Аннотация: Luminous organisms use different protein-mediated strategies to modulate light emission color. Here, we report the transient-state kinetic studies of the interaction between photoprotein clytin from Clytia gregaria and its antenna protein, cgreGFP. We propose that cgreGFP forms a transient complex with Ca2+-bound clytin before the excited singlet state of the coelenteramide product is formed. From the spectral distribution and donor-acceptor separation distance, we infer that clytin reaction intermediates may interact only with the middle side part of cgreGFP.

Аннотация: Coelenteramide-containing fluorescent proteins are products of bioluminescent reactions of marine coelenterates. They are called ‘discharged photoproteins’. Their light-induced fluorescence spectra are variable, depending considerably on external conditions. Current work studies a dependence of light-induced fluorescence spectra of discharged photoproteins obelin, aequorin, and clytin on excitation energy. It was demonstrated that photoexcitation to the upper electron-excited states (260–300 nm) of the discharged photoproteins initiates a fluorescence peak in the near UV region, in addition to the blue-green emission. To characterize the UV fluorescence, the light-induced fluorescence spectra of coelenteramide (CLM), fluorophore of the discharged photoproteins, were studied in methanol solution. Similar to photoproteins, the CLM spectra depended on photoexcitation energy; the additional peak (330 nm) in the near UV region was observed in CLM fluorescence at higher excitation energy (260–300 nm). Quantum chemical calculations by time depending method with B3LYP/cc-pVDZ showed that the conjugated pyrazine-phenolic fragment and benzene moiety of CLM molecule are responsible for the additional UV fluorescence peak. Quantum yields of CLM fluorescence in methanol were 0.028 ± 0.005 at 270–340 nm photoexcitation. A conclusion was made that the UV emission of CLM might contribute to the UV fluorescence of the discharged photoproteins. The study develops knowledge on internal energy transfer in biological structures – complexes of proteins with low-weight aromatic molecules. © 2016 Elsevier B.V.

Аннотация: 1. Aquatic systems are considered to be a main source of essential long-chain n-3 polyunsaturated fatty acids (PUFA), which are preferentially synthesized by microalgae and transferred along food chains to terrestrial consumers. Emerging aquatic insects comprise a significant part of this transfer of the essential PUFA from water to land. Quantitative data on PUFA content and composition are available mainly for rheophilic insects while taxa that are characteristic of wetlands and stagnant water bodies, such as aquatic Heteroptera, remain relatively unstudied. 2. We investigated the role of various waterbug taxa (Heteroptera: Nepomorpha and Gerromorpha) inhabiting different ecoregions in temperate Central Siberia (Russia) as potential sources of PUFA. The ecoregions were steppe, forest-steppe, hemiboreal forest and montane coniferous forest. Although these waterbugs insects are aquatic in both larval and adult stages, they can disperse through the landscape and transfer essential PUFAs from water to land so making them potentially available to terrestrial consumers. 3. Species of Naucoridae, Notonectidae and Corixidae were generally dominant in all ecoregions, attaining maximum biomass in the steppe. We showed that habitat or ecoregion played a major role in the variability of fatty acid composition of Notonectidae and Gerridae but not Corixidae. In contrast, the biochemical composition of the only naucoridae, Ilyocoris cimicoides, was largely affected by life stage. 4. Both the dominant families and species within them differed with respect to their mass-specific contents of essential long-chain PUFA of the n-3 family, namely eicosapentaenoic and docosahexaenoic acids. Corixid species had the highest content of these two essential PUFAs among the waterbug studies, and relative to literature reports for other aquatic insects. Corixids thus appear to be a potentially important vector for transfer of the essential biochemical compounds from water to land, especially in steppe ecoregions with numerous ephemeral water bodies.

Аннотация: Nowadays luciferases are effectively used as analytical instruments in a great variety of research fields. Of special interest are the studies dealing with elaboration of novel analytical systems for the purposes of medical diagnostics. The ever-expanding spectrum of clinically important analytes accounts for the increasing demand for new techniques for their detection. In this chapter we have made an attempt to summarize the results on applications of luciferases as reporters in binding assays including immunoassay, nucleic acid hybridization assay, and so on. The data over the last 15 years have been analyzed and clearly show that luciferase-based assays, due to extremely high sensitivity, low cost, and the lack of need for skilled personnel, hold much promise for clinical diagnostics.

