Аннотация: The effect of laser irradiation on the properties of the surface of films prepared from a bioresorbable polymer poly(hydroxybuturate) has been studied. To determine the spectral region of the polymer optimal for the effective action of radiation on electron molecular bonds, theoretical investigations have been performed, which have shown that, for modifying the surface of PHB scaffolds, it is expedient to use a vacuum laser at a wavelength of 160 nm. Using laser irradiation at a power from 3 to 30 W, a series of films with modified surface, from roughnesses to perforations, have been obtained. The microstructure and properties of the film surface depending on the mode of irradiation have been examined, and conditions have been found under which the contact marginal angles of film wetting with water can be decreased to 50В° (compared with 76-80В° in starting products). Thus, conditions of laser treatment of PHB scaffolds have been theoretically substantiated and experimentally realized that provide a beneficial effect on the properties of the surface without destroying the structure of the material. Аннотация: The growth, productivity, and changes in the chemical composition of the green alga Botryococcus braunii Kutz H-252 were studied at different phases of growth in a luminostat culture. Modification of the Prat medium resulted in an increase in the alga yield to 3.9 g/l and a decrease in the generation time to 3-4 days. The specific growth rate during the exponential phase was 0.235/ day. These values are comparable with the best results obtained abroad. Among the pigments of B. braunii, besides chlorophylls a and b, carotene, lutein, neoxanthin, and violaxanthin were also identified. The highest content of these pigments, based on dry weight, was achieved during the exponential growth phase. During culture aging, there was an accumulation of carbohydrates and lipids in addition to a simultaneous decrease in the concentration of nitrogen-containing substances. It was discovered that this strain can synthesize liquid hydrocarbons. These hydrocarbons were found both in the alga biomass (ca. 1% of dry weight) and among the extracellular metabolites (up to 10.3 mg/l).

Аннотация: The emission of different, diamond coatings on silicon field emitter. arrays were investigated: chemical vapor deposition diamond, high pressure and high temperature synthetic diamond, and shock-synthesized nanodiamond. Practical emission characteristics such as: emission threshold, maximum current, current stability, and reproducibility were tested. Аннотация: The process of electrophoresis/dielectrophoresis of shock-synthesized nanodiamond powders on the large-area Si field emission arrays was studied. The process was found to be very sensitive to the modification of nanodiamond powder. The growth rate and film morphology of nanodiamond coatings on Si tips were investigated. Аннотация: The recombinant coelenterazine-dependent luciferases (isoforms MLuc 164 and MLuc39) from the marine copepod Metridia longa were expressed as inclusion bodies in E. coli cells, dissolved in 6 M guanidinium chloride and folded in conditions developed for proteins containing intramolecular disulfide bonds. One of them (MLuc09) was obtained in an active monomeric form with a high yield. The luciferase bioluminescence is found to be initiated not only by free coelenterazine, but also by Ca2+-dependent coelenterazine-binding protein (CBP) of Renilla muelleri on Ca2+ addition. The use of CBP as a "substrate" provides higher light emission and simultaneously the lower level of background. The high purity MLuc39 can be detected down to attomol with a linear range extending over 5 orders of magnitude. The MLuc39 reveals also a high stability towards heating and chemical modification; the chemically synthesized biotinylated derivatives of the luciferase preserve 35-40% of the initial activity The luciferase applicability as an in vitro bioluminescent reporter is demonstrated in model tandem bioluminescent solid-phase microassay combining the Ca2+-regulated photoprotein obelin and the Metridia luciferase.

Аннотация: A unique technology of nanodiamond surface modification is suggested which allows to separation of commercial nanodiamond powders into two fractions (F1 and 172), each possessing absolutely new properties as compared to the initial powder. F1 and F2 differ in size characteristics. Initial and modified nanodiamonds contain iron impurities and two types of nondiamond carbon. The color of the powders and hydrosols does not correlate with the content of non-diamond carbon. According to the EPR data, modified nanodiamonds possess a high level of diamond matrix shielding, and the extracted fractions differ in width of the basic transition area and in the SHF energy adsorption ratio. Due to this, F1 can be applied as precursors for CVD growth of nanocrystalline diamond and as field electron emission tips.

