Аннотация: The biodegradability of oriented fibers made of polyhydroxybutyrate (P(3HB)) and its co-polymer with ?-hydroxyvalerate (P(3HB-co-3HV)) was investigated in buffer solutions and in biological media in vitro and in vivo. The fibers of both polymer types demonstrated resistance to hydrolytic degradation in buffer solutions at 38В°C and pH from 4.5 to 7.0 (for up to 180 days). It has been found that the biodegradation of the fibers in vitro in blood and serum and in vivo is accompanied by weight losses and minor changes in the microstructure with no significant losses in the tensile strength over a long time (up to 180 days). The biodegradation rate of the less crystalline co-polymer P(3HB-co-3HV) fibers was 1.4-2.0-times higher than that of the homopolymer P(3HB). It has also been shown that the degradation of the fibers in vivo is influenced both by tissue fluid enzymes and cells (macrophages and foreign-body giant cells). The fibers were eroded on the surface only with no gross defects and no dramatic effects on their mechanical performance. В© VSP 2005.

Аннотация: Kinetic indices of growth, polyhydroxyalkanoate (PHA) accumulation, and gas exchange have been studied in a culture of the carbon monoxide-resistant hydrogen strain Ralstonia eutropha B-5786 grown on a gaseous substrate (GS) obtained by lignite gasification. The GS was shown to be suitable for PHA production. To increase the degree of GS consumption, various modes of gas supply to the culture were tested. Based on the results, an algorithm was developed for calculating and controlling gas-exchange parameters in the PHA-accumulating culture of Ralstonia eutropha, grown on a new GS allowing high polymer yields (up to 75%) and degrees of the substrate utilization (up to 90%).

Аннотация: The main objective of a life support system for space missions is to supply a crew with food, water and oxygen, and to eliminate their wastes. The ultimate goal is to achieve the highest degree of closure of the system using controlled processes offering a high level of reliability and flexibility. Enhancement of closure of a biological life support system (BLSS) that includes plants relies on increased regeneration of plant waste, and utilization of solid and liquid human wastes. Clearly, the robustness of a BLSS subjected to stress will be substantially determined by the robustness of the plant components of the phototrophic unit. The aim of the present work was to estimate the heat resistance of two plants (wheat and lettuce) grown on human wastes. Human exometabolites mineralized by hydrogen peroxide in an electromagnetic field were used to make a nutrient solution for the plants. We looked for a possible increase in the heat tolerance of the wheat plants using changes in photosynthetically active radiation (PAR) intensity during heat stress. At age 15 days, plants were subjected to a rise in air temperature (from 23 В± 1 В°C to 44 В± 1 В°) under different PAR intensities for 4 h. The status of the photosynthetic apparatus of the plants was assessed by external 2 gas exchange and fluorescence measurements. The increased irradiance of the plants during the high temperature period demonstrated its protective action for both the photosynthetic apparatus of the leaves and subsequent plant growth and development. The productivity of the plants subjected to temperature changes at 250 W m-2 of PAR did not differ from that of controls, whereas the productivity of the plants subjected to the same heat stress but in darkness was halved. At ages 15 and 25 days, plants were subjected to a rise in air temperature (from 22-24В°C to 44В°C) under different PAR intensities for 4 hours. The status of the photosynthetic apparatus of the plants was assessed by external CO2 gas exchange and fluorescence measurements. The increased irradiance of the plants during the high temperature period demonstrated its protective action for both the photosynthetic apparatus of the leaves and subsequent plant growth and development. The productivity of the plants subjected to temperature changes at 250 W/m2 of PAR did not differ from that of controls, whereas the productivity of the plants subjected to the same heat stress but in darkness was halved. The heat resistance of the reproductive organs of 25-day-old plants was significantly lower than that of 15-day-old plants subjected to similar light and temperature conditions.

