Аннотация: The study addresses the effect of humic substances on marine luminous bacteria Photobacterium phosphoreum exposed to Am-241 (3,000 Bq L -1, water solution). Luminescent intensity of the bacteria was applied as a marker of their physiological activity. Humic substances have been found to reduce the effect of Am-241 on luminescence, decrease damage to cells, and change distribution of Am-241 between bacterial cells and intercellular media. It was shown that water-soluble humic substances, being products of natural transformation of organic substances in soil and bottom sediments, can serve as protecting agents for water microorganisms exposed to alpha radionuclides. Аннотация: A large number and wide variety of lake ecosystem models have been developed and published during the past four decades. We identify two challenges for making further progress in this field. One such challenge is to avoid developing more models largely following the concept of others ('reinventing the wheel'). The other challenge is to avoid focusing on only one type of model, while ignoring new and diverse approaches that have become available ('having tunnel vision'). In this paper, we aim at improving the awareness of existing models and knowledge of concurrent approaches in lake ecosystem modelling, without covering all possible model tools and avenues. First, we present a broad variety of modelling approaches. To illustrate these approaches, we give brief descriptions of rather arbitrarily selected sets of specific models. We deal with static models (steady state and regression models), complex dynamic models (CAEDYM, CE-QUAL-W2, Delft 3D-ECO, LakeMab, LakeWeb, MyLake, PCLake, PROTECH, SALMO), structurally dynamic models and minimal dynamic models. We also discuss a group of approaches that could all be classified as individual based: super-individual models (Piscator, Charisma), physiologically structured models, stage-structured models and trait-based models. We briefly mention genetic algorithms, neural networks, Kalman filters and fuzzy logic. Thereafter, we zoom in, as an in-depth example, on the multi-decadal development and application of the lake ecosystem model PCLake and related models (PCLake Metamodel, Lake Shira Model, IPH-TRIM3D-PCLake). In the discussion, we argue that while the historical development of each approach and model is understandable given its 'leading principle', there are many opportunities for combining approaches. We take the point of view that a single 'right' approach does not exist and should not be strived for. Instead, multiple modelling approaches, applied concurrently to a given problem, can help develop an integrative view on the functioning of lake ecosystems. We end with a set of specific recommendations that may be of help in the further development of lake ecosystem models. В© 2010 The Author(s).

Аннотация: The biodegradation patterns of two types of PHA, a 3-hydroxybutyrate (3-PHB) polymer and a 3-hydroxybutyrate and 3-hydroxyvalerate (3-PHB/3-PHV) copolymer, were studied in tropical marine environments (Dam Bay, South China Sea, Nha Trang, Vietnam). No reliable differences in the degradation of 3-PHB and 3-PHB/3-PHV were revealed. It was shown that the degradation process depended mainly on the shape of a polymer product and its production method: the degradation of polymer films was found to be more active than that of molded solids. A decrease in the molecular mass of both types of PHA was detected in the course of the degradation of PHA samples. However, the degree of PHA crystallinity did not change; that is, the levels of degradation of both the amorphous and crystalline phases of PHA were almost the same. Among microbial PHA degraders, three bacterial strains, Bacillus sp. IBP-V002, Enterobacter cloacae sp. IBP-V001, and Gracilibacillus sp. IBP-V003, were identified based on the results of morphological, biochemical, and molecular phylogenetic analyses. The ability of the representatives of the genera Gracilibacillus and Enterobacter to degrade PHA was revealed for the first time. В© 2011 Pleiades Publishing, Ltd.

