Аннотация: In a brackish, temperate, 24-m-deep Lake Shira, the profiles of salinity, temperature, oxygen and sulfide concentrations were measured on a seasonal basis from 2002 to 2009. The lake was shown to be meromictic with autumnal overturn restricted to mixolimnion. The depth of mixolimnion and position of oxic-anoxic interface varied annually. The spring mixing processes contribute to the formation of mixolimnion in autumn. The exceptionally windy spring of 2007 caused the deepening of mixolimnion in the winter of 2008. The winter position of oxic-anoxic interface was affected by the position of lower boundary of mixolimnion in all winters. The salinity in the winter mixolimnion increased compared with the autumn because of freezing out of salts from the upper water layers meters during ice formation and their dissolution in water below. The profiles of salinity and temperature were simulated by the mathematical 1-D model of temperature and salinity conditions taking into account ice formation. The simulated profiles generally coincided with the measured ones. The coincidence implies that simplified one-dimensional model can be applied to roughly describe salinity and density profiles and mixing behavior of Lake Shira. В© 2010 The Author(s).

Аннотация: Microspheres have been prepared from the resorbable linear polyester of ?-hydroxybutyric acid (polyhydroxybutyrate, PHB) by the solvent evaporation technique and investigated in vitro and in vivo. Biocompatibility of the microspheres has been proved in tests in the culture of mouse fibroblast cell line NIH 3T3 and in experiments on intramuscular implantation of the microspheres to Wistar rats for 3 months. Tissue response to the implantation of polymeric microspheres has been found to consist in a mild inflammatory reaction, pronounced macrophage infiltration that increases over time, involving mono- and poly-nuclear foreign body giant cells that resorb the polymeric matrix. No fibrous capsules were formed around polymeric microparticles; neither necrosis nor any other adverse morphological changes and tissue transformation in response to the implantation of the PHB microparticles were recorded. The results of the study suggest that polyhydroxybutyrate is a good candidate for fabricating prolonged-action drugs in the form of microparticles intended for intramuscular injection. Аннотация: It is shown experimentally that 241Am activity accumulated by elodea is mainly contained in leaves (up to 80%) and about 20% - in the stem. Specific capacity of leaves in respect to americium is considerably dependent on the plant age that is due to formed on elodea leaf surface organo-mineral sediment consisting of mineral salts, epiphytic microorganisms and exometabolites. Am accumulation is larger in the older leaves (located lower on the stem) - nearly 70% of activity of americium accumulated by the whole plant biomass.