Аннотация: Growth of Cupriavidus eutrophus B-10646, fatty acid (FA) composition of the lipids in the cytoplasmic membrane and cell wall, and polyhydroxyalkanoate (PHA) accumulation by the cells of this new promising producer of PHAs were studied in batch culture, using various carbon sources (fructose, glucose, oleic acid, sunflower seed oil, glycerol, CO2:O-2:H-2). In cells grown on sugars and in autotrophic culture, during the phase of active cell growth, the major FAs of the lipids of cytoplasmic membrane (CMLs) were palmitic, palmitoleic, and cis-vaccenic acids. In the stationary phase of cultivation, cyclopropane FAs were also detected. When carbohydrate substrate was replaced by oleic acid or sunflower seed oil, the proportion of oleic acid in the total FAs of the cytoplasmic membrane increased considerably. In addition to that, the lipid FAs of bacterial cells grown on sunflower seed oil also contained linoleic acid, which is the major acid of sunflower seed oil. The major acids of strongly bound lipids of cell wall (SBLs) were myristic acid and long-chain-length beta-hydroxy acids. Molecular weight of the polymer was measured during its accumulation in cells; for the first time, the presence of 2 polymer fractions (high-molecular weight and low-molecular-weight ones) was recorded in cells with a low polymer content. Аннотация: An idea of the general evolution through the long-term response of the Earth to the external flow of radiant energy from the Sun is proposed. Due to the finiteness of matter on Earth, as well as on any other planet, the continuous pumping flow of radiant energy has been shown to lead to cyclization of transformations and mass transfer along the emerging gradients. The evolution of the energy–matter interaction follows the pathway of capturing and transferring more energy by a smaller quantity of matter, i.e., the pathway of the increase in the amount of energy used by each unit mass. According to this parameter, the least effective mass transfer is a simple transfer as vortices of gases along the gradients of temperature and pressure, which took place on the primary surface of the planet. Long-term natural selection towards water accumulation on the planet has played a special role in the development of the interaction between energy and matter. Phase transitions (ice, water, and vapor) and mechanical transfers are the most common energy–matter processes. Chemical transformation of substances became possible based on water cycles, cyclic transfers, and transformations and developed with time into biological transformation. This type of energy–matter interaction is the most efficient. In particular, the energy of our star is captured during photosynthesis and utilized in the most active region of its radiation spectrum. During the biological evolution of heterotrophs, a increase in the coefficient that characterizes the energy exchange intensity from protozoa to mammals by several hundred times is most illustrative. The development and current dominance of humans as the species that is most active in the capturing of energy and meaningful organization of its new flows, in particular, based on the organic debris of former biospheres, is amazing but quite natural from the energy standpoint. During the technological evolution of humankind, the energy-exchange intensity for homoiotherms (warm-blooded animals) has increased by 20 times if it is recalculated for the technological energy that is used by the average inhabitant of the Earth. Thus, the victory of our species in planetary evolution fits well into the mainstream of the general evolution through energy–matter interactions: a multiple increase in star energy has been used to transform the matter on the surface of the irradiated planet. © 2015, Pleiades Publishing, Inc.

Аннотация: Optically enhanced biosensing strategies are prerequisites for developing miniature and highly sensitive multiplexed analytical platforms. Such smart biosensing systems are highly promising for use in the fields of biomedicine and environmental monitoring. Optical signal enhancement during bioassays is attributed to the complex opto-electronic interactions of incoming photonic signals at the nanomaterial interface. Research on the use of metals other than gold and silver for such purposes tends to extend the spectral window to observe luminescence enhancement effects. Such manifold increase in luminescence may be explained by the principles of plasmon coupling, directional emission led high collection efficiency, Rayleigh scattering and related opto-electronic events. The present review begins with a mechanistic description of important phenomena associated with metal-induced luminescence enhancement, particularly focusing on the origin of metal-enhanced luminescence. This review further analyses the hybrid nanostructure capabilities responsible for maintaining unique opto-electronic properties during bio-functionalisation. Current research trends in this area, future scope of this field for designing useful bioassays and concluding remarks are then discussed. (C) 2017 Elsevier B.V. All rights reserved.

