Аннотация: A set of polyhydroxyalkanoates are synthesized, and a comparative study of their physicochemical properties is performed. The molecular masses and polydispersities of polyhydroxyalkanoates are found to be independent of their chemical structures. It is shown that the temperature characteristics and degrees of crystallinity of polyhydroxyalkanoates are affected by the chemical compositions of the monomers and their quantitative contents in the polymers. The incorporation of 4-hydroxybutyrate, 3-hydroxyvalerate, and 3-hydroxyhexanoate units into the chain of poly(3-hydroxybutyrate) decreases its melting point and thermal degradation temperature relative to these parameters of a homogeneous poly(3-hydroxybutyrate) sample (175 В± 5 C and 275 В± 5 C, respectively). The higher the content of the second monomer units in the poly(3-hydroxybutirate) chain, the greater the changes. The degrees of crystallinity of polyhydroxyalkanoate copolymers are generally lower than that of poly(3-hydroxybutyrate) (75 В± 5%). The effect on the ratio of the amorphous and crystalline phases of the copolymer samples becomes more pronounced in the series 3-hydroxy-valerate-3-hydroxyhexanoate-4- hydroxybutyrate. Аннотация: The study addresses consequences of subcutaneous implantation of film matrices prepared from different PHAs to laboratory animals. No negative effects of subcutaneous implantation of PHA matrices on physiological and biochemical characteristics of the animals were determined. Independently of the matrices composition and duration of the contact with the internal environment of the organism we did not observe any deviations in the behavior of animals, their growth and development, as well as blood functions. Response of the tissues to PHA matrices was comparable with the response to polylactide, but substantially less expressed at the earlier time periods after implantation. Tissues response to implantation of PHA of all types is characterized by short-term (up to 2 weeks) post-traumatic inflammation with formation of fibrous capsules by 30th-60th days with the thickness less than 100 microns, which get thinner down to 40-60 microns by 180th day as the result of involution. No differences in response of tissues and the whole organism were observed for the matrices produced from the homopolymer of 3-hydroxybutyric acid (P3HB), copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids (P3HB/4HB), 3-hydroxybutyric acid and 3-hydroxyvalerianic acids (P3HB/3HV), 3-hydroxybutyric and 3-hydroxyhexanoate acids (P3HB/3HH). Macrophages and foreign-body giant cells actively participate in the response of the tissues to PHAs. In the studied conditions matrices from the copolymers containing 3-hydroxyhexanoate and 4 hydroxybutyrate were determined as more actively degraded PHA. The next less degraded matrices were matrices from the copolymer of P3HB/3HV and the most resistant were P3HB matrices. The slower degradation of PHA matrices was accompanied by delayed development of giantcells response. The studied PHA matrices can be placed in the following range by their degradation: P3HB/3HH - P3HB/4HB - P3HB/HV - P3HB.

Аннотация: The authors have constructed and characterized a series of membranes based on resorbable polyhydroxyalkanoates of different compositions. Five PHA types have been studied: a homopolymer of 3-hydroxybutyric acid, copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids, 3-hydroxybutyric and 3-hydroxyvaleric acids, 3-hydroxybutyric and 3-hydroxyhexanoic acids. Scanning electron microscopy and atomicforce microscopy were used to examine the microstructure of membrane surfaces, showing that membranes based on the copolymer of 3-hydroxybutyrate and 3-hydroxyhexanoate had the roughest surface, while membranes based on the copolymer of 3-hydroxybutyrate and 3-hydroxyvalerate had the smoothest surface. The contact angle for water in air was smaller and hydrophilic properties better in the copolymer membranes than in the membranes based on the high-crystallinity homopolymer of 3-hydroxybutyric acid. The culture of mouse fibroblast cell line NIH 3,3 was used to test PHAbased membranes; results of fluorescent probes of DNA DAPI and the MTT assay show that membranes based on studied PHAs are not cytotoxic on direct contact with cells and are highly biocompatible; their adhesive properties and ability to maintain fibroblast proliferation are similar to those of polystyrene and better than those of polylactic acid membranes.

