bacteriochlorophyll a -- chemocline -- meromictic lake -- purple sulfur bacteria -- seasonal dynamics -- Bacteria (microorganisms) -- Chromatiaceae
Аннотация: Phototrophic sulfur bacteria form dense accumulations in the chemocline zones of stratified lakes where light reaches the sulfide-containing layers of water. Many works are dedicated to the ecophysiology of these microorganisms in meromictic lakes. However, the role of these microorganisms in the trophic network of these ecosystems, the ways of biomass utilization, and the contribution to the turnover of biogenic elements have so far been insufficiently understood. This work deals with the analysis of many years' seasonal dynamics of the biomass of purple sulfur bacteria and the physicochemical conditions of their environment in Lake Shunet (Siberia, Khakassia, Russia), unraveling the causes of their anomalous development in the chemocline of this lake, as well as the comparative analysis of such type of ecosystems. Lake Shunet is characterized by markedly pronounced stratification and the high density of purple sulfur bacteria (PSB) in the chemocline, which is comparable to that of Lake Mahoney (Canada) where the number of PSB is the greatest among those known in the world. It was shown that, in the period 2002-2009, the total amount of bacterio-chlorophyll a in the water column of Lake Shunet increased and did not correlate with the seasonal variations in temperature and illumination in the chemocline. It was established that PSB cells in the purple layer experienced the effect of self-shading. The sedimentation rate of purple sulfur bacteria in Lake Shunet was low due to the pronounced density gradient in the chemocline zone. Thus, the high number of PSB in the chemocline was due to the combination of strong illumination, a high sulfide concentration, and a high water density gradient, which was responsible for stable stratification and contributed to the accumulation of the cells in a narrow layer. The data obtained could be useful for the paleoreconstruction of climatically deter-mined changes in the level of the lake and its periods of meromixis by the presence of carotenoids and bacte-riochlorophylls in the bottom sediments. Аннотация: Microbiological and isotopic-geochemical investigations of the brackish meromictic lakes Shira and Shunet were performed in the steppe region of Khakasia in winter. Measurements made with a submersed sensor demonstrated that one-meter ice transmits light in a quantity sufficient for oxygenic and anoxygenic photosynthesis. As in the summer season, in the community of phototrophic bacteria found in Lake Shira, the purple sulfur bacteria Amoebobacter purpureus dominated, whereas, in Lake Shunet, the green sulfur bacteria Pelodictyon luteolum were predominant. Photosynthetic production, measured using the radioisotopic method, was several times lower than that in summer. The rates of sulfate reduction and production and oxidation of methane in the water column and bottom sediments were also lower than those recorded in summer. The process of anaerobic methane oxidation in the sediments was an exception, being more intense in winter than in summer. The data from radioisotopic measurements of the rates of microbial processes correlate well with the results of determination of the isotopic composition of organic and mineral carbon (? 13C) and hydrogen sulfide and sulfate (? 34S) and suggest considerable seasonal variations in the activity of the microbial community in the water bodies investigated.

Аннотация: The year-to-year variations of vertical distribution and biomass of anoxic phototrophic bacteria were studied during ice periods 2003-2005 and 2007-2008 in meromictic lakes Shira and Shunet (Southern Siberia, Russian Federation). The bacterial layers in chemocline of both lakes were sampled with a thin-layer hydraulic multi-syringe sampler. In winter, biomass of purple sulphur bacteria varied considerably depending on the amount of light penetrating into the chemocline through the ice and snow cover. In relatively weakly stratified, brackish Shira Lake, the depth of chemocline varied between winters, so that light intensity for purple sulphur bacteria inhabiting this zone differed. In Shira Lake, increased transparency of mixolimnion in winter, high chemocline position and absence of snow resulted in light intensity and biomass of purple sulphur bacteria exceeding the summer values in the chemocline of the lake. We could monitor snow cover at the lake surface using remote sensing and therefore estimate dynamics and amount of light under ice and its availability for phototrophic organisms. In Shunet Lake, the light intensities in the chemocline and biomasses of purple sulphur bacteria were always lower in winter than in summer, but the biomasses of green sulphur bacteria were similar. Аннотация: The paper summarises the results of a three-year research study (European Union Grant: INTAS 97-0519) aimed at investigating the planktonic populations and trophic organization of the Lake Shira ecosystem - a saline lake in Khakasia, Siberia. The lake exhibits a stable summer-autumn stratification of the chemical-biological components. The mechanisms responsible for the 'blooming' of phytoplankton in the deeper layers were investigated in greater detail, using data from both field and laboratory experiments. The spectra of nutrition were examined to estimate the relationships between the specific growth rates of the hydrobionts and the influence of the limiting factors: light, nutrients. The observed heterotrophic capability of a metalimnetic phytoplankton population might help explain the development in the deeper waters of Lyngbya contorta. The scheme of trophic interactions was put up, based on the assessment of the carbon pools and carbon flows in the pelagic zone of the lake. A mathematical model of the vertical structure of the lake's plankton