Аннотация: AsLn2, an unusual modified peptide, was isolated from the bioluminescent earthworm Fridericia heliota (Enchytraeidae). Its structure, elucidated by NMR and mass spectrometry, includes residues of tyrosine, CompX (a novel tyrosine modification product, reported in the accompanying paper), and N(omega)-acylated lysine. Chromatography, UV, and 1H NMR data imply a close structural similarity of AsLn2 with F. heliota luciferin. AsLn2 appears to be an intermediate or by-product in F. heliota luciferin biosynthesis. © 2013 Elsevier Ltd. All rights reserved.

Аннотация: Previous studies have stated that aequorin loses most of its bioluminescence activity upon modification of the C-terminus, thus limiting the production of photoprotein fusion proteins at its N-terminus. In the present work, we investigate the importance of the C-terminal proline and the hydrogen bonds it forms for photoprotein active complex formation, stability and functional activity. According to the crystal structures of obelin and aequorin, two Ca2+-regulated photoproteins, the carboxyl group of the C-terminal Pro forms two hydrogen bonds with the side chain of Arg21 (Arg15 in aequorin case) situated in the first a-helix. Whereas, deletion or substitution of the C-terminal proline could noticeably change the bioluminescence activity, stability or the yield of an active photoprotein complex. Therefore, modifications of the first alpha-helix Arg has a clear destructive effect on the main photoprotein properties. A C-terminal hydrogen-bond network is proposed to be important for the stability of photoprotein molecules towards external disturbances, when taking part in the formation of locked protein conformations and isolation of coelenterazine-binding cavities.

Аннотация: Detonation nanodiamond powders produced by different manufacturers were surface modified to reduce the amount of surface impurities and impart to the particles a high colloidal stability in aqueous suspensions. Modified DNDs (MDNDs) were separated into fractions and evaluated by the zeta potential, particles size distribution and elemental composition. MDNDs retain colloidal stability after the hydrosols are dried and the resulting powders are resuspended. Sedimentation stability and fraction composition are proposed as parameters for standardization of commercial nanodiamonds. © 2015 Copyright © Taylor & Francis Group, LLC.

Аннотация: The kinetics of inactivation of luciferases from four species of luminescent bacteria by the thiol reagent N-ethylmaleimide were investigated The dependencies of inactivation on ionic strength differed among the enzymes. Increasing the molarity of the buffer increased the rate of inactivation of all luciferases except that of Vibrio harveyi. Modification of Photobacterium phosphoreum luciferase decreased the maximal intensity of bioluminescence, whereas modification of Photobacterium leiognathi and Vibrio fischeri luciferases in high ionic strength buffers decreased the maximal intensity of bioluminescence and changed the luminescence decay rate constant. High ionic strength apparently alters the conformational states of the luciferases.

Аннотация: The method of macro-electrophoresis is used to determine values of the zeta-potential of particles of ultradisperse diamond in solutions of AlCl3. It is shown that an increase in the concentration of aluminum ions to 0.5.10(-4) M leads to a decrease in the negative charge of the diamond particles; the dispersions coagulate in more concentrated solutions of AlCl3. Chemical modification of the surface of ultradisperse diamond by aluminum ions is used to obtain stable hydrosols with positively charged diamond particles.

Аннотация: The phase, dispersive, and morphological features of an aluminum oxide powder produced by explosive synthesis are examined experimentally. It is shown that the particle-size distribution has three distinct maxima, which are due to different combustion regimes. The relationship between the sizes and morphology of the starting powder and the product is determined. Considerable attention is given to a study of the ultrafine fraction of the product powder. The ultrafine particles are shown to have a regular spherical shape, and sinters are not revealed. Besides spherical particles, the synthesized powder contains faceted crystals. X-ray phase analysis of the ultrafine fraction of the synthesized powders shows that this fraction consists only of the metastable phases of the oxide - the delta- or oxynitride modifications. The delta -modification differs from that described in the literature.

Аннотация: The oxygen and ammonia radio-frequency (RF) plasma treatment of poly(3-hydroxybutyrate) P3HB films was performed. We revealed significant changes in the topography, a decrease in the surface zeta potential from -63 to -75 mV after the oxygen-plasma treatment and an increase after ammonia plasma treatment from -63 to -45 mV at a pH of 7.4. Investigations into the NIH 3T3 fibroblast adhesion and growth demonstrated the best cell vitality and a higher cell number for the ammonia plasma treatment at 150W. (C) 2015 Elsevier B.V. Аннотация: We tested a comparatively new method of tracing of natural food webs, compound-specific isotope analysis (CSIA) of fatty acids (FA), using laboratory culture of Daphnia galeata fed Chlorella vulgaris and Cryptomonas sp. In general, Daphnia had significantly lighter carbon stable isotope composition of most fatty acids, including essential, than those of their food, microalgae. Thus, our results did not support the pivotal premise of the FA-CSIA application for food web analysis, i.e., transmitting the isotope 'signal' of essential FAs to consumers from their food without any modification. Moreover, the values of isotope fractionation of particular FAs in the consumer relative to its food were not constant, but varied from 1.35‰ to 7.04‰. The different isotope fractionation (depletion) values of diverse FAs in consumer were probably caused by different processes of their synthesis, catabolism and assimilation. More work is evidently to be done for correct interpretation of results of FA-CSIA during field studies for tracing of natural food webs. © 2015 Elsevier GmbH.

