Аннотация: The paper summarises the results of a three-year research study (European Union Grant: INTAS 97-0519) aimed at investigating the planktonic populations and trophic organization of the Lake Shira ecosystem - a saline lake in Khakasia, Siberia. The lake exhibits a stable summer-autumn stratification of the chemical-biological components. The mechanisms responsible for the 'blooming' of phytoplankton in the deeper layers were investigated in greater detail, using data from both field and laboratory experiments. The spectra of nutrition were examined to estimate the relationships between the specific growth rates of the hydrobionts and the influence of the limiting factors: light, nutrients. The observed heterotrophic capability of a metalimnetic phytoplankton population might help explain the development in the deeper waters of Lyngbya contorta. The scheme of trophic interactions was put up, based on the assessment of the carbon pools and carbon flows in the pelagic zone of the lake. A mathematical model of the vertical structure of the lake's plankton populations was constructed, using the ecosystem description and data of vertical turbulent diffusion. The role of light and nutrient limitations and grazing mortality in forming the vertical inhomogeneities, particularly in lowering the depth of the maximal cyanobacterial biomass, has been demonstrated. The theoretical curves for the stratification of chemical and biological parameters have been brought in conformity with the field observations, e.g. for the different patterns of the peaks, and for the biomass maxima of cyanobacteria, purple and green sulphur bacteria, oxygen, and hydrogen sulphide. The calculations revealed that for an adequate assessment of the parameters for the hydrogen sulphide zone it is necessary to introduce flows of allochthonous organic matter. Based on the form of the sulphur distribution curve, the allochthonous input of organic matter and the inflow of hydrogen sulphide from the bottom have been theoretically discriminated for the first time. It has also been ascertained that irrespective of the depth the allochthonous substances limiting bacterial growth, the bacteria are uniformly distributed over depth and can serve as an indicator of the presence of limitation (the effect of autostabilisation in space). Of indisputable interest to limnology are the specific methods developed for understanding the functioning of Lake Shira ecosystem. These include the autostabilisation of the limiting factors, the on-the-spot fluorescent method of determining the three classes of microalgae, the algal mixotrophy and the planktonic population interactions and feedbacks, and development of a more sensitive, bioluminescent method for mapping the nonhomogeneities. Owing to a balanced combination of classical approaches (field observations, in situ data on production-decomposition) and the more recent ones (satellite monitoring, biophysical methods of estimating interactions of populations, mathematical models based on the field and experimental data), many of the structural-function relationships in the ecosystem can now be explained, and the models can provide 'mutual control and mutual agreement' between the data collected using different approaches.

Аннотация: The study addresses the effect of different conditions of carbon nutrition on synthesis of polyhydroxyalkanoates by the bacterium Wautersia eutropha. In experiments with two wild type strains (H16 and 65786), it has been first found that under mixotrophic growth conditions - Co2 + co-substrate (alkanoic acids) - bacteria can synthesize multi-component PHAs, consisting of short- and medium-chainlength monomers with carbon chains containing 4 to 8 atoms. It has been shown that PHA composition is determined by the type of the co-substrate. Fatty acids with odd number of carbons induce bacteria to synthesize four- and five-component PHAs with hydroxybutyrate, hydroxyvalerate as major monomers and hydroxyhexanoate, hydroxyheptanoate and hydroxyoctanoate as minor, occasionally occurring, ones. Fatty acids with even number of carbons induce synthesis of not only their respective monomers (hydroxyhexanoate and hydroxyoctanoate) but also hydroxyvalerate, making possible synthesis of four-component PHAs, containing hydroxybutyrate and hydroxyhexanoate as major components (up to 18 mol%). Аннотация: The paper deals with the study of the synthesis of 3-hydroxybutyrate (3HB) and 3-hydroxyvalerate (3HV) copolymers by the bacterium Ralstonia eutropha B-5786 grown under different carbon nutrition conditions (growth on carbon dioxide, fructose, and CO2-valerate and fructose-valerate mixtures). The parameters to be analyzed included the yield of biomass, the yield, synthesis rate, and composition of copolymers, the activity of the key enzymes of polyhydroxyalkanoate (PHA) synthesis (?-ketothiolase, acetoacetyl-CoA reductase, and PHA synthase), the maximum tolerable concentration of valerate to the bacterium, and the conditions that govern the incorporation of hydroxyvalerate to copolymers. This allowed the relationship between cultivation conditions and the proportion of monomers in the copolymers to be deduced. We were able to synthesize a range of 3HB/3HV copolymers and found that the thermal characteristics and the degree of crystallinity of these copolymers depend on the molar fraction of 3HV.

