Animalia -- Bacteria (microorganisms) -- Cupriavidus necator -- Pseudomonas carboxydohydrogena
Аннотация: The study was conducted to determine the biological value of proteins synthesized by hydrogen-oxidizing microorganisms-the hydrogen bacteria Alcaligenes eutrophus Z1 and Ralstonia eutropha B5786 and the CO-resistant strain of carboxydobacterium Seliberia carboxydohydrogena Z1062. Based on a number of significant parameters characterizing the biological value of a product, the proteins of hydrogen-oxidizing microorganisms have been found to occupy an intermediate position between traditional animal and plant proteins. The high total protein in biomass of these microorganisms, their complete amino acid content, and availability to proteolytic enzymes allow for us to consider these microorganisms as potential protein producers. Аннотация: Microparticles obtained by using 14C-labeled resorbable hydroxyaminobutyric acid polymer were injected into the caudal vein of laboratory animals without negative aftereffects for their growth and development and without changes in the macro- and microstructure of organs and tissues. The distribution of microparticles in the viscera and the dynamics of accumulation of carbon-containing polymer degradation products in the viscera were studied. The main targets for the particles are liver tissues, as well as renal and splenic tissues. The polymeric matrix of the microparticles is most actively destroyed in the spleen and liver. The presence of high-molecular- weight polymeric matrix in organs indicates the integrity of microparticles and the possibility of long-term (up to 12 weeks) functioning of polymeric particles in vivo. Аннотация: Polyhydroxyalkanoate (PHA) sutures were implanted to test animals intramuscularly, and tissue reaction was investigated and compared with the reaction to silk and catgut. Tested monofilament sutures made of PHAs of two types polyhydroxybutyrate (PHB) and a copolymer of hydroxybutyrate and hydroxyvalerate (PHV) featured the strength necessary for the healing of muscle-fascial wounds. The reaction of tissues to polymeric implants was similar to their reaction to silk and was less pronounced than the reaction to catgut; it was expressed in a transient post-traumatic inflammation (up to four weeks) and the formation of a fibrous capsule less than 200 ?m thick, which became as thin as 4060 ?m after 16 weeks, in the course of reverse development. Macrophages and foreign-body giant cells with a high activity of acid phosphatase were actively involved in this process. PHB and PHB/PHV sutures implanted intramuscularly for an extended period (up to one year) did not cause any acute vascular reaction at the site of implantation or any adverse events, such as suppurative inflammation, necrosis, calcification of the fibrous capsule or malignant tumor formation. No statistically significant differences were revealed in the tissue response to polymer sutures of the two types. Capsules around silk and catgut sutures did not become significantly thinner.

Аннотация: Specimens of 14C-labeled polymer of 3-hydroxybutyric acid, P(3-HB), with different initial molecular weights, were used to prepare microparticles, whose morphology was not influenced by the M w of the polymer. During the particle preparation process, P(3-HB) molecular weight decreased by 15-20%. Sterile microparticles (mean diameter 2.4 ?m) were injected into the tail veins of Wistar rats (5 mg/rat). The effects of the particles administered to rats were studied based on the general response of animals and local response of internal organ tissues and blood morphology; no adverse effects on growth and development of the animals or unfavorable changes in the structure of the tissues of internal organs were observed. Measurements of radioactivity in tissues showed that 14C concentrations are different in different organs, changing during the course of the experiment. The main targets for 14C-labeled microparticles were tissues of the liver, spleen, and kidneys. Comparison of radioactivity levels and residual contents of high-molecular-weight matrix in tissues suggested that the most rapid metabolism and degradation of P(3-HB) occurred in the liver and spleen. Gel-permeation chromatography showed that at 3 h after the microparticles were injected into the bloodstream, polymer degradation started in all examined organs, except the lungs; at 12 weeks, the M w of the polymer matrix was as low as 20-30% of its initial value. The presence of high-molecular-weight (undegraded) polymer in the tissues at 12 weeks after administration of the particles suggests that P(3-HB) is degraded in tissues of internal organs slowly and, hence, P(3-HB)-based microparticles can be used as sustained-release drug-delivery systems. Аннотация: The paper presents the results of biomedical investigations of sutures made of polyhydroxyalkanoates (PHAs) of two types (poly-3-hydroxybutyrate (PHB) and poly-3-hydroxybutyrate-co-poly-3-hydroxyvalerate (PHB/PHV)), synthesized by the bacteria Ralstonia eutropha B 