Аннотация: This study is devoted to investigating the content of uranium isotopes in water of the Yenisei River and its tributaries within the territories affected by the operation of Rosatom plants (mining chemical combine, and electrochemical plant). Long-term monitoring of the 238U content by mass spectrometry carried out in two institutes of the Siberian Branch of the Russian Academy of Sciences first revealed the multiple excess of 238U over the background content in different areas of the Yenisei River basin, such as the region of the Yenisei River near the effluents of the mining and chemical combine (MCC), and the territories of the Bol'shaya Tel' and Kan rivers. In these regions, the 238U content in water reaches 2.1-4.0 ?g/l, which exceeds its content upstream from the MCC (0.3-0.6 ?g/l) by almost an order of magnitude. The studies of the isotopic composition of uranium in water samples, which were carried out at the Institute of Inorganic Chemistry, Siberian Branch, Russian Academy of Sciences, showed the presence of a technogenic isotope of uranium 236U in the samples from the Bolshaya Tel' River and revealed the deviation of the isotope ratio 238U/ 235U (167 В± 3 and 177 В± 3) from the equilibrium natural ratio ( 238U/ 235U = 138). These facts attest to the technogenic origin of part of the uranium in water of the Bol'shaya Tel' River connected with the activity of MCC. The excess uranium content in the Kan River requires additional studies to ascertain the fraction of uranium of technogenic origin connected with the activity of the electrochemical plant (ECP) (Fig. 1, Table 4). Аннотация: The study addresses 242Pu accumulation by Elodea canadensis, one of the abundant species of submerged plants in the Yenisei River. 242Pu in water samples of the "Elodea - Yenisei River water" model system and in the biomass fractions was determined ?-spectrometrically, following radiochemical recovery of 242Pu using 236Pu - a chemical yield tracer. The experiments on accumulation of 242Pu by Elodea biomass showed that the activity concentration of 242Pu can reach 21 В± 2 Bq/g dry wt, with the concentration factor for 242Pu 13100 В± 2100 L/kg dry wt. Results of chemical fractionation proved that during the first few hours of the experiment 242Pu contained in Elodea was mainly concentrated in the exchangeable and the adsorbed fractions of biomass (about 100%). As Elodea biomass accumulated 242Pu, the absolute amount of 242Pu in the exchangeable and the adsorbed fractions remained almost unchanged, although the portion of 242Pu tightly bound to biomass increased. At the end of the experiment, on day 7, 242Pu tightly bound to biomass (fractions of organics and mineral residue) constituted 43-63% (in different experiments) of the total 242Pu in the biomass. Аннотация: Releases from the nuclear facility Mining-and-Chemical Combine (MCC) located at Zheleznogorsk have contributed to the radionuclide contamination of the Yenisei River since operations commenced in 1958. The aim of this study was to assess the activity concentrations of artificial radionuclides and the strength of their binding in Yenisei River sediments. Investigation of Yenisei River sediment samples revealed the presence of artificial radionuclides typical of the MCC radioactive discharge: namely, isotopes of europium, caesium, 60Co and transuranium elements. The concentrations of artificial radionuclides in the sediment layers remain relatively high as far as 200 km downstream of the MCC. In sediment cores collected upstream of the MCC, ? -spectrometric measurements registered only one artificial radionuclide, 137Cs, with a maximal activity of 8 BqВ·kg -1. Sequential extraction performed on samples of the upper layers of the sediment core showed different degrees of potential environmental availability for artificial radionuclides: the highest was recorded for 241Am and 152Eu (up to 85% of initial activity), followed by 60Co (up to 32%), and finally, 137Cs (up to 15%). In a few samples, 241Am was present in the unextractable form, which may be accounted for by the presence of reactor fuel microparticles. В© 2010 Taylor & Francis.

