Аннотация: The authors have constructed and characterized a series of membranes based on resorbable polyhydroxyalkanoates of different compositions. Five PHA types have been studied: a homopolymer of 3-hydroxybutyric acid, copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids, 3-hydroxybutyric and 3-hydroxyvaleric acids, 3-hydroxybutyric and 3-hydroxyhexanoic acids. Scanning electron microscopy and atomicforce microscopy were used to examine the microstructure of membrane surfaces, showing that membranes based on the copolymer of 3-hydroxybutyrate and 3-hydroxyhexanoate had the roughest surface, while membranes based on the copolymer of 3-hydroxybutyrate and 3-hydroxyvalerate had the smoothest surface. The contact angle for water in air was smaller and hydrophilic properties better in the copolymer membranes than in the membranes based on the high-crystallinity homopolymer of 3-hydroxybutyric acid. The culture of mouse fibroblast cell line NIH 3,3 was used to test PHAbased membranes; results of fluorescent probes of DNA DAPI and the MTT assay show that membranes based on studied PHAs are not cytotoxic on direct contact with cells and are highly biocompatible; their adhesive properties and ability to maintain fibroblast proliferation are similar to those of polystyrene and better than those of polylactic acid membranes.

Аннотация: Microparticles obtained by using 14C-labeled resorbable hydroxyaminobutyric acid polymer were injected into the caudal vein of laboratory animals without negative aftereffects for their growth and development and without changes in the macro- and microstructure of organs and tissues. The distribution of microparticles in the viscera and the dynamics of accumulation of carbon-containing polymer degradation products in the viscera were studied. The main targets for the particles are liver tissues, as well as renal and splenic tissues. The polymeric matrix of the microparticles is most actively destroyed in the spleen and liver. The presence of high-molecular- weight polymeric matrix in organs indicates the integrity of microparticles and the possibility of long-term (up to 12 weeks) functioning of polymeric particles in vivo. Аннотация: Microspheres have been prepared from the resorbable linear polyester of ?-hydroxybutyric acid (polyhydroxybutyrate, PHB) by the solvent evaporation technique and investigated in vitro and in vivo. Biocompatibility of the microspheres has been proved in tests in the culture of mouse fibroblast cell line NIH 3T3 and in experiments on intramuscular implantation of the microspheres to Wistar rats for 3 months. Tissue response to the implantation of polymeric microspheres has been found to consist in a mild inflammatory reaction, pronounced macrophage infiltration that increases over time, involving mono- and poly-nuclear foreign body giant cells that resorb the polymeric matrix. No fibrous capsules were formed around polymeric microparticles; neither necrosis nor any other adverse morphological changes and tissue transformation in response to the implantation of the PHB microparticles were recorded. The results of the study suggest that polyhydroxybutyrate is a good candidate for fabricating prolonged-action drugs in the form of microparticles intended for intramuscular injection. Аннотация: Specimens of 14C-labeled polymer of 3-hydroxybutyric acid, P(3-HB), with different initial molecular weights, were used to prepare microparticles, whose morphology was not influenced by the M w of the polymer. During the particle preparation process, P(3-HB) molecular weight decreased by 15-20%. Sterile microparticles (mean diameter 2.4 ?m) were injected into the tail veins of Wistar rats (5 mg/rat). The effects of the particles administered to rats were studied based on the general response of animals and local response of internal organ tissues and blood morphology; no adverse effects on growth and development of the animals or unfavorable changes in the structure of the tissues of internal organs were observed. Measurements of radioactivity in tissues showed that 14C concentrations are different in different organs, changing during the course of the experiment. The main targets for 14C-labeled microparticles were tissues of the liver, spleen, and kidneys. Comparison of radioactivity levels and residual contents of high-molecular-weight matrix in tissues suggested that the most rapid metabolism and degradation of P(3-HB) occurred in the liver and spleen. Gel-permeation chromatography showed that at 3 h after the microparticles were injected into the bloodstream, polymer degradation started in all examined organs, except the lungs; at 12 weeks, the M w of the polymer matrix was as low as 20-30% of its initial value. The presence of high-molecular-weight (undegraded) polymer in the tissues at 12 weeks after administration of the particles suggests that P(3-HB) is degraded in tissues of internal organs slowly and, hence, P(3-HB)-based microparticles can be used as sustained-release drug-delivery systems. Аннотация: Series of cell carriers made of resorbable polyester Bioplastotan, which is a copolymer of 3- And 4-hydroxybutyric acid, are characterized in this work. With use of various technologies membranes, 3D scaffolds, and nanomatrixes formed by ultrafine fibers produced with electrostatic spinning were constructed. All types of scaffolds provide adhesion of multipotent mesenchymal stromal cells and are suitable for the cultivation and differentiation of cells in the osteoblastes, that confirmed by detection of extracellular calcium precipitates, alkaline phosphatase activity and osteopontin production in cell culture. Аннотация: This work is a review of the results of biomedical studies of polymer devices (films, fibers, microparticles, 30 implants) made from resorbable PHAs synthesized by the bacterium Wautersia (Ralstonia) eutropha 65786, using the technology developed at the Institute of Biophysics of the Siberian Branch of the Russian Academy of Sciences. Two types of PHAs - polyhydroxybutyrate (PHB) and a hydroxybutyrate/hydroxyvalerate copolymer (PHB/PHV) - have been proven to be biocompatible in vitro in cultures of fibroblasts, endothelial cells, hepatocytes, and osteoblasts, and in short- and long-duration experiments on animals. Polymer films and membranes have been found to be usable as scaffolds for functioning cells and monofilament suture fibers - for stitching muscular-fascial wounds and in abdominal surgery. Ectopic bone formation assay and experiments with the model of segmental osteotomy showed that 30 PHB and PHB/HA implants can be used for reparative osteogenesis. Аннотация: Antitumor efficiency of an experimental form of an experimental form of anthracyclin antibiotic (doxorubicin), resorbable microparticles from Bioplastotaneв„ў, was studied on laboratory mice with transplanted Ehrlich's solid carcinoma. Use of the experimental form of the cytostatic in polymeric microparticles from resorbable Bioplastotaneв„ў in animals with solid tumor led to inhibition of the cancerous process, comparable to that in response to intravenous free doxorubicin, but without negative effects on the blood system. Аннотация: The effect of the preparation technique (chemical composition of a polymer, type and method of emulsion mixing, and molecular mass of a drug) on the yield, structure, and size of microparticles obtained from resorbable polyesters of microbiological origin, polyhydroxyalkanoates, is studied. It is found that the concentration of the polymer solution and the method of emulsion mixing are the most significant factors affecting the diameter of microparticles based on polyhydroxyalkanoates; the surface structure of particles depends to a higher extent on the chemical composition of the polymer. The family of microparticles from 100-200 nm to 50-70 mu m in diameter is synthesized. It is shown that the rate of drug release from microparticles in vitro into the medium is higher in the case of 3-hydroxybutyrate copolymers with 3-hydroxyvalerate than in the case of the homopolymer of 3-hydroxybutyrate. This parameter increases with the content of 3-hydroxyvalerate units in the copolymer and the porosity and mass fraction of the drug in particles with a decrease in their sizes. For in vitro systems containing a phosphate buffer, variation in the preparation parameters makes it possible to obtain microparticles with various characteristics suitable for deposition of drugs. For microparticles obtained from polyhydroxyalkanoates and having different diameters, the mathematical description of the kinetics of drug release from the polymer matrix is provided.

