Аннотация: The paper summarises the results of a three-year research study (European Union Grant: INTAS 97-0519) aimed at investigating the planktonic populations and trophic organization of the Lake Shira ecosystem - a saline lake in Khakasia, Siberia. The lake exhibits a stable summer-autumn stratification of the chemical-biological components. The mechanisms responsible for the 'blooming' of phytoplankton in the deeper layers were investigated in greater detail, using data from both field and laboratory experiments. The spectra of nutrition were examined to estimate the relationships between the specific growth rates of the hydrobionts and the influence of the limiting factors: light, nutrients. The observed heterotrophic capability of a metalimnetic phytoplankton population might help explain the development in the deeper waters of Lyngbya contorta. The scheme of trophic interactions was put up, based on the assessment of the carbon pools and carbon flows in the pelagic zone of the lake. A mathematical model of the vertical structure of the lake's plankton populations was constructed, using the ecosystem description and data of vertical turbulent diffusion. The role of light and nutrient limitations and grazing mortality in forming the vertical inhomogeneities, particularly in lowering the depth of the maximal cyanobacterial biomass, has been demonstrated. The theoretical curves for the stratification of chemical and biological parameters have been brought in conformity with the field observations, e.g. for the different patterns of the peaks, and for the biomass maxima of cyanobacteria, purple and green sulphur bacteria, oxygen, and hydrogen sulphide. The calculations revealed that for an adequate assessment of the parameters for the hydrogen sulphide zone it is necessary to introduce flows of allochthonous organic matter. Based on the form of the sulphur distribution curve, the allochthonous input of organic matter and the inflow of hydrogen sulphide from the bottom have been theoretically discriminated for the first time. It has also been ascertained that irrespective of the depth the allochthonous substances limiting bacterial growth, the bacteria are uniformly distributed over depth and can serve as an indicator of the presence of limitation (the effect of autostabilisation in space). Of indisputable interest to limnology are the specific methods developed for understanding the functioning of Lake Shira ecosystem. These include the autostabilisation of the limiting factors, the on-the-spot fluorescent method of determining the three classes of microalgae, the algal mixotrophy and the planktonic population interactions and feedbacks, and development of a more sensitive, bioluminescent method for mapping the nonhomogeneities. Owing to a balanced combination of classical approaches (field observations, in situ data on production-decomposition) and the more recent ones (satellite monitoring, biophysical methods of estimating interactions of populations, mathematical models based on the field and experimental data), many of the structural-function relationships in the ecosystem can now be explained, and the models can provide 'mutual control and mutual agreement' between the data collected using different approaches.

Аннотация: In 2002, a "top-down" biomanipulation (reduction of biomass of planktivorous fish Carassius auratus) had been successfully carried out in a small reservoir of the river Bugach (Krasnoyarsk, Russia), after which the cyanobacterial blooming ceased. However, the reservoir ecosystem was absolutely free of Daphnia - the main link of trophic cascade. As supposed, the reduction of blooming was the result of suppression of a direct stimulation effect of planktivorous fish on cyanobacteria, revealed earlier in laboratory experiments. The question arose as to whether the effect of stimulation of cyanobacteria revealed in laboratory may lead to the changes in biomass of cyanobacteria in the reservoir, observed after the biomanipulation. To test this supposition, a mathematical model describing growth of cyanobacteria in the reservoir was developed. The modelling results and field data on biomass of cyanobacteria in summer closely coincided. Modelling calculations showed that direct influence of planktivorous fish could cause the second summer peak of water blooming by Microcystis. On the contrary, removal of crucian carp from the reservoir will not affect the level of water blooming caused by cyanobacteria Anabaena, as this species' growth is not stimulated by fish. Аннотация: The reaction of algae and cyano-bacteria photosynthetic apparatus on atmospheric sediments, incident solar radiation and wind velocity was estimated by measuring chlorophyll fluorescence using three-beam fluorescent diagnostics. As shown, wind in the spring positively affects the chlorophyll concentration but causes negative effect on phytoplankton photosynthetic activity in other seasons. Under conditions of high-intensity solar irradiation confervoid and colonial cyanobacteria, possessing gas vacuoles, have an advantage over plankton eukaryotic algae. Other response differences weren't observed.

Аннотация: The comparison of the dynamics of phytoplankton biomass and total amino acid composition was made for two water bodies: in one the phytoplankton were dominated by prokaryotes (i.e., there was a bloom of cyanobacteria) and by eukaryotic microalgae in the other. The dynamics of phytoplankton biomass and of total amino acid composition of water were investigated during the vegetation season. It was found that the only factor that significantly changed the percentages of amino acids in water was the bloom of cyanobacteria in the "blooming" water body. During the bloom of cyanobacteria, the absolute and relative content of the Leu-Glu group increased, while the contents of other acids generally dropped. Before and after the bloom, no significant variations in the total amino acid composition were recorded. In the reservoir where eukaryotic microalgae dominated, no significant variations in amino acid composition were recorded during the season.

