Аннотация: Synthesis of polyhydroxyalkanoates (PHAs) by a new strain of Cupriavidus - Cupriavidus eutrophus B-10646 - was investigated under autotrophic growth conditions. Under chemostat, at the specific flow rate D=0.1h-1, on sole carbon substrate (CO2), with nitrogen, sulfur, phosphorus, potassium, and manganese used as growth limiting elements, the highest poly(3-hydroxybutyrate) [P(3HB)] yields were obtained under nitrogen deficiency. In batch autotrophic culture, in the fermenter with oxygen mass transfer coefficient 0.460h-1, P(3HB) yields reached 85% of dry cell weight (DCW) and DCW reached 50g/l. Concentrations of supplementary PHA precursor substrates (valerate, hexanoate, ?-butyrolactone) and culture conditions were varied to produce, for the first time under autotrophic growth conditions, PHA ter- and tetra-polymers with widely varying major fractions of 3-hydroxybutyrate, 4-hydroxybutyrate, 3-hydroxyvalerate, and 3-hydroxyhexanoate monomer units. Investigation of the high-purity PHA specimens showed significant differences in their physicochemical and physicomechanical properties. В© 2013 Elsevier Ltd.

Аннотация: A set of polyhydroxyalkanoates are synthesized, and a comparative study of their physicochemical properties is performed. The molecular masses and polydispersities of polyhydroxyalkanoates are found to be independent of their chemical structures. It is shown that the temperature characteristics and degrees of crystallinity of polyhydroxyalkanoates are affected by the chemical compositions of the monomers and their quantitative contents in the polymers. The incorporation of 4-hydroxybutyrate, 3-hydroxyvalerate, and 3-hydroxyhexanoate units into the chain of poly(3-hydroxybutyrate) decreases its melting point and thermal degradation temperature relative to these parameters of a homogeneous poly(3-hydroxybutyrate) sample (175 В± 5 C and 275 В± 5 C, respectively). The higher the content of the second monomer units in the poly(3-hydroxybutirate) chain, the greater the changes. The degrees of crystallinity of polyhydroxyalkanoate copolymers are generally lower than that of poly(3-hydroxybutyrate) (75 В± 5%). The effect on the ratio of the amorphous and crystalline phases of the copolymer samples becomes more pronounced in the series 3-hydroxy-valerate-3-hydroxyhexanoate-4- hydroxybutyrate. Аннотация: The study addresses consequences of subcutaneous implantation of film matrices prepared from different PHAs to laboratory animals. No negative effects of subcutaneous implantation of PHA matrices on physiological and biochemical characteristics of the animals were determined. Independently of the matrices composition and duration of the contact with the internal environment of the organism we did not observe any deviations in the behavior of animals, their growth and development, as well as blood functions. Response of the tissues to PHA matrices was comparable with the response to polylactide, but substantially less expressed at the earlier time periods after implantation. Tissues response to implantation of PHA of all types is characterized by short-term (up to 2 weeks) post-traumatic inflammation with formation of fibrous capsules by 30th-60th days with the thickness less than 100 microns, which get thinner down to 40-60 microns by 180th day as the result of involution. No differences in response of tissues and the whole organism were observed for the matrices produced from the homopolymer of 3-hydroxybutyric acid (P3HB), copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids (P3HB/4HB), 3-hydroxybutyric acid and 3-hydroxyvalerianic acids (P3HB/3HV), 3-hydroxybutyric and 3-hydroxyhexanoate acids (P3HB/3HH). Macrophages and foreign-body giant cells actively participate in the response of the tissues to PHAs. In the studied conditions matrices from the copolymers containing 3-hydroxyhexanoate and 4 hydroxybutyrate were determined as more actively degraded PHA. The next less degraded matrices were matrices from the copolymer of P3HB/3HV and the most resistant were P3HB matrices. The slower degradation of PHA matrices was accompanied by delayed development of giantcells response. The studied PHA matrices can be placed in the following range by their degradation: P3HB/3HH - P3HB/4HB - P3HB/HV - P3HB.

