Аннотация: Kinetic parameters of a culture of the hydrogen-oxidizing bacterium Ralstonia eutropha grown on a gas substrate under conditions favoring autotrophic biosynthesis of polyhydroxybutyrate were studied. The following parameters, making it possible to control and optimize the process in industrial situations, were determined: specific rate of substrate consumption, physical properties of the culture medium, and coefficients of heat emission and mass transfer.

Аннотация: A structural approach to studying the regularities of the population dynamics of unstable recombinant bacterial strains in a chemostat was elaborated. The approach is based on the mathematical modeling of cell distribution in a population with different numbers of plasmid copies. The effect of decreased selective preference of plasmidless variants of the recombinant strain in the chemostat, which is related to a decrease in the number of plasmid copies in cells upon long-term incubation was analyzed. It is shown that the time of half-elimination of plasmids from the bacterial population in the steady state in the chemostat T1/2 does not depend on the maximum number of plasmid copies in cells N but is determined only by the mean time of generation g and the probability of the loss of one plasmid copy tau. The dependence of the preference of bacterial plasmidless variants on the efficiency of expression of genes cloned into plasmids in chemostat was analyzed using the recombinant strain E. coli Z905, whose plasmids pPHL-7 contain cloned genes for the luminescence system of marine luminescing bacteria Photobacterium leiognathi.

Аннотация: Experimental and theoretical models of closed 'autotroph-heteretroph' (chlorella-yeast, chlorella- protozoa) ecosystems with spatially separated components have been created and studied. The chart of flows and interaction of components of gas-closed 'chlorella-yeast' system have formed the basis describe mathematically the functioning of the given system, experimental results have been found to agree with computer solution of the problem in terms of quality. Investigation of the experimental model of the 'producer-consumer' trophic chain demonstrated the role of protozoa in nitrogen turnover. 'Production-decomposition' and 'production-grazing-decomposition' cycle models has been theoretically analyzed and compared. The predator has been shown to be a more intensive mineralizer than the reducer component.

Аннотация: The CO oxidase (COX) activity of carboxydobacteria Seliberia carboxydohydrogena cells and the rate of CO oxidation by this culture were studied under various modes of fermentation. The CO oxidase activity of cells was maximal (60-80 U) during limitation of bacterial growth by the source of energy or carbon. The rate of CO oxidation by the culture was 1.4-1.71/g. The adaptive type of dependence of the CO oxidase activity of cells on the concentration of CO in the medium within the range of 0-20% was established.

Аннотация: The present study addresses the synthesis and properties of polyhydroxyalkanoates (PHA) of different composition synthesized by Cupriavidus eutrophus B-10646 using glycerol as a carbon substrate. Poly(3-hydroxybutyrate) [P(3HB)] was effectively synthesized in fed-batch culture in a 30-L fermenter on glycerol of various purification degrees, with 99.5, 99.7, and 82.1% content of the main component. Purified glycerol (99.7%) was used for 150-L pilot scale fermentation. The total biomass and P(3HB) concentration reached 110 and 85.8 g/L, respectively, after 45 h of fed-batch fermentation. An average volumetric productivity of P(3HB) was 1.83 g/(L h). The degree of crystallinity and molecular weight of P(3HB) synthesized on glycerol were lower than and temperature characteristics were the same as those of P(3HB) synthesized on sugars. © 2018, Springer-Verlag GmbH Germany, part of Springer Nature.
