing the proportions of species in the mixture and FlowCam - in assessing their abundance. To study mixtures of algae and/or natural phytoplankton communities, it is advisable to use jointly a flow cytometer and a multichannel fluorimeter. The images of algae saved by the flow cytometer, if necessary, can be used to identify them, with a certain accuracy, to the species. Information on cells size and spectral characteristics obtained by two methods will be detailed enough to perform such common tasks as studying trophic interactions between phyto- and zooplankton or creating warning systems to inform of unwanted blooms of phytoplankton and their individual groups (for example, cyanobacteria). © Siberian Federal University. Аннотация: The world map of anthropogenic atmospheric nitrogen deposition and its effects on natural ecosystems is not described with equal precision everywhere. In this paper, we report atmospheric nutrient, sulfate and spheroidal carbonaceous particle (SCP) deposition rates, based on snowpack analyses of a formerly unexplored Siberian mountain region. Then, we discuss their potential effects on lake phytoplankton biomass limitation. We estimate that the nutrient depositions observed in the late-season snowpack (40 +/- 16 mgNO(3)-Nm(-2) and 0.58 +/- 0.13 mg TP-Pm-2; TP for total phosphorous) would correspond to yearly depositions lower than 119 +/- 71 mgNO(3)-Nm(-2) yr(-1) and higher than 1.71 +/- 0.91 mg TP-Pm-2 yr(-1). These yearly deposition estimates would approximately fit the predictions of global deposition models and correspond to the very low nutrient deposition range, although they are still higher than world background values. In spite of the fact that such a low atmospheric nitrogen deposition rate would be enough to induce nitrogen limitation in unproductive mountain lakes, phosphorus deposition was also extremely low, and the resulting lake water N: P ratio was unaffected by atmospheric nutrient deposition. In the end, the studied lakes' phytoplankton appeared to be split between phosphorus and nitrogen limitation. We conclude that these pristine lakes are fragile sensitive systems exposed to the predicted climate warming, increased winter precipitation, enhanced forest fires and shifts in anthropogenic nitrogen emissions that could finally couple their water chemistry to that of atmospheric nutrient deposition and unlock temperature-inhibited responses of phytoplankton to nutrient shifts.

Аннотация: The application of detonation nanodiamonds (DNDs) in delivery systems has been demonstrated many times for a variety of substances, including anti-inflammatory drugs, antibiotics, antineoplastic agents, vitamins etc. The applicability of detonation nanodiamonds as a carrier for delivery of corticosteroids is investigated in this study. Commercial DNDs were modified to obtain particles with high colloidal stability in aqueous suspensions. Physical adsorption was then used to conjugate the DNDs with methylprednisolone sodium succinate and dexamethasone sodium phosphate. The experimental adsorption isotherms were fit with Langmuir and Freundlich adsorption models. The results of in vitro release study demonstrated an explicit prolonged effect of DNDs, due to which a sustained release of the adsorbed drugs was provided.

Аннотация: Productivity values, sodium accumulation in aboveground biomass, and photosynthetic indices of watercress (Nasturtium officinale) leaves were investigated under conditions resembling artificial closed ecological systems (CES). The seedlings were grown on nutrient media with various NaCl concentrations (0.7, 1.4, and 1.8 g/L) for 7, 14, and 19 days after transferring them to saline solutions. The productivity of plants on the seventh day of their growth on saline media did not differ from that of control plants. The decrease in plant productivity was noted in all the treatments starting from the 14th day after transferring the plants to saline solutions. When NaCl concentration in the nutrient solution was raised from 0.7 to 1.8 g/L, a significant increase in relative Na+ content in plant tissues was observed, regardless of the duration of NaCl treatment. A substantial decrease in chlorophyll (a + b) to carotenoid content ratio was noted on the seventh and 14th days in plants grown at 1.8 g/L NaCl. In plants treated for 7 days with 0.7 and 1.4 g/L NaCl, the content of chlorophylls a and b and carotenoids was found to increase, which indicates the tolerance of N. officinale to CES conditions. The relative content of chlorophylls a and b in the light-harvesting chlorophyll (a + b) complex was independent of the extent of salinity. The maximum quantum yield of photosystem II reaction in N. officinale plants had typically high values (Y(II)(max) of 0.755 +/- 0.007). Using the Imaging Maxi version of the pulse amplitude-modulated (PAM) fluorometer, it was found that light curves for the effective quantum yield of photochemical and nonphotochemical fluorescence quenching (Y(II) and Y(NPQ), respectively) differed appreciably between the salt-treated and untreated plants in the case of long-term cultivation (19 days) at 0.7 and 1.4 g/L NaCl. The treatment with 1.8 g/L NaCl for the period from 14 to 19 days had no effect on light curves of Y(II) and Y(NPQ). It is argued that N. officinale can be used as a source of NaCl for humans under CES conditions.