Аннотация: The dynamics of accumulation of polyhydroxybutyrate (PHB) and the activities of key enzymes of PHB metabolism (?-ketothiolase, acetoacetyl-CoA reductase, PHB synthase, D-hydroxybutyrate dehydrogenase, and PHB depolymerase) in the hydrogen bacterium Ralstonia eutropha B5786 were studied under various conditions of carbon nutrition and substrate availability. The highest activities of ?-ketothiolase, acetoacetyl-CoA reductase, and PHB synthase were recorded during acceleration of PHB synthesis. The activities of enzymes catalyzing PHB depolymerization (PHB depolymerase and D-hydroxybutyrate dehydrogenase) were low, being expressed only upon stimulated endogenous PHB degradation. The change of carbon source (CO2 or fructose) did not affect the time course of the enzyme activity significantly.

Аннотация: The molecular structure of polyhydroxybutyrate from hydrogen-oxidizing bacteria Alcaligenes eutrophus was studied by X-ray diffraction analysis. It was shown that the degree of crystallinity of various samples depends little on the conditions of their preparation and is equal to 0.62-0.76. The molecular structure of solid samples and solution of polyhydroxybutyrate in chloroform were studied by the NMR and EPR methods. The conclusion is made that the molecular structure of polyhydroxybutyrate does not depend on the features of the strain and conditions of carbon nutrition of microorganisms producing polyhydroxybutyrate. Defects induced by gamma-radiation in polyhydroxybutyrate were studied. Free radicals were isolated, and their structure was decoded.

Аннотация: We studied the fatty acid (FA) content and composition of ten zoobenthic species of several taxonomic groups from different freshwater bodies. Special attention was paid to essential polyunsaturated fatty acids, eicosapentaenoic acid (EPA, 20:5n-3), docosahexaenoic acid (DHA, 22:6n-3), and arachidonic acid (ARA, 20:4n-6); and the n-3/n-6 and DHA/ARA ratios, which are important for consumers of higher trophic levels, i.e., fish. The content and ratios of these FA varied significantly in the studied zoobenthic species, consequently, the invertebrates were of different nutritional quality for fish. Eulimnogammarus viridis (Crustacea) and Dendrocoelopsis sp. (Turbellaria) had the highest nutrition value for fish concerning the content of EPA and DHA and n-3/n-6 and DHA/ARA ratios. Using canonical correspondence analysis we compared the FA profiles of species of the studied taxa taking into account their feeding strategies and habitats. We gained evidence that feeding strategy is of importance to determine fatty acid profiles of zoobenthic species. However, the phylogenetic position of the zoobenthic species is also responsible and may result in a similar fatty acid composition even if species or populations inhabit different water bodies or have different feeding strategies. Аннотация: It is shown that in the area inhabited by a colony of the common heron in a high-trophic shallow lake a number of structure parameters of plankton undergoes changes similar to those observed at anthropogenic eutrophication and indicating an increase in biogenic and organic load. At the same time a number of changes is revealed that are usually not registered when human influence increases. The composition and content of polyunsaturated fatty acids in seston and zooplankton show that diatoms and detritus of land genesis play relatively smaller role in the zooplankton nutrition in the lake area influenced by the birds. The zooplankton developing in this zone is a better food rich in essential docosahexaenoic acid necessary for the growth and development of the fish. В© 2011 Pleiades Publishing, Ltd.

Аннотация: Three canned fish species - Pacific saury (Cololabis saira), Pacific herring (Clupea harengus) and Baltic sprat (Sprattus sprattus) - most common and popular in Russia, were analyzed for fatty acids. Special attention was paid to long-chain essential polyunsaturated fatty acids: eicosapentaenoic acid (20:5?3) and docosahexaenoic acid (22:6?3). Sums of eicosapentaenoic acid and docosahexaenoic acid in saury, herring and sprat were, on average, 2.42, 1.80 and 1.43 g/100 g product, respectively. Contents of these essential acids in all the canned fish species were found to be very high compared with many other fish reported in the available literature. All the canned fish appeared to be highly valuable products for human nutrition concerning the content of eicosapentaenoic and docosahexaenoic acids.