Аннотация: Magnetic Ni2+-zeolite/ferrosphere and Ni2+-silica/ferrosphere beads (Ni-ferrosphere beads - NFB) of a core-shell structure were synthesized starting from coal fly ash ferrospheres having diameters in the range of 0.063-0.050 mm. The strategy of NFB fabrication is an oriented chemical modification of the outer surface preserving the magnetic core of parent beads with the formation of micro-mesoporous coverings. Two routes of ferrosphere modification were realized, such as (i) hydrothermal treatment in an alkaline medium resulting in a NaP zeolite layer and (ii) synthesis of micro-mesoporous silica on the glass surface using conventional methods. Immobilization of Ni2+ ions in the siliceous porous shell of the magnetic beads was carried out via (i) the ion exchange of Na+ for Ni2+ in the zeolite layer or (ii) deposition of NiO clusters in the zeolite and silica pores. The final NFB were tested for affinity in magnetic separation of the histidine-tagged green fluorescent protein (GFP) directly from a cell lysate. Results pointed to the high affinity of the magnetic beads towards the protein in the presence of 10 mM EDTA. The sorption capacity of the ferrosphere-based Ni-beads with respect to GFP was in the range 1.5-5.7 mg cm-3. © The Royal Society of Chemistry.

Аннотация: The surface properties of poly-3-hydroxybutyrate (P3HB) membranes were modified using oxygen and an ammonia radio-frequency (RF, 13.56 MHz) plasma. The plasma treatment procedures used in the study only affected the surface properties, including surface topography, without inducing any significant changes in the crystalline structure of the polymer, with the exception being a power level of 250 W. The wettability of the modified P3HB surfaces was significantly increased after the plasma treatment, irrespective of the treatment procedure used. It was revealed that both surface chemistry and surface roughness changes caused by the plasma treatment affected surface wettability. A treatment-induced surface aging effect was observed and resulted in an increase in the water contact angle and a decrease in the surface free energy. However, the difference in the water contact angle between the polymers that had been treated for 4 weeks and the untreated polymer surfaces was still significant. A dependence between cell adhesion and proliferation and the polar component of the surface energy was revealed. The increase in the polar component after the ammonia plasma modification significantly increased cell adhesion and proliferation on biodegradable polymer surfaces compared to the untreated P3HB and the P3HB modified using an oxygen plasma. © 2016 Elsevier B.V.

Аннотация: Color variants of Ca2+-regulated photoprotein obelin were shown to be an important tool for dual-analyte binding assay. To provide site-directed conjugation with biospecific molecules, several obelin color mutants carrying unique cysteine residues were obtained and characterized for their novel properties. A pair of obelins Y138F,A5C and W92F,H22E,D12C was found to be most suitable (in terms of high bioluminescent activity and stability) as reporters in simultaneous assay of two targets in a sample. Availability of SH-groups, accessible for chemical modification, essentially simplifies the synthesis of biospecific conjugates, increases their yield and conserves obelins' bioluminescence activity. Conjugates with immunoglobulin and oligonucleotide were produced and successfully applied in single nucleotide polymorphism genotyping. © 2017 The American Society of Photobiology

Аннотация: Parameters for sample introduction, dried reagents dissolution and mixing with sample for bienzyme system NAD(H):FMN-oxidoreductase and luciferase immobilised in microfluidic chip were successfully determined. Numerical simulations of reaction chamber geometry, flavin mononucleotide (FMN) escape from starch gel and mixing options were conducted to achieve higher sensitivity of bioluminescent reaction. Results of numerical simulations were verified experimentally. The active mixer for dried reagents was made from an electro-mechanical speaker's membrane which was connected to the input of the chip. Such a mixer provided better efficiency than a passive mixing, and it is simple enough for use in point-of-care devices with any systems based on immobilised enzymes in chips.