Аннотация: Thorium is one of the most widespread radioactive elements in natural ecosystems, along with uranium, it is the most important source of nuclear energy. However, the effects of thorium on living organisms have not been thoroughly studied. Marine luminescent bacteria and their enzymes are optimal bioassays for studying low-dose thorium exposures. Luminescent bioassays provide a quantitative measure of toxicity and are characterized by high rates, sensitivity, and simplicity. It is known that the metabolic activity of bacteria is associated with the production of reactive oxygen species (ROS). We studied the effects of thorium-232 (10(-11)-10(-3) M) on Photobacterium phosphoreum and bacterial enzymatic reactions; kinetics of bacterial bioluminescence and ROS content were investigated in both systems. Bioluminescence activation was revealed under low-dose exposures (<0.1 Gy) and discussed in terms of "radiation hormesis". The activation was accompanied by an intensification of the oxidation of a low-molecular reducer, NADH, during the enzymatic processes. Negative correlations were found between the intensity of bioluminescence and the content of ROS in bacteria and enzyme systems; an active role of ROS in the low-dose activation by thorium was discussed. The results contribute to radioecological potential of bioluminescence techniques adapted to study low-intensity radioactive exposures.

Аннотация: The wide use of pesticides in agriculture has caused uncontrolled distribution of these chemicals in the environment, calling for the development and investigation of new environmentally friendly formulations, which would reduce human impact on nature. In the present study, the metribuzin (MET), tribenuron-methyl (TBM), and fenoxaprop-P-ethyl (FPE) herbicides were encapsulated in microparticles of degradable microbial polymers – polyhydroxyalkanoates (PHAs) – of two types – poly-3-hydroxybutyrate [P(3HB)] and poly(3-hydroxybutyrate-co-3-hydroxyvalerate [P(3HB/3HV)]. The use of P(3HB) resulted in higher yields of microparticles (63% to 79%) and larger sizes of the particles, whose average diameter was 0.60 ± 0.06–0.75 ± 0.11 ?m, while the average diameter of copolymer particles varied between 0.43 ± 0.12 and 0.55 ± 0.05 ?m. Encapsulation efficiency was rather determined by the type of herbicide and its solubility, varying from 24.7% to 48.2%. In vitro herbicide release from microparticles to water was affected by herbicide solubility and PHA chemical composition. The readily soluble MET showed the highest release rate, and over 30 days, 64% and 78% of the encapsulated amounts were released from P(3HB) and P(3HB/3HV) microparticles, respectively. High herbicidal activity of microparticles loaded with metribuzin and tribenuron-methyl was demonstrated in the laboratory stands of the Elsholtzia ciliata weed plant. © 2021 Wiley Periodicals LLC.