Аннотация: The usage of chlorophyll fluorescence induction (CFI) for estimating various types of plant resistance (primary, general, initial, adaptive) to stress factors is reviewed. The necessity of ontogenetic approach (considering the age-specific properties of the photosynthetic apparatus) in determining general and adaptive resistance of plants to prolonged action of stress factors by the CFI method is argued. In the plant Cucumbis sativus L., the possibility is shown of using age-specific qualitative and quantitative traits of leaf CFI (changes in the shape of chlorophyll fluorescence induction curves and in the dynamics of CFI parameters in the course of leaf ontogeny) for comparative study of differences between fully active and stressed plants. Possible criteria are suggested for estimating the effect of outer stress factors by the presence or absence of a steady-state phase in the dynamics of CFI parameters during leaf ontogeny. It is also suggested to use the duration of the steady-state phase following the termination of leaf growth (estimated by the dynamics of the slow phase of CFI as the ratio of fluorescence intensity at the peak P and the steady-state fluorescence intensity, FP/FS, or as the viability index Rfd) and the variability of CFI parameters during this period as qualitative estimates of plant resistance to prolonged action of stress factors.

Аннотация: The review deals with objective reasons that limit the use of chlorophyll fluorescence induction kinetics (Chl FIK) method in plant ecology. Based on the ontogenetic approach (analysis and comparison of the dynamics of the Chl fluorescence ratio Fp/Fs and physiological characteristics of plant leaves in ontogeny) possible criteria for the estimation of general plant resistance of photosynthetic apparatus (PSA) to prolonged stress affects are proposed. One of these criteria is the presence or absence of a steady-state phase in the dynamics of Chl fluorescence ratio Fp/Fs (or Rfd) of plant leaves after they stop growing. We also propose to use the duration of the steady-state phase and variability of Chl fluorescence ratios Rfd and Fp/Fs in this period for quantitative assessment of plant PSA resistance to prolonged stress during plant leaf ontogeny.

Аннотация: The effect of elevated temperatures of 35 and 45В°C (at the intensities of photosynthetically active radiation 322, 690 and 1104 ?mol-m-2-s-1) on the photosynthesis, respiration, and qualitative and quantitative composition of the volatiles emitted by wheat (Triticum aestuvi L., cultivar 232) crops was investigated in growth chambers. Identification and quantification of more than 20 volatile compounds (terpenoids - ?-pinene, ?3 carene, limonene, benzene, ?-and transcaryophyllene, ?- and ?-terpinene, their derivatives, aromatic hydrocarbons, etc.) were conducted by gas chromatograph/mass spectrometry. Under light intensity of 1104 ?mol-m-2-s-1, heat resistance of photosynthesis and respiration increased at 35В°C and decreased at 45В°C. The action of elevated temperatures brought about variations in the rate and direction of the synthesis of volatile metabolites. The emission of volatile compounds was the greatest under a reduced irradiation of 322 ?mol-m-2-s-1 and the smallest under 1104 ?mol-m-2-s-1 at 35В°C. During the repair period, the contents and proportions of volatile compounds were different from their initial values, too. The degree of disruption and the following recovery of the functional state depended on the light intensity during the exposure to elevated temperatures. The investigation of the atmosphere of the growth chamber without plants has revaled the substances that were definitely technogenic in origin: tetramethylurea, dimethylsulfide, dibutylsulfide, dibutylphthalate, and a number of components of furan and silane nature. Аннотация: Tomato (Lycopersicon esculentum Mill), sunflower (Helianthus annuus L.), and cucumber (Cucumis sativus L.) plants were grown under red or white light at a radiance of 115 W/m2 under controlled conditions. The red spectra were characterized by either two well-distinguished spectral lines (RL1) or only one (RL2). Apparent photosynthesis (Pa), gross photosynthesis (Pg), dark (Rd), and light (R1) respiration, the content of pigments, and the biochemical composition of plants were estimated. Enhancing the destructive red light influence led to a rise in the Pa/R1 and Pa/Pg ratios and to a decrease in chlorophyll a, nitrate, and reduced nitrogen. RL1 was more inhibitory for plants than RL2. It is concluded that declined energy efficiency of respiration contributes to the destructive effect of RL in sunflower and cucumber plants. The rearrangement of metabolic processes, at which energy efficiency of respiration is maintained, is responsible for adaptation of tomato plants to red light. The obtained results allow one to outline the ways to study the species-specific mechanisms of plant responses to prolonged RL action.

