Аннотация: Biodegradability patterns of two PHAs: a polymer of 3-hydroxybutyric acid (3-PHB) and a copolymer of 3-hydroxybutyric and 3-hydroxyvaleric acids (3-PHB/3-PHV) containing 11 mol% of hydroxyvalerate, were studied in the tropical marine environment, in the South China Sea (Nha Trang, Vietnam). No significant differences have been observed between degradation rates of 3-PHB and 3-PHB/3-PHV specimens; it has been found that under study conditions, biodegradation is rather influenced by the shape of the polymer item and the preparation technique than by the chemical composition of the polymer. Biodegradation rates of polymer films in seawater have been found to be higher than those of compacted pellets. As 3-PHB and 3-PHB/3-PHV are degraded and the specimens lose their mass, molecular weight of both polymers is decreased, i.e. polymer chains get destroyed. The polydispersity index of the PHAs grows significantly. However, the degree of crystallinity of both PHAs remains unchanged, i.e. the amorphous phase and the crystalline one are equally disintegrated. PHA-degrading microorganisms were isolated using the clear-zone technique, by inoculating the isolates onto mineral agar that contained PHA as sole carbon source. Based on the 16S rRNA analysis, the PHA-degrading strains were identified as Enterobacter sp. (four strains), Bacillus sp. and Gracilibacillus sp.В© 2010 Elsevier Ltd. All rights reserved.

Аннотация: During the summers of 1999-2001 the dynamics of polyhydroxyalkanoate degradation in a small recreational eutrophic reservoir was studied experimentally. It has been shown that biodegradation of polyhydroxyalkanoates in the environment is determined by the structure and physicochemical properties of the polymer and by local weather conditions, which influence the state of the aquatic ecosystem. Species (clones) of bacteria able to utilize polyhydroxyalkanoates in the reservoir were identified using molecular phylogenetic analysis of 16S rRNA genes. В© 2007 Elsevier Ltd. Аннотация: The paper analyzes the stability of areas in the Global Ocean, which are quasistationary with respect to the seasonal dynamics of chlorophyll concentration. Such areas were first discovered based on the CZCS data. They occur almost in all regions of the Global Ocean. They reflect typical seasonal dynamics of phytoplankton, which depends on both large ocean currents and local hydrological instability. It is related to the displacement of frontal zones and to local hydrological anomalies. Since the completion of the CZCS work there has been a 10 years gap in monitoring chlorophyll spatial distribution dynamics in the ocean. The reason is the long absence of equipment on the orbit, which would be suitable for the measurement of chlorophyll concentration. Recently, SeaWiFS data have been accumulated and their quantity is sufficient for the crude statistical analysis of image time series for 25 years. Thus, it has become possible to obtain data on the long-term stability of quasistationary areas and, therefore, on the stability of large-scale hydrological phenomena in the Atlantic Ocean. Аннотация: The purpose of this study is to estimate the possible effect produced on plant productivity by inedible plant biomass added to soil-like substrate (SLS). Results of the experiments with radish plants grown on the SLS with inedible biomass of carrot and beet plants added in the amounts roughly equal to their yields harvested from the same area showed a significant reduction in productivity of radish plants. The yield of radish plants grown on the SLS with added radish tops was almost the same as the yield of the radish grown on the neutral substrate. Experiments with addition of dry wheat straw to the SLS and growing of wheat and radish plants on that substrate also showed that the productivity of the plants grown in that way was decreased. Attempts to negate the adverse effect of plant wastes proved that the most effective way was to mineralize the wastes using the technique of "wet incineration". В© 2008 Elsevier Ltd. Аннотация: Stability of LSS based on biological regeneration of water, air and food subject to damaging factors is largely dependent on the behavior of the photosynthesizing component represented, mainly, by higher plants. The purpose of this study is to evaluate the tolerance of uneven-aged wheat and radish cenoses to temperature effects different in time and value. Estimation of thermal tolerance of plants demonstrated that exposure for 20 h to the temperature increasing to 45В°C brought about irreversible damage both in photosynthetic processes (up to 80% of initial value) and the processes of growth and development. Kinetics of visible photosynthesis during exposure to elevated temperatures can be used to evaluate critical exposure time within the range of which the damage of metabolic processes is reversible. With varying light intensity and air temperature it is possible to find a time period admissible for the plants to stay under adverse conditions without considerable damage of metabolic processes. Аннотация: Functional, regulatory and indicator features of microorganisms in development and functioning of the systems and sustaining stability of three man-made ecosystem types has been studied. 1) The functional (metabolic) feature was studied in aquatic ecosystems of biological treatment of sewage waters for the reducer component. 2) The regulatory feature of bacteria for plants (producer component) was studied in simple terrestrial systems "wheat plants-rhizospheric microorganisms - artificial soil" where the behavior of the system varied with activity of the microbial component. For example with atmospheric carbon dioxide content elevated microbes promote intensification of photosynthesis processes, without binding the carbon in the plant biomass. 3) The indicator feature for the humans (consumer component) was studied in Life Support Systems (LSS). High sensitivity of human microflora to system conditions allowed its use as an indicator of the state of both system components and the entire ecosystem. В© 2001 COSPAR. Аннотация: Изучена стабильность рекомбинантного штамма Escherichia coli Z905, несущего гибридную плазмиду pPHL(Ap{r} Lux{+}), которая содержит гены люминесцентной системы Photobacterium letognathi, клонированные в векторе pUC18. Люминесценция этого штамма подвержена сАМР зависимой катаболитной репрессии. При лимитировании роста популяции бактерий в хемостате как глюкозой, так и глицерином в присутствии антибиотика (50 мкг/мл) доля плазмидсодержащих клеток в популяции за 8-10 генераций резко снижается до 1% и ниже. После 20 генераций обнаружены клетки, образующие несветящиеся, но устойчивые к ампициллину колонки (Ap{r} Lux{-}). При лимитировании роста глюкозой их доля не превышает 0,01%, а глицерином - 18%. Аннотация: Electron microscopical studies of the nucleoid structure of hydrogen bacteria using ultrahin sections and spread DNA from bacterial cell lysates revealed a different DNA packaging in the cell. A compact state of the major part of DNA at all growth stages and stability of nucleosome-like structures were shown. The use of antibodies to HU protein of E. coli labelled by protein A-colloidal gold demonstrated the immunological relationship between HU protein of E. coli and histone-like proteins of Alcaligenes eutrophus and their possible role in the nucleosome-like DNA packaging in procariotic genome.