Аннотация: Biodegradability patterns of two PHAs: a polymer of 3-hydroxybutyric acid (3-PHB) and a copolymer of 3-hydroxybutyric and 3-hydroxyvaleric acids (3-PHB/3-PHV) containing 11 mol% of hydroxyvalerate, were studied in the tropical marine environment, in the South China Sea (Nha Trang, Vietnam). No significant differences have been observed between degradation rates of 3-PHB and 3-PHB/3-PHV specimens; it has been found that under study conditions, biodegradation is rather influenced by the shape of the polymer item and the preparation technique than by the chemical composition of the polymer. Biodegradation rates of polymer films in seawater have been found to be higher than those of compacted pellets. As 3-PHB and 3-PHB/3-PHV are degraded and the specimens lose their mass, molecular weight of both polymers is decreased, i.e. polymer chains get destroyed. The polydispersity index of the PHAs grows significantly. However, the degree of crystallinity of both PHAs remains unchanged, i.e. the amorphous phase and the crystalline one are equally disintegrated. PHA-degrading microorganisms were isolated using the clear-zone technique, by inoculating the isolates onto mineral agar that contained PHA as sole carbon source. Based on the 16S rRNA analysis, the PHA-degrading strains were identified as Enterobacter sp. (four strains), Bacillus sp. and Gracilibacillus sp.В© 2010 Elsevier Ltd. All rights reserved.

Аннотация: The ability of marine luminescent bacteria to synthesize polyesters of hydroxycarboxylic acids (polyhydroxyalkanoates, PHA) as reserve macromolecules was studied. Twenty strains from the collection of the luminescent bacteria CCIBSO (WDCM839) of the Institute of Biophysics, Siberian Branch, Russian Academy of Sciences, assigned to different taxa (Photobacterium leiognathi, Ph. phosphoreum, Vibrio harveyi, and V. fischeri) were analyzed. The most productive strains were identified, and the conditions ensuring high polymer yields in batch culture (40-70% of the cell dry mass weight) were determined. The capacity for synthesizing two-and three-component polymers containing hydroxybutyric acid as the main monomer and hydroxyvaleric and hydroxyhexanoic acids was revealed in Ph. leiognathi and V. harveyi strains. The results allow luminescent microorganisms to be regarded as new producers of multicomponent polyhydroxyalkanoates. В© 2008 MAIK Nauka.

Аннотация: Abstract: Fatty acid composition and content of 2 forms of sockeye salmon Oncorhynchus nerka from lakes in Kamchatka Peninsula (Russia) were compared. One form of sockeye salmon was anadromous ("marine"), that is, adult fish migrated in ocean to feed and grow and than return in the lake to breed. Fish of another form, kokanee, never migrate in the ocean. Per cent levels of the main indicators of nutritive value, eicosapentaenoic acid (EPA, 20:5n-3), and docosahexaenoic acid (DHA, 22:6n-3), were significantly higher in the landlocked O. nerka. However, concentrations of EPA and DHA per wet weight of filets were higher in the marine form, because of the relatively higher content of sum of fatty acids in their muscle tissue. As concluded, fish fed in marine environment had higher contents of long-chain n-3 fatty acids per wet weight than fish of the same species, fed in fresh waters. In general, both the anadromous sockeye salmon and the landlocked kokanee salmon can be recommended for human diet as a valuable product concerning contents of EPA and DHA. Аннотация: We studied four-link food chain, periphytic microalgae and water moss (producers), trichopteran larvae (consumers I), gammarids (omnivorous - consumers II) and Siberian grayling (consumers III) at a littoral site of the Yenisei River on the basis of three years monthly sampling. Analysis of bulk carbon stable isotopes and compound specific isotope analysis of fatty acids (FA) were done. As found, there was a gradual depletion in 13C contents of fatty acids, including essential FA upward the food chain. In all the trophic levels a parabolic dependence of ? 13C values of fatty acids on their degree of unsaturation/chain length occurred, with 18:2n-6 and 18:3n-3 in its lowest point. The pattern in the ? 13C differences between individual fatty acids was quite similar to that reported in literature for marine pelagic food webs. Аннотация: Recruitment of pelagic populations of cyanobacteria from littoral bottom sediments is assumed to be one of the key factors for the development of harmful blooms. Therefore, it is necessary to obtain more information on the possible factors influencing the spatial distribution of the resting stages in littoral bottom sediments. To test the hypothesis that akinete abundances in littoral bottom sediments are associated with silt and the presence of higher water plants, we sampled spatial and vertical distribution and seasonal dynamics of akinetes of cyanobacterium Anabaena flos-aquae in a small Siberian reservoir. We found that akinetes accumulate in silt at stations located in macrophyte beds. Two experiments were carried out to test if there is relationship between size of inoculum and size of the pelagic population of A. flos-aquae. The proportion of A. flos-aquae in total phytoplankton biomass depended on the initial abundance of akinetes in bottom sediments. The model based on laboratory experiments gave good qualitative description of the field data. These findings can be useful to predict the location of the main akinete accumulation zones in a water body and plan the removal of sediment to prevent the cyanobacterial blooms. В© CSIRO 2011.