5786 in the experiment on test animals in vivo, in comparison with silk and catgut sutures. PHB and PHB/PHV (PHBV) implants produced no adverse effect on physiological, biochemical and functional parameters of the animals during the post-surgery period. The tested PHA sutures featured the necessary strength throughout the healing period of the muscle-fascial cuts. The reaction of tissues to the implantation of PHB and PHB/PHV fibers fitted into the usual scheme characteristic of the wound process and of the reaction to a foreign-body invasion. This reaction and the reaction of tissues to silk had the similar nature and period of inflammation, but it was much less pronounced than the reaction to catgut. The tissue response to the implantation of PHAs consisted in a short-duration (up to 2 weeks) post-traumatic inflammation and the formation of a fibrous capsule less than 200 ?m thick during weeks 4-8, which in 4-6 months was reduced to 40-60 ?m in the course of reverse development. There were no adverse changes, such as suppurative inflammation, necrosis, calcification, and malignization of the cicatrice, at the site of implantation of PHA filaments, unlike in the cases with silk and catgut. In the case of PHA implantation there was a typical prolonged (throughout the post-surgery monitoring period) pronounced macrophagal stage with a large number of macrophages present. The macrophages were of the phagocytic type and multinucleate giant foreign cells, with a high activity of acid phosphomonoesterase that correlated with the activity of the enzyme in blood. Throughout the period of monitoring no differences in the tissue response to the implantation of the polymer filaments of two PHA types were recorded. В© 2003 Elsevier Science B.V. Аннотация: The study was done to investigate the content of manmade radionuclides in aquatic organisms of the Yenisei River near the Mining-and-Chemical Combine (MCC) and to estimate the exposure dose rates to organisms from various sources. The results of the investigation and calculations suggest that the main source of radioactive contamination of aquatic organisms is the coolant of the third MCC reactor, which is still being released into the Yenisei. Gamma-spectrometric analysis revealed 23 manmade radionuclides in the biomass of aquatic plants. The aquatic animal Phylolimnogammarus viridis and diatoms also contain manmade radionuclides. Among aquatic organisms, the highest dose rate is received by aquatic plants (up to 39 ?Gy/day). For most aquatic organisms under study, the dose received from the technogenic irradiation is an order of magnitude higher than the dose received from natural irradiation. The water moss (Fontinalis antipyretica) features the highest capacity to accumulate manmade radionuclides; hence, it accumulates the largest technogenic exposure dose among the study aquatic organisms.

Аннотация: In 11 lakes from cold and warm regions we studied the content of highly unsaturated fatty acids (HUFA) in seston (<130?m) and crustacean zooplankton using gas chromatography-mass spectrometry. An increase of temperature correlated with a decrease of HUFA content in zooplankton. A multivariate canonical correlation analyses revealed, that the decrease of HUFA content was related with a decrease of per cent of copepods in zooplankton communities, which are known to have higher HUFA levels in their biomass, than cladocerans. This means that temperature primarily affected the HUFA levels indirectly, via changing of taxonomic structure of zooplankton community, while the homeoviscous adaptation of zooplankton individuals had comparatively lower importance. As found, water temperature was better predictor of HUFA contents of zooplankton, than the fatty acid composition of seston. Thus, it can be predicted, that a probable climate warming will decrease the content of the essential HUFA in freshwater zooplankton with possible negative consequences for animals of higher trophic levels. Аннотация: Bryophytes are abundant in streams and are a habitat for many invertebrates, but their contribution to the diet of fluvial zoobenthos is still debated. To estimate the amount of bryophyte-derived organic matter assimilated by benthic invertebrates, we used a combination of fatty acid and stable isotope analyses during a four-year monthly study of a littoral site in the Yenisei River (Siberia, Russia). Acetylenic acids, which are highly specific biomarkers of the water moss Fontinalis antipyretica, were found in lipids of all dominant benthic animals: gammarids, ephemeropterans, chironomids and trichopterans. The dominant zoobenthic species, Eulimnogammarus viridis, had maximum levels of the biomarkers in its biomass during winter, and minimum levels in summer. The zoobenthos in the studied site regularly consume and assimilate bryophyte-derived organic matter as a minor supplemental food. This consumption increases in winter, when the main food source of the zoobenthos, epilithic biofilms, are probably scarce. В© 2010 Versita Warsaw and Springer-Verlag Wien.

