Аннотация: To create a life support system based on biological and physical-chemical processes is the optimum solution providing full-valued condidtions for existence and efficient work of people at a lunar base. Long-standing experinece in experimental research or closed ecosystems and their components allows us to suggest a realistic functional structure of the lunar base and to estimate qualitatively its parameters. The original restrictions are as follows: 1) the basic source of energy to support the biological processes has to be the solar radiation; 2) the initial amount of basic biological elelments forming the turnover of substances (C, O, H, P, K, N) has to be delivered from Earth; 3). Moon materials are not to be used in the biological turnover inside the base; 4) the base is to supply the crew fully with atmosphere and water, and with 90% (A scenario) or 40% (B scenario) of food. Experimental data about the plant productivity under the "Moon" rhythm of light and darkness allow us to suggest that the A scenario requires per one human: plant area - 40 m2 irradiated during the lunar day by 250-300 W/m2 PAR producing 1250 g of dry biomass a terrestrial day; a heterotrophic component of "biological incineration" of inedible plant biomass (800 g/day) including the aquaculture of fish to produce animal products and contaminating the environment less than birds and mammals, and the culture of edible mushrooms; a component of physical-chemical correction for the LSS envi ronment including the subsystems of: deep oxidation of organic impurities in the atmosphere and of water, organic wastes of human activity and that biological components (420 g/day) Co2 concentration in "Moon" nights, damping O2 in "Moon" days, etc. The stock of presotred or delivered from Earth substances (food additions, seeds, etc.) to be involved in biological turnover is to be about 50 kg/year per man. Increase of the mass of prestored substances per man up to 220 kg/year would reduce twice the plant area and consumed amount of radiant energy to exclude the components of "biological incineration" and physical-chemical destruction of organic wastes. Аннотация: In the paper an attempt is made to combine three important criteria of LSS comparison: minimum mass, maximum safety and maximum quality of life. Well-known types of BLSS were considered: with higher plant, higher plants and mushrooms, microalgae, and hydrogen-oxidizing bacteria. These BLSSs were compared in terms of "integrated" mass for the case of a vegetarian diet and a "normal" one (with animal proteins and fats). It was shown that the BLSS with higher plants and incineration of wastes becomes the best when the exploitation period is more than 1 yr. The dependence of higher plants' LSS structure on operation time was found. Comparison of BLSSs in terms of integral reliability (this criterion includes mass and quality of life criteria) for a lunar base scenario showed that BLSSs with higher plants are advantageous in reliability and comfort. Аннотация: Material support of all manned space flights so far has been provided from a prestored stock of substances or replenished from the Earth's biosphere. Exploration of space will, however, become real only when man is able to break away from Earth completely, when he will be accompanied by a system providing everything necessary to sustain full-valued life for an unlimited time. The only known system to date meeting this requirement is the Earth's biosphere. To break away from his cradle, as K.E. Tsiolkovsky called Earth, it is necessary to devise a life support system functionally similar to the natural biosphere. This need not be similar in structure to the vast diversity of trophic relationships available on Earth, but requires the solution of a multitude of various problems of an ecological, physiological, engineering and social-psychological nature. Human life-support systems based on biological regeneration of environments in small volumes have been studied at the Institute of Biophysics (Siberian Branch of the Russian Academy of Sciences) over many years. This work has resulted in the design of Bios-3, a biologically-based self-sustained human life support system.

Аннотация: Waters of the Yenisei River, certain rivers and lakes of the Altai Territory, and effluents of some industrial factories in Krasnoyarsk were studied by luminous bacteria biotest Microbiosensor B17 677F. The lyophilized luminous bacteria Photobacterium phosphoreum from the IBSO collection were used to design this biotest. The bioluminescent test is based on bioluminescence quenching resulting from the action of water samples on luminous bacteria. The test results indicated locations and zones of impaired water quality. The heaviest pollution of water in the Yenisei River was recorded in the zones 0-116 km downstream from Krasnoyarsk (Krasnoyarsk and satellite towns). The effluents of most factories were found to be toxic. Underground and surface waters of some areas of the Altai Territory had different toxicity levels; there were deviations from the norm in most water samples taken from the different lakes and rivers. The data from this study show that the luminous bacteria biotest is simple and convenient, and that the results obtained are within acceptable levels of accuracy for the evaluation of the toxicity of effluent and river water. It takes no more than 30 min to do the biotest. It can be used in ecological monitoring like the Microtox toxicity test. В© 1998 John Wiley & Sons, Inc.

