Аннотация: Microparticles made from degradable polyhydroxyalkanoates of different chemical compositions a homopolymer of 3-hydroxybutyric acid, copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids (P3HB/4HB), 3-hydroxybutyric and 3-hydroxyvaleric acids (P3HB/3HV), 3-hydroxybutyric and 3-hydroxyhexanoic acids (P3HB/3HHx) were prepared using the solvent evaporation technique, from double emulsions. The study addresses the influence of the chemical compositions on the size and ?-potential of microparticles. P3HB microparticles loaded with doxorubicin have been prepared and investigated. Their average diameter and ?-potential have been found to be dependent upon the level of loading (1, 5, and 10 % of the polymer mass). Investigation of the in vitro drug release behavior showed that the total drug released from the microparticle into the medium increased with mass concentration of the drug. In this study mouse fibroblast NIH 3T3 cells were cultivated on PHA microparticles, and results of using fluorescent DAPI DNA stain, and MTT assay showed that microparticles prepared from PHAs of different chemical compositions did not exhibit cytotoxicity to cells cultured on them and proved to be highly biocompatible. Cell attachment and proliferation on PHA microparticles were similar to those on polystyrene. The cytostatic drug encapsulated in P3HB/3HV microparticles has been proven to be effective against HeLa tumor cells. В© 2013 Springer Science+Business Media New York.

Аннотация: Synthesis of 3- and 4-hydroxybutyrate copolymer (3HB-co-4HB), the most promising member of the biodegradable polyhydroxyalkanoate (PHA) family, has been studied. Cultivation conditions of naturally occurring strains of hydrogen-oxidizing bacteria Ralstonia eutropha B5786 and Cupriavidus eutrophus B10646 have been optimized to ensure efficient synthesis of the 3HB-co-4HB copolymer. A set of highly pure samples of the 3HB-co-4HB copolymer with 4HB content varying from 8.7 to 24.3 mol% has been obtained. Incorporation of 4HB into the copolymer was shown to cause a more pronounced decrease in polymer crystallinity than the incorporation of 3-hydroxyvalerate or 3-hydroxyhexanoate; samples with a degree of crystallinity below 30% have been obtained. The weight average molecular mass of the 3HB-co-4HB copolymers was shown to be independent on the monomer ratio and to vary broadly (from 540 to 1110 kDa). В© 2010 Pleiades Publishing, Ltd.

Аннотация: Biodegradability patterns of two PHAs: a polymer of 3-hydroxybutyric acid (3-PHB) and a copolymer of 3-hydroxybutyric and 3-hydroxyvaleric acids (3-PHB/3-PHV) containing 11 mol% of hydroxyvalerate, were studied in the tropical marine environment, in the South China Sea (Nha Trang, Vietnam). No significant differences have been observed between degradation rates of 3-PHB and 3-PHB/3-PHV specimens; it has been found that under study conditions, biodegradation is rather influenced by the shape of the polymer item and the preparation technique than by the chemical composition of the polymer. Biodegradation rates of polymer films in seawater have been found to be higher than those of compacted pellets. As 3-PHB and 3-PHB/3-PHV are degraded and the specimens lose their mass, molecular weight of both polymers is decreased, i.e. polymer chains get destroyed. The polydispersity index of the PHAs grows significantly. However, the degree of crystallinity of both PHAs remains unchanged, i.e. the amorphous phase and the crystalline one are equally disintegrated. PHA-degrading microorganisms were isolated using the clear-zone technique, by inoculating the isolates onto mineral agar that contained PHA as sole carbon source. Based on the 16S rRNA analysis, the PHA-degrading strains were identified as Enterobacter sp. (four strains), Bacillus sp. and Gracilibacillus sp.В© 2010 Elsevier Ltd. All rights reserved.

