Аннотация: The paper presents the results of biomedical investigations of sutures made of polyhydroxyalkanoates (PHAs) of two types (poly-3-hydroxybutyrate (PHB) and poly-3-hydroxybutyrate-co-poly-3-hydroxyvalerate (PHB/PHV)), synthesized by the bacteria Ralstonia eutropha B 5786 in the experiment on test animals in vivo, in comparison with silk and catgut sutures. PHB and PHB/PHV (PHBV) implants produced no adverse effect on physiological, biochemical and functional parameters of the animals during the post-surgery period. The tested PHA sutures featured the necessary strength throughout the healing period of the muscle-fascial cuts. The reaction of tissues to the implantation of PHB and PHB/PHV fibers fitted into the usual scheme characteristic of the wound process and of the reaction to a foreign-body invasion. This reaction and the reaction of tissues to silk had the similar nature and period of inflammation, but it was much less pronounced than the reaction to catgut. The tissue response to the implantation of PHAs consisted in a short-duration (up to 2 weeks) post-traumatic inflammation and the formation of a fibrous capsule less than 200 ?m thick during weeks 4-8, which in 4-6 months was reduced to 40-60 ?m in the course of reverse development. There were no adverse changes, such as suppurative inflammation, necrosis, calcification, and malignization of the cicatrice, at the site of implantation of PHA filaments, unlike in the cases with silk and catgut. In the case of PHA implantation there was a typical prolonged (throughout the post-surgery monitoring period) pronounced macrophagal stage with a large number of macrophages present. The macrophages were of the phagocytic type and multinucleate giant foreign cells, with a high activity of acid phosphomonoesterase that correlated with the activity of the enzyme in blood. Throughout the period of monitoring no differences in the tissue response to the implantation of the polymer filaments of two PHA types were recorded. В© 2003 Elsevier Science B.V. Аннотация: The composition and content of fatty acids (FAs) in total lipids, triacylglycerols (TAG) and polar lipids (PL) in dominant groups of benthic invertebrates: gammarids (Gammaridae, Amphipoda), chironomid larvae (Chironomidae, Diptera), caddisfly larvae (Trichoptera) and mayfly larvae (Ephemeroptera) were studied in the Yenisei river. For the first time data on the FA composition of species belonging to Trichoptera (Insecta) are presented. The groups of aquatic insect larvae and gammarids weakly differed in total content of essential polyunsaturated fatty acids (PUFAs). Hence, the strong invasion of gammarids which occurred in the last decades in the Yenisei river should not result in a decrease in potential yield of essential PUFA in the ecosystem and corresponding decrease in food resource quality for fish in respect to PUFA content. Significant differences in biomarker FAs in TAG were found which correlated to specific food sources. Different levels of long-chain PUFA in PL of the invertebrates are discussed in relation to the genetic ability of particular taxa to form these FAs. (C) 2002 Elsevier Science Inc. Аннотация: Understanding the factors that regulate the abundance, size structure and community structure of cladocerans is an important goal of aquatic ecologists. While both top-down and bottom-up factors help to structure cladoceran communities, there may be interactions between these factors. We conducted a mesocosm study to determine how alien large-bodied Daphnia, zebra mussels and fish affected cladoceran community and size structure. We found that large-bodied Daphnia reduced algal resources and the fecundity of smaller bodied cladocerans. Fish removed the large-bodied Daphnia magna from the mesocosms and shifted the cladoceran community to a smaller body size. Fish also appeared to promote increases in cladoceran diversity through the coexistence of several smaller bodied taxa. In contrast, zebra mussels increased cyanobacteria and helped to promote the success of the alien Daphnia, but reduced the biomass of small-bodied cladocerans. Zebra mussels reduced the carbon (C):phosphorus (P) ratio of the phytoplankton in the mesocosms which may have favored the relatively P-limited Daphnia. Combined, our results highlight the complex interactions of multiple factors that help to regulate cladoceran community and size structure.

Аннотация: Content of eicosapentaenoic acid (EPA, 20 : 5n-3) and docosahexaenoic acid (DHA, 22 : 6n-3) and the n-3/n-6 ratio are important indicators of nutritive value of aquatic invertebrates as food for fish. We studied fatty acid (FA) content and composition of 68 zoobenthic species. Benthic invertebrates differed significantly in their contents of EPA and DHA and n-3/n-6 ratios. The most valuable food for fish were Insecta, especially Ephemeroptera and Trichoptera, Gammaridae, and Turbellaria (Dendrocaelopsis sp.). The invertebrates of low food quality for fish were Hirudinea, Mollusca, Oligochaeta, Bryozoa (Plumatella emarginata), and Coleoptera (Dytiscus lapponicus). Our data suggest that the recent and ongoing global replacement of native species, mainly Insecta, by invaders, mostly represented by Mollusca, Crustacea, Polychaeta, Oligochaeta, Bryozoa, and Hirudinea, probably will reduce the nutritive value of food sources for benthivorous fish. In addition, some of the taxa studied have peculiar FAs, which may be useful as their markers in trophic webs. Thus, 20 : 1n-13, 20 : 2n-6, 22 : 5n-3, and 22 : 6n-3 are likely to be considered markers of Mollusca, Hirudinea, Turbellaria, and Gammaridae, respectively, for tracing food webs in freshwater ecosystems. © 2016 by The Society for Freshwater Science.

Аннотация: MicroRNAs epigenetically regulate physiological and pathological processes. Previously, we found that miR-204-5p is expressed at low levels in melanoma cells, and an increase in its level leads to a change in proliferation, migration, and invasion of these cancer cells. Now, using bioinformatics analysis, it has been shown that the target of miR-204-5p is FOXC1 transcription factor, which is implicated in carcinogenesis. Using the luciferase reporter assay, it was found that miR-204-5p suppresses expression of the FOXC1 gene by binding to its 3' non-coding region. Transfection of small interfering RNA (siRNA) targeting FOXC1 into melanoma cells caused a decrease in miR-204-5p levels, which is consistent with the generally accepted concept of feedback regulation of miRNA expression by target genes. According to the results of the MTT test and fluorescence microscopy, the proliferation level of melanoma cells under the influence of siRNA to FOXC1 decreased 72 h after transfection. Changes in the ratio of cells by cell cycle phase were analyzed using flow cytometry. Regulatory relationships between FOXC1 and miR-204-5p, and an inhibitory effect of FOXC1 knockdown on melanoma cell proliferation were revealed. Based on the results, it can be assumed that miR-204-5p regulates proliferation of melanoma cells by affecting FOXC1 expression.