Аннотация: The authors have constructed and characterized a series of membranes based on resorbable polyhydroxyalkanoates of different compositions. Five PHA types have been studied: a homopolymer of 3-hydroxybutyric acid, copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids, 3-hydroxybutyric and 3-hydroxyvaleric acids, 3-hydroxybutyric and 3-hydroxyhexanoic acids. Scanning electron microscopy and atomicforce microscopy were used to examine the microstructure of membrane surfaces, showing that membranes based on the copolymer of 3-hydroxybutyrate and 3-hydroxyhexanoate had the roughest surface, while membranes based on the copolymer of 3-hydroxybutyrate and 3-hydroxyvalerate had the smoothest surface. The contact angle for water in air was smaller and hydrophilic properties better in the copolymer membranes than in the membranes based on the high-crystallinity homopolymer of 3-hydroxybutyric acid. The culture of mouse fibroblast cell line NIH 3,3 was used to test PHAbased membranes; results of fluorescent probes of DNA DAPI and the MTT assay show that membranes based on studied PHAs are not cytotoxic on direct contact with cells and are highly biocompatible; their adhesive properties and ability to maintain fibroblast proliferation are similar to those of polystyrene and better than those of polylactic acid membranes.

Аннотация: Synthesis of 3- and 4-hydroxybutyrate copolymer (3HB-co-4HB), the most promising member of the biodegradable polyhydroxyalkanoate (PHA) family, has been studied. Cultivation conditions of naturally occurring strains of hydrogen-oxidizing bacteria Ralstonia eutropha B5786 and Cupriavidus eutrophus B10646 have been optimized to ensure efficient synthesis of the 3HB-co-4HB copolymer. A set of highly pure samples of the 3HB-co-4HB copolymer with 4HB content varying from 8.7 to 24.3 mol% has been obtained. Incorporation of 4HB into the copolymer was shown to cause a more pronounced decrease in polymer crystallinity than the incorporation of 3-hydroxyvalerate or 3-hydroxyhexanoate; samples with a degree of crystallinity below 30% have been obtained. The weight average molecular mass of the 3HB-co-4HB copolymers was shown to be independent on the monomer ratio and to vary broadly (from 540 to 1110 kDa). В© 2010 Pleiades Publishing, Ltd.

Аннотация: The study addresses the effect of different conditions of carbon nutrition on synthesis of polyhydroxyalkanoates by the bacterium Wautersia eutropha. In experiments with two wild type strains (H16 and 65786), it has been first found that under mixotrophic growth conditions - Co2 + co-substrate (alkanoic acids) - bacteria can synthesize multi-component PHAs, consisting of short- and medium-chainlength monomers with carbon chains containing 4 to 8 atoms. It has been shown that PHA composition is determined by the type of the co-substrate. Fatty acids with odd number of carbons induce bacteria to synthesize four- and five-component PHAs with hydroxybutyrate, hydroxyvalerate as major monomers and hydroxyhexanoate, hydroxyheptanoate and hydroxyoctanoate as minor, occasionally occurring, ones. Fatty acids with even number of carbons induce synthesis of not only their respective monomers (hydroxyhexanoate and hydroxyoctanoate) but also hydroxyvalerate, making possible synthesis of four-component PHAs, containing hydroxybutyrate and hydroxyhexanoate as major components (up to 18 mol%). Аннотация: The effect of carbon supply on polyhydroxyalkanoate (PHA) synthesis by bacteria Wautersia eutropha was studied. Synthesis of multicomponent PHA composed of short-and long-chain monomers (C4-C8) by two natural strains (H16 and B5786) under mixotrophic conditions (CO2 + alkanoic acids as cosubstrates) was demonstrated for the first time. The PHA composition was shown to be dependent on the cosubstrate type. In the presence of odd fatty acids, four-and five-component polymers were synthesized; hydroxybutyrate, hydroxyvalerate, and hydroxyheptanoate were the major monomers, while hydroxyhexanoate and hydroxyoctanoate were minor. In the presence of even fatty acids, PHA contained not only the corresponding molecules (hydroxyhexanoate and hydroxyoctanoate), but also hydroxyvalerate; synthesis of four-component PHA which contain mainly hydroxybutyrate and hydroxyhexanoate (up to 18 mol %) is therefore possible. Аннотация: The properties of new five-component poly(hydroxyalkanoates) (PHA) formed by short-and medium-chain monomers synthesized by the bacterium Wautersia eutropha B5786 were studied by X-ray diffraction, IR spectroscopy, differential thermal analysis, and viscometry. The degree of crystallinity of PHA decreased from 72 to 57% as the molar fraction of hydroxyhexanoate increased from 2.5 to 18.0 mol%. The melting temperature (T m) and decomposition temperature (T d) of the multicomponent PHA are lower than those for poly(hydroxybutyrate), whose T m and T d are 168-170 and 260-265В°C, respectively. Both parameters of the multicomponent PHA decrease to 156 and 252В°C, respectively, as the hydroxyhexanoate mole fraction is raised. The effect of hydroxyhexanoate on the physicochemical properties of the PHA is similar to that of hydroxyvalerate observed previously. В© 2007 Pleiades Publishing, Inc.

