Аннотация: The surface properties of diamond nanoparticles (40-50 A in diameter) have been investigated by X-ray photoelectron spectroscopy (XPS), UV photoelectron spectroscopy (UPS) and Raman spectroscopy. The diamond nanoparticles have been deposited on flat Si(100) substrates by electrophoresis/dielectrophoresis. The as-deposited films are strongly oxidized and present a 1-2% nitrogen content. After treatment at 850 В°C in H2 plasma for 60 min, the oxygen is removed, and the position of the C 1s core-level peak indicates a n-type electronic comportment of the diamond nanoparticles. Raman spectroscopy of the as-deposited film shows a sp3 contribution at 1321 cm-1 and a sp2 contribution around 1620 cm-1. The 12 cm-1 shift of the sp3 contribution with respect to the bulk diamond peak at 1333 cm-1 is attributed to a phonon confinement effect due to the size of the diamond particles. The H2 plasma treatment induces a size decrease of the nanocrystallites confirmed by Raman and scanning electron microscopy (SEM) measurements. UPS spectroscopy shows a negative electron affinity of -0.2 eV of the hydrogenated nanodiamond film.

Аннотация: The purpose of this study was to develop micro and nano sized drug carriers from poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV), and study the cell and skin penetration of these particles. PHBV micro/nanospheres were prepared by o/w emulsion method and were stained with a fluorescent dye, Nile Red. The particles were fractionated by centrifugation to produce different sized populations. Topography was studied by SEM and average particle size and its distribution were determined with particle sizer. Cell viability assay (MTT) was carried out using L929 fibroblastic cell line, and particle penetration into the cells were studied. Transdermal permeation of PHBV micro/nanospheres and tissue reaction were studied using a BALB/c mouse model. Skin response was evaluated histologically and amount of PHBV in skin was determined by gas chromatography-mass spectrometry. The average diameters of the PHBV micro/nanosphere batches were found to be 1.9 mu m, 426 and 166 nm. Polydispersity indices showed that the size distribution of micro sized particles was broader than the smaller ones. In vitro studies showed that the cells had a normal growth trend. MTT showed no signs of particle toxicity. The 426 and 166 nm sized PHBV spheres were seen to penetrate the cell membrane. The histological sections revealed no adverse effects. In view of this data nano and micro sized PHBV particles appeared to have potential to serve as topical and transdermal drug delivery carriers for use on aged or damaged skin or in cases of skin diseases such as psoriasis, and may even be used in gene transfer to cells.

Аннотация: Polymer poly(3-hydroxybutyrate) [P(3HB)] has been used as a matrix in slow-release formulations of the herbicide metribuzin (MET). Physical P(3HB)/MET mixtures in the form of solutions, powders, and emulsions were used to construct different metribuzin formulations (films, granules, pellets, and microparticles). SEM, X-Ray, and DSC proved the stability of these formulations incubated in sterile water in vitro for long periods of time (up to 49 days). Metribuzin release from the polymer matrix has been also studied. By varying the shape of formulations (microparticles, granules, films, and pellets), we were able to control the release time of metribuzin, increasing or decreasing it. © 2016 Taylor & Francis Group, LLC.

Аннотация: The long-term operation of three reactors and the radiochemical plant of the Mining-and-Chemical Combine (MCC), Russia's largest producer of weapons-grade plutonium, has resulted in radioactive contamination of the Yenisei River floodplain. From 1995 to 2016, we found more than 200 radioactive particles (RP) in the Yenisei floodplain, downstream of the MCC. Analytical characterization showed that most of the RP were fuel particles, which were carried into the river after incidents at the MCC reactors. Having compared the Cs-137/Cs-134 ratios in the particles, we determined three time intervals when the RP were formed. The plutonium isotope ratios (Pu-238/Pu-239,Pu-240) vary substantially between the particles and indicate several different source terms. In addition to fuel RP, we found particles that only contained activation products (Co-60 or europium isotopes). SEM and gamma-spectrometry showed that the cobalt particles could have originated from the corrosion of the reactor coolant system and the europium particles -from the damaged compensating rods. No europium particles have been found anywhere else in the world. The presence of RP from different sources (fuel, cobalt, and europium particles) in the Yenisei River floodplain makes this region a unique site for studying environmental effects of the particles. These RP represent point sources of radioecological significance.