populations was constructed, using the ecosystem description and data of vertical turbulent diffusion. The role of light and nutrient limitations and grazing mortality in forming the vertical inhomogeneities, particularly in lowering the depth of the maximal cyanobacterial biomass, has been demonstrated. The theoretical curves for the stratification of chemical and biological parameters have been brought in conformity with the field observations, e.g. for the different patterns of the peaks, and for the biomass maxima of cyanobacteria, purple and green sulphur bacteria, oxygen, and hydrogen sulphide. The calculations revealed that for an adequate assessment of the parameters for the hydrogen sulphide zone it is necessary to introduce flows of allochthonous organic matter. Based on the form of the sulphur distribution curve, the allochthonous input of organic matter and the inflow of hydrogen sulphide from the bottom have been theoretically discriminated for the first time. It has also been ascertained that irrespective of the depth the allochthonous substances limiting bacterial growth, the bacteria are uniformly distributed over depth and can serve as an indicator of the presence of limitation (the effect of autostabilisation in space). Of indisputable interest to limnology are the specific methods developed for understanding the functioning of Lake Shira ecosystem. These include the autostabilisation of the limiting factors, the on-the-spot fluorescent method of determining the three classes of microalgae, the algal mixotrophy and the planktonic population interactions and feedbacks, and development of a more sensitive, bioluminescent method for mapping the nonhomogeneities. Owing to a balanced combination of classical approaches (field observations, in situ data on production-decomposition) and the more recent ones (satellite monitoring, biophysical methods of estimating interactions of populations, mathematical models based on the field and experimental data), many of the structural-function relationships in the ecosystem can now be explained, and the models can provide 'mutual control and mutual agreement' between the data collected using different approaches.

Аннотация: Data on the seasonal changes in vertical heterogeneity of the physical-chemical and biological parameters of the thermally stratified Shira Lake ecosystem (Khakasia, Siberia) in 1996-2000 have been analyzed. The interaction mechanisms involving: (1) The plankton populations in aerobic and anaerobic zones, involving the cycling of carbon and sulphur, (2) the primary production limitation (by light and phosphorus) and inhibition (by light), and (3) the kinetic characteristics of plankton populations have been elucidated. A mathematical model of the vertical structure of the lake's plankton populations, based on the ecosystem description and on vertical turbulent diffusion of the matter, has been constructed. The green alga Dictyosphaerium tetrachotomum (Chlorophyta) and the cyanobacterium Lyngbya contorta (Cyanophyta), which dominated the phytoplankton biomass, were taken as oxygen producers. Arctodiaptomus salinus (a calanoid copepod) has been assumed as the main grazer in Shira Lake as it dominated the zooplankton biomass. Four groups of microorganisms involved in the sulphur cycle formation have been distinguished: sulphur, sulphur purple, sulphur green and SRB. H2S is oxidized to sulphate (only the green sulphur bacteria oxidize it to sulphur), and sulphate is reduced to H2S, forming neither sulphur nor its water-soluble compounds. The role of grazing, light and nutrient limitation, in forming the vertical inhomogeneities, particularly in lowering the depth of the maximal cyanobacterial biomass, has been demonstrated. When the model takes into account both light limitation and nutrient limitation of algal growth by P and consumption of algae by crustaceans: (a) in the scenario where the P is formed only by the cycling and decomposition of autochthonous organic matter, both the green algae and cyanobacteria are eliminated; (b) in the scenario involving an additional P flux in the deep water layers the peak of the cyanobacteria is at a depth of 10 m, and its amplitude is close to the one observed in the lake. The position of the peak remains stable owing to the 'double' limitation mechanism: light 'from above' and P 'from below'. Another mechanism responsible for the deep position of the peak of cyanobacteria was analyzed mathematically based on the model involving the experimentally proven assumption of the growth inhibition by light in the epilimnion and the light limitation in the hypolimnion. The main result is: the peak is positioned stable at its depth and does not change with time. The analytical and numerical calculations made for this positioning mechanism yielded the formulae relating the depth of the maximum of algal biomass, the 'width' of the peak base and the peak amplitude and a number of parameters (algae elimination, turbulent diffusion coefficient, sedimentation rate, light extinction coefficient and light intensity). The theoretical curves for the stratification of chemical and biological parameters have been brought in conformity with field observations, e.g. for the different patterns for the peaks, and the biomass maxima of cyanobacteria, purple and green sulphur bacteria, oxygen, and hydrogen sulphide. The calculations revealed that for an adequate assessment of the parameters for the hydrogen sulphide zone it is necessary to introduce flows of allochthonous organic matter. For the first time, theoretically, based on the form of the sulphur distribution curve, the allochthonous input of organic matter and the inflow of hydrogen sulphide from the bottom have been discriminated. The theoretical limit for the depth up to which the hydrogen-sulphide zone can ascend under the impact of allochthonous organic loading, has been determined.