Аннотация: Use of halophytes (salt-tolerant vegetation), in a particular vegetable Salicornia europaea plants which are capable of utilizing NaCl in rather high concentrations, is one of possible means of NaCl incorporation into mass exchange of bioregenerative life support systems. In preliminary experiments it was shown that S. europaea plants, basically, could grow on urine pretreated with physicochemical processing and urease-enzyme decomposing of urea with the subsequent ammonia distillation. But at the same time inhibition of the growth process of the plants was observed. The purpose of the given work was to find out the influence of excessive quantities of some mineral elements contained in products of physicochemical processing of urine on the production process and NaCl accumulation by S. europaea plants. As the content of mineral salts in the human liquid wastes (urine) changed within certain limits, two variants of experimental solutions were examined. In the first variant, the concentration of mineral salts was equivalent to the minimum salt content in the urine and was: K - 1.5 g/l, P - 0.5 g/l, S - 0.5 g/l, Mg - 0.07 g/l, Ca - 0.2 g/l. In the second experimental variant, the content of mineral salts corresponded to the maximum salt content in urine and was the following: K - 3.0 g/l, P - 0.7 g/l, S - 1.2 g/l, Mg - 0.2 g/l, Ca - 0.97 g/l. As the control, the Tokarev nutrient solution containing nitrogen in the form of a urea, and the Knop nutrient solution with nitrogen in the nitrate form were used. N quantity in all four variants made up 177 mg/l. Air temperature was 24 В°C, illumination was continuous. Light intensity was 690 ?mol/m2s of photosynthetically active radiation. NaCl concentration in solutions was 1%. Our researches showed that the dry aboveground biomass of an average plant of the first variant practically did not differ from the control and totaled 11 g. In the second variant, S. europaea productivity decreased and the dry aboveground biomass of an average plant totaled 8 g. The increase of K quantity in the experimental solutions resulted in an elevated content of the element in the plants. The increase of K uptake in the second experimental variant was accompanied by a 30-50% decrease of Na content in comparison with the other variants. Comparative Na content in the other variants was practically identical. N, Mg and P content in the control and experimental variants was also practically identical. The increase of S quantity in the second experimental variant also increased S uptake by the plants. But Ca quantity, accumulated in aboveground plants biomass in the experimental variants was lower than in the control. NaCl uptake by plants, depending on the concentration of mineral salts in the experimental solutions, ranged from 8 g (maximum salt content) up to 15 g (minimum salt content) on a plant growth area that totaled 0.032 m2. Thus, high concentrations of mineral salts simulating the content of mineral salts contained in urine did not result in a significant decrease of S. europaea productivity. The present work also considers the influence of higher light intensity concentrations on productivity and NaCl accumulation by S. europaea plants grown on experimental solutions with high salt content. Аннотация: The life support systems (LSS) for long-term missions are to use cycling-recycling systems, including biological recycling. Higher plants are the traditional regenerator of air and producer of food. They should be used in many successive generations of their reproduction in LSS. Studies of influence of microbial associations on germination of wheat seeds and on growth of seedlings under impact of heavy metals are necessary because of migration of heavy metals in LSS. Microbial associations are able to stimulate growth of plants, to protect them from pathogenic organisms and from toxicity of heavy metal salts. The goal of this work was to investigate effect of microbial associations on the germination of wheat seeds and on the growth of seedlings under impact of different concentrations of ZnSO4. The results of investigations showed that:(1)Zinc salt had an adverse effect on germination of wheat seeds, beginning with concentrations of 8 MPC (Maximum Permissible Concentration) and higher.(2)Microbial associations (concentrations -104 to 107 cells/ml) were able to decrease (partly or completely) the adverse effect of ZnSO4 on germination of wheat seeds.(3)Concentrations (104-107 cells/ml) of microbial associations were able to decrease partly the adverse effect of zinc salts (intervals: from 1 to 32 MPS{cyrillic}) on the growth and development of wheat plantlets during heterotrophic phase.(4)The root system of plants was more sensitive to the adverse effect of ZnSO4 than shoots of plants. Аннотация: Populations of Escherichia coli Z905/pPHL7, a transgenic microorganism, were heterogenic in the expression of plasmid genes when adapting to the conditions of water microcosms of various mineralization levels and structure of microbial community. This TM has formed two subpopulations (ampicillin- resistant and ampicillin-sensitive) in every microcosm. Irrespective of mineralization level of a microcosm, when E. coli Z905/pPHL7 alone was introduced, the ampicillin-resistant subpopulation prevailed, while introduction of the TM together with indigenous bacteria led to the dominance of the ampicillin-sensitive subpopulation. A high level of lux gene expression maintained longer in the freshwater microcosms than in sterile saline lake water microcosms. A horizontal gene transfer has been revealed between the jointly introduced TM and Micrococcus sp. 9/pSH1 in microcosms with the Lake Shira sterile water. В© 2005 COSPAR. Published by Elsevier Ltd. Аннотация: It has been demonstrated that the transgenic microorganism Escherichia coli Z905/pPHL7 (Ap'Lux+) can exist for a long time at an elevated concentration of mineral salts. The microorganism was introduced into microcosms with sterile brackish water (salinity variable from 21 to 22 g 1-1) taken from Lake Shira (Khakasia, Russia). The survivof the microorganism was estimated both by measuring the growth of the colonies on solid nutrient media and by the bioluminescence exhibited by the transgenic strain in samples from the microcosms and in the enrichment culture with the added selective factor - ampicillin (50 ?g/ml). In the enrichment culture, the bioluminescent signal was registered through the 160-day experiment. It has been shown that in the closed microcosms with brackish water the E. coli strain becomes heterogeneous in its ampicillin resistance. The populations of the transgenic strain were mainly represented by isolates able to persist in the medium containing 50 ?g/ml, but there were also the cells (about 10%) with the threshold of ampicillin resistance not more than 0.05 ?g/ml. Thus, it was shown that in the microcosms with brackish water and in the absence of the selective factor the transgenic strain survives and retains the recombinant plasmid. В© 2003 COSPAR. Аннотация: The effect of different concentrations of salts on natural and recombinant strains of Bacillus subtilis and Escherichia coli was studied. The recombinant strain of B. subtilis was found to be more osmotolerant than the wild-type strain of this bacterium, whereas the opposite situation was observed for the recombinant and wild-type strains of E. coli. Some salts exerted a bacteriostatic effect on E. coli and B. subtilis. The adaptive capacity of recombinant strains depended on the number of plasmid copies in the cells. Аннотация: In monocultures of micro-organisms, growth is controlled by feedback mechanisms involving chemical factors such as limiting substrates and inhibitory metabolic products. The role of such feedback in the growth regulation of Escherichia coli O-124 was investigated by growing cells in batch culture using a medium containing glucose and mineral salts. In various phases of growth, portions of the native culture were diluted with culture filtrate, so that although cell density decreased, the chemical composition of the growth medium was unaltered. As the diluted cultures grew, variations in growth acceleration were calculated and compared with those of native (undiluted) cultures. Towards the end of the exponential phase and in the growth deceleration phase, the specific feedback level (FBL) was between -20 and -200 (h g l-1)-1. The feedback components resulting from changes in glucose concentration were calculated using experimentally determined values of mu(max) (0.55 +/- 0.05 h-1) and K(s) (2.5 +/- 0.7 mg l-1). Only 0.1-40% of FBL could be accounted for by changes in glucose concentration, indicating the presence of additional growth regulators. The method developed may become a new tool for determination of growth-regulating cell-density factors in microbial cultures.