Аннотация: To preserve and utilize the biosphere it is imperative that we understand the long-term dynamics of the primary production process on our planet. Variability of chlorophyll concentration in the ocean is one of the most important components of this process. However, size and access make its investigation labour-intensive. Dependence of primary production on high variability of hydrophysical phenomena in the ocean (fluctuations of currents, frontal zones, etc.) makes it difficult to locate points to measure chlorophyll concentration dynamics. To locate such quasistationary zones with respect to seasonal dynamics of chlorophyll concentration by satellite data the Institute of Biophysics (Russian Academy of Sciences, Siberian Branch) has developed a geoinformation system. Its use made it possible to locate such zones over the entire ocean. The work shows zones on the Central and Northern Atlantic. They form the ground to analyse perennial dynamics of chlorophyll concentration. The quasistationary zones are proposed for systematic monitoring of phytopigment concentration by space-borne and marine craft.

Аннотация: To preserve the biosphere and to use it efficiently, it is necessary to gain a deep insight into the dynamics of the primary production process on our planet. Variability of chlorophyll concentration in the ocean is one of the most important components of this process. These investigations are, however, very labor-consuming, because of the difficulties related to the accessibility of the water surface and its large size. In this work long-term changes in chlorophyll concentration in the surface layer of the ocean have been analyzed on the basis of the CZCS data for 7.5 years from 1979 to 1986 and the SeaWiFS data from 1997 to 2004. It has been shown that the average chlorophyll concentration calculated in all investigated areas varies moderately. However, when analyzing spatially local trends, the areas have been detected that have significant rise and fall of chlorophyll concentrations. Some interesting features of the long-term dynamics of chlorophyll concentration have been found. The opposite directions of long-term trends (essential increase or decrease) cannot be explained only by large-scale hydrological phenomena in the ocean (currents, upwellings, etc.). The measured chlorophyll concentration results from the balance between production and destruction processes. Which process dominates is determined by various hydrophysical, hydrobiological, and climatic processes, leading to sharp rises or falls of the concentration. It is important to estimate the scale of the areas in which this or that process dominates. Therefore, the study addresses not only the dynamics of the mean value but also the dynamics of the areas in which the dominance of certain factors has led to a sharp fall or rise in chlorophyll concentration. Areas such as these, found in the Indian Ocean, are described in this work. These areas form the basis for the analysis of long-term dynamics of chlorophyll concentration. In these quasistationary zones systematic monitoring of phytopigment concentration is conducted by space-borne and marine craft. The work presents long-term satellite-based data on the space distribution of anomalous deviations of chlorophyll concentration in the ocean. Аннотация: Polyunsaturated fatty acids (PUFA) of ?3 family are of crucial physiological importance for the most animals and they are an essential and deficient component of human nutrition. These compounds are most effectively synthesized by some groups of algae, hence, aquatic ecosystems are considered to be the main source of these PUFA for human nutrition. Factors controlling the content and distribution of ?3 PUFA in freshwater organisms of basic trophic levels and determined PUFA final production in freshwater ecosystems are considered in the review. PUFA biosynthesis is known to be tightly related to basic fatty acid metabolic pathways. Hence, fatty composition and the PUFA content of major freshwater hydrobiont groups, including bacteria, algae, invertebrates and vertebrates, and environmental and population age effects are described. The peculiarities of PUFA transfer between organisms of various trophic levels are discussed. The essential ?3 PUFA is one of the important parameter of food quality of aquatic consumers and they can determine the rate of energy and matter transfer between producers and primary consumers and, as a result, in a whole freshwater food chain. Analysis of PUFA content and its regulation in biomass of various fish populations indicates that freshwater ecosystems are of the same value in respect of PUFA sources as marine ecosystems. Despite the great practical importance, the studies focused on production and whole pools of ?3 PUFA in different freshwater ecosystems are still scarce and need to be continued.