Аннотация: The study addresses the effect of different conditions of carbon nutrition on synthesis of polyhydroxyalkanoates by the bacterium Wautersia eutropha. In experiments with two wild type strains (H16 and 65786), it has been first found that under mixotrophic growth conditions - Co2 + co-substrate (alkanoic acids) - bacteria can synthesize multi-component PHAs, consisting of short- and medium-chainlength monomers with carbon chains containing 4 to 8 atoms. It has been shown that PHA composition is determined by the type of the co-substrate. Fatty acids with odd number of carbons induce bacteria to synthesize four- and five-component PHAs with hydroxybutyrate, hydroxyvalerate as major monomers and hydroxyhexanoate, hydroxyheptanoate and hydroxyoctanoate as minor, occasionally occurring, ones. Fatty acids with even number of carbons induce synthesis of not only their respective monomers (hydroxyhexanoate and hydroxyoctanoate) but also hydroxyvalerate, making possible synthesis of four-component PHAs, containing hydroxybutyrate and hydroxyhexanoate as major components (up to 18 mol%). Аннотация: During the summers of 1999-2001 the dynamics of polyhydroxyalkanoate degradation in a small recreational eutrophic reservoir was studied experimentally. It has been shown that biodegradation of polyhydroxyalkanoates in the environment is determined by the structure and physicochemical properties of the polymer and by local weather conditions, which influence the state of the aquatic ecosystem. Species (clones) of bacteria able to utilize polyhydroxyalkanoates in the reservoir were identified using molecular phylogenetic analysis of 16S rRNA genes. В© 2007 Elsevier Ltd. Аннотация: Physiological-biochemical, genetic, and cultural properties of the glucose-utilizing mutant strain Ralstonia eutropha B8562 have been compared with those of its parent strain R. eutropha B5786. It has been shown that growth characteristics of the strain cultured on glucose as the sole carbon and energy source are comparable with those of the parent strain. Strain B8562 is characterized by high polyhydroxyalkanoate (PHA) yields on different carbon sources (CO2, fructose, and glucose). PHA accumulation in the strain batch cultured on glucose under nitrogen deficiency reaches 90 %. The major monomer in the PHA is ?-hydroxybutyric acid (more than 99 mol %); the identified minor components are ?-hydroxyvaleric acid (0.25-0.72 mol %) and ?-hydroxyhexanoic acid (0.08-1.5 mol %). The strain is a promising PHA producer on available sugar-containing media with glucose. В© Springer-Verlag 2006.

Аннотация: Samples of a hybrid composite of polyhydroxybutyrate (PHB), a biodegradable polyester, and hydroxyapatite (HA), with different PHB/HA ratios, have been prepared using mechanical-physical method. Electron microscopy, X-ray structure analysis and differential thermal analysis have been used to investigate the structure and physicochemical properties of the composite, depending on the PHB/HA ratio. The properties of the surface of the HA-loaded composite are significantly different from those of the pure polymer. As the HA percentage in the composite increases, free interface energy, the cohesive force, i.e., the strength of the adhesive bond between the composite surface and the water phase, and surface wettability increase. The HA percentage of the composite does not influence its melting temperature, but affects the temperature for the onset of decomposition: as the HA content increases from 0 to 10% (w/w), Td decreases from 260В°C to 225В°C. The degree of crystallinity of PHB/HA increases from 77% to 89% with an increase in the HA fraction from 10% to 50%. Functional properties of the composites have been investigated in vitro and in vivo. The best parameters of growth and differentiation of murine marrow osteoblasts are registered on PHB/HA samples containing 10% and 20% HA. In ectopic bone formation assay it has been proven that the hybrid PHB/HA composites can function as scaffolds and that bone tissue develops on their surface and in pores. В© VSP 2006.

Аннотация: The paper presents the results of biomedical investigations of sutures made of polyhydroxyalkanoates (PHAs) of two types (poly-3-hydroxybutyrate (PHB) and poly-3-hydroxybutyrate-co-poly-3-hydroxyvalerate (PHB/PHV)), synthesized by the bacteria Ralstonia eutropha B 5786 in the experiment on test animals in vivo, in comparison with silk and catgut sutures. PHB and PHB/PHV (PHBV) implants produced no adverse effect on physiological, biochemical and functional parameters of the animals during the post-surgery period. The tested PHA sutures featured the necessary strength throughout the healing period of the muscle-fascial cuts. The reaction of tissues to the implantation of PHB and PHB/PHV fibers fitted into the usual scheme characteristic of the wound process and of the reaction to a foreign-body invasion. This reaction and the reaction of tissues to silk had the similar nature and period of inflammation, but it was much less pronounced than the reaction to catgut. The tissue response to the implantation of PHAs consisted in a short-duration (up to 2 weeks) post-traumatic inflammation and the formation of a fibrous capsule less than 200 ?m thick during weeks 4-8, which in 4-6 months was reduced to 40-60 ?m in the course of reverse development. There were no adverse changes, such as suppurative inflammation, necrosis, calcification, and malignization of the cicatrice, at the site of implantation of PHA filaments, unlike in the cases with silk and catgut. In the case of PHA implantation there was a typical prolonged (throughout the post-surgery monitoring period) pronounced macrophagal stage with a large number of macrophages present. The macrophages were of the phagocytic type and multinucleate giant foreign cells, with a high activity of acid phosphomonoesterase that correlated with the activity of the enzyme in blood. Аннотация: The time series of various parameters of satellite imagery (NDVI/EVI, temperature) during the growing season were considered in this work. This means that satellite images were considered not like a number of single scenes but like temporal sequences. Using time series enables estimating the integral phenological properties of vegetation. The basis of the developed technique is to use one of the methods of transformation of the multidimensional space in order to get the principal components. The technique is based on considering each dimension of the multidimensional space as satellite imagery for a specific date range. The technique automatically identifies spatial patterns of vegetation that are similar by phenology and growing conditions. Subsequent analysis allowed identification of the belonging of derived classes. Thus, the technique of revealing the spatial distribution of different dynamical vegetation patterns based on the phenological characteristics has been developed. The technique is based on a transformation of the multidimensional space of states of vegetation. Based on the developed technique, areas were obtained with similar interannual trends. В© 2011 SPIE.