Measurements made with a submersed sensor demonstrated that one-meter ice transmits light in a quantity sufficient for oxygenic and anoxygenic photosynthesis. As in the summer season, in the community of phototrophic bacteria found in Lake Shira, the purple sulfur bacteria Amoebobacter purpureus dominated, whereas, in Lake Shunet, the green sulfur bacteria Pelodictyon luteolum were predominant. Photosynthetic production, measured using the radioisotopic method, was several times lower than that in summer. The rates of sulfate reduction and production and oxidation of methane in the water column and bottom sediments were also lower than those recorded in summer. The process of anaerobic methane oxidation in the sediments was an exception, being more intense in winter than in summer. The data from radioisotopic measurements of the rates of microbial processes correlate well with the results of determination of the isotopic composition of organic and mineral carbon (? 13C) and hydrogen sulfide and sulfate (? 34S) and suggest considerable seasonal variations in the activity of the microbial community in the water bodies investigated.

Аннотация: The integrated study addressing biodegradation of microbial linear polyesters of hydroxyalkanoic acids (polyhydroxyalkanoates, PHAs) in tropical conditions by microbial communities of Vietnamese soils was performed in locations close to Hanoi and Nha Trang, which differed in their weather conditions and microbial communities. It shows that PHA degradation in tropical soils is influenced by polymer chemical composition, specimen shape, and microbial characteristics. The homopolymer of 3-hydroxybutyric acid is degraded at higher rates than the copolymer of 3-hydroxybutyric and 3-hydroxyvaleric acids. The average rates of mass loss were 0.04-0.33% per day for films and 0.02-0.18% for compact pellets. PHA degradation was accompanied by a decrease in the polymer molecular mass and, usually, an increase in the degree of crystallinity, suggesting preferential degradation of the amorphous phase. Under the study conditions, representatives of the bacterial genera Burkholderia, Bacillus, Cupriavidus, Mycobacterium, and Nocardiopsis and such micromycetes as Acremonium, Gongronella, Paecilomyces, and Penicillium, Trichoderma have been identified as major PHA degraders. Аннотация: Microparticles made from degradable polyhydroxyalkanoates of different chemical compositions a homopolymer of 3-hydroxybutyric acid, copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids (P3HB/4HB), 3-hydroxybutyric and 3-hydroxyvaleric acids (P3HB/3HV), 3-hydroxybutyric and 3-hydroxyhexanoic acids (P3HB/3HHx) were prepared using the solvent evaporation technique, from double emulsions. The study addresses the influence of the chemical compositions on the size and ?-potential of microparticles. P3HB microparticles loaded with doxorubicin have been prepared and investigated. Their average diameter and ?-potential have been found to be dependent upon the level of loading (1, 5, and 10 % of the polymer mass). Investigation of the in vitro drug release behavior showed that the total drug released from the microparticle into the medium increased with mass concentration of the drug. In this study mouse fibroblast NIH 3T3 cells were cultivated on PHA microparticles, and results of using fluorescent DAPI DNA stain, and MTT assay showed that microparticles prepared from PHAs of different chemical compositions did not exhibit cytotoxicity to cells cultured on them and proved to be highly biocompatible. Cell attachment and proliferation on PHA microparticles were similar to those on polystyrene. The cytostatic drug encapsulated in P3HB/3HV microparticles has been proven to be effective against HeLa tumor cells. В© 2013 Springer Science+Business Media New York.