Аннотация: The physiological, biochemical, genetic, and cultural characteristics of the glucose-utilizing mutant strain Ralstonia eutropha B8562 were investigated in comparison with the parent strain R. eutropha B5786. The morphological, cultural, and biochemical characteristics of strain R. eutropha B8562 were similar to those of strain R. eutropha B5786. Genetic analysis revealed differences between the 16S rRNA gene sequences of these strains. The growth characteristics of the mutant using glucose as the sole carbon and energy source were comparable with those of the parent strain grown on fructose. Strain B8562 was characterized by high yields of polyhydroxyalkanoate (PHA) from different carbon sources (CO 2, fructose, and glucose). In batch culture with glucose under nitrogen limitation, PHA accumulation reached 90% of dry weight. In PHA, ?-hydroxybutyrate was predominant (over 99 mol %); ?-hydroxyvalerate (0.25-0.72 mol %) and ?-hydroxyhexanoate (0.008-1.5 mol %) were present as minor components. The strain has prospects as a PHA producer on glucose-containing media.

Аннотация: Two chemolithoautotrophs - the hydrogen-oxidizing bacterium Alcaligenes eutrophus and the carboxydobacterium Seliberia carboxydohydrogena - were investigated with respect to their ability to produce heteropolymeric polyhydroxyalkanoates (PHA) during mixotrophic growth on CO2 and medium-chain-length (C6 to C9) alkanoic acids. Hydroxy derivatives of the alkanoic acids were incorporated into PHA synthesized by the bacteria studied in proportions dependent on the time of cultivation. Irrespective of the nature of the acid added to the medium, PHAs were found to contain hydroxybutyrate as the main constituent and also hydroxyvalerate and hydroxyhexanoate, whose maximal incorporation was observed 4 to 6 h after the addition of an alkanoic acid to the medium. During further cultivation, the fractions of hydroxyvalerate and hydroxyhexanoate in PHA decreased, and the fraction of hydroxybutyrate and the total intracellular content of the polymer increased. Three-component polyhydroxyalkanoates with different proportions of hydroxybutyrate, hydroxyvalerate, and hydroxyhexanoate were obtained, and some of their properties were analyzed. Аннотация: The study addresses the effect of different conditions of carbon nutrition on synthesis of polyhydroxyalkanoates by the bacterium Wautersia eutropha. In experiments with two wild type strains (H16 and 85786), it has been first found that under mixotrophic growth conditions -CO2+ co-substrate (alkanoic acids) - bacteria can synthesize multi-component PHAs, consisting of short- and medium-chain-length monomers with carbon chains containing 4 to 8 atoms. It has been shown that PHA composition is determined by the type of the co-substrate. Fatty acids with odd number of carbons induce bacteria to synthesize four- and five-component PHAs with hydroxybutyrate, hydroxyvalerate as major monomers and hydroxyhexanoate, hydroxyheptanoate and hydroxyoctanoate as minor, occasionally occurring, ones. Fatty acids with even number of carbons induce synthesis of not only their respective monomers (hydroxyhexanoate and hydroxyoctanoate) but also hydroxyvalerate, making possible synthesis of four-component PHAs, containing hydroxybutyrate and hydroxyhexanoate as major components (up to 18 mol%). A family of short- and medium-chain-length four- and five-component PHAs were synthesized and their physicochemical properties examined.