Аннотация: The present study describes production of bacterial cellulose composites with silver nanoparticles and antibiotics and compares their properties. Bacterial cellulose (BC) composites synthesized in the culture of the strain of acetic acid bacterium Komagataeibacter xylinus VKPM B-12068 with silver nanoparticles, BC/AgNps, were produced hydrothermally, under different AgNO3 concentrations (0.0001, 0.001, and 0.01 M) in the reaction medium. The presence of silver in the BC/AgNp composites was confirmed by elemental analysis conducted using scanning electron microscopy with a system of X-ray spectral analysis. Analysis showed that the average atomic number of silver particles in composite samples depended on the concentration of AgNO3: as AgNO3 concentration in the reaction solution was increased, silver content in the composites increased from 0.044 to 0.37 mg/cm2. BC composites with amikacin and ceftriaxone were prepared by immersing dry BC films in solutions containing different concentrations of the antibiotics. The surface structure and properties and physicochemical and mechanical characteristics of composites were investigated using SEM, DSC, X-ray analysis, the system for measuring water contact angles, and electromechanical tensile testing machine. The disk-diffusion method and the shake-flask culture method used in this study showed that all experimental composites had pronounced antibacterial activity against E. coli, Ps. eruginosa, K. pneumoniae, and St. aureus, and the BC/antibiotic composites were more active than BC/AgNp ones; S. aureus was the most susceptible to the effect of BC composites. No potential cytotoxicity was detected in any of the BC/AgNp composites in the NIH 3T3 mouse fibroblast cell culture, in contrast to the BC/antibiotic composites. These results suggest that BC composites constructed in the present study hold promise as dressings for managing wounds, including contaminated ones. © 2017 Elsevier Ltd

Аннотация: A strain of acetic acid bacteria, Komagataeibacter xylinus B-12068, was studied as a source for bacterial cellulose (BC) production. The effects of cultivation conditions (carbon sources, temperature, and pH) on BC production and properties were studied in surface and submerged cultures. Glucose was found to be the best substrate for BC production among the sugars tested; ethanol concentration of 3% (w/v) enhanced the productivity of BC. Optimization of medium and cultivation conditions ensures a high production of BC on glucose and glycerol, up to 2.4 and 3.3 g/L/day, respectively. C/N elemental analysis, emission spectrometry, SEM, DTA, and X-ray were used to investigate the structure and physical and mechanical properties of the BC produced under different conditions. MTT assay and SEM showed that native cellulose membrane did not cause cytotoxicity upon direct contact with NIH 3T3 mouse fibroblast cells and was highly biocompatible. © 2018 Springer-Verlag GmbH Germany, part of Springer Nature

Screening of biopolymeric materials for cardiovascular surgery toxicity—Evaluation of their surface relief with assessment of morphological aspects of monocyte/macrophage polarization in atherosclerosis patients / N. G. Menzyanova [et al.] // Toxicol. Rep. - 2019. - Vol. 6. - P74-90, DOI 10.1016/j.toxrep.2018.11.009 . - ISSN 2214-7500