Аннотация: A set of polyhydroxyalkanoates are synthesized, and a comparative study of their physicochemical properties is performed. The molecular masses and polydispersities of polyhydroxyalkanoates are found to be independent of their chemical structures. It is shown that the temperature characteristics and degrees of crystallinity of polyhydroxyalkanoates are affected by the chemical compositions of the monomers and their quantitative contents in the polymers. The incorporation of 4-hydroxybutyrate, 3-hydroxyvalerate, and 3-hydroxyhexanoate units into the chain of poly(3-hydroxybutyrate) decreases its melting point and thermal degradation temperature relative to these parameters of a homogeneous poly(3-hydroxybutyrate) sample (175 В± 5 C and 275 В± 5 C, respectively). The higher the content of the second monomer units in the poly(3-hydroxybutirate) chain, the greater the changes. The degrees of crystallinity of polyhydroxyalkanoate copolymers are generally lower than that of poly(3-hydroxybutyrate) (75 В± 5%). The effect on the ratio of the amorphous and crystalline phases of the copolymer samples becomes more pronounced in the series 3-hydroxy-valerate-3-hydroxyhexanoate-4- hydroxybutyrate. Аннотация: The study addresses consequences of subcutaneous implantation of film matrices prepared from different PHAs to laboratory animals. No negative effects of subcutaneous implantation of PHA matrices on physiological and biochemical characteristics of the animals were determined. Independently of the matrices composition and duration of the contact with the internal environment of the organism we did not observe any deviations in the behavior of animals, their growth and development, as well as blood functions. Response of the tissues to PHA matrices was comparable with the response to polylactide, but substantially less expressed at the earlier time periods after implantation. Tissues response to implantation of PHA of all types is characterized by short-term (up to 2 weeks) post-traumatic inflammation with formation of fibrous capsules by 30th-60th days with the thickness less than 100 microns, which get thinner down to 40-60 microns by 180th day as the result of involution. No differences in response of tissues and the whole organism were observed for the matrices produced from the homopolymer of 3-hydroxybutyric acid (P3HB), copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids (P3HB/4HB), 3-hydroxybutyric acid and 3-hydroxyvalerianic acids (P3HB/3HV), 3-hydroxybutyric and 3-hydroxyhexanoate acids (P3HB/3HH). Macrophages and foreign-body giant cells actively participate in the response of the tissues to PHAs. In the studied conditions matrices from the copolymers containing 3-hydroxyhexanoate and 4 hydroxybutyrate were determined as more actively degraded PHA. The next less degraded matrices were matrices from the copolymer of P3HB/3HV and the most resistant were P3HB matrices. The slower degradation of PHA matrices was accompanied by delayed development of giantcells response. The studied PHA matrices can be placed in the following range by their degradation: P3HB/3HH - P3HB/4HB - P3HB/HV - P3HB.

Аннотация: The effect of 0.3 and 0.7 M NaCl on biomass yield, total nitrogen content, intracellular lipid content, and fatty acid profile of the lipids of the alga Botryococcus braunii IPPAS H-252 in different phases of the culture cycle was studied. The presence of sodium chloride in the medium inhibited the growth of algal cells for the first 3 days of the experiment, causing a decrease in total nitrogen, enhanced synthesis of triacylglycerols, and considerable changes in the lipid fatty acid profile: decreases in polyenoic acid contents (from 68.34% to 29.38% and 12.8%) and proportions of long-chain saturated acids (from 0.53% to 5.3% and 14.13% of the total fatty acids) at 0.3 M NaCl and 0.7 M NaCl, respectively. In later phases of the culture, at 0.3 M NaCl, the content of polyenoic acids rose to the values characteristic of the active growth phase of this alga. At 0.7 M NaCl, the proportion of polyenoic acids grew less significantly, but biomass concentration and total nitrogen increased, similarly to the experiment with 0.3 M NaCl. Аннотация: Biodegradability patterns of two PHAs: a polymer of 3-hydroxybutyric acid (3-PHB) and a copolymer of 3-hydroxybutyric and 3-hydroxyvaleric acids (3-PHB/3-PHV) containing 11 mol% of hydroxyvalerate, were studied in the tropical marine environment, in the South China Sea (Nha Trang, Vietnam). No significant differences have been observed between degradation rates of 3-PHB and 3-PHB/3-PHV specimens; it has been found that under study conditions, biodegradation is rather influenced by the shape of the polymer item and the preparation technique than by the chemical composition of the polymer. Biodegradation rates of polymer films in seawater have been found to be higher than those of compacted pellets. As 3-PHB and 3-PHB/3-PHV are degraded and the specimens lose their mass, molecular weight of both polymers is decreased, i.e. polymer chains get destroyed. The polydispersity index of the PHAs grows significantly. However, the degree of crystallinity of both PHAs remains unchanged, i.e. the amorphous phase and the crystalline one are equally disintegrated. PHA-degrading microorganisms were isolated using the clear-zone technique, by inoculating the isolates onto mineral agar that contained PHA as sole carbon source. Based on the 16S rRNA analysis, the PHA-degrading strains were identified as Enterobacter sp. (four strains), Bacillus sp. and Gracilibacillus sp.В© 2010 Elsevier Ltd. All rights reserved.