Аннотация: Continuous monitoring of phytopigment concentrations and sea surface temperature in the ocean by space-borne methods makes possible to estimate ecological condition of biocenoses in critical areas. In the papers of the authors (Shevyrnogov A.P., Vysotskaya G.S., Gitelzon J.I. 1996) existence of zones, which are quasi-stationary with similar seasonal dynamics of chlorophyll concentration at surface layer of ocean, was shown. Results were obtained on the base of processing of time series of satellite images SeaWiFS. It was shown that fronts and frontal zones coincide with dividing lines between quasi-stationary areas, especially in areas of large oceanic streams. The usage of the seasonal dynamics gives a possibility to circumvent influence of high-frequency component in investigation of dynamics of spatial distribution of surface streams. In addition, an analyses of unstable ocean productivity phenomena, stood out time series of satellite images, showed existence of areas with different types of instability in the all Global ocean. They are observed as adjacent nonstationary zones of different size, which are associated by different ways with known oceanic phenomena. It is evident that dynamics of a spatial distribution of biological productivity and sea surface temperature can give an additional knowledge of complicated picture of surface oceanic layer hydrology. Аннотация: The new data on anomalies in seasonal dynamics of chlorophyll concentration in the ocean have altered the approach to the analysis of long-term trends of phytopigments in the surface layer of the ocean. Zones have been found in which at various times there was a many-fold increase in chlorophyll concentration versus its usual level. These changes are local in time and space. If anomalies in the long-term tendencies in the change of phytopigment surface distribution in the ocean are ignored in the analysis, the resulting picture will inadequately reflect the real processes. The work presents the results of the analysis of long-term trends of chlorophyll concentration on the basis of the CZCS satellite data in the Atlantic Ocean. The results are given as maps representing different variants of trends. The data obtained can be used to analyze the consequences of long-term changes of climate and changes in the interaction between the atmosphere and the ocean. В©2002 COSPAR. Аннотация: The work presents long-standing CZCS-satellite-based data about spatial distribution of anomalies in chlorophyll concentration and temperature of the ocean. An anomaly criterion is proposed. Maps of Atlantic ocean areas with elevated probability of anomalies are given. Properties of anomalous zones in different parts of the Atlantic ocean are discussed. Satellite-based maps of anomaly coefficients for chlorophyll concentration and temperature in the Atlantic ocean have been shown to feature substantial differences, the zones of elevated anomaly of chlorophyll concentration and temperature - to be close only in the vicinity of the Amazon outflow into the Atlantic ocean. Along with fundamental knowledge about dynamics of biological phenomena in the ocean the methods described may be used to reveal deviations due to anthropogenic impact. The methods presented may be used to process SeaWiFS prograna data. The data derived may be used to analyze long-standing time series to estimate randomness of cunent changes. В©2002 COSPAR. Аннотация: One of the ways of solving the problem of the human liquid wastes utilization in bioregenerative life support systems (BLSS) can be the use of halophytic vegetable plant Salicornia europaea capable of accumulating sodium chloride in rather high concentrations. Since the most specific higher plant function in BLSS, which at present cannot be substituted by physicochemical processes, appears to be the biosynthesis of a wide spectrum of nutritive substances necessary for a human, the object of the given work was the investigation of the S. europaea productivity, biochemical and mineral composition when grown under close to optimal BLSS vegetative component conditions. As the use of human urine after its preliminary physicochemical processing is supposed to be the mineral solution basis for the S. europaea cultivation, it is necessary to clear up the effect of reduced nitrogen on plants growth. Ground research was carried out. Biochemical composition of the S. europaea edible part showed that crude protein was contained in the highest degree. At that the content of crude protein (24% per dry weight) and cellulose (4.7% per dry weight) was higher in the plants grown on solutions containing amide nitrogen in comparison with the plants grown on solutions with nitrate nitrogen (15.4%-3.1% correspondingly). The water-soluble sugar contents were not high in the S. europaea edible part and depending on the nitrogen nutrition form they amounted to 1.1% (amide nitrogen) and 1.5% (nitrate nitrogen). The polysaccharide number (except cellulose) was rather higher and varied from 7.7% to 8.2%. Although the lipid content in the S. europaea plants was relatively low (7% per dry weight), it was shown that the plant lipids are characterized by a high nonsaturation degree mainly due to alpha linolenic and linoleic acids. Nitrogen nutrition form did not significantly affect the S. europaea productivity, and dry edible biomass of one plant was 8.6 g. Sodium and its concentrations predominated in the plant mineral composition and amounted in average to 9% per dry weight. Thus the S. europaea being the vegetable plant it can be the source of several biochemical substances and essential fatty acids. The present work also considers the influence of nitrate and amide forms of nitrogen on S. europaea biochemical and mineral composition. В© 2008 Elsevier Ltd. All rights reserved.

