Аннотация: The study addresses the ability of luminous bacteria of different taxa (Photobacterium leiognathi, Photobacterium phosphoreum, Vibrio harveyi, Vibrio fischeri) to synthesize polyesters of hydrocarbon acids (polyhydroxyalkanoates, PHAs) as storage macromolecules. The screened strains widely varied in their PHA productivity. Conditions for attaining high polymer yields (including two- and three-component polymers) in batch culture have been determined. The attained polymer yields reached 40-70% of dry cell biomass. The results suggest a conclusion that luminous microorganisms can be considered as producers of multi-component PHAs. Аннотация: The structure of liquid hydrocarbons and fatty acids produced by the green alga Botryococcus was identified. Two representatives of this alga, Botryococcus braunii Kutz, strain IPPAS H-252, introduced into culture earlier and an organism isolated for the first time from the Shira Lake, were used for this identification. Fatty acid composition of B. braunii, strain H-252, lipids was characterized by a high content of trienoic acids of C16-C18 series. The hydrocarbon composition of this strain was represented by straight-chain and branched-chain C14-C28 components; long-chain linear aliphatic C20-C27 hydrocarbons (54.4%) and 2,6,10,14-tetramethylhexadecane (20.5%) predominated among them. The strain H-252 differed in its fatty acid and hydrocarbon composition from the strains described earlier as Botryococcus braunii. The fatty acid composition of the Botryococcus isolate was represented mainly by C12-C32 saturated and monoenoic acids. The hydrocarbons formed by this isolate were represented by dienoic and trienoic components. C29 (48.9-56.3%) and C31 (11.1-16.3%) hydrocarbons predominated among the C23-C31 dienoic hydrocarbons, and C27, C29, and C31 trienoic hydrocarbons comprised 2.5-2.6% of total hydrocarbons. This type of hydrocarbons and the lipid fatty acid composition were characteristic for the race A of B. braunii.

Аннотация: Lipid composition and hydrocarbon structure of two colonial green algae of the genus Botryococcus, i.e., a mu seum strain and a field sample collected for the first time from Lake Shira (Khakasia, Siberia), have been compared. Polar lipids, diacylglycerols, alcohols, triacylglycerols, sterols, sterol esters, free fatty acids and hydrocarbons have been identified among lipids in the laboratory culture. The dominant fraction in the museum strain was formed by polar lipids (up to 50% of the lipids) made up of fatty acids from C12 to C24. Palmitic, oleic, C16 - C18 dienoic and trienoic acids were the main fatty acids of the museum strain. Aliphatic hydrocarbons were found in the lipid of the museum strain. However, these amounted maximally to about 1% of the dry biomass at the end of exponential growth phase. The qualitative and quantitative compositions of FAs and hydrocarbons of the museum strain of Botryococcus, (registered at the Cambridge collection as Botryococcus braunii Kutz No LB 807/1 Droop 1950 H-252) differed from those of the Botryococcus strain described in the literature as Botryococcus braunii. The Botryococcus sp. found in Lake Shira is characterized by a higher lipid content (< 40% of the dry weight). Polar lipids, sterols, triacylglycerols, free fatty acids and hydrocarbons have been identified among lipids in the field sample. The main lipids in this sample were dienes and trienes (hydrocarbons < 60% of total lipid). Monounsaturated and very long chain monounsaturated fatty acids, including C28:1 and C32:1 acids, were identified in the Botryococcus found in Lake Shira. The chemo-taxonomic criteria allow us to unequivocally characterize the organism collected from Lake Shira as Botryococcus braunii race A.