Efficiency of transfer of essential polyunsaturated fatty acids versus organic carbon from producers to consumers in a eutrophic reservoir / M. I. Gladyshev [et al.] // Oecologia. - 2011. - Vol. 165, Is. 2. - P521-531, DOI 10.1007/s00442-010-1843-6 . - ISSN 0029-8549

Аннотация: One of the central paradigms of ecology is that only about 10% of organic carbon production of one trophic level is incorporated into new biomass of organisms of the next trophic level. Many of energy-yielding compounds of carbon are designated as 'essential', because they cannot be synthesized de novo by consumers and must be obtained with food, while they play important structural and regulatory functions. The question arises: are the essential compounds transferred through trophic chains with the same efficiency as bulk carbon? To answer this question, we measured gross primary production of phytoplankton and secondary production of zooplankton and content of organic carbon and essential polyunsaturated fatty acids of ?-3 family with 18-22 carbon atoms (PUFA) in the biomass of phytoplankton and zooplankton in a small eutrophic reservoir during two summers. Transfer efficiency between the two trophic levels, phytoplankton (producers) and zooplankton (consumers), was calculated as ratio of the primary production versus the secondary (zooplankton) production for both carbon and PUFA. We found that the essential PUFA were transferred from the producers to the primary consumers with about twice higher efficiency than bulk carbon. In contrast, polyunsaturated fatty acids with 16 carbon atoms, which are synthesized exclusively by phytoplankton, but are not essential for animals, had significantly lower transfer efficiency than both bulk carbon, and essential PUFA. Thus, the trophic pyramid concept, which implicitly implies that all the energy-yielding compounds of carbon are transferred from one trophic level to the next with the same efficiency of about on average 10%, should be specified for different carbon compounds. Аннотация: Contents of fatty acids in filets of unfrozen (control), boiled, fried, roasted and boiled in a small amount of water humpback salmon, collected from a wholesale market in Krasnoyarsk city (Siberia, Russia) were analyzed. Special attention was paid to essential polyunsaturated fatty acids of ?3 family: eicosapentaenoic, 20:5?3 (EPA) and docosahexaenoic, 22:6?3 (DHA). Heat treatment in general did not decrease content of EPA and DHA in humpback, except a modest reduction during frying. Cooked humpback appeared to be the valuable source of essential ?3 PUFAs, namely EPA and DHA. It was hypothesized that the absence of significant reduction of PUFAs' contents in red flesh of fishes of Salmonidae family during heat treatment may be due to a high level of natural antioxidants which formed in the course of evolution as adaptation to their ecological niche. Аннотация: We studied fatty acid (FA) composition in samples from bodies and intestinal contents of the littoral amphipod Gammarus lacustris Sars, from the Bugach freshwater reservoir. Simultaneously, samples of seston and bottom sediments were also collected from the reservoir during early August. There were no differences in FA composition of gut contents, seston and sediments of pebbly bottom. Seston was the main food source of Gammarus but some FAs Gammarus got from sediments. The FA composition of G. lacustris and seston from the Bugach freshwater reservoir were compared with those of the animals from the saltwater Lake Shira (Siberia). While FA composition of the two Gammarus populations differed significantly, those of seston were practically similar: the composition of long-chain unsaturated fatty acids, 20:5?3, 22:6?3 and 20:4?6, were significantly higher in animals from saline Shira Lake, whereas 16:1 and 16:0 were higher in the freshwater populations of amphipods from the Bugach freshwater reservoir. Taking into account the relevant literature data, we hypothesise that this difference in C16 acid might be a distinguishing characteristic of FA composition of freshwater and saltwater crustaceans.