Atherosclerosis -- Cell morphology -- Intravascular stenting -- Macrophages -- Monocytes -- Polyhydroxyalkanoates
Аннотация: The morphotypes of human macrophages (MPh) were studied in the culture on nano-structured biopolymer substrates, made from polyhydroxyalcanoates (PHAs) of five various monomer compositions, followed by the solvent evaporation. Its surface relief, which was further in direct contact with human cells in vitro, was analyzed by atomic force microscopy (AFM) and scanning electron microscopy (SEM). It was shown, that the features of the micro/nano relief depend on the monomeric composition of the polymer substrates. Monocytes (MN) of patients with atherosclerosis and cardiac ischemia, undergoing stenting and conventional anti-atherosclerotic therapy, were harvested prior and after stenting. MN were isolated and cultured, with the transformation into MPh in direct contact with biopolymer culture substrates with different monomer composition and nano-reliefs, and transformed into MPh, in comparison with the same process on standard culture plastic. Sub-populations of cells with characteristic morphology in each phenotypic class were described, and their quantitative ratios for each sample of polymers were counted as an intermediate result in the development of “smart” material for cardiovascular devices. The results obtained allow us to assume, that the processes of MPh differentiation and polarization in vitro depend not only on the features of the micro/nano relief of biopolymer substrates, but also on the initial state of MN in vivo and general response of patients. © 2018 The Authors

antioxidant -- dispersion -- freeze-drying -- Grape seed extract -- proanthocyanidins -- soluplus
Аннотация: Objective: The main objective of this work was to formulate a nanodispersion containing grape seed extract and analyzed its release profile, antioxidant potential of the prepared formulations. Methods: The grape seed extract (GSE) containing proanthocyanidins (PC’s) has been dispersed in polymer matrix soluplus (SOLU) by the freeze-drying method. The morphological analysis was carried out using atomic force microscopy (AFM), scanning electron microscopy (SEM) and Transmission electron microscopy (TEM). The in-vitro release of the nanodispersion formulations was evaluated by simulated intestinal fluid (SIF). The antioxidant activity of GSE and the formulation were evaluated by employing various in-vitro assays such as 2, 2’-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), 2, 2-diphenyl-1- picrylhydrazyl (DPPH), Ferric reducing antioxidant power (FRAP) and peroxidation inhibiting activity. Results: The formulation FIII (1:5) resulted in a stable formulation with a higher loading efficiency of 95.36%, a particle size of 69.90 nm, a polydispersity index of 0.154 and a zeta potential value of ?82.10 mV. The antioxidant efficiency of GSE-SOLU evaluated by DPPH was found to be 96.7%. The ABTS and FRAP model exhibited a dose-dependent scavenging activity. Linoleic model of FIII formulation and GSE exhibited a 66.14 and 86.58% inhibition respectively at 200 µg/l. Conclusions: The main reason for excellent scavenging activity of the formulations can be attributed to the presence of monomeric, dimeric, oligomeric procyanidins and the phenolic group. The present work denotes that GSE constitutes a good source of PC’s and will be useful in the prevention and treatment of free radical related diseases. © 2020, © 2020 Informa UK Limited, trading as Taylor & Francis Group.