Аннотация: The influence of some mineral salts on the recombinant strain Escherichia coli Z9051 was investigated. It was shown that the composition (NaCl, Na2SO4, MgCl2 and MgSO4) and concentration (5 and 10%) of the salts substantially affect the expression of genes for the luminescence system of fight-emitting bacteria cloned in the plasmid under the control of the lac-promoter. In some cases, the luminescence level of the microorganism in the presence of salts was similar to the luminescence level under catabolite repression by glucose, the more strong influence of the salts exceeding the effect of catabolite repression. The possibility of adaptation of the genetically modified microorganism to the salinity factor is discussed.

Аннотация: The vertical structure of the ecosystem in the euphotic zone of the western Equatorial Pacific has been studied. The studies were based on a continuous sounding of the bioluminescence field, with simultaneous, vertically aimed sampling made with the aid of a 5l water bottle and plankton nets. The bioluminescence field has a two-maxima structure with a more pronounced and permanent lower maximum found in the oligotrophic regions as deep as 60 to 100 m. In the narrow (10 m) layer of maximum bioluminescence, the concentration of zooplankton, as well as the concentration, activity and production of bacteria and phytoplankton, are several times higher than those in immediately adjacent waters at greater or lesser depth. At the same time, the concentration of nutrient salts in that layer diminishes sharply and approaches zero in the overlying water. Analysis of the data obtained shows that the production processes above the lower maximum layer are dependent not only on the turbulent influx of nutrient salts from the underlying layers, but also on the horizontal supply from the zone of upwelling. A pelagic community, in its different time aspects from the moment of water ascending until its sinking in the convergence zone, is suggested as a common system for modelling. Аннотация: The study addresses the effect produced by different inorganic salts and detergents (SDS, Triton X-100, the Tween series) on the ATP-dependent bioluminescent reaction catalyzed by the luciferase of the new earthworm species Fridericia heliota (Annelida: Clitellata: Oligochaeta: Enchytraeidae). It has been shown that the effect of divalent metal salts on luminescence is determined by the action of cations. Three of them - Mg2+, Mn2+ and Ca2+ - can stimulate luciferase activity at concentrations varying within a wide range, and Mn2+ can act as a 100%-effective substitute for Mg2+ in F. heliota luminescence reaction in vitro. The inhibitory effect of monovalent metal salts on luminescence is largely determined by the action of the anion part of the molecule. The effectiveness of the inhibitory effect of anions increases in the following order: {Mathematical expression}. Of the sodium salts, dodecyl sulfate, which is an anionic detergent, produces the strongest inhibitory effect on luciferase. On the contrary, nonionic detergents produce a stimulatory effect on the F. heliota luciferase. The action of the most effective of them - Triton X-100 - is determined by its ability to reduce the actual concentration of lipid inhibitors in the reaction mixture. В© 2006 Elsevier B.V. Аннотация: Action of metallic salts on the bacterial bioluminescent systems (1 - water-soluble coupled system luciferase - NADH:FMN-oxydoreductase; 2 - immobilized in starch gel coupled system luciferase - NADH:FMN-oxydoreductase; 3 - intact bacteria) has been studied. The action parameters have been calculated. The effects have been shown to depend on nature of cations but not anions. Electronic structure of cations (nature of external orbitals, charge, radii) has been taken into account. Cations with similar electronic shells nature affect on the row of the bioluminescent systems mentioned above in a similar way. Differences in effects have been explained with diffusion rate, cellular shell and respiration of bacteria.