Аннотация: The wide use of pesticides in agriculture has caused uncontrolled distribution of these chemicals in the environment, calling for the development and investigation of new environmentally friendly formulations, which would reduce human impact on nature. In the present study, the metribuzin (MET), tribenuron-methyl (TBM), and fenoxaprop-P-ethyl (FPE) herbicides were encapsulated in microparticles of degradable microbial polymers - polyhydroxyalkanoates (PHAs) - of two types - poly-3-hydroxybutyrate [P(3HB)] and poly(3-hydroxybutyrate-co-3-hydroxyvalerate [P(3HB/3HV)]. The use of P(3HB) resulted in higher yields of microparticles (63% to 79%) and larger sizes of the particles, whose average diameter was 0.60 +/- 0.06-0.75 +/- 0.11 mu m, while the average diameter of copolymer particles varied between 0.43 +/- 0.12 and 0.55 +/- 0.05 mu m. Encapsulation efficiency was rather determined by the type of herbicide and its solubility, varying from 24.7% to 48.2%. In vitro herbicide release from microparticles to water was affected by herbicide solubility and PHA chemical composition. The readily soluble MET showed the highest release rate, and over 30 days, 64% and 78% of the encapsulated amounts were released from P(3HB) and P(3HB/3HV) microparticles, respectively. High herbicidal activity of microparticles loaded with metribuzin and tribenuron-methyl was demonstrated in the laboratory stands of the Elsholtzia ciliata weed plant.

Аннотация: Carbon turnover in aquatic environments is dependent on biochemical properties of organic matter (OM) and its degradability by the surrounding microbial community. Non-additive interactive effects represent a mechanism where the degradation of biochemically persistent OM is stimulated by the provision of bioavailable OM to the degrading microbial community. Whilst this is well established in terrestrial systems, whether it occurs in aquatic ecosystems remains subject to debate. We hypothesised that OM from zooplankton carcasses can stimulate the degradation of biochemically persistent leaf material, and that this effect is influenced by the daphnia:leaf OM ratio and the complexity of the degrading microbial community. Fresh Daphnia magna carcasses and C-13-labelled maize leaves (Zea mays) were incubated at different ratios (1:1, 1:3 and 1:5) alongside either a complex microbial community (50 mu m) or solely bacteria (0.8 mu m). C-13 stable-isotope measurements of CO2 analyses were combined with phospholipid fatty acids (PLFA) analysis and DNA sequencing to link metabolic activities, biomass and taxonomic composition of the microbial community. Our experiments indicated a significantly higher respiration of leaf-derived C when daphnia-derived OM was most abundant (i.e. daphnia:leaf OM ratio of 1:1). This process was stronger in a complex microbial community, including eukaryotic microorganisms, than a solely bacterial community. We concluded that non-additive interactive effects were a function of increased C-N chemodiversity and microbial complexity, with the highest net respiration to be expected when chemodiversity is high and the degrading community complex. This study indicates that identifying the interactions and processes of OM degradation is one important key for a deeper understanding of aquatic and thus global carbon cycle.

Аннотация: This work is dedicated to developing enzyme biosensor software to solve problems regarding soil pollution analysis. An algorithm and specialised software have been developed which stores, analyses and visualises data using JavaScript programming language. The developed software is based on matching data of 51 non-commercial standard soil samples and their inhibitory effects on three enzyme systems of varying complexity. This approach is able to identify the influence of chemical properties soil samples, without toxic agents, on enzyme biosensors. Аннотация: The salinization of freshwaters due to the use of deicing road salts is a serious anthropogenic threat. We investigated the effects of the road deicer, which is mostly composed (ca. 70%) of NaCl, on the life cycle parameters of the cladoceran Moina macrocopa in acute and chronic toxicity tests and on the hatching success of resting eggs exposed to sediments contaminated with the road salt. The negative effects of the road salt on survival and life cycle parameters of animals were observed at concentrations above 5 g L?1. The 6-month exposure of resting eggs to contaminated sediments had a consistent but relatively weak effect on the postexposure hatching of resting eggs. Experiments demonstrated that the concentration of the deicer in the top water layer in the water-sediment systems is more important for the hatching success of resting eggs than the salt content in the sediment. Only 2.2 ± 1.9% of resting eggs hatched when the deicer content in the top water layer was equal to 12 g L?1. Lethal effects on hatchlings were observed starting from the deicer content in the water equal to 1 g L?1, and 97.0 ± 0.8% of hatchlings were dead at the deicer content in the water equal to 5 g L?1. Thus, the resilience of resting eggs to the contaminated sediments can ensure the replenishment of the population from the egg bank after the salinity disturbance is diminished but the negative effects of the elevated salt content in surface waters on active population will be manifested at lower salinities. © 2021, The Author(s), under exclusive licence to Springer Nature B.V. part of Springer Nature.