Аннотация: The circadian rhythm of grazing of microalgae by a laboratory culture of Ceriodaphnia quadrangula under continuous illumination was studied by continuous registration of chlorophyll fluorescence at the outlet of a flow-through cultivator. A culture of green alga Chlorella vulgaris was used as a feed. The data obtained were treated by the statistical spectral analysis. It was found that animals preliminarily grown under a 12 h light : 12 h dark regime and transferred to constant light showed two maxima in the circadian rhythm of grazing with periods of 0.7 and 1.1 h. Animals preliminarily grown under constant light showed no circadian rhythm of grazing. It was concluded that the circadian rhythm of grazing of C. quadrangula has endogenous nature and can change according to light conditions.

Аннотация: The study addresses reflection and spectral-polarization characteristics of forest stands and farm crops obtained under field conditions. The study of the reflection properties of farm crops shows that during the summer plant growing season, the major factors influencing the plant canopy reflectance are morpho-physiological parameters, plant architectonics, solar elevation h0, and viewing angle. The crop reflectance minimum was recorded at viewing angles 25-30В° with respect to the nadir. Coniferous and broadleaf forest stands had similar reflectance spectra of polarized light. The polarized component was smaller for all coniferous stands than for broadleaf ones. For broad-leaved farm crops (wheat and corn), the polarized component of the spectral brightness coefficients had a greater influence on the plant reflectance in the red and near-infrared spectral regions, ?>720nm. chlorophyll fluorescence induction -- Cyperus esculentus -- mineral nutrition -- ontogeny -- PAR irradiance -- Cyperus -- Cyperus esculentus
Аннотация: Application of earlier proposed ontogenetic approach to assessment of chufa (Cyperus esculentus L.) response to artificial-light culture growing conditions differing in illuminance and type of mineral nutrition is described. It was shown that, on biological soil-like substrate, plant productivity did not increase as a result of PAR level rising, and life time of chufa leaves was reduced to 11 days as compared with 18 days on the neutral substrate. Changes in the parameters of chlorophyll fluorescence induction (Fv/Fm, Yield = (Fm - Ft)/Fm, and ETR = 0.5 ? 0.84 ? Yield ? PAR) analyzed on the basis of ontogenetic approach show that it can disclose nonoptimal culture conditions. В© 2011 Pleiades Publishing, Ltd.

Аннотация: The main objective of an LSS is to supply a crew with food, water and oxygen, and to eliminate its waste. The ultimate goal is to achieve the highest degree of closure of the system using controlled processes offering a high level of reliability and flexibility. Enhancement of closedness of biological life support systems (BLSS) including plants relies on increased regeneration of gas, water and plant waste, and utilization of solid and liquid human wastes. Clearly, the robustness of an LSS subjected to stress will be substantially determined by the robustness of the plant components of the phototrophic unit. The aim of the present work was to estimate the heat resistance of plants grown on human wastes. Human exometabolites mineralized by hydrogen peroxide in an alternating electromagnetic field were used to make a nutrient solution for the plants. We looked for a possible increase in the heat resistance of the plants using changes in photosynthetically active radiation (PAR) intensity during heat stress. At ages 15 and 25 days, plants were subjected to a rise in air temperature (from 22-24В°C to 44В°C) under different PAR intensities for 4 hours. The status of the photosynthetic apparatus of the plants was assessed by external CO2 gas exchange and fluorescence measurements. The increased irradiance of the plants during the high temperature period demonstrated its protective action for both the photosynthetic apparatus of the leaves and subsequent plant growth and development. The productivity of the plants subjected to temperature changes at 250 W/m2 of PAR did not differ from that of controls, whereas the productivity of the plants subjected to the same heat stress but in darkness was halved. The heat resistance of the reproductive organs of 25-day-old plants was significantly lower than that of 15-day-old plants subjected to similar light and temperature conditions.