Аннотация: The effect of laser irradiation on the properties of the surface of films prepared from a bioresorbable polymer poly(hydroxybuturate) has been studied. To determine the spectral region of the polymer optimal for the effective action of radiation on electron molecular bonds, theoretical investigations have been performed, which have shown that, for modifying the surface of PHB scaffolds, it is expedient to use a vacuum laser at a wavelength of 160 nm. Using laser irradiation at a power from 3 to 30 W, a series of films with modified surface, from roughnesses to perforations, have been obtained. The microstructure and properties of the film surface depending on the mode of irradiation have been examined, and conditions have been found under which the contact marginal angles of film wetting with water can be decreased to 50В° (compared with 76-80В° in starting products). Thus, conditions of laser treatment of PHB scaffolds have been theoretically substantiated and experimentally realized that provide a beneficial effect on the properties of the surface without destroying the structure of the material. Аннотация: The possibility of use of polyhydroxyalkanoates (PHAs), biodegradable microbial polyesters, as a carrier for pesticides (?-hexachlorcyclohexane and lindane) for targeted and controlled delivery of these compounds to soil was investigated. The kinetics of polymer degradation and the dynamics of pesticide release from the extended-release formulations was studied. It is shown that pesticides embedded in a degradable polymer (PHA) carrier are released gradually and slowly, without surges, as the polymer is degraded by the soil micro-flora. The microbial soil component actively responded to the addition of the polymer as an additional nutrient substrate: the latter was degraded and then utilized. The rate of the pesticide release to the soil can be regulated by varying the polymer-pesticide ratio. В© 2009 Pleiades Publishing, Ltd.

Аннотация: The kinetics of polyhydroxyalkanoate (PHA) degradation in natural environment in two water reservoirs having various ecological characteristics was studied. It was shown that biodegradation of polymer essentially depends on the environment temperature and inorganic composition of water. Аннотация: Samples of a hybrid composite of polyhydroxybutyrate (PHB), a biodegradable polyester, and hydroxyapatite (HA), with different PHB/HA ratios, have been prepared using mechanical-physical method. Electron microscopy, X-ray structure analysis and differential thermal analysis have been used to investigate the structure and physicochemical properties of the composite, depending on the PHB/HA ratio. The properties of the surface of the HA-loaded composite are significantly different from those of the pure polymer. As the HA percentage in the composite increases, free interface energy, the cohesive force, i.e., the strength of the adhesive bond between the composite surface and the water phase, and surface wettability increase. The HA percentage of the composite does not influence its melting temperature, but affects the temperature for the onset of decomposition: as the HA content increases from 0 to 10% (w/w), Td decreases from 260В°C to 225В°C. The degree of crystallinity of PHB/HA increases from 77% to 89% with an increase in the HA fraction from 10% to 50%. Functional properties of the composites have been investigated in vitro and in vivo. The best parameters of growth and differentiation of murine marrow osteoblasts are registered on PHB/HA samples containing 10% and 20% HA. In ectopic bone formation assay it has been proven that the hybrid PHB/HA composites can function as scaffolds and that bone tissue develops on their surface and in pores. В© VSP 2006.

Аннотация: Polyhydroxyalkanoate (PHA) sutures were implanted to test animals intramuscularly, and tissue reaction was investigated and compared with the reaction to silk and catgut. Tested monofilament sutures made of PHAs of two types polyhydroxybutyrate (PHB) and a copolymer of hydroxybutyrate and hydroxyvalerate (PHV) featured the strength necessary for the healing of muscle-fascial wounds. The reaction of tissues to polymeric implants was similar to their reaction to silk and was less pronounced than the reaction to catgut; it was expressed in a transient post-traumatic inflammation (up to four weeks) and the formation of a fibrous capsule less than 200 ?m thick, which became as thin as 4060 ?m after 16 weeks, in the course of reverse development. Macrophages and foreign-body giant cells with a high activity of acid phosphatase were actively involved in this process. PHB and PHB/PHV sutures implanted intramuscularly for an extended period (up to one year) did not cause any acute vascular reaction at the site of implantation or any adverse events, such as suppurative inflammation, necrosis, calcification of the fibrous capsule or malignant tumor formation. No statistically significant differences were revealed in the tissue response to polymer sutures of the two types. Capsules around silk and catgut sutures did not become significantly thinner.

Results of the study of tissue response to biodegradable polyoxyalkanoate suture threads in the postoperative period are analyzed. These suture threads have been shown to have the required strength and meet a number of other requirements for currently available materials.