Аннотация: Class I polyhydroxyalkanoic acid (PHA) synthase gene (phaC) of Ralstonia eutropha strain B5786 was cloned and characterized. R. eutropha B5786 features the ability to synthesize multicomponent PHAs with short- and medium-chain-length monomers from simple carbohydrate substrate. A correlation was made between the molecular structure of PHA synthase and substrate specificity and the ability of strain-producers to accumulate PHAs of this or that structure. A strong similarity of PHA synthase of R. eutropha strain B5786 with PHA synthase of R. eutropha strain H16, which, as opposed to strain B5786, enables to incorporate medium chain length PHAs if hexanoate is used as carbon source, exhibited 99%. A correlation between the structure of PHA synthase of B5786 strain with synthases of microorganisms which synthesize short and medium chain length PHAs similarly to B5786 strain, showed an identity level from 26 to 41% (homology with synthase of Rhodospirillum rubrum makes 41%, Ectothiorhodospira shaposhnikovii makes 26%, Aeromonas punctata makes 40%, Thiococcus pfennigii makes 28%, Rhodococcus ruber makes 38%, and with PhaCl and PhaC2 synthases of Pseudomonas sp. 61-3 makes 34 and 37%, respectively). This allows for speaking about the absence of a direct connection between the molecular organization of PHA synthases and their functional abilities, namely, the ability to synthesize PHAs of a particular composition. Аннотация: The study addresses the effect of different conditions of carbon nutrition on synthesis of polyhydroxyalkanoates by the bacterium Wautersia eutropha. In experiments with two wild type strains (H16 and 65786), it has been first found that under mixotrophic growth conditions - Co2 + co-substrate (alkanoic acids) - bacteria can synthesize multi-component PHAs, consisting of short- and medium-chainlength monomers with carbon chains containing 4 to 8 atoms. It has been shown that PHA composition is determined by the type of the co-substrate. Fatty acids with odd number of carbons induce bacteria to synthesize four- and five-component PHAs with hydroxybutyrate, hydroxyvalerate as major monomers and hydroxyhexanoate, hydroxyheptanoate and hydroxyoctanoate as minor, occasionally occurring, ones. Fatty acids with even number of carbons induce synthesis of not only their respective monomers (hydroxyhexanoate and hydroxyoctanoate) but also hydroxyvalerate, making possible synthesis of four-component PHAs, containing hydroxybutyrate and hydroxyhexanoate as major components (up to 18 mol%). Аннотация: The effect of carbon supply on polyhydroxyalkanoate (PHA) synthesis by bacteria Wautersia eutropha was studied. Synthesis of multicomponent PHA composed of short-and long-chain monomers (C4-C8) by two natural strains (H16 and B5786) under mixotrophic conditions (CO2 + alkanoic acids as cosubstrates) was demonstrated for the first time. The PHA composition was shown to be dependent on the cosubstrate type. In the presence of odd fatty acids, four-and five-component polymers were synthesized; hydroxybutyrate, hydroxyvalerate, and hydroxyheptanoate were the major monomers, while hydroxyhexanoate and hydroxyoctanoate were minor. In the presence of even fatty acids, PHA contained not only the corresponding molecules (hydroxyhexanoate and hydroxyoctanoate), but also hydroxyvalerate; synthesis of four-component PHA which contain mainly hydroxybutyrate and hydroxyhexanoate (up to 18 mol %) is therefore possible. Аннотация: The fatty acid composition of the lipids of a Wautersia eutropha polyhydroxyalkanoate-producing strain was studied by chromato-mass spectrometry. A total of 27 fatty acids were identified; their distribution in the cell fractions was determined. In the cytoplasmic membrane, palmitic, palmitoleic, and cis-vaccenic acids were the major components. Long-chain ?-hydroxy acids and myristic acids (components of the lipopolysaccharides of the cell envelope) predominated in the fraction of strongly bound lipids. When the polymer was actively synthesized, the content of cyclopropane acids in the easily extracted lipids increased and the content of the corresponding monoenoic acids decreased. The strongly bound lipids had a high content of long-chain ?-hydroxy acids (more than 50% of the total fatty acids). These results made it possible to determine the source of polyhydroxyalkanoate (PHA) contamination and to choose the strategy for their purification. В© 2007 Pleiades Publishing, Ltd.

