Аннотация: This review examines the general principles of bioluminescent enzymatic toxicity bioassays and describes the applications of these methods and the implementation in commercial biosensors. Bioluminescent enzyme system technology (BEST) has been proposed in the bacterial coupled enzyme system, wherein NADH: FMN-oxidoreductase-luciferase substitutes for living organisms. BEST was introduced to facilitate and accelerate the development of cost-competitive enzymatic systems for use in biosensors for medical, environmental, and industrial applications. For widespread use of BEST, the multicomponent reagent "Enzymolum'' has been developed, which contains the bacterial luciferase, NADH: FMN-oxidoreductase, and their substrates, co-immobilized in starch or gelatin gel. Enzymolum is the central part of Portable Laboratory for Toxicity Detection (PLTD), which consists of a biodetector module, a sampling module, a sample preparation module, and a reagent module. PLTD instantly signals chemical-biological hazards and allows us to detect a wide range of toxic substances. Enzymolum can be integrated as a biological module into the portable biodetector-biosensor originally constructed for personal use. Based on the example of Enzymolum and the algorithm for creating new enzyme biotests with tailored characteristics, a new approach was demonstrated in biotechnological design and construction. The examples of biotechnological design of various bioluminescent methods for ecological monitoring were provided. Possible applications of enzyme bioassays are seen in the examples for medical diagnostics, assessment of the effect of physical load on sportsmen, analysis of food additives, and in practical courses for higher educational institutions and schools. The advantages of enzymatic assays are their rapidity (the period of time required does not exceed 3-5 min), high sensitivity, simplicity and safety of procedure, and possibility of automation of ecological monitoring; the required luminometer is easily available.

Аннотация: Background Mortality is a main driver in zooplankton population biology but it is poorly constrained in models that describe zooplankton population dynamics, food web interactions and nutrient dynamics. Mortality due to non-predation factors is often ignored even though anecdotal evidence of non-predation mass mortality of zooplankton has been reported repeatedly. One way to estimate non-predation mortality rate is to measure the removal rate of carcasses, for which sinking is the primary removal mechanism especially in quiescent shallow water bodies. Objectives and Results We used sediment traps to quantify in situ carcass sinking velocity and non-predation mortality rate on eight consecutive days in 2013 for the cladoceran Bosmina longirostris in the oligo-mesotrophic Lake Stechlin; the outcomes were compared against estimates derived from in vitro carcass sinking velocity measurements and an empirical model correcting in vitro sinking velocity for turbulence resuspension and microbial decomposition of carcasses. Our results show that the latter two approaches produced unrealistically high mortality rates of 0.58-1.04 d(-1), whereas the sediment trap approach, when used properly, yielded a mortality rate estimate of 0.015 d(-1), which is more consistent with concurrent population abundance data and comparable to physiological death rate from the literature. Ecological implications Zooplankton carcasses may be exposed to water column microbes for days before entering the benthos; therefore, non-predation mortality affects not only zooplankton population dynamics but also microbial and benthic food webs. This would be particularly important for carbon and nitrogen cycles in systems where recurring mid-summer decline of zooplankton population due to non-predation mortality is observed.

Аннотация: 1. Aquatic systems are considered to be a main source of essential long-chain n-3 polyunsaturated fatty acids (PUFA), which are preferentially synthesized by microalgae and transferred along food chains to terrestrial consumers. Emerging aquatic insects comprise a significant part of this transfer of the essential PUFA from water to land. Quantitative data on PUFA content and composition are available mainly for rheophilic insects while taxa that are characteristic of wetlands and stagnant water bodies, such as aquatic Heteroptera, remain relatively unstudied. 2. We investigated the role of various waterbug taxa (Heteroptera: Nepomorpha and Gerromorpha) inhabiting different ecoregions in temperate Central Siberia (Russia) as potential sources of PUFA. The ecoregions were steppe, forest-steppe, hemiboreal forest and montane coniferous forest. Although these waterbugs insects are aquatic in both larval and adult stages, they can disperse through the landscape and transfer essential PUFAs from water to land so making them potentially available to terrestrial consumers. 3. Species of Naucoridae, Notonectidae and Corixidae were generally dominant in all ecoregions, attaining maximum biomass in the steppe. We showed that habitat or ecoregion played a major role in the variability of fatty acid composition of Notonectidae and Gerridae but not Corixidae. In contrast, the biochemical composition of the only naucoridae, Ilyocoris cimicoides, was largely affected by life stage. 4. Both the dominant families and species within them differed with respect to their mass-specific contents of essential long-chain PUFA of the n-3 family, namely eicosapentaenoic and docosahexaenoic acids. Corixid species had the highest content of these two essential PUFAs among the waterbug studies, and relative to literature reports for other aquatic insects. Corixids thus appear to be a potentially important vector for transfer of the essential biochemical compounds from water to land, especially in steppe ecoregions with numerous ephemeral water bodies.