Аннотация: The circadian rhythm of grazing of microalgae by a laboratory culture of Ceriodaphnia quadrangula under continuous illumination was studied by continuous registration of chlorophyll fluorescence at the outlet of a flow-through cultivator. A culture of green alga Chlorella vulgaris was used as a feed. The data obtained were treated by the statistical spectral analysis. It was found that animals preliminarily grown under a 12 h light : 12 h dark regime and transferred to constant light showed two maxima in the circadian rhythm of grazing with periods of 0.7 and 1.1 h. Animals preliminarily grown under constant light showed no circadian rhythm of grazing. It was concluded that the circadian rhythm of grazing of C. quadrangula has endogenous nature and can change according to light conditions.

Аннотация: The vertical distribution and abundance of Gammarus lacustris in the pelagic zone of two fishless meromictic lakes, L. Shira and L. Shunet, in Southern Siberia (Russia), was studied with the underwater video recording system and using vertical hauls. In both lakes, during summer stratification, Gammarus was distributed non-homogenously, with a stable peak in the metalimnion. The average depth of Gammarus population in the pelagic zone was significantly correlated with the depth of the thermocline. Gammarus abundances obtained using vertical plankton hauls with net were quite comparable with those obtained from video records. The peak abundance of Gammarus in the pelagic zone of the lakes observed with underwater video amounted up to 400 individuals m-2, while the peak animal densities in the metalimnion reached 50 ind. m-3. The data are compared with previously published abundances of Gammarus in the littoral of Lake Shira. Both littoral and pelagic can be equally important habitats for amphipods in meromictic lakes. The absence of fish in the pelagic zone, high oxygen concentration, low water temperature, increased seston concentration, elevated water density in the metalimnion and the anoxic hypolimnion can be the most probable combination of factors that are responsible for the peak of Gammarus in the metalimnion of these lakes. Аннотация: An individual-based model of cyclic development of Cladocera populations was developed on the basis of experimental data. The model takes into account the following processes describing the development of an individual animal: maturation, transition into other reproductive classes, selection of the reproduction mode (parthenogenetic or gamogenetic), release of parthenogenetic progeny and death. The model assumes that switching from asexual to sexual reproduction is controlled by the concentration of food and metabolic by-products of the animal population. Verification of the model by independent experiments demonstrated that (1) during population growth, metabolic by-products build up in the medium, and (2) the effect of metabolic by-products on gamogenesis induction depends on concentration. The hypothesis that the effect of regulating reproductive switching factors should synchronise the development of population with the change of environmental conditions in order to ensure production of the maximum number of diapausing eggs was tested. It is shown that combination of regulating reproductive switching factors maximises the production of diapausing eggs. В© 2002 Elsevier Science B.V. All rights reserved.