Аннотация: A number of approaches have been proposed and tested to transfer enzymatic reactions into the functional elements of microfluidic chips on the example of the bienzyme bioluminescent reaction involving NAD(P)H:FMN-oxidoreductase and luciferase. Measurement of the catalytic activity of these enzymes (under the influence of pollutants) is the basis of enzymatic bioassay of various liquids. It was found that all of the components of the reaction must be placed in the same cell of the chip to improve the reproducibility of the measurements. The use of starch gel as a carrier for immobilization and gelatin as a scaffold in the reactor of the chip enables the preservation of enzyme activity in the course of sealing the chip at room temperature. It is shown that the components of the reaction should be vigorously stirred in a microfluidic chip reactor to improve the efficiency of the analysis. As a result of the studies, a prototype of microfluidic chip based on the enzymatic bioluminescent reaction is proposed. It is characterized by a detection limit of copper sulfate of 3 mu M that corresponds to the sensitivity of traditional lux-biosensors based on living cells. The analysis time is reduced to 1 min, and the analysis can be performed by individuals without special laboratory skills.

Аннотация: Two fish species reared in aquaculture (pink salmon Oncorhynchus gorbuscha and whitefish Coregonus lavaretus) and ten fish species from natural habitats (whitefish C. lavaretus, tugun Coregonus tugun, broad whitefish Coregonus nasus, least cisco Coregonus sardinella, vendace Coregonus albula, boganid charr Salvelinus boganidae, charr Salvelinus alpinus complex, northern pike Esox lucius, sharp-snouted lenok Brachymystax lenok, and taimen Hucho taimen) have been studied. The content of two long-chain polyunsaturated omega-3 fatty acids (PUFAs), eicosapentaenoic acid (20:5n-3, EPA) and docosahexaenoic acid (22:6n-3, DHA), in the muscle tissue of the fish and in their food (intestine contents) are compared. In the aquacultures of whitefish and pink salmon, the total content of EPA and DHA is significantly higher in feed than in the muscle tissue of the fish, which indicates losses of PUFA in the two-link food chain of the aquaculture during their transfer to the upper trophic level. EPA and DHA losses in aquaculture, which are confirmed by numerous literature data, mean an inefficient usage of the available sources of PUFAs and the aggravation of the global deficit of these biochemicals in the human diet. A study of natural fish populations reveals the accumulation of EPA and DHA in their biomass compared to food in many cases, although opposite phenomena are also observed. An assumption on the presence of an optimal, physiologically adequate species-specific level of PUFA in the fish muscle tissue has been made based on our data and literature data. If the level of PUFAs in the muscles is lower than optimal, their accumulation (bioaccumulation) from food and/or de novo synthesis are observed. When the optimal level is exceeded, the content of EPA and DHA in biomass approaches maximum species-specific values; however, part of these PUFAs entering from food is not digested or is catabolized. According to the obtained data, the species of the order Salmoniformes have an optimal level of 2 to 6 mg/g of wet weight. It has been found that in aquaculture approaching to maximum values of EPA + DHA content was accompanied by their losses (scattering) in the food chains, while in natural ecosystems the maximum values of PUFA content in the fish biomass are achieved by their accumulation from the lower trophic level. Boganid charr S. boganidae had the highest content of EPA + DHA in the muscle tissue among all known fish species (32.78 mg/g of wet weight).

Аннотация: Effect of dimethyl sulfoxide (DMSO), a widespread biomedical agent, on spectral-luminescent characteristics of coelenteramide-containing fluorescent protein - discharged obelin - is investigated. Contributions of violet and blue-green spectral components to fluorescence of discharged obelin are elucidated and characterized at different photoexcitation energies. Dependences of these contributions on the DMSO concentration are presented. Spectral changes are related to the destructive effect of DMSO on fluorescent protein and decreasing efficiency of proton transfer to electronically excited states of fluorophore.

Аннотация: Two groups of factors, phylogenetic and ecological, are presently regarded as controlling fatty acid composition of fish, including essential eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids. Environmental effects, e.g., trophic position, temperature and/or seasonality, were previously studied using sums of fatty acids or only their level data. We tested the hypothesis that differences in trophic levels of piscivorous (pike and perch) and omnivorous (roach and bream) fish from a mesotrophic reservoir allow discriminating levels and contents of individual fatty acids, especially EPA and DHA. The more established measurements, i.e., stomach contents and carbon and nitrogen stable isotopes in fish muscles, were also carried out to provide linkages between the different ecological tracers, fatty acids versus stable isotopes, and matching the methods for long-term food sources (fatty acids and stable isotopes) and recent foraging (stomach content analysis). We also studied a putative influence of seasonality. Similar to other studies, there were seasonal changes in fatty acid composition and contents of two fish, perch and roach, due to direct and indirect effects of water temperature. Meanwhile, the piscivorous and omnivorous species captured in the same month, were explicitly differentiated on a base of stable isotopes and fatty acids. Significantly higher percentages and contents of DHA in piscivorous fish, perch and pike, relatively to those in roach and bream, likely indicated a higher trophic transfer efficiency for this essential fatty acid. All the fishes have commercial importance for regional fishery and are harvested from the studied reservoir for human nutrition. Regarding content of EPA + DHA (mg g?1 fish) as the indicator of nutritive value for humans, pike had the highest nutritive value, roach and perch had intermediate overlapped values, and bream was of the least benefit. © 2016 Elsevier B.V.