Chlorophyll fluorescence -- Leaf -- Ontogeny -- Stress -- Cucumis sativus
Аннотация: The causes that limit the applicability of the chlorophyll fluorescence induction method to evaluating the state of the photosynthetic apparatus of leaves under long-term stress were analyzed. It was substantiated that it is necessary to use an ontogenetic approach, which consists in more accurately taking into account the age state of the plant leaves studied. Comparison of the calendar and ontogenetic methods for determining the leaf age by the example of cucumber plant under controlled conditions revealed significant differences in the results of evaluating the photosynthetic apparatus of plant leaves by the chlorophyll fluorescence induction method in two variants with contrasting illumination conditions (in white light at 400-700 nm and in stressful red light at 600-700 nm). It was shown that, under long-term stress, the unequivocal interpretation of the chlorophyll fluorescence induction parameters in evaluating the photosynthetic apparatus of plants depends significantly on the choice of the ontogeny period of the leaves of plants being compared and on the accuracy of determining their age. Аннотация: We studied soil-like substrate (SLS) as a potential candidate for plant cultivation in bioregenerative life support systems (BLSS). The SLS was obtained by successive conversion of wheat straw by oyster mushrooms and worms. Mature SLS contained 9.5% humic acids and 4.9% fulvic acids. First, it was shown that wheat, bean and cucumber yields as well as radish yields when cultivated on mature SLS were comparable to yields obtained on a neutral substrate (expanded clay aggregate) under hydroponics. Second, the possibility of increasing wheat and radish yields on the SLS was assessed at three levels of light intensity: 690, 920 and 1150 ?mol m-2 s-1 of photosynthetically active radiation (PAR). The highest wheat yield was obtained at 920 ?mol m-2 s-1, while radish yield increased steadily with increasing light intensity. Third, long-term SLS fertility was tested in a BLSS model with mineral and organic matter recycling. Eight cycles of wheat and 13 cycles of radish cultivation were carried out on the SLS in the experimental system. Correlation coefficients between SLS nitrogen content and total wheat biomass and grain yield were 0.92 and 0.97, respectively, and correlation coefficients between nitrogen content and total radish biomass and edible root yield were 0.88 and 0.87, respectively. Changes in hormone content (auxins, gibberellins, cytokinins and abscisic acid) in the SLS during matter recycling did not reduce plant productivity. Quantitative and species compositions of the SLS and irrigation water microflora were also investigated. Microbial community analysis of the SLS showed bacteria from Bacillus, Pseudomonas, Proteus, Nocardia, Mycobacterium, Arthrobacter and Enterobacter genera, and fungi from Trichoderma, Penicillium, Fusarium, Aspergillus, Mucor, Botrytis, and Cladosporium genera. Аннотация: Use of halophytes (salt-tolerant vegetation), in a particular vegetable Salicornia europaea plants which are capable of utilizing NaCl in rather high concentrations, is one of possible means of NaCl incorporation into mass exchange of bioregenerative life support systems. In preliminary experiments it was shown that S. europaea plants, basically, could grow on urine pretreated with physicochemical processing and urease-enzyme decomposing of urea with the subsequent ammonia distillation. But at the same time inhibition of the growth process of the plants was observed. The purpose of the given work was to find out the influence of excessive quantities of some mineral elements contained in products of physicochemical processing of urine on the production process and NaCl accumulation by S. europaea plants. As the content of mineral salts in the human liquid wastes (urine) changed within certain limits, two variants of experimental solutions were examined. In the first variant, the concentration of mineral salts was equivalent to the minimum salt content in the urine and was: K - 1.5 g/l, P - 0.5 g/l, S - 0.5 g/l, Mg - 0.07 g/l, Ca - 0.2 g/l. In the second experimental variant, the content of mineral salts corresponded to the maximum salt content in urine and was the following: K - 3.0 g/l, P - 0.7 g/l, S - 1.2 g/l, Mg - 0.2 g/l, Ca - 0.97 g/l. As the control, the Tokarev nutrient solution containing nitrogen in the form of a urea, and the Knop nutrient solution with nitrogen in the nitrate form were used. N quantity in all four variants made up 177 mg/l. Air temperature was 24 В°C, illumination was continuous. Light intensity was 690 ?mol/m2s of photosynthetically active radiation. NaCl concentration in solutions was 1%. Our researches showed that the dry aboveground biomass of an average plant of the first variant practically did not differ from the control and totaled 11 g. In the second variant, S. europaea productivity decreased and the dry aboveground biomass of an average plant totaled 8 g. The increase of K quantity in the experimental solutions resulted in an elevated content of the element in the plants. The increase of K uptake in the second experimental variant was accompanied by a 30-50% decrease of Na content