Аннотация: Characteristics of the diurnal rhythm of the bioluminescence field are analyzed on the basis of experimental data obtained in the region around Kamchatka peninsula (50В°3 1'N; 163В°50'E). The feasibility of reconstructing the nocturnal structure of the bioluminescence field by measurements made at other times of the day has been demonstrated. Diurnal rhythm parameters for the bioluminescent field of several other studied areas of the ocean have been defined. The temporal parameters feature linear latitudinal dependence. At the equator the boundaries of morning and evening transition periods are 05:06 to 07:30 and 17:48 to 19:54 hrs, respectively. In the Barents Sea in spring these boundaries are 02:42 to 04:06 and 19:48 to 21:12 hrs. Boundary times of diurnal rhythm do not deviate from the linear regression by more than 10%.

Аннотация: A structural approach to studying the regularities of the population dynamics of unstable recombinant bacterial strains in a chemostat was elaborated. The approach is based on the mathematical modeling of cell distribution in a population with different numbers of plasmid copies. The effect of decreased selective preference of plasmidless variants of the recombinant strain in the chemostat, which is related to a decrease in the number of plasmid copies in cells upon long-term incubation was analyzed. It is shown that the time of half-elimination of plasmids from the bacterial population in the steady state in the chemostat T1/2 does not depend on the maximum number of plasmid copies in cells N but is determined only by the mean time of generation g and the probability of the loss of one plasmid copy tau. The dependence of the preference of bacterial plasmidless variants on the efficiency of expression of genes cloned into plasmids in chemostat was analyzed using the recombinant strain E. coli Z905, whose plasmids pPHL-7 contain cloned genes for the luminescence system of marine luminescing bacteria Photobacterium leiognathi.

Аннотация: Manifestation of pleiotropic effects in the isogenic variants of the luminescent bacterium Photobacterium leiognathi 54 was investigated. The decrease or increase of the expression level of bioluminescence was caused by changes in lux operon regulation. The dynamics of the bioluminescence of dark and dim variants did not differ from the dynamics of the initial luminescent variant, but dependence of the level of luminescence intensity on the exogenous autoinducer of the lux operon was revealed. The investigated variants of P. leiognathi 54 inherited fairly stable morphological characteristics, colony architectonics, level of luminescence, and activity of some enzymes; variants with reduced bioluminescence formed colonies of the S type. Stable bright variants with S-and R-type colonies appeared both in the initial strain population and in the dark variant population, but with smaller frequency. Populations of the bright variant with R-type colonies were most heterogeneous; this can be determined by the lack of glucose repression of the bioluminescence in contrast to other investigated inherited variants of P. leiognathi. Аннотация: The Institute of Biophysics SB RAS hosts and maintains a specialized collection of luminous bacteria (CCIBSO 836) containing over 700 strains isolated in various regions of the world's oceans. The culture collection is a source of lux genes and biologically active substances. The wide application of bioluminescence in medicine and ecology has given importance to analys-ing information on the structure and functioning of bioluminescence systems in natural and transgenic microorganisms, as well as on their features that are closely interrelated with bioluminescence. The aims of our BIOLUMBASE database are: gathering information on microorganisms with lux genes, their analysis and free access, and distribution of this data throughout the global network. The database includes two sections, natural and transgenic luminous microorganisms, and is updated by our own experimental results, the published literature and internet resources. For the future, a publicly available internet site for BIOLUMBASE is planned. This will list the strains and provide comprehensive information on the properties and functions of luminous bacteria, the mechanisms of regulation of bioluminescence systems, constructs with lux genes, and applications of bioluminescence in microbiology, ecology, medicine and biotechnology. It is noteworthy that this database will also be useful for evaluation of biological hazards of transgenic strains. Аннотация: The culture collection maintained at the Institute of Biophysics (IB), Siberian Division, Russian Academy of Sciences, is the only deposit in Russia and CIS which contains about 700 strains of five species of marine luminous bacteria sampled in various sites of the World Ocean. The strains collected are used for identification of luminous bacteria; studying their physiology, biochemistry, and cytology; and developing concepts of organization and metabolism of luminous bacteria under natural and laboratory conditions. Methods of their storage and practical use are developed. The results of these studies are summarized in a data bank, which will be incorporated into the general network of data banks on microbial cultures kept in Russia and into the international network of data banks on microorganisms (MSDN). Systematization of information on luminous bacteria possessing a unique visual marker allows their extensive use in basic research and genetic engineering, for environmental and medical applications, and for biotechnological purposes.