Аннотация: The study addresses interaction of bacteria and phages in the host-parasite system in batch and continuous cultures. The study system consists of the auxotrophic strain of Brevibacterium - Brevibacterium sp. 22L - and the bacteriophage of Brevibacterium sp., isolated from the soil by the enrichment method.1.Closed system. In the investigation of the relationship between the time of bacterial lysis and multiplicity of phage infection it has been found that at a lower phage amount per cell it takes a longer time for the lysis of the culture to become discernible. Another important factor determining cytolysis in liquid medium is the physiological state of bacterial population. Specific growth rate of bacteria at the moment of phage infection has been chosen as an indicator of the physiological state of bacteria. It has been shown that the shortest latent period and the largest output of the phage are observed during the logarithmic growth phase of bacteria grown under favorable nutrient conditions. In the stationary phase, bacterial cells become "a bad host" for the phage, whose reproduction rate decreases, and the lysis either slows down significantly or does not occur at all.2.Open system. It has been found that in continuous culture, the components of the host-parasite system can coexist over a long period of time. After phage infection, the sizes of the both populations vary for some time and then the density of the host population reaches the level close to that of the uninfected culture. The phage population copies the variations in the density of the host population, but in antiphase. It has been proven that the bacterium becomes resistant to the phage rather soon. It has been supposed that primary resistance is of physiological origin, because the percentage of cells that have survived lysis - about 0.2% of the initial bacterial population - is too high for phage-resistant mutants. Bacteria and phages cultured over extended periods of time in the host-parasite system have been found to co-evolve. The stressful effect of the phage causes development of bacteria resistant to this phage, then a mutant phage capable of lysing these bacteria evolves, etc. В© 2007 COSPAR.