Аннотация: Two-section enclosures were designed for the investigation of the effect of various physicochemical and biological factors on vertical distribution of zooplankton in situ. The framework of the enclosure was a cylindrical polyethylene column without any partitions inside, in which the isolation of animals in different sections after in situ exposure was achieved by pinching the flexible central part of the column. Enclosures were tested at the brackish stratified meromictic Lake Shira (Russia, Khakasia). The absence of fish and carnivorous zooplankton in the lake suggests that the vertical distribution of zooplankton is mainly determined by physicochemical gradients in the water column. Experiments and field observations demonstrated that all age and size groups of Arctodiaptomus salinus and Brachionus plicatilis strongly avoided surface layers during the daylight. The escape of zooplankton from the anoxic hypolimnion was less active. Statistically significant avoidance was observed only for copepodites C4-C5 and females of A. salinus. The relatively simple construction of the columns and easy handling during the experiment were the factors that favoured the use of this device to perform in situ basic tests of the effect of different factors on the vertical distribution of zooplankton. Аннотация: The change in reproductive mode from parthenogenesis to gametogenesis in Cladocera is controlled by multiple environmental cues. Maternal effects are involved in the control of reproductive switching. In this study, we estimated the readiness of Moina macrocopa females to change reproduction mode under the effect of conspecific chemicals on maternal and offspring generations. The results demonstrated that information about the chemical environment was not transmitted between generations (none of the females produced diapausing eggs in the control medium irrespective of their mothers' environment). Differences in maternal energetic investments were not significant, hence the maternal environment did not affect the fecundity. However, tested animals adapted to the effect of the diapause inducing factor. When offspring of mothers cultured in crowded water were also cultured in crowded water (the constant effect of conspecific chemicals), they switched less readily to gametogenesis than offsprings of mothers cultured in fresh medium.

Аннотация: During two vegetation seasons (2004-2005), we compared feeding spectra of Arctodiaptomus salinus (Calanoida, Copepoda) populations inhabiting two neighboring salt lakes, Shira and Shunet, Khakasia, Russia, using fatty acid (FA) trophic markers. Sestonic FA composition in two lakes moderately differed, whereas levels of diatom FA markers were higher in Lake Shunet and of Cyanobacteria and green algae markers in Lake Shira. In general, markers in storage lipids-triacylglycerols (TAG) of A. salinus-reflected the differences in sestonic composition of the two lakes. Nevertheless, TAG fraction was also enriched by FA trophic markers of the minor components of seston, which were selectively ingested by the animals. In Lake Shira, A. salinus had significantly higher concentrations of bacterial FA markers in TAG. In Lake Shunet, TAG of A. salinus contained significantly higher relative amounts of 18:4?3, 18:5?3 and C22 polyunsaturated fatty acids (PUFA), which indicated marked contribution of cryptophytes or (and) flagellates into the diet. Laboratory experiments showed feeding on Cryptomonas and sulfur purple bacteria in Lake Shunet and ciliates and colonial picoplankton in both lakes, and generally confirmed the differences in FA trophic markers in A. salinus between the lakes. The two populations of A. salinus markedly differed in levels of essential long-chain PUFA, eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids, although the levels of these FA in seston were quite similar between the two lakes. The higher levels of the essential ?3 PUFA in A. salinus in Lake Shunet may be an adaptive response of the animals to a vertical stratification of physico-chemical conditions and significantly higher salinity levels at the boundary of adjacent bottom layer in this lake. Аннотация: Polyunsaturated fatty acids (PUFA) of ?3 family are of crucial physiological importance for the most animals and they are an essential and deficient component of human nutrition. These compounds are most effectively synthesized by some groups of algae, hence, aquatic ecosystems are considered to be the main source of these PUFA for human nutrition. Factors controlling the content and distribution of ?3 PUFA in freshwater organisms of basic trophic levels and determined PUFA final production in freshwater ecosystems are considered in the review. PUFA biosynthesis is known to be tightly related to basic fatty acid metabolic pathways. Hence, fatty composition and the PUFA content of major freshwater hydrobiont groups, including bacteria, algae, invertebrates and vertebrates, and environmental and population age effects are described. The peculiarities of PUFA transfer between organisms of various trophic levels are discussed. The essential ?3 PUFA is one of the important parameter of food quality of aquatic consumers and they can determine the rate of energy and matter transfer between producers and primary consumers and, as a result, in a whole freshwater food chain. Analysis of PUFA content and its regulation in biomass of various fish populations indicates that freshwater ecosystems are of the same value in respect of PUFA sources as marine ecosystems. Despite the great practical importance, the studies focused on production and whole pools of ?3 PUFA in different freshwater ecosystems are still scarce and need to be continued.