Аннотация: Six basic phase portraits differing in character and in the number of their equilibrium behaviour regimes were suggested in the classification of forest insect population dynamics. The portraits graphically illustrate six types of dynamic behavior of numbers of the system 'phytophage-entomophage': two types of stable equilibrium dynamics (stationary and oscillatory) and four types corresponding to outbreaks of mass reproduction - fixed, permanent, reversive and true outbreaks, respectively. In this paper we consider the single parameter model, with its parametric domains realizing a wide spectrum of phase dynamics, both basic and transitional. The changes in the phase portraits are accompanied by the bifurcations in the model. The results of these analyses are in agreement with the 'stability principle of mobile ecological systems' and may supply its parameter substantiation at the model level.

Аннотация: The emission of different, diamond coatings on silicon field emitter. arrays were investigated: chemical vapor deposition diamond, high pressure and high temperature synthetic diamond, and shock-synthesized nanodiamond. Practical emission characteristics such as: emission threshold, maximum current, current stability, and reproducibility were tested. Аннотация: The temperature dependence of bioluminescent activity of the Ca2+-activated photoprotein obelin from the hydroid polyp Obelia longissima and thermoinactivation of this protein at different concentrations of (NH4)2SO4 have been studied. The maximal intensity of luminescence of obelin was observed at 4-15-degrees-C. The activity of the photoprotein is completely stable to storage for 3 days at room temperature. Increasing the temperature to 40-degrees-C resulted in a 25-30% loss of enzyme activity in 1 h. The presence of ammonium sulfate during heating stabilizes the activity of obelin. Two breaks, at 11 +/- 3-degrees-C and 47 +/- 3-degrees-C, are observed in the Arrhenius plot of the first-order rate constant of the luminescence decay. The bioluminescent curves of obelin are biphasic in the temperature range 10-40-degrees-C. It is assumed that obelin may exist in two kinetically distinct conformers (active and inactive) whose ratio is temperature dependent.