Аннотация: The purpose of this study is to estimate the possible effect produced on plant productivity by inedible plant biomass added to soil-like substrate (SLS). Results of the experiments with radish plants grown on the SLS with inedible biomass of carrot and beet plants added in the amounts roughly equal to their yields harvested from the same area showed a significant reduction in productivity of radish plants. The yield of radish plants grown on the SLS with added radish tops was almost the same as the yield of the radish grown on the neutral substrate. Experiments with addition of dry wheat straw to the SLS and growing of wheat and radish plants on that substrate also showed that the productivity of the plants grown in that way was decreased. Attempts to negate the adverse effect of plant wastes proved that the most effective way was to mineralize the wastes using the technique of "wet incineration". В© 2008 Elsevier Ltd. Аннотация: For systematic human Mars exploration, meeting crew safety requirements, it seems perspective to assemble into a spacecraft: an electrical rocket, a well-shielded long-term life support system, and a manipulator-robots operating in combined "presence effect" and "master-slave" mode. The electrical spacecraft would carry humans to the orbit of Mars, providing short distance (and low signal time delay) between operator and robot-manipulators, which are landed on the surface of the planet. Long-term hybrid biological and physical/chemical LSS could provide environment supporting human health and well being. Robot-manipulators operating in "presence effect" and "master-slave" mode exclude necessity of human landing on Martian surface decreasing the level of risk for crew. Since crewmen would not have direct contact with the Martian environment then the problem of mutual biological protection is essentially reduced. Lightweight robot-manipulators, without heavy life support systems and without the necessity of returning to the mother vessel, could be sent as scouts to different places on the planet surface, scanning the most interesting for exobiological research site. Some approximate estimations of electric spacecraft, long-term hybrid LSS, radiation protection and mission parameters are conducted and discussed. В© 2002 COSPAR. Published by Elsevier Science Ltd. Аннотация: Material support of all manned space flights so far has been provided from a prestored stock of substances or replenished from the Earth's biosphere. Exploration of space will, however, become real only when man is able to break away from Earth completely, when he will be accompanied by a system providing everything necessary to sustain full-valued life for an unlimited time. The only known system to date meeting this requirement is the Earth's biosphere. To break away from his cradle, as K.E. Tsiolkovsky called Earth, it is necessary to devise a life support system functionally similar to the natural biosphere. This need not be similar in structure to the vast diversity of trophic relationships available on Earth, but requires the solution of a multitude of various problems of an ecological, physiological, engineering and social-psychological nature. Human life-support systems based on biological regeneration of environments in small volumes have been studied at the Institute of Biophysics (Siberian Branch of the Russian Academy of Sciences) over many years. This work has resulted in the design of Bios-3, a biologically-based self-sustained human life support system.

Аннотация: Genes encoding luminescence of Photobacterium leiognathi have been cloned in Escherichia coli. The luminescent clones were readily apparent. Among them, a clone containing a recombinant plasmid with a 13.5-kb insertion was identified. This DNA fragment contained all of the luminescence-encoding genes. The luciferase-encoding genes (lux) in this DNA fragment were localized. We have sequenced a part of the cloned lux region and identified the luxA, luxB and luxG genes encoding the ? and ? subunits of luciferase and a ? protein with an Mr of 26 180, respectively. The analysis of deduced amino acid sequences and comparison with known luciferase sequences from Vibrio harveyi, indicate the common origin of these proteins. В© 1990.

Аннотация: The copy number of R plasmids weakly depends on the selective pressure of the respective antibiotic but does depend on the physiology of the host species and the type of plasmids and cloned genes, whose expression leads to a further load on the biosynthetic apparatus of cells. The last factor is critical in the maintenance of recombinant plasmids in transgenic microorganisms.