Аннотация: In several Russian northern lakes and rivers, Arctic cisco Coregonus autumnalis, least cisco C. sardinella, peled C. peled, tugun C. tugun, broad whitefish C. nasus, whitefish C. lavaretus and vendace C. albula were sampled in periods of officially permitted commercial fishery. Special attention was paid to contents (mg g(-1) of wet weight) of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in muscle tissues (filets), which are essential for human nutrition. The highest values of EPA + DHA content in semi-anadromous fish and freshwater fish were recorded for C. autumnalis from the Yenisei River, 17.60 mg g(-1) wet weight, and for C. lavaretus from the Sobachye Lake, 16.61 mg g(-1) wet weight, respectively. Intra-genus variations of EPA + DHA contents of Coregonus species were from 1.87 to 17.60 mg g(-1) wet weight. Since the congeneric species were genetically close to each other, the variations in EPA and DHA contents were thought to be caused primarily by ecological factors: migrational capability, type of feeding and trophic status of aquatic ecosystems. In general, the majority of studied species appeared to be of a high nutritive value for humans, although unfavorable environmental conditions could considerably diminish this value.

Аннотация: The large, deep, freshwater Norilsk lakes (Lake Lama, Lake Sobachye, Lake Glubokoe and Lake Keta) are located beyond the Arctic Circle, at the northwestern edge of the Putorana Plateau. They underwent a Pleistocene boreal marine transgression. A relict of that transgression, Limnocalanus macrurus Sars 1863 (Copepoda, Calanoida), presently dominates the crustacean zooplankton of these lakes. Sympatric with the native relict species L. macrurus, Senecella siberica Vyshkvartzeva 1994, was found in our zooplankton samples taken from the deep parts of lakes Lama and Sobachye in the summer-autumn of 2014-2016. Sampling was performed using a Juday net hauled vertically from a depth of 15, 20(25/30) and/or 50 m to the surface. All Senecella specimens were older copepodites, mainly CIV stage in Lama and Sobachye lakes in August and CV in Sobachye Lake in September; their abundance was low (25-2000 ind. m(-2)). They inhabited the central deeper regions of the lakes, contributing up to 7-14% to the total wet biomass of net zooplankton in 0-20 and 0-50 m strata. Previous sporadic and rather superficial samplings in these lakes missed Senecella due to its deep locations, low abundance levels and the absence of both the oldest copepodites and adults in summer.

Аннотация: Contents of highly unsaturated long-chain fatty acids of omega-3 family (HUFA); eicosapentaenoic acid (EPA; 20:5n-3); and docosahexaenoic acid (DHA; 22:6n-3) are the principal indicators of the nutritive quality of fish for humans. Effects of environmental factors, first of all water temperature, on EPA and DHA contents in fish tissue are not currently completely understood. To reveal the putative effect of water temperature, fatty acid composition and contents were studied for 6 fish species, inhabiting the cold waters of the Yenisei River (Siberia, Russia) downstream of a dam (hypolimnetic release from reservoir) and its adjacent warm water tributaries (the Mana River and the Kacha River). It was hypothesized that (a) fish species from the cold river would have higher HUFA contents than fish from the warm rivers and (b) temperature would be negatively correlated with HUFA content in fish species. Using gas chromatography-mass spectrometry, distinct species-specific fatty acid profiles were observed, whereas contents of the essential fatty acids, EPA and DHA, in fish species from the cold Yenisei River were in general similar to that from warm tributaries. Thus, in contrast to the first hypothesis, phylogenetic factors overweighed the effect of water temperature and food (benthic invertebrate) composition, on fatty acid composition and HUFA content in fish. For the second hypothesis, for the 2 species inhabiting both cold and warm rivers, only one had higher EPA and DHA content at lower temperatures. Consequently, the response of EPA and DHA content in fish tissue to temperature variations may be species-specific.

