Аннотация: The effect of light sources with discrete or continuous spectra of red light (50 W/m2, 600-700 nm) on the structural and functional characteristics of chloroplasts were studied in leaves of cucumber (Cucumis sativum L.) plants, which die under strong red light, and pea (Pisum sativum L.) plants, which tolerate red light under the same conditions. Leaf condition was assessed by measuring thermo-and photoinduced changes in the chlorophyll fluorescence yield and the photochemical and photophosphorylating activities of the chloroplasts. Different responses of the pigment apparatus of pea and cucumber plants to red light with continuous or line spectra were revealed. Pea plants responded to discrete-spectrum light by changing P700 content, the ratio of antenna chlorophyll to P700, and the position of the low-temperature peak of the temperature-induced chlorophyll fluorescence yield. In cucumber plants, disturbances in the energy transfer from the short-wavelength pigments to chlorophyll a were observed. In both plants, the effects of line spectrum light on the pigment apparatus were reversible; the ratio of cyclic to noncyclic photophosphorylation, photosynthetic control, and the extent of the coupling of photosynthetic electron transport to photophosphorylation did not change compared to the control light. The need for examining the line spectra of light sources for growing plants under moderate intensities of artificial light (about 50 W/m2 of photosynthetically active radiation) is discussed.

Аннотация: New results obtained in the detection of rhythms in the low frequency range in the light signal of a small number of photo bacteria are reported. The luminescence of a small number of bacteria was recorded with an apparatus the sensitivity of which was 12 quanta/pulse. To check the possible rhythms in the luminescence of the bacteria, correlation analysis was made of the interpulse intervals of the detection of the signal with a computer. This revealed a definite rhythm in the bacterial luminescence the fundamental frequency of which lies in the region 8 c/s. Profound modulation of a bioluminescent signal at this and multiple frequencies was demonstrated. The harmonic components found in the spectrum of the signal refute the existing view of the continuous character of bacterial luminescence.

Аннотация: The crystal structure of the photoprotein obelin (22.2 kDa) from Obelia longissima has been determined and refined to 1.7 Angstrom resolution. Contrary to the prediction of a peroxide, the noncovalently bound substrate, coelenterazine, has only a single oxygen atom bound at the C2-position. The protein-coelenterazine 2-oxy complex observed in the crystals is photo-active because, in the presence of calcium ion, bioluminescence emission within the crystal is observed. This structure represents only the second de novo protein structure determined using the anomalous scattering signal of the sulfur substructure in the crystal. The method used here is theoretically different from that used for crambin in 1981 (4.72 kDa) and represents a significant advancement in protein crystal structure determination.

Аннотация: The paper describes the principal limitations of the traditional methods used to construct atmospheric models. These limitations would not allow any fundamental improvement of atmospheric modeling. Ways are proposed to overcome the current limitations, based oil the methodology of constructing adaptive models and neuroinformatics. Algorithms of self-adaptation for neural networks intended for the construction of atmospheric models are given. Essentially, the developed algorithms are adaptive shells and can be easily transferred to other models.

Аннотация: Spontaneous assembling of a simplest bacterial cell even if all necessary molecules are present in a solution seems to be extremely rare event and from the scientific standpoint has to be considered as impossible. Therefore, a predecessor of a living cell has to be very simple for providing its self-assembling and at the same time it should be able of progressive increase in complexity. Now phase-separated particles, first of all micelles, are put forward as possible predecessors of living cell. According to the offered working concept only phase-separated particles possessing autocatalytic properties can be considered as predecessors of living cells. The first stage of evolution of these phase-separated autocatalytic systems is the appearance of pre-biotic metabolism providing synthesis of amphiphiles for formation of capsules of these systems. This synthesis is maintained by the energy of a base reaction being a component of a planet-chemical cycle. Catalytic system providing functioning of pre-biotic metabolism is based on multivariate oligomeric autocatalyst, which reproduces itself from monomers, penetrating the particles from the outside. Since the autocatalyst realizes random polymerization then a collection of other oligomers possessing different catalytic functions is produced. Аннотация: Closed ecological systems are of two-fold interest - as models of the Earth's biosphere explorable in experiments and as a facility for long-term autonomous human life support beyond the Earth. Theoretical analysis and experimental implementation of highly closed manned systems has been the subject of studies at the Institute of Biophysics (Russian Academy of Sciences, Siberian Branch) for many years. BIOS systems of increasing complexity with complete regeneration of atmosphere, water and partially food have been realized. In BIOS-3 experiments the system inhabited by 2-3 researchers for 4-6 months maintained its metabolic equilibrium without any negative physiological effect on the crew, which proves its sustainable condition. Specific for BIOS-3 is internal control by the people inhabiting the system. So, BIOS-3 is the first experimental implementation of V.l. Vernadsky's idea about the noosphere - habitable Biosphere controlled by human intelligence. Contrary to predictions of many environmentalists the closedness of the ecosystem is a factor that does not reduce, but increases its sustainability and makes its use for reliable life support outside the Earth realistic. The system is sustainable owing to permanent feedback between the monitoring of few key parameters of the system and automatic corrective actions on them. Main object of control is photo-biosynthesis regenerating parameters of human habitat disturbed by his vital activities. This principle has been realized in BIOS system and proved its reliability in long-term experiments. A new challenge is specified-optimal increase of trophic closedness of the system by reproduction within it essential proteins (peptides and amino acids), lipids, vitamins and other essential compounds. Alternative lines of attack on this problem by state-of-the-art biotechnological methods, GMO including, are under analysis. Reduced BIOS version - without complete closure - can be a breakthrough instrument to improve the quality of life of people living under extreme conditions on the Earth - in polar latitudes (Arctic, Antarctic), in deserts, in high mountains.