Аннотация: BACKGROUND: P(3HB-co-3HHx) copolymers are very promising biomaterials. The main challenge in the production of these polymers is to simultaneously achieve high cell biomass; high P(3HB-co-3HHx) content; and high molar fraction of 3HHx in P(3HB-co-3HHx). The most common approach to production of these copolymers is the use of recombinant bacterial strains. The purpose of this study was to optimize the process of production of P(3HB-co-3HHx) copolymers containing high molar fractions of 3HHx by using the wild-type strain Cupriavidus eutrophus B10646. RESULTS: Kinetic properties of C. eutrophus B10646 were studied during cultivation of the cells on substrates necessary for P(3HB-co-3HHx) synthesis: glucose, nitrogen, sodium hexanoate, and sodium acrylate. The physiological ranges of their effects were determined experimentally, and C. eutrophus B10646 was grown in culture media with different dosages of these substrates. P(3HB-co-3HHx) copolymers with different molar fractions of 3HHx, including high ones (12 to 68%), were synthesized, and their physicochemical and mechanical properties were investigated. CONCLUSION: For the first time, cultivation conditions of Cupriavidus eutrophus B10646 enabled production of high biomass yields (5-6gL-1) and high content of the polymer (60-75%) that contained high 3HHx molar fraction. By varying the 3HB/3HHx ratio, one can change physicochemical and mechanical properties of P(3HB-co-3HHx) copolymers. © 2014 Society of Chemical Industry © 2016 Society of Chemical Industry.

Аннотация: Cupriavidus eutrophus В10646 was used to synthesize a series of polyhydroxyalkanoate (PHA) quaterpolymers composed of the short-chain-length 3-hydroxybutyrate (3HB), 4-hydroxybutyrate (4HB), and 3-hydroxyvalerate (3HV) and the medium-chain-length 3-hydroxyhexanoate (3HHx). The molar fraction of 3HB in the quaterpolymers varied between 63.5 and 93.1 mol.%, 3HV – between 1.1 and 24.6 mol.%, 4HB – between 2.4 and 15.6 mol.%, and 3HHx – between 0.4 and 4.8 mol.%. The properties of PHA quaterpolymers were significantly different from those of the P(3HB) homopolymer: they had much lower degrees of crystallinity (up to 30–45%), and lower melting points and thermal decomposition temperatures, with the interval between these temperatures remaining practically unchanged. Films prepared from PHA quaterpolymers were rougher and more porous than P(3HB) films; they showed higher values of elongation at break (up to 6–113%), i.e. were more elastic. Films prepared from PHA quaterpolymers were biocompatible and had no toxic effect on mouse fibroblast NIH 3T3 cells. © 2016 Elsevier Ltd

Аннотация: The physicochemical properties such as the degree of crystallinity and temperature and molecularmass characteristics of a number of polyhydroxyalkanoates of various chemical composition synthesized on a complex carbon substrate by bacteria Cupriavidus eutrophus В10646 have been investigated. Two-, three-, and four-component copolymer samples have different sets and ratios of monomers with various lengths of carbon chains: 3-hydroxybutyrate (3HB), 4-hydroxybutyrate (4HB), 3-hydroxyvalerate (3HV), 3-hydroxyhexanoate (3HH), 3-hydroxy-4-methyl valerate (3H4MV), and diethylene glycol (DEG). It has been shown that weight-average molar mass Мw and polydispersity vary in a wide range with no correlation existing with the composition of copolymer polyhydroxyalkanoates and that thermal stability is preserved in the temperature interval between the melting temperature and the thermal degradation temperature from 100 to 120–140°С. The composition and ratio of monomers most notably affect the degree of crystallinity of polyhydroxyalkanoates. Significant differences between the degrees of crystallinity of three- and four-component polyhydroxyalkanoates have been found for the first time. The degree of crystallinity for copolymers P(3HB/3HV/4HB) is 9–22%, and the degree of crystallinity for copolymers P(3HB/3HV/3HH) and P(3HB/3GV/3H4MV) is 41–63%; this value is close to the degree of crystallinity for diblock copolymers P(3HB)/DEG, which is 56–69%. For the four-component copolymers P(3HB/3GV/4HB/3HH), the degree of crystallinity is 30–41%. The values of Мw for the copolymers P(3HB/DEG) are inhomogeneous and the polymers contain fractions uneven with respect to molecular mass: a high-molecular-mass polymer (Мw from 2700 to 4900 kDa) and a low-molecular-mass polymer (Мw = 46–167 kDa). For the copolymers P(3HB)/DEG and P(3HB/3HV/3H4MV), two peaks are observed in the region of melting with the gap between these peaks being 4–20°С. All of the types of copolymer samples, regardless of the monomer ratio, show an increase in elongation at break against the background of a decrease in tensile stress and Young’s modulus, with these effects being pronounced to different extents. On the whole, the properties of multicomponent polyhydroxyalkanoates differ appreciably. © 2017, Pleiades Publishing, Ltd.