Аннотация: The paper deals with the study of the synthesis of 3-hydroxybutyrate (3HB) and 3-hydroxyvalerate (3HV) copolymers by the bacterium Ralstonia eutropha B-5786 grown under different carbon nutrition conditions (growth on carbon dioxide, fructose, and CO2-valerate and fructose-valerate mixtures). The parameters to be analyzed included the yield of biomass, the yield, synthesis rate, and composition of copolymers, the activity of the key enzymes of polyhydroxyalkanoate (PHA) synthesis (?-ketothiolase, acetoacetyl-CoA reductase, and PHA synthase), the maximum tolerable concentration of valerate to the bacterium, and the conditions that govern the incorporation of hydroxyvalerate to copolymers. This allowed the relationship between cultivation conditions and the proportion of monomers in the copolymers to be deduced. We were able to synthesize a range of 3HB/3HV copolymers and found that the thermal characteristics and the degree of crystallinity of these copolymers depend on the molar fraction of 3HV.

Аннотация: The effect of nitrogen limitation in a medium on the composition of intracellular lipids in the alga Botryococcus braunii Kutz IPPAS H-252 in the course of culture development was investigated. Under the conditions of nitrogen limitation, the alga under investigation accumulated lipids as triacylglycerols, and this process was accompanied by substantial changes in the total fatty acid (FA) composition, which were manifested in a decrease in trienoic acids (from 52.8-57.2 to 19.5-24.7% of total FAs) and an increase in the content of oleic (from 1.1-1.2 to 17.1-24.4%) and saturated (from 23.7-26.0 to 32.9-46.1%) acids. In the control culture, the directionality of FA redistribution was less marked, and these changes were noticed at the later stages of culture development. Under nitrogen limitation, marked changes in the FA composition of polar lipids occurred by the 13th day, and they were characterized by an increase in the content of saturated acids (up to 76.8%) and a dramatic decrease in the content of all polyenoic acids (up to 6.8%). The changes in the FA composition of triacylglycerols were noticed as early as by the 7th day; these changes consisted in an increase in the content of oleic acid, and its high content (28.4-38.4%) was maintained up to the end of culturing. In the control culture, triacylglycerols with a high content of oleic acid were found by the 13th day, although, by this time, the content of total lipids and triacylglycerols did not change. Аннотация: A study has been carried out to investigate the influence of nitrogen deficiency on intracellular lipid composition, including total fatty acid composition of lipids, polar lipids, and triacylglycerols, of the alga Botryococcus braunii Kutz IPPAS H-252 in batch culture. Under nitrogen limitation, the alga accumulates lipids as triacylglycerols and the total fatty acid (FA) composition changes: trienoic acids decrease (from 52.8-57.2 to 19.5-24.7% of the total FAs) and the oleic acid increases (from 1.1-1.2 to 17.1-24.4%) as does the saturated acids (from 23.7-26 to 32.9-46.1%). A similar rearrangement in the FA spectrum occurs at later times in the control culture, but it is less pronounced. Under nitrogen limitation, considerable changes in the polar lipid FAs are registered at day 13: saturated acids increase (from 28.6-35.5 to 76.8%) and all polyenoic acids markedly decrease (from 56.9-64.1 to 6.8%). Changes in the triacylglycerol fatty acid spectrum are seen on day 7: the oleic acid increases (from 14.7 to 34.2%) and remains at a high level till the end of the culture. In the control, triacylglycerols with large contents of oleic acid are detected at day 13, the total lipids and triacylglycerols still remaining unchanged. В© Springer 2005.