Аннотация: Nowadays, physicochemical approach to understanding toxic effects remains underdeveloped. A proper development of such mode would be concerned with simplest bioassay systems. Coelenteramide-Containing Fluorescent Proteins (CLM-CFPs) can serve as proper tools for study primary physicochemical processes in organisms under external exposures. CLM-CFPs are products of bioluminescent reactions of marine coelenterates. As opposed to Green Fluorescent Proteins, the CLM-CFPs are not widely applied in biomedical research, and their potential as colored biomarkers is undervalued now. Coelenteramide, fluorophore of CLM-CFPs, is a photochemically active molecule; it acts as a proton donor in its electron-excited states, generating several forms of different fluorescent state energy and, hence, different fluorescence color, from violet to green. Contributions of the forms to the visible fluorescence depend on the coelenteramide microenvironment in proteins. Hence, CLM-CFPs can serve as fluorescence biomarkers with color differentiation to monitor results of destructive biomolecule exposures. The paper reviews experimental and theoretical studies of spectral-luminescent and photochemical properties of CLM-CFPs, as well as their variation under different exposures – chemicals, temperature, and ionizing radiation. Application of CLM-CFPs as toxicity bioassays of a new type is justified. © 2017

Аннотация: Quenching of bioluminescence of the bienzyme system bacterial luciferase-NAD В· H:FMN-oxidoreductase† † NADВ·H is reduced nicotinamide adenine dinucleotide and FMN is flavine manonucleotide. is considered in terms of the patterns of intermolecular transfer of an electron and hydrogen between donor (bioluminescent centre) and acceptor (compound to be analysed) taking into account the structure of the acceptor molecule. The link between the induction period of bioluminescence and the redox potential of the quinone-acceptor has been confirmed. It is shown that the induction period and the time of passage to the bioluminescence maximum in the presence of quinones are determined by the size of the aromatic and aliphatic fragments of the quinone molecule; the intensity of bioluminescence depends on the efficiency of the processes of intermolecular electron transfer (donor-acceptor) and the competition of the compound being analysed with aldehyde for the binding site on luciferase. Аннотация: To study further the factors providing for cellular quiescence, we used okadaic acid (OA) at concentrations (0.1, 1, 10 or 100 nM) inhibiting type 1 and/or type 2A protein phosphatases in mammalian cell cultures. Brief (2 h) exposure of resting (0.2% serum for 72 h) NIH 3T3 mouse fibroblasts to OA with subsequent incubation of cells in a medium with 0.2% serum, stimulated DNA synthesis at all concentrations studied. Maximal stimulation was observed following pre-incubation of resting cells with 10 nM OA. Treatment of cycling cells (10% serum) with OA (2 h pulses at 12 h intervals for 72 h) prevented their exit to the resting state on transfer to a medium with 0.2% serum. Brief exposures of resting cells to OA did not affect the rate of protein synthesis. OA pulses in the late pre-replicative period had no effect on the entry of serum-stimulated cells into the S phase. Cell fusion experiments with resting (serum-deprived) and proliferating (serum-stimulated) NIH 3T3 cells, using radioautography with a double-labelling technique, revealed that pre-incubation of resting cells with OA for 2 h before and after fusion abrogates their ability to suppress the onset of DNA synthesis in the nuclei of proliferating cells in heterodikaryons. The results indicate that protein phosphatases of type 1 and/or 2A may be involved in the growth-arrest machinery that provides for cellular quiescence.

Аннотация: We have studied the fatty acid (FA) contents and composition of cladocerans and copepods from warm and cold lakes. We have found no significant differences in FA percent levels and per carbon contents of cladocerans from warm and cold lakes or of copepods from warm and cold lakes. A discriminant analysis showed that all cladocerans differed from all copepods mainly due to the content of docosahexaenoic acid. Compared to cladocerans, copepods had significantly higher levels of all C22 polyunsaturated FA. Thus, we conclude that cladocerans and copepods in all environments had comparatively invariant taxon-specific compositions and contents of long-chain highly unsaturated fatty acids (HUFAs). According to HUFA content, all studied copepods can be regarded as a valuable food for fish.

