Аннотация: The influence of some chemical substances on luminous bacteria was studied to elucidate the interrelation between the xenobiotics action on bacterial luminescence and cell ultrastructure. Such substances as quinones, phenols, chlorides of heavy metals (in concentrations of substances inhibiting luminescence by 50%) resulted in damaging effects upon bacteria: a lot of cells had damage of membranes due to changes in their permeability. It was found that the high concentration of EDTA and toluene decreased the luminescence and caused the condensation of DNA-fibrils and the cell damage after long-term and short-term action. The low concentration of EDTA and toluene did not decrease the bacterial luminescence; the noticeable damage of cell membranes did not take place during short-term treatment. However, the long action of these substances changed the membrane permeability resulting in increased sensitivity of bacterial luminescence to some toxic substances. Аннотация: The study deals with the effects of herbicides (metribuzin, tribenuron-methyl, fenoxaprop-P-ethyl) and fungicides (tebuconazole, epoxiconazole, azoxystrobin) applied to soil as free pesticides or as slow release formulations embedded in a biodegradable composite matrix on the structure of the soil microbial community. The matrix consisted of a natural biopolymer poly-3-hydroxybutyrate [P(3HB)] and a filler—one of the natural materials (peat, clay, and wood flour). The soil microbial community was characterized, including the major eco-trophic groups of bacteria, dominant taxa of bacteria and fungi, and primary P(3HB)-degrading microorganisms, such as Pseudomonas, Bacillus, Pseudarthrobacter, Streptomyces, Penicillium, and Talaromyces. The addition of free pesticides adversely affected the abundance of soil microorganisms; the decrease varied from 1.4 to 56.0 times for different types of pesticides. The slow release pesticide formulations, in contrast to the free pesticides, exerted a much weaker effect on soil microorganisms, no significant inhibition in the abundance of saprotrophic bacteria was observed, partly due to the positive effects of the composite matrix (polymer/natural material), which was a supplementary substrate for microorganisms. The slow release fungicide formulations, like the free fungicides, reduced the total abundance of fungi and inhibited the development of the phytopathogens Fusarium and Alternaria. Thus, slow release formulations of pesticides preserve the bioremediation potential of soil microorganisms, which are the main factor of removing xenobiotics from the biosphere. © 2020, Springer-Verlag GmbH Germany, part of Springer Nature.

Аннотация: Major advances in the development and application of the bioluminescent analysis to detect certain biologically active substances are discussed. The main merit of the method lies in its high sensitivity and specificity along with its simplicity and rapid performance. The available methodologies allow for detection of substances of varying nature: Ca2+, ATP, FMN, NAD(P), long-chain aldehydes, ATP- and NAD(P)-dependent enzymes and their substrates, many xenobiotics and antibiotics, and mutagens. The bioluminescence methodologies may be widely applied in clinical laboratory diagnosis.

Dyes, non-fluorescent chemically inert organic compounds, redox-active organic compounds and metallic salts were investigated. The influence of the different xenobiotics depends in a complex way on physicochemical characteristics of the xenobiotic molecules (spectral-luminescent characteristics, electron-acceptation energy and redox potential). Аннотация: The presence of potentially toxic xenobiotics in complex matrices has become rather the rule than the exception. Therefore, there is a need for highly sensitive inexpensive techniques for analyzing environmental and food matrices for toxicants. Enzymes are selectively sensitive to various toxic compounds, and, thus, they can be used as the basis for detection of contaminants in complex matrices. There are, however, a number of difficulties associated with the analysis of complex matrices using enzyme assays, including the necessity to take into account properties and effects of the natural components of the test media for accurate interpretation of results. The present study describes the six-stage procedure for designing new enzyme sensors intended for assessing the quality of complex matrices. This procedure should be followed both to achieve the highest possible sensitivity of the biosensor to potentially toxic substances and to minimize the effect of the uncontaminated components of complex mixtures on the activity of the biosensor. The proposed strategy has been tested in designing a bioluminescent biosensor for integrated rapid assessment of the safety of fruits and vegetables. The biosensor is based on the coupled enzyme system NAD(P)H:FMN-oxidoreductase and luciferase as the biorecognition element. The study describes methods and techniques for attaining the desired result in each stage. The proposed six-stage procedure for designing bioluminescent enzyme biosensors can be used to design the enzymatic biosensors based on other enzymes. © 2021 Elsevier B.V.

Аннотация: The bioluminescent bacterial enzyme system NAD(P)H:FMN-oxidoreductase-luciferase has been used as a test system for ecological monitoring. One of the modes to quench bioluminescence is the interaction of xenobiotics with the enzymes, which inhibit their activity. The use of endogenous flavin fluorescence for investigation of the interactions of non-fluorescent compounds with the bacterial luciferase from Photobacterium leiognathi and NAD(P)H:FMN-oxidoreductase from Vibrio fischeri has been proposed. Fluorescence spectroscopy methods have been used to study characteristics of endogenous flavin fluorescence (fluorophore lifetime, the rotational correlation time). The fluorescence anisotropy behaviour of FMN has been analysed and compared to that of the enzyme-bound flavin. The fluorescence characteristics of endogenous flavin of luciferase and NAD(P)H:FMN-oxidoreductase have been shown to be applicable in studying enzymes' interactions with non-fluorescent compounds.