Аннотация: Some terrestrial consumers may be limited by food quality, namely by contents of essential polyunsaturated fatty acids (PUFA), eicosapentaenoic acid (20:5n - 3, EPA) and docosahexaenoic acid (22:6n - 3, DHA) in their food. Since EPA and DHA are mainly produced in aquatic ecosystems, for future estimating of the potential limitation by food quality in global scale, the water-land fluxes of these PUFA with the biomass of emergent aquatic insects in several biomes were calculated. The water /land area ratios for each biome were calculated by dividing the water area of each biome by its terrestrial area. Data on insect emergence from water bodies (g of dry mass m(-2) year(-1)), were summarized and averaged for each biome. From available data, EPA and DHA contents (mg g(-1) dry mass), in the biomass of emergent aquatic insects were calculated first so that annual fluxes of PUFA to land area via aquatic insect emergence could be estimated for each biome. PUFA fluxes occurred between the biomes, ranging from 0.04 to 4.39 mg m(-2) year(-1). In this study, the aquatic PUFA supply to land area appeared to be significantly lower than estimated earlier. This suggests that terrestrial consumers may experience food quality limitations mediated by shortage of PUFA compounds.

Аннотация: In cold smoked species of genus Coregonus, identified by molecular genetic analysis, contents of fatty acids and heavy metals and arsenic were measured. The highest content of sum of long-chain polyunsaturated fatty acids of omega-3 family (LC-PUFA), namely eicosapentaenoic (EPA) and docosahexaenoic (DHA) fatty acids, 6.53 +/- 0.78 mg g(-1) wet weight, was characteristic of tugun Coregonus tugun. This is the first quantitative estimation (mg LC-PUFA per g of product) of the nutritive value of smoked fish. Thus, to obtain a daily personal doze of EPA + DHA of 1 g, recommended for prevention of cardiovascular diseases, one needs to consume 153 g of the smoked tugun. Metals contents did not exceed standards for fish meat except Pb in least cisco Coregonus sardinella. Accordingly, values of hazard quotients, which estimate benefit-risk ratio of fish intake, indicate that most of the smoked fish species are safe product for human nutrition, except least cisco regarding Pb content.

Аннотация: The efficiency of the transfer of physiologically valuable substances including polyunsaturated fatty acids (PUFA), nitrogen, and phosphorus from phytoplankton to zooplankton is extremely important for the functioning of the whole food web. The bivalve mollusk Dreissena polymorpha has a great impact on planktonic communities. However, its effects on the efficiency of transfer of physiologically valuable substances from phytoplankton to zooplankton have not been assessed so far. The goal of our study was to assess the effect of zebra mussels on the transfer efficiency of carbon, fatty acids including PUFA, nitrogen, and phosphorus from phytoplankton to zooplankton under mesotrophic conditions. In the experimental mesocosms filled with water from a mesotrophic lake containing phyto- and zooplankton, we manipulated the presence/absence of zebra mussels. The experiments showed that zebra mussels decreased the efficiency of the transfer of carbon, PUFA, nitrogen, and phosphorus from phytoplankton to zooplankton, thus reducing the quality of zooplankton as a food resource for higher trophic levels. However, zooplankton revealed the ability to increase the content of physiologically valuable substances in their tissues relative to their contents in the diet. We believe that the mechanism of accumulation of physiologically valuable substances by zooplankton enables to at least partially resist the negative influence of zebra mussels on the quality of zooplankton as a food resource.