Аннотация: The elemental and fatty acid composition of seston was studied for 3 years, from May to October, in a small Reservoir. Under comparatively low C:P ratio, multivariate canonical analysis revealed no straightforward simple correlations between phosphorus and single ?3 PUFA species, but complex significant interaction between elemental composition (stoichiometry) of seston and total sestonic ?3 PUFA as a whole. Since sestonic C, P and N were found to originate mostly from phytoplankton, the contents of particulate elements and PUFA were attributed to single species in periods of their pronounced dominance. Phytoplankton species of genera of Stephanodiscus, Peridinium, Gomphosphaeria, Planktothrix and Anabaena in periods of their pronounced dominance had relatively constant species-specific elemental and PUFA composition. Phytoplankton species significantly differed in their elemental and PUFA composition, as well as in ratios of C:N, N:P, PUFA:P and partly C:P that indicate food quality for zooplankton. Hence, there were no phytoplankton species of clearly high or low nutritional value. All of phytoplankters, or at least detritus, that originated from them, may meet specific elemental and biochemical requirements of specific groups of zooplankton. Dividing phytoplankton on basis of their elemental and biochemical composition, i.e., nutrition quality, into large taxa (cyanobacteria, diatoms, etc.) appeared to be too coarse for assessing nutritional value for zooplankton. Аннотация: 1. We studied variation in the composition of fatty acids in the seston of a small freshwater reservoir with changes in phytoplankton composition during four growth seasons. We focused on the dynamics of the ?3 fatty acids because of their potential importance for zooplankton nutrition. 2. Total diatoms were related to the 20:5?3 fatty acid (eicosapentaenoic, EPA) content in seston. Among two dominant diatom genera, Cyclotella was not associated with EPA content. In contrast, there was a significant correlation between Stephanodiscus and the percentage contribution and content of EPA throughout the study. Hence, freshwater diatoms can differ strongly in content of the essential EPA. 3. We considered abundant cyanobacteria as a potential source of 18:3?3 fatty acid (linolenic, ALA) to aquatic food webs. Among four dominant cyanobacteria species, two (Anabaena flos-aquae and Planktothrix agardhii) showed significant correlation with the ALA content of the seston, while the other two (Aphanizomenon flos-aquae and Microcystis aeruginosa) did not. 4. Dinophyta had a relatively high level of 22:6?3 (docosahexaenoic, DHA) for freshwater species and can be also a source of EPA to aquatic food webs. 5. Our results show that various species of diatoms as well as cyanobacteria can be of contrasting nutritional value for zooplankton because of their different content of the essential PUFAs. Diatoms, which are low in EPA, could not be considered as a valuable food, while some field populations of cyanobacteria might be valuable sources of essential ALA.

Аннотация: Application of earlier proposed ontogenetic approach to assessment of chufa (Cyperus esculentus L.) response to artificial-light culture growing conditions differing in illuminance and type of mineral nutrition is described. It was shown that, on biological soil-like substrate, plant productivity did not increase as a result of PAR level rising, and life time of chufa leaves was reduced to 11 days as compared with 18 days on the neutral substrate. Changes in the parameters of chlorophyll fluorescence induction (Fv/Fm, Yield = (Fm - Ft)/Fm, and ETR = 0.5 ? 0.84 ? Yield ? Аннотация: Bioregenerative life-support systems (BLSS) are studied for developing the technology for a future biological life-support system for long-term manned space missions. Ways to utilize human liquid and solid wastes to increase the closure degree of BLSS were investigated. First, urine and faeces underwent oxidation by Kudenko's physicochemical method. The products were then used for root nutrition of wheat grown by the soil-like substrate culture method. Two means of eliminating sodium chloride, introduced into the irrigation solution together with the products of urine oxidation, were investigated. The first was based on routine electrodialysis of irrigation water at the end of wheat vegetation. Dialysis eliminated about 50% of Na from the solution. This desalinization was performed for nine vegetations. The second method was new: after wheat cultivation, the irrigation solution and the solution obtained by washing the substrate containing mineral elements not absorbed by the plants were used to grow salt-tolerant Salicornia europaea L. plants (saltwort). The above-ground biomass of this plant can be used as a food, and roots can be added to the soil-like substrate. Four consecutive wheat and Salicornia vegetations were cultivated. As a result of this wheat and Salicornia cultivation process, the soil-like substrate salinization by NaCl were considerably decreased. В© 2008 Humana Press.