Аннотация: The vertical structure of the ecosystem in the euphotic zone of the western Equatorial Pacific has been studied. The studies were based on a continuous sounding of the bioluminescence field, with simultaneous, vertically aimed sampling made with the aid of a 5l water bottle and plankton nets. The bioluminescence field has a two-maxima structure with a more pronounced and permanent lower maximum found in the oligotrophic regions as deep as 60 to 100 m. In the narrow (10 m) layer of maximum bioluminescence, the concentration of zooplankton, as well as the concentration, activity and production of bacteria and phytoplankton, are several times higher than those in immediately adjacent waters at greater or lesser depth. At the same time, the concentration of nutrient salts in that layer diminishes sharply and approaches zero in the overlying water. Analysis of the data obtained shows that the production processes above the lower maximum layer are dependent not only on the turbulent influx of nutrient salts from the underlying layers, but also on the horizontal supply from the zone of upwelling. A pelagic community, in its different time aspects from the moment of water ascending until its sinking in the convergence zone, is suggested as a common system for modelling. Аннотация: For analytical purposes bioluminescence can be used in three main ways: 1. luminescence measurement of bioluminescent system components isolated in vitro; 2. determination of luminous organisms' reaction to the in vivo test-action; 3. measurement of bioluminescence in marine ecological systems. The majority of the reports of this Symposium are dealing with the first two topics. The aim of our presentation is to draw attention to the third one. The possibilities of bioluminescent analysis are wider than its traditional scheme of applications in the laboratory, when the emitting system is withdrawn from a native source and is placed in a cuvette of the light measuring device. The reverse scheme is also possible, i.e. the device can be introduced into light emitting system such as a marine biocenosis--the community of the sea inhabitants--where we obtain a highly sensitive and rapid means of gaining the information on the vital activity of marine ecosystems, i.e. their spatial structure, rhythms, man's influence upon them, etc. The present communication will consider the possibilities of this form of bioluminescent analysis.

Аннотация: A cDNA clone encoding the Ca2+-activated photoprotein, obelin (Obl), from Obelia longissima was sequenced. The nucleotide (nt) sequence contained two long overlapping open reading frames (ORFs), one of which encoded apoobelin (apoObl). The deduced amino acid (aa) sequence of apoObl revealed that this 195-aa protein has three EF-hand structures that are characteristic for Ca2+-binding domains. Strong aa homology was shown among apoObl, apoaequorin and apoclytin. The second ORF present in the obl cDNA consists of 139 codons and encodes a very basic protein with a calculated pI of 10.56 and a molecular mass of 16153 Da.