Аннотация: The study addresses degradation of polyhydroxyalkanoates (PHA) with different chemical compositions-the polymer of 3-hydroxybutyric acid [P(3HB)] and copolymers of P(3HB) with 3-hydroxyvalerate [P(3HB/3HV)], 4-hydroxybutyrate [P(3HB/4HB)], and 3-hydroxyhexanoate [P(3HB/3HHx)] (10-12 mol%)-in the agro-transformed field soil of the temperate zone. Based on their degradation rates at 21 and 28 A degrees C, polymers can be ranked as follows: P(3HB/4HB) > P(3HB/3HHx) > P(3HB/3HV) > P(3HB). The microbial community on the surface of the polymers differs from the microbial community of the soil with PHA specimens in the composition and percentages of species. Thirty-five isolates of bacteria of 16 genera were identified as PHA degraders by the clear zone technique, and each of the PHA had both specific and common degraders. P(3HB) was degraded by bacteria of the genera Mitsuaria, Chitinophaga, and Acidovorax, which were not among the degraders of the three other PHA types. Roseateles depolymerans, Streptomyces gardneri, and Cupriavidus sp. were specific degraders of P(3HB/4HB). Roseomonas massiliae and Delftia acidovorans degraded P(3HB/3HV), and Pseudoxanthomonas sp., Pseudomonas fluorescens, Ensifer adhaerens, and Bacillus pumilus were specific P(3HB/3HHx) degraders. All four PHA types were degraded by Streptomyces.

Аннотация: The present study investigates physicochemical, mechanical, and biological properties of polyhydroxyalkanoate (PHA) copolymers containing 4-hydroxybutyrate (4HB) synthesized in Cupriavidus eutrophus B10646 culture. In poly(3-hydroxybutyrate/4-hydroxybutyrate) [P(3HB/4HB)]bipolymers, 4HB varied between 10.4 and 75.0 mol%; in poly(3-hydroxybutyrate/3-hydroxyvalerate/4-hydroxybutyrate) terpolymers, 4HB constituted 28.7-55.6 mol%; and in poly(3-hydroxybutyrate/3-hydroxyvalerate/4-hydroxybutyrate/3-hydroxyhexanoate) quaterpolymers, 4HB varied between 9.3 and 13.3 mol%. The degree of crystallinity of P(3HB/4HB) copolymers decreased consistently with an increase in 4HB content, reaching 38%. The incorporation of 3-hydroxyvalerate and 3-hydroxyhexanoate into copolymers enhanced that effect. The effect of 4HB monomer units on temperature properties of copolymers was exhibited as lowering of the melting temperature and crystallization temperature, which improved the processing-related properties of the copolymers. All copolymers containing 4HB showed enhanced elongation at break compared to poly(3-hydroxybutyrate). Polymer films prepared from PHA5 with different chemical composition had similar microstructure and porosity and had no toxic effect on mouse fibroblast NIH 3 T3 cells, proving their high biocompatibility. Аннотация: Four PHA types were synthesized in the culture of Cupriavidus eutrophus B-10646 under special conditions, poly(3?hydroxybutyrate) [P(3HB)] and of copolymers, which contained 3HB monomers and 4?hydroxybutyrate (4HB), 3?hydroxyvalerate (3HV), or 3?hydroxyhexanoate (3HHx). All copolymers had the Mw of about 550–670 kDa, and the homopolymer P(3HB) had a significantly higher Mw – 920 kDa. P(3HB?co?4HB) and P(3HB?co?3HHx) had the lowest Cx (42 and 49%) while P(3HB?co?3HV) and P(3HB) exhibited higher Cx values (76%). Polymer films were prepared from different PHAs. Electron microscopy showed differences in the surface microstructure of the films. Films prepared from the P(3HB) were more hydrophobic and the arithmetic mean surface roughness of 71–75 nm, than the copolymer films, which were hydrophilic (57–60°) and had considerably higher roughness (158–177 nm). Blood parameters (hemoglobin and hemolysis) and response of the cells (erythrocytes, platelets, and monocytes) were studied in experiments with blood directly contacting the surface of the films of PHAs with different compositions. Cultivation of blood cells on polymer films did not cause any adverse effects on adhesion and morphology of all cell types. Results of studying blood cell response suggested that the films made from low-crystallinity copolymers containing 4?hydroxybutyrate and 3?hydroxyhexanoate were the best for contact with blood. © 2019 Elsevier B.V.