Аннотация: The lipid fraction of the green alga Botryococcus cultured in a batch mode was found to contain polar lipids (more than 50% of the total lipids), di- and triacylglycerols, sterols and their esters, free fatty acids, and hydrocarbons. In aging culture, the content of polar lipids somewhat decreased and that of triacylglycerols increased by more than four times. The content of hydrocarbons in the algal biomass did not exceed 0.9% and depended little on the culture age. Intracellular lipids contained saturated and unsaturated (mono-, di-, and trienoic) fatty acids. The maximum content of C16:3 and ?-C18:3 fatty acids (up to 35% of the total fatty acids) was detected in the phase of active growth. Аннотация: The effect of elevated temperatures of 35 and 45В°C (at the intensities of photosynthetically active radiation 322, 690 and 1104 ?mol-m-2-s-1) on the photosynthesis, respiration, and qualitative and quantitative composition of the volatiles emitted by wheat (Triticum aestuvi L., cultivar 232) crops was investigated in growth chambers. Identification and quantification of more than 20 volatile compounds (terpenoids - ?-pinene, ?3 carene, limonene, benzene, ?-and transcaryophyllene, ?- and ?-terpinene, their derivatives, aromatic hydrocarbons, etc.) were conducted by gas chromatograph/mass spectrometry. Under light intensity of 1104 ?mol-m-2-s-1, heat resistance of photosynthesis and respiration increased at 35В°C and decreased at 45В°C. The action of elevated temperatures brought about variations in the rate and direction of the synthesis of volatile metabolites. The emission of volatile compounds was the greatest under a reduced irradiation of 322 ?mol-m-2-s-1 and the smallest under 1104 ?mol-m-2-s-1 at 35В°C. During the repair period, the contents and proportions of volatile compounds were different from their initial values, too. The degree of disruption and the following recovery of the functional state depended on the light intensity during the exposure to elevated temperatures. The investigation of the atmosphere of the growth chamber without plants has revaled the substances that were definitely technogenic in origin: tetramethylurea, dimethylsulfide, dibutylsulfide, dibutylphthalate, and a number of components of furan and silane nature. Аннотация: The growth, productivity, and changes in the chemical composition of the green alga Botryococcus braunii Kutz H-252 were studied at different phases of growth in a luminostat culture. Modification of the Prat medium resulted in an increase in the alga yield to 3.9 g/l and a decrease in the generation time to 3-4 days. The specific growth rate during the exponential phase was 0.235/ day. These values are comparable with the best results obtained abroad. Among the pigments of B. braunii, besides chlorophylls a and b, carotene, lutein, neoxanthin, and violaxanthin were also identified. The highest content of these pigments, based on dry weight, was achieved during the exponential growth phase. During culture aging, there was an accumulation of carbohydrates and lipids in addition to a simultaneous decrease in the concentration of nitrogen-containing substances. It was discovered that this strain can synthesize liquid hydrocarbons. These hydrocarbons were found both in the alga biomass (ca. 1% of dry weight) and among the extracellular metabolites (up to 10.3 mg/l).

Аннотация: This study concerns the formation of structured communities by monocultures and binary associations of Pseudomonas fluorescens transgenic strains and natural heterotrophic bacterial species in naphthalene-containing media with various osmotic pressures. It was shown that cells of P. fluorescens strain 5RL, harboring a recombinant construct in the chromosome, were more resistant to the combined action of the stress factors under study than P. fluorescens 82/pUTK21, harboring a recombinant construct within a plasmid. Natural P. fluorescens 1 strains, particularly Vibrio sp. 14, were more viable at high osmotic pressures and naphthalene concentrations. Experiments with the combined introduction of transgenic and natural bacterial strains at high osmotic pressures demonstrated the stable coexistence of bacterial associations in biofilms, independent of naphthalene concentration. Strains considered for introduction into the environment for bioremediation should be assessed with regard to their susceptibility to the combined effect of anthropogenic and natural stress factors. The design of bacterial associations for the same purpose should take into account the effect of factors important for their survival in polluted areas.

Аннотация: The analysis of the literature on the methods of soluble organosolv lignins isolation, their physical-chemical study and on the method of their processing to porous aerogels and liquid hydrocarbons was carried out. A review of the literature allowed us to choice of the most important areas of research. For isolation from wood the soluble lignins free from sulfur the methods of catalytic peroxide delignification at mild conditions (temperature <= 100 degrees C, atmospheric pressure) and methods of lignin extraction by supercritical organic solvents were used. Molecular mass and molecular-mass distribution of ethanol-lignin samples isolated from aspenwood and abies-wood were studied by gel-permeation chromatography. Weighted molecular mass of ethanol-lignin from abies wood is 478 Da and that from aspen wood ethanol-lignin - 750 Da. Thus, the studied samples of ethanol-lignin have rather low molecular mass, what should facilitate their further processing to liquid hydrocarbons and aerogels. For the depolymerization of organosolv lignins to liquid hydrocarbons the processes of their catalytic conversion in supercritical alcohols have good prospects for the use. In the processes of lignin thermal conversion alcohols can to extract the products of lignin depolymerization and to alkylate these products, preventing their repolymerization to high molecular mass substances. To obtain a new class of nanoporous materials based on lignin the methods of organic aerogels synthesis from mixtures of lignin with other natural polymers and crosslinking agents were applied. It was found that the structure and properties of porous materials of aerogel type depend not only from the reaction mixture composition but from the method of drying. Drying in subcritical conditions leads to the formation of xerogels, in supercritical conditions - to the formation aerogels and the freezdrying - of cryogels. Obtained porous materials can have very low density (around 0.2 g/cm(3)), high specific surface area (to 500 m(2)/g) and the pore volume near 5 cm(3)/g.