Аннотация: Matrices based on resorbable polyhydroxyalkanoates (PHAs) of five types {a homopolymer of 3- hydroxybutyric acid, copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids [P(3HB/4HB)], 3-hydroxybutyric and 3-hydroxyvaleric acids [P(3HB/3HV)], 3-hydroxybutyric and 3-hydroxyhexanoic acids [P(3HB/3HHx)]} have been constructed and characterised. No significant differences have been found in tissue response to implantation of these PHAs. Non-coarse fibrous capsules that formed around PHA matrices reached their maximum thickness (60-90 ?m) 90 days after implantation; by day 180, the average thickness of the capsules had decreased by 1·5-2·3 times. The number of foreign body giant cells, resorbing PHAs, remained high. In vivo biodegradation behaviour of polymer matrices is related to the chemical composition of the PHA. Matrices prepared from copolymers P(3HB/4HB) and P(3HB/3HHx) exhibited the fastest degradation rates. P3HB/3HV matrices were degraded more slowly, and P3HB matrices were the most durable. In the PHA matrices that were degraded more slowly, giant cell reaction developed later. © 2014 Institute of Materials, Minerals and Mining.

Аннотация: A series of 3D implants and filling materials prepared from powdered biodegradable polymers, polyhydroxyalkanoates (PHAs), have been designed for the purposes of reparative osteogenesis. The 3D implants are made of resorbable polymer of hydroxybutyric acid (poly-3-hydroxybutyrate, P3HB) and a composite of this polymer with hydroxyapatite (HA) (P3HB/HA). The properties of the implants were studied in vivo in a model of segmental osteotomy and compared with commercial material Bio-Oss®. All implants containing P3HB as the main component facilitate reconstructive osteogenesis. P3HB and P3HB/HA show pronounced osteoplastic properties; their in vivo degradation is slow and corresponds to the growth of a new bone tissue, facilitating normal reparative osteogenesis. Also, powdered P3HB and P3HB/tienam can be used as filling materials for osteoplasty of bone cavities infected by Staphylococcus aureus. Biodegradable 3D implants and P3HB-based filling materials show pronounced osteoplastic properties and degrade in vivo at a slow rate, enabling normal reparative osteogenesis.

P(3HB?co?4HB) and P(3HB?co?3HHx) had the lowest Cx (42 and 49%) while P(3HB?co?3HV) and P(3HB) exhibited higher Cx values (76%). Polymer films were prepared from different PHAs. Electron microscopy showed differences in the surface microstructure of the films. Films prepared from the P(3HB) were more hydrophobic and the arithmetic mean surface roughness of 71–75 nm, than the copolymer films, which were hydrophilic (57–60°) and had considerably higher roughness (158–177 nm). Blood parameters (hemoglobin and hemolysis) and response of the cells (erythrocytes, platelets, and monocytes) were studied in experiments with blood directly contacting the surface of the films of PHAs with different compositions. Cultivation of blood cells on polymer films did not cause any adverse effects on adhesion and morphology of all cell types. Results of studying blood cell response suggested that the films made from low-crystallinity copolymers containing 4?hydroxybutyrate and 3?hydroxyhexanoate were the best for contact with blood. © 2019 Elsevier B.V.