in comparison with the other variants. Comparative Na content in the other variants was practically identical. N, Mg and P content in the control and experimental variants was also practically identical. The increase of S quantity in the second experimental variant also increased S uptake by the plants. But Ca quantity, accumulated in aboveground plants biomass in the experimental variants was lower than in the control. NaCl uptake by plants, depending on the concentration of mineral salts in the experimental solutions, ranged from 8 g (maximum salt content) up to 15 g (minimum salt content) on a plant growth area that totaled 0.032 m2. Thus, high concentrations of mineral salts simulating the content of mineral salts contained in urine did not result in a significant decrease of S. europaea productivity. The present work also considers the influence of higher light intensity concentrations on productivity and NaCl accumulation by S. europaea plants grown on experimental solutions with high salt content. Аннотация: To increase the degree of closure of biological life support systems of a new generation, we used vermicomposting to involve inedible phytomass in the intra-system mass exchange. The resulting product was a soil-like substrate, which was quite suitable for growing plants (Manukovsky et al. 1996, 1997). However, the soil-like substrate can be regarded as a candidate for inclusion in a system only after a comprehensive examination of its physical, chemical, and other characteristics. An important criterion is the ability of the soil-like substrate to supply the necessary mineral elements to the photosynthesizing component under the chosen cultivation conditions. Thus, the purpose of this work was to study the feasibility of enhancing the production activity of wheat and radish crops by varying the intensity of photosynthetically active radiation, without decreasing the harvest index. The increase of light intensity from 920 to 1150 ?molВ·m -2В·s -1 decreased the intensity of apparent photosynthesis of the wheat crops and slightly increased the apparent photosynthesis of the radish crops The maximum total and grain productivity (kg/m 2) of the wheat crops was attained at the irradiance of 920 ?molВ·m -2В·s -1. Light intensity of 1150 ?molВ·m -2В·s -1 decreased the productivity of wheat plants and had no significant effect on the productivity of the radish crops (kg/m 2) as compared to 920 ?molВ·m 2В·s -1. The qualitative and quantitative composition of microflora of the watering solution and substrate was determined by the condition of plants, developmental phase and light intensity. By the end of wheat growth under 1150 ?molВ·m -2В·s -1 the numbers of bacteria of the coliform family and phytopathogenic bacteria in the watering solution and substrate were an order of magnitude larger than under other illumination conditions. The obtained data suggest that the cultivation of plants in a life support system on soil-like substrate from composts has a number of advantages over the cultivation on neutral substrates, which require continual replenishment of the plant nutrient solution from the system's store to complement the macro- and microelements. Yet, a number of problems arise, including those related to the controlling of the production activity of the plants by the intensity of photosynthetically active radiation. It is essential to understand why the intensity of production processes is limited at higher irradiation levels and to overcome the factors responsible for this, so that the soil-like substrate could have an even better chance in the competition for the best plant cultivation technology to be used in biological life support systems. Аннотация: The effect of elevated temperatures of 35 and 45В°C (at the intensities of photosynthetically active radiation 322, 690 and 1104 ?mol-m-2-s-1) on the photosynthesis, respiration, and qualitative and quantitative composition of the volatiles emitted by wheat (Triticum aestuvi L., cultivar 232) crops was investigated in growth chambers. Identification and quantification of more than 20 volatile compounds (terpenoids - ?-pinene, ?3 carene, limonene, benzene, ?-and transcaryophyllene, ?- and ?-terpinene, their derivatives, aromatic hydrocarbons, etc.) were conducted by gas chromatograph/mass spectrometry. Under light intensity of 1104 ?mol-m-2-s-1, heat resistance of photosynthesis and respiration increased at 35В°C and decreased at 45В°C. The action of elevated temperatures brought about variations in the rate and direction of the synthesis of volatile metabolites. The emission of volatile compounds was the greatest under a reduced irradiation of 322 ?mol-m-2-s-1 and the smallest under 1104 ?mol-m-2-s-1 at 35В°C. During the repair period, the contents and proportions of volatile compounds were different from their initial values, too. The degree of disruption and the following recovery of the functional state depended on the light intensity during the exposure to elevated temperatures. The investigation of the atmosphere of the growth chamber without plants has revaled the substances that were definitely technogenic in origin: tetramethylurea, dimethylsulfide, dibutylsulfide, dibutylphthalate, and a number of components of furan and silane nature.