Аннотация: This work is a review of the results of biomedical studies of polymer devices (films, fibers, microparticles, 30 implants) made from resorbable PHAs synthesized by the bacterium Wautersia (Ralstonia) eutropha 65786, using the technology developed at the Institute of Biophysics of the Siberian Branch of the Russian Academy of Sciences. Two types of PHAs - polyhydroxybutyrate (PHB) and a hydroxybutyrate/hydroxyvalerate copolymer (PHB/PHV) - have been proven to be biocompatible in vitro in cultures of fibroblasts, endothelial cells, hepatocytes, and osteoblasts, and in short- and long-duration experiments on animals. Polymer films and membranes have been found to be usable as scaffolds for functioning cells and monofilament suture fibers - for stitching muscular-fascial wounds and in abdominal surgery. Ectopic bone formation assay and experiments with the model of segmental osteotomy showed that 30 PHB and PHB/HA implants can be used for reparative osteogenesis. Аннотация: Procedure for microencapsulation providing stable formation of high quality microspheres was designed. Conditions for deposition of the anthracycline antibiotic daunorubicin to polymer matrix were developed and microsheres loaded with various quantities of the drug were prepared. The kinetics of the in vitro and in vivo release of daunorubicin from the microsheres was studied. The rate of the rubomycin release to the medium in vitro (balanced phosphate buffer at 37.8В°C) in a 300-hour experiment directly depended on the quantity of the incorporated drug and averaged 0.81 В· 10 -4 to 2.3 В· 10 -4 mcg/mlВ·h. The experiment on laboratory animals with intraperitoneal administration of the rubomycin microsperes showed that the drug remained in the blood and abdominal liquid for a long time (up to 10 days). Possible control of the quantity of the rubomycin encapsulation to the polymer matrix, no sharp efflux of the drug at the early stages of the observation and low rate of the drug release to the medium allowed to conclude that the use of the biodegradable polymer microsperes as carriers of the high toxic antibiotic providing its prolonged action was prospective.

Аннотация: A procedure for preparation of microspheres from biodegradable linear polyether of microbiological origin (polyhydroxybutirate, PHB) with using the technology of solvent evaporation was developed considering a specific example of two- and three-component emulsions. The procedure provided permanent preparation of the microspheres of high quality. The influence of the procedure (emulsion type, dispersion process and medium temperature) on the yield of the microspheres, their structure and size was shown. The temperature had a significant impact on incorporation of the antitumor anthracycline antibiotic daunorubicin (rubomycin) to the polymer matrix. The microspheres with various levels of the drug load (29 and 90% of the initial content in the emulsion) were prepared and the kinetics of the in vitro rubomycin release was studied. The dynamics of the highly toxic rubomycin release from the microspheres was on the whole even with the curve profile reaching the plateau in 20-22 hours of the observation period. The rate of the rubomycin release to the medium depended on the value of the antibiotic incorporation and was maximum within the first two hours (3.3 and 13.0 mcg/mlВ·h) that corresponded to the release of 0.97 and 3.89 of the incorporated antibiotic. The average rate of rubomycin release during 300 hours was 0.81В·10-4 and 2.3В·10-4 mcg/mlВ·h. The release constituted respectively 3.9 and 13.11% of the antibiotic incorporated to the microspheres.

Аннотация: The kinetics of polyhydroxyalkanoate (PHA) degradation in natural environment in two water reservoirs having various ecological characteristics was studied. It was shown that biodegradation of polymer essentially depends on the environment temperature and inorganic composition of water. The processes of polymer degradation under aerobic and anaerobic conditions were compared. The polymer degradation was slower under anaerobic conditions.

Аннотация: The paper describes the production and investigation of flexible films made of high-purity polyhydroxyalkanoates (PHAs) - polyhydroxybutyrate [poly-(3HB)] and poly-3-hydroxybutyrate-co-poly-3-hydroxyvalerate [poly(3Hl3-co-3HV)], containing 4-30 mol % hydroxyvalerate. Poly(3HB-co-3HV) films have a more porous structure than poly-(3HB) films, which are more compact, but their surface properties, such as wettability and surface and interface energies, are the same. Sterilisation of the PHA films by conventional methods (heat treatment and gamma-irradiation) did not impair their strength. Cells cultured on PHA films exhibited high levels of cell adhesion. Cell morphology, protein synthesis and DNA synthesis were estimated by extent of H-3-thymidine incorporation into the animal cell cultures of various origins (fibroblasts, endothelium cells, and isolated hepatocytes) in direct contact with PHAs. The investigation showed that this material can be used to make matrices for in vitro proliferous cells. The investigated properties of poly-(3HB) and poly(3HB-co-3HV) films proved to be fundamentally similar. (C) 2004 Kluwer Academic Publishers.