Аннотация: One of the ways of solving the problem of the human liquid wastes utilization in bioregenerative life support systems (BLSS) can be the use of halophytic vegetable plant Salicornia europaea capable of accumulating sodium chloride in rather high concentrations. Since the most specific higher plant function in BLSS, which at present cannot be substituted by physicochemical processes, appears to be the biosynthesis of a wide spectrum of nutritive substances necessary for a human, the object of the given work was the investigation of the S. europaea productivity, biochemical and mineral composition when grown under close to optimal BLSS vegetative component conditions. As the use of human urine after its preliminary physicochemical processing is supposed to be the mineral solution basis for the S. europaea cultivation, it is necessary to clear up the effect of reduced nitrogen on plants growth. Ground research was carried out. Biochemical composition of the S. europaea edible part showed that crude protein was contained in the highest degree. At that the content of crude protein (24% per dry weight) and cellulose (4.7% per dry weight) was higher in the plants grown on solutions containing amide nitrogen in comparison with the plants grown on solutions with nitrate nitrogen (15.4%-3.1% correspondingly). The water-soluble sugar contents were not high in the S. europaea edible part and depending on the nitrogen nutrition form they amounted to 1.1% (amide nitrogen) and 1.5% (nitrate nitrogen). The polysaccharide number (except cellulose) was rather higher and varied from 7.7% to 8.2%. Although the lipid content in the S. europaea plants was relatively low (7% per dry weight), it was shown that the plant lipids are characterized by a high nonsaturation degree mainly due to alpha linolenic and linoleic acids. Nitrogen nutrition form did not significantly affect the S. europaea productivity, and dry edible biomass of one plant was 8.6 g. Sodium and its concentrations predominated in the plant mineral composition and amounted in average to 9% per dry weight. Thus the S. europaea being the vegetable plant it can be the source of several biochemical substances and essential fatty acids. Аннотация: Wheat was cultivated on soil-like substrate (SLS) produced by the action of worms and microflora from the inedible biomass of wheat. After the growth of the wheat crop, the inedible biomass was restored in SLS and exposed to decomposition ("biological" combustion) and its mineral compounds were assimilated by plants. Grain was returned to the SLS in the amount equivalent to human solid waste produced by consumption of the grain. Human wastes (urine and feces) after physicochemical processing turned into mineralized form (mineralized urine and mineralized feces) and entered the plants' nutrient solution amounts equal to average daily production. Periodically (once every 60-70 days) the nutrient solution was partly (up to 50%) desalinated by electrodialysis. Due to this NaCl concentration in the nutrient solution was sustained at a fixed level of about 0.26%. The salt concentrate obtained could be used in the human nutrition through NaCl extraction and the residuary elements were returned through the mineralized human liquid wastes into matter turnover. The control wheat cultivation was carried out on peat with use of the Knop nutrient solution. Serial cultivation of several wheat vegetations within 280 days was conducted during the experiment. Grain output varied and yield/harvest depended, in large part, upon the amount of inedible biomass returned to SLS and the speed of its decomposition. After achieving a stationary regime, (when the quantity of wheat inedible biomass utilized during vegetation in SLS is equal to the quantity of biomass introduced into SLS before vegetation) grain harvest in comparison with the control was at most 30% less, and in some cases was comparable to the control harvest values. The investigations carried out on the wheat example demonstrated in principle the possibility of long-term functioning of the LSS photosynthesizing link based on optimizations of biological and physicochemical methods of utilization of the human and plants wastes. The possibilities for the use of these technologies for the creation integrated biological-physicochemical LSS with high closure degree of internal matter turnover are discussed in this paper. Аннотация: Use of halophytes (salt-tolerant vegetation), in a particular vegetable Salicornia europaea plants which are capable of utilizing NaCl in rather high concentrations, is one of possible means of NaCl incorporation into mass exchange of bioregenerative life support systems. In preliminary experiments it was shown that S. europaea plants, basically, could grow on urine pretreated with physicochemical processing and urease-enzyme decomposing of urea with the subsequent ammonia distillation. But at the same