Аннотация: Populations of Escherichia coli Z905/pPHL7, a transgenic microorganism, were heterogenic in the expression of plasmid genes when adapting to the conditions of water microcosms of various mineralization levels and structure of microbial community. This TM has formed two subpopulations (ampicillin- resistant and ampicillin-sensitive) in every microcosm. Irrespective of mineralization level of a microcosm, when E. coli Z905/pPHL7 alone was introduced, the ampicillin-resistant subpopulation prevailed, while introduction of the TM together with indigenous bacteria led to the dominance of the ampicillin-sensitive subpopulation. A high level of lux gene expression maintained longer in the freshwater microcosms than in sterile saline lake water microcosms. A horizontal gene transfer has been revealed between the jointly introduced TM and Micrococcus sp. 9/pSH1 in microcosms with the Lake Shira sterile water. В© 2005 COSPAR. Published by Elsevier Ltd. Аннотация: The problem of interaction between man and microorganisms in closed habitats is an inextricable part of the whole problem of co-existence between macro- and microorganisms. Concerning the support of human life in closed habitat, we can, conventionally, divide microorganisms, acting in life support system (LSS) into three groups: useful, neutral and harmful. The tasks, for human beings for optimal coexistence with microhabitants seem to be trivial: (1) to increase the activity of useful forms, (2) decrease the activity harmful forms, (3) not allow the neutral forms to become the harmful ones and even to help them to gain useful activity. The task of efficient management and control of microbial population's development in LSS highly depends on mission duration. As for short-term missions without recycling, the proper hygienic procedures are developed. For longer missions, the probability of transformation of the neutral forms into the harmful ones is becoming more dangerous. The LSS for long-term missions are to use cycling-recycling systems, including system with biological recycling. In these systems, microbial populations as regenerative link should be useful and active agents. Some problems of microbial populations control and management are discussed in the paper. Аннотация: It has been demonstrated that the transgenic microorganism Escherichia coli Z905/pPHL7 (Ap'Lux+) can exist for a long time at an elevated concentration of mineral salts. The microorganism was introduced into microcosms with sterile brackish water (salinity variable from 21 to 22 g 1-1) taken from Lake Shira (Khakasia, Russia). The survivof the microorganism was estimated both by measuring the growth of the colonies on solid nutrient media and by the bioluminescence exhibited by the transgenic strain in samples from the microcosms and in the enrichment culture with the added selective factor - ampicillin (50 ?g/ml). In the enrichment culture, the bioluminescent signal was registered through the 160-day experiment. It has been shown that in the closed microcosms with brackish water the E. coli strain becomes heterogeneous in its ampicillin resistance. The populations of the transgenic strain were mainly represented by isolates able to persist in the medium containing 50 ?g/ml, but there were also the cells (about 10%) with the threshold of ampicillin resistance not more than 0.05 ?g/ml. Thus, it was shown that in the microcosms with brackish water and in the absence of the selective factor the transgenic strain survives and retains the recombinant plasmid. В© 2003 COSPAR. Аннотация: The processes resulting from the introduction of the transgenic microorganism (TM) E. coli Z905/pPHL7 into aquatic microcosms have been modeled experimentally. It has been shown that the TM E. coli is able to adapt to a long co-existence with indigenous heterotrophic microflora in variously structured microcosms. In more complex microcosms the numerical dynamics of the introduced E. coli Z905/pPHL7 population is more stable. In the TM populations staying in the microcosms for a prolonged time, changes are recorded in the phenotypic expression of plasmid genes (ampicillin resistance and the luminescence level) and chromosome genes (morphological and physiological traits). However, in our study microcosms, the recombinant plasmid persisted in the TM cells for 6 years after the introduction, and as the population adapts to the conditions of the microcosms, the efficiency of the cloned gene expression in the cells is restored. In the microcosms with high microalgal counts (107 cells/ml), cells with a high threshold of sensitivity to ampicillin dominate in the population of the TM E. coli Z905/pPHL7. В© 2003 COSPAR. Аннотация: From 1996 to 1999 heterotrophic bacteria of the brackish-water Lake Shira (Republic of Khakasia, Russia) were studied to understand the seasonal dynamics of their antibiotic resistance. During the winter, these bacteria were represented primarily by forms that could not be cultured and were psychrotolerant. In the summer period, heterotrophic, mesophilic bacteria increased in number. The percentages of isolates with multiple, antibiotic resistance isolated from the lake region near the resort area of the lake were 2-3 times higher than those from the central part of the lake. A decline in the bacterial numbers with multiple antibiotic resistance was observed during the cold period (February-March). Various mechanisms of multiple, antibiotic resistance of heterotrophic bacteria isolated from Shira lake are discussed.

Аннотация: The effect of different concentrations of salts on natural and recombinant strains of Bacillus subtilis and Escherichia coli was studied. The recombinant strain of B. subtilis was found to be more osmotolerant than the wild-type strain of this bacterium, whereas the opposite situation was observed for the recombinant and wild-type strains of E. coli. Some salts exerted a bacteriostatic effect on E. coli and B. subtilis. The adaptive capacity of recombinant strains depended on the number of plasmid copies in the cells. Аннотация: The population heterogeneity of recombinant and plasmid-free Bacillus subtilis strains introduced into aquatic microcosms was studied. After introduction, the population of the plasmid-free strain B. subtilis 2335 in microcosms has long been represented by both vegetative cells and spores, whereas, already ten days after introduction, the population of the recombinant strain B. subtilis 2335/105 (KmI)Inf+) was represented only by spores. The number of plasmid copies in the spore isolates of the recombinant strain was the same as before introduction, but the plasmid abundance in the vegetative isolates of this strain decreased. The isolates of B. subtilis 2335/105 obtained from microcosms and the variants of this strain obtained by ten successive subcultures on M9 and 0.1?M9 media with and without kanamycin (Km) differed in the number of plasmid copies, Km resistance, and maximum biomass yield during batch cultivation. Irrespective of the presence of Km, more than 50% of the variants subcultured on M9 medium showed reduced plasmid abundance. At the same time, about 70% of the variants subcultured on 0.1 ? M9 medium with Km and 90% of the variants subcultured on the same medium without Km retained the initial number of plasmid copies. The variants subcultured on media with Km retained the initial biomass level. In more than 70% of the variants isolated from media without Km, the biomass yield increased.