Аннотация: Mathematical model is constructed for studying the effect of photosynthesis stimulation in cyanobacteria after their passing through crucian intestines on cyanobacteria biomass changes after bio-manipulations. At good agreement of model and natural data an explanation is found for observed differences in the influence of planktivorous fish on the two species of cyanobacteria. The calculations showed that one of the reasons for appearance of the peak of water blooming with a species Microcystic may be direct metabolic influence of planktivorous fish. Elimination of crucian from the pond will not affect the level of water blooming with Anabaena species.

Аннотация: A set of bioluminescent tests was developed to monitor water quality in natural and laboratory ecosystems. It consisted of four bioluminescent systems: luminous bacteria, coupled enzyme system NADH:FMN-oxidoreductase-luciferase and triplet enzyme systems with alcohol dehydrogenase and trypsin. The set of biotests was applied for a small forest pond (Siberia, Russia), laboratory microecosystems polluted with benzoquinone and a batch culture of blue-green algae. Thereby effects of natural water compared to those of models of heavy pollution and "bloom" of blue-greens on the bioluminescent tests were revealed. The set of biotests was not affected by a natural seasonal variability of water quality in the unpolluted pond, but responded to the heavy pollution and the "bloom" of blue-greens. The set of biotests could be recommended as the alarm test to control the acute toxicity of natural water bodies. В© 2001 Elsevier Science Ltd.

Аннотация: Using experimental microecosystems the kinetics of phenol disappearance in small forest pond waters (Siberia, Russia) in the summer of 1995-96 were investigated. Despite of high variability of components of the ecosystem (plankton biomass and species composition) and two pronounced 'blooms' of green algae Volvox aureus the same kinetics of the disappearance took place over the investigated period. Half-lives of the pollutant depended on water temperature only. A comparison of the self-purification of the pond with that of the Krasnoyarsk reservoir, 'blooming' with blue-greens was carried out. Half-lives in the pond were significantly lower than that in the reservoir. During the periods of 'blooms' of the green algae in the pond the concentrations of inorganic nutrients were comparatively high and the phenol-degrading bacteria likely were not limited by these nutrients, in contrast to the periods of 'bloom' of the blue-green algae in the reservoir.

Аннотация: The possibility of introducing genetically engineered microorganisms (GEM) into simple biotic cycles of laboratory water microcosms was investigated. The survival of the recombinant strain Escherichia coli Z905 (Ap(r), Lux+) in microcosms depends on the type of model ecosystems. During the absence of algae blooming in the model ecosystem, the part of plasmid-containing cells E.coli decreased fast, and the structure of the plasmid was also modified. In conditions of algae blooming (Ankistrodesmus sp.) an almost total maintenance of plasmid-containing cells was observed in E.coli population. A mathematics model of GEM's behavior in water ecosystems with different level of complexity has been formulated. Mechanisms causing the difference in luminescent exhibition of different species are discussed, and attempts are made to forecast the GEM's behavior in water ecosystems.

Аннотация: The behavior of Escherichia coil Z905, carrying a recombinant plasmid pPHL7 with genes determining ampicillin resistance and bacterial luminescence, and the efficiency of expression of cloned genes were studied after introduction of the strain into model aqueous ecosystems with different trophic chain lengths. The E. coli Z905 variants isolated from ecosystems after different periods of time were found to vary in their resistance to ampicillin (from 50 to 0.05 mu g/ml) and in the intensity of bioluminescence. An increase in the concentration of the selective factor (ampicillin) or in the extent of the aqueous microcosm blooming restored the expression of the recombinant plasmid genes in some clones.

Аннотация: The comparison of the dynamics of phytoplankton biomass and total amino acid composition was made for two water bodies: in one the phytoplankton were dominated by prokaryotes (i.e., there was a bloom of cyanobacteria) and by eukaryotic microalgae in the other. The dynamics of phytoplankton biomass and of total amino acid composition of water were investigated during the vegetation season. It was found that the only factor that significantly changed the percentages of amino acids in water was the bloom of cyanobacteria in the "blooming" water body. During the bloom of cyanobacteria, the absolute and relative content of the Leu-Glu group increased, while the contents of other acids generally dropped. Before and after the bloom, no significant variations in the total amino acid composition were recorded. In the reservoir where eukaryotic microalgae dominated, no significant variations in amino acid composition were recorded during the season.

Аннотация: The possibility of symbiotic, mutualistic (++) interrelations between epibiontic microalgae (Characidiopsis ellipsoidea, Colacium vesiculosum) and cyclopoids (Cyclops vicinus, Mesocyclops leukarti) in the Bugach recreational reservoir with a bloom of cyanobacteria is demonstrated. Overgrown cyclopoids, compared to those free from microalgae, have advantages in oxygen supply, which may be insufficient in the period of intensive algal bloom. Calculations have shown that oxygen requirements of cyclopoids may be fully satisfied due to C. vesiculosum photosynthetic activity in the dysphotic zone if the density of overgrowth exceeds 1000 algal cells per individual. There is no significant correlation between the natural mortality of crustaceans and the indices of their overgrowth by microalgae.