time inhibition of the growth process of the plants was observed. The purpose of the given work was to find out the influence of excessive quantities of some mineral elements contained in products of physicochemical processing of urine on the production process and NaCl accumulation by S. europaea plants. As the content of mineral salts in the human liquid wastes (urine) changed within certain limits, two variants of experimental solutions were examined. In the first variant, the concentration of mineral salts was equivalent to the minimum salt content in the urine and was: K - 1.5 g/l, P - 0.5 g/l, S - 0.5 g/l, Mg - 0.07 g/l, Ca - 0.2 g/l. In the second experimental variant, the content of mineral salts corresponded to the maximum salt content in urine and was the following: K - 3.0 g/l, P - 0.7 g/l, S - 1.2 g/l, Mg - 0.2 g/l, Ca - 0.97 g/l. As the control, the Tokarev nutrient solution containing nitrogen in the form of a urea, and the Knop nutrient solution with nitrogen in the nitrate form were used. N quantity in all four variants made up 177 mg/l. Air temperature was 24 В°C, illumination was continuous. Light intensity was 690 ?mol/m2s of photosynthetically active radiation. NaCl concentration in solutions was 1%. Our researches showed that the dry aboveground biomass of an average plant of the first variant practically did not differ from the control and totaled 11 g. In the second variant, S. europaea productivity decreased and the dry aboveground biomass of an average plant totaled 8 g. The increase of K quantity in the experimental solutions resulted in an elevated content of the element in the plants. The increase of K uptake in the second experimental variant was accompanied by a 30-50% decrease of Na content in comparison with the other variants. Comparative Na content in the other variants was practically identical. N, Mg and P content in the control and experimental variants was also practically identical. The increase of S quantity in the second experimental variant also increased S uptake by the plants. But Ca quantity, accumulated in aboveground plants biomass in the experimental variants was lower than in the control. NaCl uptake by plants, depending on the concentration of mineral salts in the experimental solutions, ranged from 8 g (maximum salt content) up to 15 g (minimum salt content) on a plant growth area that totaled 0.032 m2. Thus, high concentrations of mineral salts simulating the content of mineral salts contained in urine did not result in a significant decrease of S. europaea productivity. The present work also considers the influence of higher light intensity concentrations on productivity and NaCl accumulation by S. europaea plants grown on experimental solutions with high salt content. Аннотация: This study addresses the possibility of growing different halophytic plants on mineralized human urine as a way to recycle NaCl from human wastes in a bioregenerative life support system (BLSS). Two halophytic plant species were studied: the salt-accumulating Salicornia europaea and the salt-secreting Limonium gmelinii. During the first two weeks, plants were grown on Knop's solution, then an average daily amount of urine produced by one human, which had been preliminarily mineralized, was gradually added to the experimental solutions. Nutrient solutions simulating urine mineral composition were gradually added to control solutions. NaCl concentrations in the stock solutions added to the experimental and control solutions were 9 g/L in the first treatment and 20 g/L in the second treatment. The mineralized human urine showed some inhibitory effects on S. europaea and L. gmelinii. The biomass yield of experimental plants was lower than that of control ones. If calculated for the same time period (120 d) and area (1 m 2), the amount of sodium chloride taken up by S. europaea plants would be 11.7 times larger than the amount taken up by L. gmelinii plants (486 g/m 2 vs. 41 g/m 2). Аннотация: Effects of various combinations of nutrient solution salinity (0.3, 171, and 342 mM NaCl), photosynthetically active radiation (PAR) of 600 or 1150 ?mol/(m2 s), and type of nitrogen nutrition (amide-N or nitrate-N) on the productivity and the content of accumulated mineral nutrients and free amino acids were studied in Salicornia europaea plants. At PAR of 600 ?mol/(m2 s), plant productivity increased with elevation of salinity level; at 1150 ?mol/(m2 s), the maximum productivity was observed in the plants grown at 171 mM of NaCl. The content of free amino acids in shoots, regardless of PAR, decreased with growing salinity level, whereas Na content, on the opposite, increased. Glutamic acid, rather than proline, was shown to be the main organic osmolyte in this plant species. Comparison of the productivity of plants grown on solutions with amide (urea) or nitrate nitrogen showed that higher biomass accumulation was achieved in the former case.