Аннотация: Many proteins require a non-covalently bound ligand to be functional. How ligand binding affects protein conformation is often unknown. Here we address thermal unfolding of the free and ligand-bound forms of photoprotein obelin. Fluorescence and far-UV circular dichroism ( CD) data show that the various ligand-dependent conformational states of obelin differ significantly in stability against thermal unfolding. Binding of coelenterazine and calcium considerably stabilizes obelin. In solution, all obelin structures are similar, except for apo-obelin without calcium. This latter protein is an ensemble of conformational states, the populations of which alter upon increasing temperature. (C) 2012 Federation of European Biochemical Societies. Аннотация: It has been shown that the coelenterazine analog, coelenterazine-v, is an efficient substrate for a reaction catalyzed by Renilla luciferase. The resulting bioluminescence emission maximum is shifted to a longer wavelength up to 40 nm, which allows the use of some "yellow" Renilla luciferase mutants for in vivo imaging. However, the utility of coelenterazine-v in small-animal imaging has been hampered by its instability in solution and in biological tissues. To overcome this drawback, we ligated coelenterazine-v to Ca2+-triggered coelenterazine-binding protein from Renilla muelleri, which apparently functions in the organism for stabilizing and protecting coelenterazine from oxidation. The coelenterazine-v bound within coelenterazine-binding protein has revealed a greater long-term stability at both 4 and 37 degrees C. In addition, the coelenterazine-binding protein ligated by coelenterazine-v yields twice the total light over free coelenterazine-v as a substrate for the red-shifted R. muelleri luciferase. These findings suggest the possibility for effective application of coelenterazine-v in various in vitro assays.

Аннотация: The recombinant coelenterazine-dependent luciferases (isoforms MLuc 164 and MLuc39) from the marine copepod Metridia longa were expressed as inclusion bodies in E. coli cells, dissolved in 6 M guanidinium chloride and folded in conditions developed for proteins containing intramolecular disulfide bonds. One of them (MLuc09) was obtained in an active monomeric form with a high yield. The luciferase bioluminescence is found to be initiated not only by free coelenterazine, but also by Ca2+-dependent coelenterazine-binding protein (CBP) of Renilla muelleri on Ca2+ addition. The use of CBP as a "substrate" provides higher light emission and simultaneously the lower level of background. The high purity MLuc39 can be detected down to attomol with a linear range extending over 5 orders of magnitude. The MLuc39 reveals also a high stability towards heating and chemical modification; the chemically synthesized biotinylated derivatives of the luciferase preserve 35-40% of the initial activity The luciferase applicability as an in vitro bioluminescent reporter is demonstrated in model tandem bioluminescent solid-phase microassay combining the Ca2+-regulated photoprotein obelin and the Metridia luciferase.

Аннотация: Two kinds of Ca(2+)-regulated photoprotein obelin with altered color of bioluminescence were obtained by active-center amino acid substitution. The mutant W92F-H22E emits violet light (lambda(max)=390 nm) and the mutant Y139F emits greenish light (lambda (max)=498 nm), with small spectral overlap, both display high activity and stability and thus may be used as reporters. For demonstration, the mutants were applied in dual-color simultaneous immunoassay of two gonadotropic hormones-follicle-stimulating hormone and luteinizing hormone. Bioluminescence of the reporters was simultaneously triggered by single injection of Ca(2+) solution, divided using band-pass optical filters and measured with a two-channel photometer. The sensitivity of simultaneous bioluminescence assay was close to that of a separate radioimmunoassay.