Аннотация: The bioaccumulation of actinide Am-241 from food and water in bodies of freshwater fish (Carassius auratus gibelio, silver crucian carp) has been investigated in laboratory experiments. Homogenized biomass of submerged macrophytes labeled with Am-241 was injected into the fish gullet. Internal organs and tissues of crucian carp could be ranked according to activity concentration of dietary Am-241 after depuration of the digestive tract as follows: liver > gonads > bones > muscles. Accumulation of waterborne Am-241 in internal organs and tissues of crucian carp mainly occurred via the digestive tract. While the concentration of Am-241 in liver of crucian carps decreased during depuration time, its concentration in bones increased indicating slower transfer of Am-241 to skeleton compared to muscles and liver. The retention of dietary Am-241 in the bodies of crucian carp reached 35%-46% of ingested Am-241: 20%-31% was retained in liver, 0.6%-0.8% in skeleton, and 1.4%-2.0% in muscles. The concentration factor of Am-241 from water was 0.4 for the whole body, 0.3 for liver, 0.01 for muscles, and 0.01 for skeleton. Trace amounts of Am-241 were recorded in viscera and muscles of a wild population of silver crucian carp inhabiting a radioactively contaminated part of the Yenisei River.

Аннотация: Following the Fukushima accident in March 2011, samples of pine trees (Pinus sylvestris) were collected from three sites near the city of Krasnoyarsk (Siberia, Russia) during 2011-2012 and analyzed for artificial radionuclides. Concentrations of Fukushima-derived radionuclides in the samples of pine needles in April 2011 reached 5.51 ± 0.52 Bq kg-1 131I, 0.92 ± 0.04 Bq kg-1 134Cs, and 1.51 ± 0.07 Bq kg-1 137Cs. An important finding was the detection of 134Cs from the Fukushima accident not only in the pine needles and branches but also in the new shoots in 2012, which suggested a transfer of Fukushima cesium isotopes from branches to shoots. In 2011 and 2012, the 137Cs/134Cs ratio for pine needles and branches collected in sampling areas Krasnoyarsk-1 and Krasnoyarsk-2 was greater than 1 (varying within a range of 1.2-2.6), suggesting the presence of “older“, pre-Fukushima accident 137Cs. Calculations showed that for pine samples growing in areas of the Krasnoyarskii Krai unaffected by contamination from the nuclear facility, the activity of the Fukushima-derived cesium isotopes was two-three times higher than the activity of the pre-accident 137Cs.

Аннотация: The hypothesis of activity of the upper electron-excited states of the bacterial bioluminescent emitter was verified using dye molecules as foreign energy acceptors. Six compounds were selected having fluorescent state energies ranging from 25 700 to 32 000 cm(-1) (anthracene, pyrene, 1.4-bis(5-phenyloxasol-2-yl)benzene (POPOP), p-bis(o-methylstyryl)benzene (MSB), 2-methoxy-naphtalene, p-terphenyl), exceeding that of the bioluminescent emitter (22 000 cm(-1)). Their absorption spectra do not overlap with the bioluminescence spectrum; the trivial light absorption and the intermolecular resonance S-S energy transfer were excluded. Bacterial bioluminescent spectra of the coupled enzyme system NADH:FMN-oxidoreductase-luciferase in the presence of MSB were presented as an example. The weak sensitized fluorescence of MSB was registered. The results obtained have confirmed the activity of the energetic precursor in the bacterial bioluminescence. Its energy can be located in the interval of 26 000-27 000 cm(-1). (C) 2002 Published by Elsevier Science B.V.

Аннотация: The time-resolved and steady-state fluorescence techniques,were employed to elucidate possible interactions of four aromatic compounds (anthracene, POPOP, MSB and 1,4-naphthalendiol) with bacterial luciferase. Fluorescence spectra and fluorescence anisotropy decays of these compounds were studied in ethanol, water-ethanol solutions and in the presence of bacterial luciferase. Shifts of fluorescent spectra and differences in rotational correlation times are interpreted in terms of weak (hydrophobic) interactions of the molecules with the enzyme. These interactions suggest the feasibility of intermolecular energy transfer by an exchange resonance mechanism with a collision-interaction radius as a way of excitation of these compounds in the reaction catalysed by bacterial luciferase.
