Аннотация: The problem of correlation between rheological properties in macro- and micro- scales of media with biopolymers of polypeptide (gelatin) and polysaccharide (starch) nature is investigated. The viscosity of the biopolymer solutions with concentrations 0.5-5 wt% was estimated by standard rotational rheometry technique and with fluorescent molecular rotor at 15-50 A degrees C. Opposite trends were observed for relationship between microviscosity eta (m) and macroviscosity eta for two biopolymers: eta (m) eta for gelatin and eta (m) eta for starch solutions. The temperature dependence of eta (m) followed the monoexponential decay law in all samples over the whole temperature range indicating insensitivity of microviscosity to gel mesh melting under heating. The dissimilarity of macro- and micro-rheological properties of gelatin and starch-containing media is discussed in terms of difference in architecture of the gels.

Аннотация: Experimental data have been presented on the influence of silver on the viscosity and thermal conductivity of a dispersion of diamond nanoparticles. A stable dispersion (5 wt %) of detonation nanodiamond particles has been used in the experiments. Silver ions have been introduced electrolytically into the dispersion of diamond nanoparticles. Silver concentration was not higher than 0.05 wt %. It has been shown that the introduction of silver ions significantly affects the thermal conductivity and viscosity of the dispersion. © 2017, Pleiades Publishing, Ltd.

Аннотация: One of the current trends of the modern biology figures out cellular enzyme behaviour. Numerous researches look more closely at the chemical composition of creating in vivo simulated media conditions. The aim of this work was to find out a thermodynamic cooperativity of enzymes in a triple-enzyme chain (lactate dehydrogenase + NAD(P)H: FMN-oxidoreductase + bacterial luciferase) under in vivo simulated condition. The thermodynamic cooperativity effects were found out based on the influence of the viscogens (glycerol and sucrose) on the thermal stability of the triple-enzyme system. The results showed that the viscogens do not lead to an increase in the thermal stability of the triple-enzyme system. In addition, organic solvents (sucrose and glycerol) added as viscous agents to the reaction medium altered the kinetics of this triple-enzyme chain, including changing the light emission decay constant (kdec) and quantum yield of luminescence (Q). Plus, sucrose was found to be more efficient in limiting the flexibility of enzymes than glycerol. The high sensitivity of the triple-enzyme system to the viscogens may be connected with a fact that lactate dehydrogenase does not bound with couple enzyme system NAD(P)H: FMN-oxidoreductase + bacterial luciferase inside the real cell. Since this approach may be used as a method to understand the real connection between enzymes in cellular multi-enzyme metabolic chains inside the luminous bacteria cell. © 2018 Elsevier B.V.

It was found that at low concentrations of glycerol or sucrose (viscosity 1.1–1.3 cP) the decay rate rises, whereas a further increase in viscosity to 6.2 cP leads to a decrease in the decay rate following a power function with an exponent of 0.82–0.84. Using molecular dynamics methods, it was shown that the presence of glycerol and sucrose molecules causes a change in the mobility of the amino acid residues in the active center of luciferase, particularly those responsible for binding of flavin. The results obtained are indicative of two opposite effects of viscous media with glycerol and sucrose: (1) destabilization of 4a-hydroperoxyflavin due to a change in the structural and dynamic properties of the protein and (2) stabilization of this intermediate by the decrease in the diffusion rate of its decay products. © 2020, Pleiades Publishing, Ltd.

Аннотация: Enzymes activity in a cell is determined by many factors, among which viscosity of the microenvironment plays a significant role. Various cosolvents can imitate intracellular conditions in vitro, allowing to reduce a combination of different regulatory effects. The aim of the study was to analyze the media viscosity effects on the rate constants of the separate stages of the bacterial biolumi-nescent reaction. Non-steady-state reaction kinetics in glycerol and sucrose solutions was measured by stopped-flow technique and analyzed with a mathematical model developed in accordance with the sequence of reaction stages. Molecular dynamics methods were applied to reveal the effects of cosolvents on luciferase structure. We observed both in glycerol and in sucrose media that the stages of luciferase binding with flavin and aldehyde, in contrast to oxygen, are diffusion-limited. More-over, unlike glycerol, sucrose solutions enhanced the rate of an electronically excited intermediate formation. The MD simulations showed that, in comparison with sucrose, glycerol molecules could penetrate the active-site gorge, but sucrose solutions caused a conformational change of functionally important ?Glu175 of luciferase. Therefore, both cosolvents induce diffusion limitation of substrates binding. However, in sucrose media, increasing enzyme catalytic constant neutralizes viscosity effects. The activating effect of sucrose can be attributed to its exclusion from the catalytic gorge of luciferase and promotion of the formation of the active site structure favorable for the catalysis. © 2021 by the authors. Аннотация: The cladoceran Daphnia magna inhabits lakes with salinities up to 10 g L-1. We compared the effects of different salinities (up to 9 g L-1) on the survival, specific grazing rate, and size selective feeding of Daphnia females adapted to fresh or saline waters (3-4 g L-1). The freshwater population was more sensitive to high salinity (LC50 = 5.3 g L-1), while the survival of the saline water population also decreased in fresh water. Freshwater population demonstrated a higher grazing rate in fresh water, while the saline water population had a higher grazing rate at the salinity above 3 g L-1. A decrease in the grazing rate of the freshwater population was observed at the salinity above 4-5 g L-1. Populations differed in food selectivity. The saline water population consumed particles of larger sizes than the freshwater population. The average size of phytoplankton particles grazed in fresh water was larger than in saline water. This size selective salinity-dependent grazing may be related to the dependence of the feeding efficiency of cladocerans on the viscosity of water and size of phytoplankton particles. Our results indicate that Daphnia populations adapted to a certain salinity can temporarily lose the ability to control phytoplankton because of salinity fluctuations.