Аннотация: The biodegradable polymer poly(3-hydroxybutyrate-co-3-hydroxyvalerate) has been used to design experimental sustained-release formulations of the herbicide Zellek Super in the form of films and microgranules. The kinetics of polymer degradation and the dynamics of herbicide release show that the rate and extent of herbicide release from the polymer matrix into the soil depends on the geometry of the carrier and the proportion of the pesticide loaded into it (polymer/pesticide mass ratio). Experiments with the creeping bentgrass (Agrostis stolonifera L.) show that the formulations of the herbicide Zellek Super constructed as microgranules and films can be successfully used to suppress the growth of grasses. This study is the first to demonstrate that biodegradable polyhydroxyalkanoates can be used effectively to construct environmentally friendly sustained-release PHA-herbicide systems that can be placed into the soil together with seeds.

Аннотация: An "aluminum oxide film-adhesive layer-nanodiamond-luciferase" supramolecular structure is prepared on a flat plate. It is demonstrated that, in this structure, the enzyme retains the catalytic activity. The structure prepared can be treated as a luminescent biochip prototype for use in bioluminescent analysis. Аннотация: We have studied the effects of a gel-like environment on the characteristics of enzyme preparations based on the coupled enzyme system of luminous bacteria, NADH:FMN-oxidoreductase-luciferase, to design a stable immobilizing reagent for bioluminescent analysis. Natural polymers, gelatin and starch, were used to create a viscous, structured microenvironment. The stability of the coupled enzyme system to such physical and chemical environmental factors as temperature, pH, and ionic strength in gelatin and starch-containing media was examined. It was shown that both gelatin and starch have a stabilizing effect on the enzymes of luminous bacteria under specific conditions. In particular, the enzymes' activity is increased twofold in the presence of 1 and 5 % of gelatin at 20 °C and 25 °C, respectively (temperatures lower than the gel point). Also, the acceptable pH range of the coupled enzyme system expands into the alkaline region and becomes 6.8-8.1. Stabilization at low ionic strength (0.01-0.06 mol L-1) is observed. At the same time, microenvironments based on either gelatin or starch do not change the enzymes' thermal inactivation rate constants in the temperature range from 25 to 43 °C. Finally, gelatin and starch are suitable for development of a reagent for immobilization of enzymes which would be stable and resistant to physical and chemical environmental conditions. © 2014 Springer-Verlag Berlin Heidelberg.

Аннотация: Any attempt to create LSS for practical applications must take into account the possibility of catastrophic consequences if the problem of LSS reliability and stability is not solved. An integrated conception of CELSS studies development as a possible way to increase its reliability is considered. The BIOS-4 facility project is developed in the context of the conception. Three principles of highly effective experimental CELSS facility design are proposed. Some details of BIOS-4 design and its exploitation features are presented.