Аннотация: The optical limiting (OL) of detonation nanodiamond (DND) suspensions in engine oil was studied at a temperature range of 20°C to 100°C. Oil suspensions were prepared on the basis of the DNDs with an average nanoparticle cluster size in hydrosols (Daver) of 50 and 110 nm. Raman spectroscopy was used to characterize the samples. The OL investigation was carried out by the z-scan technique. The fundamental (1064 nm) and second (532 nm) harmonic radiations of YAG:Nd3 laser with passive Q-switching as an excitation source were used. The OL thresholds for both suspensions at 532 and 1064 nm were determined. It is shown that a decrease in the average nanoparticle cluster size as well as an increase of the wavelength of the incident radiation leads to the OL threshold increase. It is established that the OL performance is not influenced by increasing the temperature from 20°C to 100°C. The results obtained show the possibility of using the DNDs suspensions in engine oil as an optical limiter in a wide temperature range. © 2017 Society of Photo-Optical Instrumentation Engineers (SPIE).

Аннотация: We report on a saturable absorption in aqueous dispersions of nanodiamonds with femtosecond laser pulse excitation at a wavelength of 795 nm. The open aperture Z-scan experiments reveal that in a wide range of nanodiamond particle sizes and concentrations, a lightinduced increase of transmittance occurs. The transmittance increase originates from the saturation of light absorption and is associated with a light absorption at 1.5 eV by graphite and dimer chains (Pandey dimer chains). The obtained key nonlinear parameters of nanodiamond dispersions are compared with those of graphene and carbon nanotubes, which are widely used for the mode-locking. © 2017 Society of Photo-Optical Instrumentation Engineers (SPIE).

Аннотация: Presently, the most promising way of studying, forecasting, and enhancing of organisms' tolerance to harsh environmental impacts is considered to be the estimation of initial functional state of an organism's regulatory systems. To resolve the problem of photosynthetic apparatus (PSA) tolerance to harsh impacts at the level of such a complicated functional system as a plant leaf, it is necessary to assess integral responses of the leaf's PSA to the impact. At that, simple and versatile traits may have certain advantages. At present, chlorophyll fluorescence seems to be one of the main indices of PSA activity, which can be measured relatively fast and easy. One of the possible approaches to operational integrative assessment of PSA activity may consist in usage of the parameters introduced for the curves of chlorophyll fluorescence induction (CFI) slow phase. Temporal patterns of CFI are of special interest. The simplest index T-0.5 (i.e., half-time of chlorophyll fluorescence intensity decrease during the slow phase of CFI) appears to be an integral characteristic of activation rate with regard to a number of photo-assimilation and photo-protective processes in leaves. On basis of the studies, conducted earlier, and published data, we have analyzed the behavior of T-0.5 parameter with comparison to other CFI traits (namely: qN - non-photochemical quenching coefficient, ETR - electron transport rate, Phi(PSII) - effective quantum yield of PSII photochemistry, F-p/F-T ratio) under different conditions. The influence of leaf senescence, changes in intensity of excitation light, slight dehydration of plants and their recovery from water deficiency have been examined. The pattern of T-0.5 behavior, observed in laboratory experiments, and the results of its comparison with other indices of CFI give occasion to propose the usage of T-0.5 for indirect estimation of PSA activity when operational integrative monitoring of PSA state is required. Further studies are necessary for establishing quantitative relationships between PSA activity and fluorescence parameter T-0.5 under specific stress conditions.

Аннотация: Ca2+-regulated photoproteins of ctenophores lose bioluminescence activity when exposed to visible light. Little is known about the chemical nature of chromophore photo-inactivation. Using a total synthesis strategy, we have established the structures of two unusual coelenterazine products, isolated from recombinant berovin of the ctenophore Beroe abyssicola, which are Z/E isomers. We propose that during light irradiation, these derivatives are formed from 2-hydroperoxycoelenterazine via the intermediate 8a-peroxide by a mechanism reminiscent of that previously described for the auto-oxidation of green-fluorescent-protein-like chromophores.

Аннотация: Bioluminescence of a number of marine organisms is conditioned by Ca2+-regulated photoprotein (CaRP) with coelenterazine as the reaction substrate. The reaction product, coelenteramide, at the first singlet excited state (S-1) is the emitter of CaRP. The S-1-state coelenteramide is produced via the decomposition of coelenterazine dioxetanone. Experiments suggested that the neutral S-1-coelenteramide is the primary emitter species. This supposition contradicts with theoretical calculations showing that the anionic S-1-coelenteramide is a primary product of the decomposition of coelenterazine dioxetanone. In this study, applying molecular dynamic (MD) simulations and the hybrid quantum mechanics/molecular mechanics (QM/MM) method, we investigated a proton-transfer (PT) process taking place in CaRP obelin from Obelia longissima for emitter formation. Our calculations demonstrate a concerted PT process with a water molecule as a bridge between anionic S-1-coelenteramide and the nearest histidine residue. The low activation barrier as well as the strong hydrogen-bond network between the proton donor and the proton acceptor suggests a fast PT process comparable with that of the lifetime of excited anionic S-1-coelenteramide. The existence of the PT process eliminates the discrepancy between experimental and theoretical studies. The fast PT process at emitter formation can also take place in other CaRPs.