Аннотация: The study reports results of using a CO2-laser in continuous wave (3 W; 2 m/s) and quasi-pulsed (13.5 W; 1 m/s) modes to treat films prepared by solvent casting technique from four types of polyhydroxyalkanoates (PHAs), namely poly-3-hydroxybutyrate and three copolymers of 3-hydroxybutyrate: with 4-hydroxybutyrate, 3-hydroxyvalerate, and 3-hydroxyhexanoate (each second monomer constituting about 30 mol.%). The PHAs differed in their thermal and molecular weight properties and degree of crystallinity. Pristine films differed in porosity, hydrophilicity, and roughness parameters. The two modes of laser treatment altered these parameters and biocompatibility in diverse ways. Films of P(3HB) had water contact angle and surface energy of 92? and 30.8 mN/m, respectively, and average roughness of 144 nm. The water contact angle of copolymer films decreased to 80–56? and surface energy and roughness increased to 41–57 mN/m and 172–290 nm, respectively. Treatment in either mode resulted in different modifications of the films, depending on their composition and irradiation mode. Laser-treated P(3HB) films exhibited a decrease in water contact angle, which was more considerable after the treatment in the quasi-pulsed mode. Roughness parameters were changed by the treatment in both modes. Continuous wave line-by-line irradiation caused formation of sintered grooves on the film surface, which exhibited some change in water contact angle (76–80? ) and reduced roughness parameters (to 40–45 mN/m) for most films. Treatment in the quasi-pulsed raster mode resulted in the formation of pits with no pronounced sintered regions on the film surface, a more considerably decreased water contact angle (to 67–76? ), and increased roughness of most specimens. Colorimetric assay for assessing cell metabolic activity (MTT) in NIH 3T3 mouse fibroblast culture showed that the number of fibroblasts on the films treated in the continuous wave mode was somewhat lower; treatment in quasi-pulsed radiation mode caused an increase in the number of viable cells by a factor of 1.26 to 1.76, depending on PHA composition. This is an important result, offering an opportunity of targeted surface modification of PHA products aimed at preventing or facilitating cell attachment. © 2021 by the authors. The water contact angle of copolymer films decreased to 80-56 degrees and surface energy and roughness increased to 41-57 mN/m and 172-290 nm, respectively. Continuous wave line-by-line irradiation caused formation of sintered grooves on the film surface, which exhibited some change in water contact angle (76-80 degrees) and reduced roughness parameters (to 40-45 mN/m) for most films. Treatment in the quasi-pulsed raster mode resulted in the formation of pits with no pronounced sintered regions on the film surface, a more considerably decreased water contact angle (to 67-76 degrees), and increased roughness of most specimens.

Аннотация: The application of nanobiotechnology is an emerging area of nanoscience and nanotechnology. Nanodi-amond has been a potent antibacterial, antifungal, antioxidant, and antiplatelet agent. In the present study, nanodiamonds were reduced by green synthesis and characterization was done through SEM, TEM, FTIR, and XRD. Further they were tested for their biological applications. The antimicrobial activity was investigated/studied/examined? through well diffusion method. The best activity was observed against Trichoderma reesei (16 mm) at 140 ?g/ml. The antioxidant activity was investigated through DPPH and FRAPS method. It was observed that the biologically reduced nanodiamonds reduce the Fe3+ ions to Fe2+ ions at 600 mM/l/g concentration. In DPPH assay, inhibitory concentration was found to be 4.58 ?g/ml. Further the antiplatelet activity was investigated by prothrombin time (PT) and activated partial throm-boplastin time (APTT) assay, and it was observed that biologically reduced nanodiamonds have potent an-tiplatelet activity. © 2021 Sumy State University

Аннотация: The development of inexpensive, easy-to-produce, and easy-to-use analytical tools for detection of harmful and toxic substances is a relevant research problem with direct applications in environmental monitoring and protection. In this work, we propose a novel composite material based on alumina nanofibers and detonation nanodiamonds for detection of phenol in aqueous medium. The composite material was obtained by mixing an aqueous suspension of alumina nanofibers with a diameter of 10-15 nm and a length of several microns and a hydrosol of nanodiamonds with an average cluster size of 70 nm. The mechanisms underlying the interaction of these nanomaterials are clarified and the physicochemical properties of the composite are investigated. The SEM and TEM studies show that the obtained composite has a network structure, in which clusters of nanodiamonds (10-20 nm in diameter) are distributed over the surface of nanofibers. Coupling of nanomaterials occurs due to opposite signs of their zeta potentials, which results in electrostatic attraction and subsequent chemical bonding as indicated by the X-ray photoelectron spectroscopy and simultaneous thermal analysis. The bonding apparently occurs between functional groups (mainly carboxyl) on the surface of nanodiamonds and amphoteric hydroxyl groups on the surface of alumina nanofibers. The proposed composite allows an easy-to-perform colorimetric analysis for qualitative and quantitative determination of phenol in aqueous samples with linear response over a wide range of concentrations (0.5-106 mu M). Multiple tests have shown that the composite is reusable and retains its catalytic function for at least 1 year during storage at room temperature.