Аннотация: Scytalone dehydratase is a molecular target of inhibitor design efforts aimed at preventing the fungal disease caused by Magnaporthe grisea. A method for cocrystallization of enzyme with inhibitors at neutral pH has produced several crystal structures of enzyme-inhibitor complexes at resolutions ranging from 1.5 to 2.2 Angstrom Four high resolution structures of different enzyme-inhibitor complexes are described. In contrast to the original X-ray structure of the enzyme, the four new structures have well-defined electron density for the loop region comprising residues 115-119 and a different conformation between residues 154 and 160. The structure of the enzyme complex with an aminoquinazoline inhibitor showed that the inhibitor is in a position to form a hydrogen bond with the amide of the Asn131 side chain and with two water molecules in a fashion similar to the salicylamide inhibitor in the original structure, thus confirming design principles. The aminoquinazoline structure also allows for a more confident assignment of donors and accepters in the hydrogen bonding network, The structures of the enzyme complexes with two dichlorocyclopropane carboxamide inhibitors showed the two chlorine atoms nearly in plane with the amide side chain of Asn131. The positions of Phe53 and Phe158 are significantly altered in the new structures in comparison to the two structures obtained from crystals grown at acidic pH, The multiple structures help define the mobility of active site amino acids critical for catalysis and inhibitor binding. Аннотация: To develop and design Life Support Systems it is necessary to evaluate their reliability. However direct experiments take much time, are very expensive, and therefore are practically impossible. Promising way is to use approximate estimates of reliability, which need essentially fewer amounts of experimental data. Two types of estimates of Life Support System reliability - additive and multiplicative ones are considered in the paper. Additive estimate is based on the assumption that total system failure probability is low and therefore it can be considered as the sum of failure probability of separate units. Additive approach allows obtaining near lower-bounded estimate of failure probability. Multiplicative estimate allows evaluating the possibility of system catastrophe due to simultaneous effect of several factors when each of them separately is not dangerous. Evaluation shows that the possible error of reliability forecast increases with the increasing of number of external factors faster than exponential function. An illustration of the ecological similarity approach as promising tool for providing estimation of full-scale system reliability by means the set of small similar experimental models. Аннотация: This paper is devoted to the consideration of possible viewpoint on CELSS development and design. If the aim to create practically applicable CELSS is accepted then the task to optimize the process of CELSS research and development in terms of minimum cost, hours, maximum applicability, scientific contribution, etc. becomes actual. Requirements of applicability and scientific significance are synergetic since understanding of general properties of CELSS gives an ability to create CELSS for different applications. To accomplish the task three main groups of parameters have to be optimized: i) configuration and operating parameters of developing CELSS itself, ii) organizational management of research and development of CELSS; iii) features of an area where CELSS is planned to be used (space missions, terrestrial applications, or biosphere investigation) and where requirements to CELSS characteristic come from. Given paper is a brief review presented some attempts to arrange mentioned above into some set of formalized and interacting criteria, and some progression of research stages derived from these criteria. Аннотация: This study concerns the formation of structured communities by monocultures and binary associations of Pseudomonas fluorescens transgenic strains and natural heterotrophic bacterial species in naphthalene-containing media with various osmotic pressures. It was shown that cells of P. fluorescens strain 5RL, harboring a recombinant construct in the chromosome, were more resistant to the combined action of the stress factors under study than P. fluorescens 82/pUTK21, harboring a recombinant construct within a plasmid. Natural P. fluorescens 1 strains, particularly Vibrio sp. 14, were more viable at high osmotic pressures and naphthalene concentrations. Experiments with the combined introduction of transgenic and natural bacterial strains at high osmotic pressures demonstrated the stable coexistence of bacterial associations in biofilms, independent of naphthalene concentration. Strains considered for introduction into the environment for bioremediation should be assessed with regard to their susceptibility to the combined effect of anthropogenic and natural stress factors. The design of bacterial associations for the same purpose should take into account the effect of factors important for their survival in polluted areas.

Аннотация: This review examines the general principles of bioluminescent enzymatic toxicity bioassays and describes the applications of these methods and the implementation in commercial biosensors. Bioluminescent enzyme system technology (BEST) has been proposed in the bacterial coupled enzyme system, wherein NADH: FMN-oxidoreductase-luciferase substitutes for living organisms. BEST was introduced to facilitate and accelerate the development of cost-competitive enzymatic systems for use in biosensors for medical, environmental, and industrial applications. For widespread use of BEST, the multicomponent reagent "Enzymolum'' has been developed, which contains the bacterial luciferase, NADH: FMN-oxidoreductase, and their substrates, co-immobilized in starch or gelatin gel. Enzymolum is the central part of Portable Laboratory for Toxicity Detection (PLTD), which consists of a biodetector module, a sampling module, a sample preparation module, and a reagent module. PLTD instantly signals chemical-biological hazards and allows us to detect a wide range of toxic substances. Enzymolum can be integrated as a biological module into the portable biodetector-biosensor originally constructed for personal use. Based on the example of Enzymolum and the algorithm for creating new enzyme biotests with tailored characteristics, a new approach was demonstrated in biotechnological design and construction. The examples of biotechnological design of various bioluminescent methods for ecological monitoring were provided. Possible applications of enzyme bioassays are seen in the examples for medical diagnostics, assessment of the effect of physical load on sportsmen, analysis of food additives, and in practical courses for higher educational institutions and schools. The advantages of enzymatic assays are their rapidity (the period of time required does not exceed 3-5 min), high sensitivity, simplicity and safety of procedure, and possibility of automation of ecological monitoring; the required luminometer is easily available.

Аннотация: CO2 laser radiation was used to process poly(3-hydroxybutyrate) constructs - films and 3D pressed plates. Laser processing increased the biocompatibility of unperforated films treated with moderate uniform radiation, as estimated by the number and degree of adhesion of NIH 3T3 mouse fibroblast cells. The biocompatibility of perforated films modified in the pulsed mode did not change significantly. At the same time, pulsed laser processing of the 3D plates produced perforated scaffolds with improved mechanical properties and high biocompatibility with bone marrow-derived multipotent, mesenchymal stem cells, which show great promise for bone regeneration. © 2015 Taylor & Francis.