Аннотация: The biodegradable polymer poly(3-hydroxybutyrate-co-3-hydroxyvalerate) has been used to design experimental sustained-release formulations of the herbicide Zellek Super in the form of films and microgranules. The kinetics of polymer degradation and the dynamics of herbicide release show that the rate and extent of herbicide release from the polymer matrix into the soil depends on the geometry of the carrier and the proportion of the pesticide loaded into it (polymer/pesticide mass ratio). Experiments with the creeping bentgrass (Agrostis stolonifera L.) show that the formulations of the herbicide Zellek Super constructed as microgranules and films can be successfully used to suppress the growth of grasses. This study is the first to demonstrate that biodegradable polyhydroxyalkanoates can be used effectively to construct environmentally friendly sustained-release PHA-herbicide systems that can be placed into the soil together with seeds.

Аннотация: Matrices based on resorbable polyhydroxyalkanoates (PHAs) of five types {a homopolymer of 3- hydroxybutyric acid, copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids [P(3HB/4HB)], 3-hydroxybutyric and 3-hydroxyvaleric acids [P(3HB/3HV)], 3-hydroxybutyric and 3-hydroxyhexanoic acids [P(3HB/3HHx)]} have been constructed and characterised. No significant differences have been found in tissue response to implantation of these PHAs. Non-coarse fibrous capsules that formed around PHA matrices reached their maximum thickness (60-90 ?m) 90 days after implantation; by day 180, the average thickness of the capsules had decreased by 1·5-2·3 times. The number of foreign body giant cells, resorbing PHAs, remained high. In vivo biodegradation behaviour of polymer matrices is related to the chemical composition of the PHA. Matrices prepared from copolymers P(3HB/4HB) and P(3HB/3HHx) exhibited the fastest degradation rates. P3HB/3HV matrices were degraded more slowly, and P3HB matrices were the most durable. In the PHA matrices that were degraded more slowly, giant cell reaction developed later. © 2014 Institute of Materials, Minerals and Mining.

Аннотация: This study investigates kinetic and production parameters of a glucose-utilizing bacterial strain, C. eutrophus B-10646, and its ability to synthesize PHA terpolymers. Optimization of a number of parameters of bacterial culture (cell concentration in the inoculum, physiological activity of the inoculum, determined by the initial intracellular polymer content, and glucose concentration in the culture medium during cultivation) provided cell concentrations and PHA yields reaching 110 g/L and 80%, respectively, under two-stage batch culture conditions. Addition of precursor substrates (valerate, hexanoate, propionate, c-butyrolactone) to the culture medium enabled synthesis of PHA terpolymers, P(3HB/3HV/4HB) and P(3HB/3HV/3HHx), with different composition and different molar fractions of 3HB, 3HV, 4HB, and 3HHx. Different types of PHA terpolymers synthesized by C. eutrophus B-10646 were used to prepare films, whose physicochemical and physicalmechanical properties were investigated. The properties of PHA terpolymers were significantly different from those of the P3HB homopolymer: they had much lower degrees of crystallinity and lower melting points and thermal decomposition temperatures, with the difference between these temperatures remaining practically unchanged. Films prepared from all PHA terpolymers had higher mechanical strength and elasticity than P3HB films. In spite of dissimilar surface structures, all films prepared from PHA terpolymers facilitated attachment and proliferation of mouse fibroblast NIH 3T3 cells more effectively than polystyrene and the highly crystalline P3HB.

Аннотация: A series of 3D implants and filling materials prepared from powdered biodegradable polymers, polyhydroxyalkanoates (PHAs), have been designed for the purposes of reparative osteogenesis. The 3D implants are made of resorbable polymer of hydroxybutyric acid (poly-3-hydroxybutyrate, P3HB) and a composite of this polymer with hydroxyapatite (HA) (P3HB/HA). The properties of the implants were studied in vivo in a model of segmental osteotomy and compared with commercial material Bio-Oss®. All implants containing P3HB as the main component facilitate reconstructive osteogenesis. P3HB and P3HB/HA show pronounced osteoplastic properties; their in vivo degradation is slow and corresponds to the growth of a new bone tissue, facilitating normal reparative osteogenesis. Also, powdered P3HB and P3HB/tienam can be used as filling materials for osteoplasty of bone cavities infected by Staphylococcus aureus. Biodegradable 3D implants and P3HB-based filling materials show pronounced osteoplastic properties and degrade in vivo at a slow rate, enabling normal reparative osteogenesis.