Аннотация: The efficiency of the transfer of carbon, polyunsaturated fatty acids (PUFA), nitrogen, and phosphorus from phytoplankton to zooplankton determines the functioning of the entire ecosystem. However, this parameter depends on environmental conditions. Fish as a very important factor regulating planktonic communities very likely affect the efficiency of the transfer of basic elements and substances from phytoplankton to zooplankton. In experimental mesocosms filled with water from a eutrophic lake and containing phyto- and zooplankton, we investigated how fish affect both primary and secondary production and the efficiency of transfer of carbon, nitrogen, phosphorus, PUFA, and fatty acids (FAs) from phytoplankton to zooplankton. Two treatments (control and fish treatment) were repeated in three replicates. The transfer efficiency of substances from phytoplankton to zooplankton was measured as the ratio of secondary production to primary production expressed per L and per unit of biomass. The efficiency expressed per L characterizes the productivity of the water body, while the efficiency expressed per unit of biomass indicates the effectiveness of aquatic species to transfer biologically valuable substances from one trophic level to another. We found that phytoplankton-zooplankton-fish interface is determined not only by predator-prey relationships, but are also affected by the quality of both phytoplankton and zooplankton in terms of contents of phosphorus, nitrogen, PUFA, and FAs in their biomass. We showed that, in the presence of fish, the transfer efficiency of carbon, phosphorus, nitrogen, eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), and FAs per unit of biomass from phytoplankton to zooplankton was increased twofold, 12.4-fold, 2.5-fold, 12.4-fold, 7.4-fold, and tenfold, respectively, relative to the control. This facilitates sustainable functioning of the ecosystem under fish pressure. Such a mechanism prevents zooplankton over-exploitation by fish and enlarges the food chain.

Аннотация: Carbon turnover in aquatic environments is dependent on biochemical properties of organic matter (OM) and its degradability by the surrounding microbial community. Non-additive interactive effects represent a mechanism where the degradation of biochemically persistent OM is stimulated by the provision of bioavailable OM to the degrading microbial community. Whilst this is well established in terrestrial systems, whether it occurs in aquatic ecosystems remains subject to debate. We hypothesised that OM from zooplankton carcasses can stimulate the degradation of biochemically persistent leaf material, and that this effect is influenced by the daphnia:leaf OM ratio and the complexity of the degrading microbial community. Fresh Daphnia magna carcasses and C-13-labelled maize leaves (Zea mays) were incubated at different ratios (1:1, 1:3 and 1:5) alongside either a complex microbial community (50 mu m) or solely bacteria (0.8 mu m). C-13 stable-isotope measurements of CO2 analyses were combined with phospholipid fatty acids (PLFA) analysis and DNA sequencing to link metabolic activities, biomass and taxonomic composition of the microbial community. Our experiments indicated a significantly higher respiration of leaf-derived C when daphnia-derived OM was most abundant (i.e. daphnia:leaf OM ratio of 1:1). This process was stronger in a complex microbial community, including eukaryotic microorganisms, than a solely bacterial community. We concluded that non-additive interactive effects were a function of increased C-N chemodiversity and microbial complexity, with the highest net respiration to be expected when chemodiversity is high and the degrading community complex. This study indicates that identifying the interactions and processes of OM degradation is one important key for a deeper understanding of aquatic and thus global carbon cycle.

We found that phytoplankton-zooplankton-fish interactions are determined not only by predator-prey relationships, but also are affected by the quality of both phytoplankton and zooplankton, measured as the contents of phosphorus, nitrogen, PUFA and FA in their biomass. We showed that, in the presence of fish, the transfer efficiency of carbon, phosphorus, nitrogen, eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), FA per biomass unit from phytoplankton to zooplankton was increased two-fold, 12.4-fold, 2,5-fold, 12.4-fold, 7.4-fold, and 10-fold, respectively, relative to control. This provides the sustainability of the functioning of the ecosystem under fish pressure. Such a mechanism prevents zooplankton over-exploitation by fish and enables to prolong the food chain.