Аннотация: The application of detonation nanodiamonds (DNDs) in delivery systems has been demonstrated many times for a variety of substances, including anti-inflammatory drugs, antibiotics, antineoplastic agents, vitamins etc. The applicability of detonation nanodiamonds as a carrier for delivery of corticosteroids is investigated in this study. Commercial DNDs were modified to obtain particles with high colloidal stability in aqueous suspensions. Physical adsorption was then used to conjugate the DNDs with methylprednisolone sodium succinate and dexamethasone sodium phosphate. The experimental adsorption isotherms were fit with Langmuir and Freundlich adsorption models. The results of in vitro release study demonstrated an explicit prolonged effect of DNDs, due to which a sustained release of the adsorbed drugs was provided. © 2021 Elsevier B.V.

Аннотация: The world map of anthropogenic atmospheric nitrogen deposition and its effects on natural ecosystems is not described with equal precision everywhere. In this paper, we report atmospheric nutrient, sulfate and spheroidal carbonaceous particle (SCP) deposition rates, based on snowpack analyses of a formerly unexplored Siberian mountain region. Then, we discuss their potential effects on lake phytoplankton biomass limitation. We estimate that the nutrient depositions observed in the late-season snowpack (4016 mgNO3-Nm2 and 0.580.13 mg TP-Pm2; TP for total phosphorous) would correspond to yearly depositions lower than 11971 mgNO3-Nm2 yr1 and higher than 1.710.91 mg TP-Pm2 yr1. These yearly deposition estimates would approximately fit the predictions of global deposition models and correspond to the very low nutrient deposition range, although they are still higher than world background values. In spite of the fact that such a low atmospheric nitrogen deposition rate would be enough to induce nitrogen limitation in unproductive mountain lakes, phosphorus deposition was also extremely low, and the resulting lake water N: P ratio was unaffected by atmospheric nutrient deposition. In the end, the studied lakes' phytoplankton appeared to be split between phosphorus and nitrogen limitation. We conclude that these pristine lakes are fragile sensitive systems exposed to the predicted climate warming, increased winter precipitation, enhanced forest fires and shifts in anthropogenic nitrogen emissions that could finally couple their water chemistry to that of atmospheric nutrient deposition and unlock temperature-inhibited responses of phytoplankton to nutrient shifts. © Author(s) 2021. Lethal effects on hatchlings were observed starting from the deicer content in the water equal to 1 g L-1, and 97.0 +/- 0.8% of hatchlings were dead at the deicer content in the water equal to 5 g L-1. Thus, the resilience of resting eggs to the contaminated sediments can ensure the replenishment of the population from the egg bank after the salinity disturbance is diminished but the negative effects of the elevated salt content in surface waters on active population will be manifested at lower salinities.

Аннотация: This work is dedicated to developing enzyme biosensor software to solve problems regarding soil pollution analysis. An algorithm and specialised software have been developed which stores, analyses and visualises data using JavaScript programming language. The developed software is based on matching data of 51 non-commercial standard soil samples and their inhibitory effects on three enzyme systems of varying complexity. This approach is able to identify the influence of chemical properties soil samples, without toxic agents, on enzyme biosensors. Such software may find wide use in environmental monitoring.