Аннотация: Effects of temperature on bioluminescent patterns of luciferases from luminescent bacteria Vibrio harveyi, Vibrio fischeri, Photobacterium leiognathi, and Photobacterium phosphoreum were studied. The highest luminescence level was observed at 15-25 degrees C for the luciferase from P. phosphoreum, at 20-30 degrees C for the V. fischeri and P. leiognathi enzymes, and at 30-37 degrees C for the enzyme from V. harveyi. All the luciferases were significantly stabilized at increased salt concentrations, at low pH values, or in the presence of dithiothreitol (DTT) and EDTA. The addition of DTT and EDTA affected the reversible stage of enzyme inactivation, while salts reduced the rate of the irreversible stage. A peak corresponding to aggregated protein was detected by gel chromatography of irreversibly inactivated luciferase. Activation energies were calculated for each luciferase in bioluminescent reactions with decanal, dodecanal, tetradecanal, and without aldehydes. The activation energy of the reaction with tetradecanal was much lower than those with the other aldehydes. The temperature dependence of the lifetime of the long-lived reaction intermediate showed that in the 10-30 degrees C interval all the luciferases, except for the enzyme from V. harveyi, have only one active form.

Аннотация: The influence of several suites of pollutants (metallic salts, quinones, and phenols) on bacterial bioluminescence in vivo and in vitro (five test systems) was investigated. The sensitivity of bioluminescence to the different pollutants was evaluated, and inhibition constants were measured. The data obtained were shown to correlate with the physical and chemical characteristics of the substances and the structure of the bioluminescent systems. It has been found that three bioluminescent tests (water-soluble enzyme systems, immobilized enzyme systems, and bioluminescent bacteria) show higher sensitivity to pollutants and cover all types of widespread contamination. Аннотация: The action of met al salts on the luciferase-NADH: FMN-oxydoheductase bacterial luminescent system in vitro was studied. The effect is shown to depend on the nature of cations rather than anions, The type of the effect (activation, inhibition, and absence of the effect) is determined by the structure of the; cation, the nature and occupancy of its external orbitals, and charge. The results were interpreted on the basis of the hypothesis that upper electron excited states of the emitter are involved in the bioluminescence process. Based on thermodynamic calculations, the type of the effect was related to the energetic characteristics of the cation, Presumably, the mechanism of action of cations is determined by their involvement in the formation and deactivation of electron excited states in the bioluminescence process.

Аннотация: The effects of potassium halides KCl, KBr and KI on NADH:FMN-oxidoreductase-luciferase bioluminescent coupled enzyme system were studied. The influence of salt additions on bioluminescence intensity and bioluminescence light yield was investigated. The inhibition and activation parameters of the salts were calculated using their dependencies on concentration of the salts. The correlation between the inhibition of bioluminescence intensity and the halide mass was demonstrated: the inhibiting ability of the salts increases with the increase of atomic weight of the anions.
