Аннотация: The main analytical use of Ca2+-regulated photoproteins from luminous coelenterates is for real-time non-invasive visualization of intracellular calcium concentration ([Ca2+](i)) dynamics in cells and whole organisms. A limitation of this approach for in vivo deep tissue imaging is the fact that blue light emitted by the photoprotein is highly absorbed by tissue. Seven novel coelenterazine analogues were synthesized and their effects on the bioluminescent properties of recombinant obelin from Obelia longissima and aequorin from Aequorea victoria were evaluated. Only analogues having electron-donating groups (m-OCH3 and m-OH) on the C6 phenol moiety or an extended resonance system at the C8 position (1-naphthyl and alpha-styryl analogues) showed a significant red shift of light emission. Of these, only the alpha-styryl analogue displayed a sufficiently high light intensity to allow eventual tissue penetration. The possible suitability of this compound for in vivo assays was corroborated by studies with aequorin which allowed the monitoring of [Ca2+](i) dynamics in cultured CHO cells and in hippocampal brain slices. Thus, the alpha-styryl coelenterazine analogue might be potentially useful for non-invasive, in vivo bioluminescence imaging in deep tissues of small animals.

Аннотация: The integral bioluminescent biotest with lyophilized fluorescent bacteria was used for monitoring of LPO processes in tissue extracts and serum of rats exposed to stress. A relationship between the content of MDA (LPO indicator) and fluorescence of bacteria was observed in all biological samples.

Аннотация: The distribution in mice of intravenously injected modified nanodiamonds (MNDs) obtained by detonation synthesis was studied using electron paramagnetic resonance (EPR) spectrometry. It has been shown that 2.5 h after MND injection into the tail vein of mice, the nanoparticles accumulate mainly in the lungs and liver of animals; much smaller amounts of nanoparticles were found in the kidneys and heart. The presence of MNDs in the samples of blood, spleen, brain, and thigh muscles of mice was not detected within the sensitivity of the method used.

Аннотация: In vitro experiments proved the usefulness of electron paramagnetic resonance (EPR) spectrometry for detecting detonation nanodiamonds (NDs) in samples of biomaterials (blood and homogenates of organs of mice). A characteristic EPR signal (g = 2.003, Delta H similar or equal to 10 G) was detected in biomaterials containing NDs, and its intensity linearly increased at nanoparticle concentrations of between 1.6 and 200 mu g/ml. In vivo experiments demonstrated that EPR spectrometry was effective for monitoring the inter-organ distribution of NDs intravenously injected to mice. In 2.5 h after the injection of NDs, the nanoparticles mainly accumulated in the lungs and liver of the animals - about 25 and 20%, respectively, of the initially injected NDs. The amounts of NDs accumulated in the heart and kidneys were considerably lower. Also, EPR spectrometry did not detect NDs in the blood, spleen, brain, and femoral muscles of mice. Ten days after injection, EPR spectrometry detected redistribution of NDs in mice. The amounts of nanoparticles decreased approximately by a factor of 3.5 in the lungs and increased almost by a factor of 3 in the liver; NDs were detected in the spleen. This study suggests ways to use EPR spectrometry to study the distribution, accumulation, and elimination of detonation NDs injected into laboratory animals.

Аннотация: Considerable efforts have been focused on shifting the wavelength of aequorin Ca2+? dependent blue bioluminescence through fusion with fluorescent proteins. This approach has notably yielded the widely used GFP?aequorin (GA) Ca2+ sensor emitting green light, and tdTomato-aequorin (Redquorin), whose bioluminescence is completely shifted to red, but whose Ca2+ sensitivity is low. In the present study, the screening of aequorin mutants generated at twenty?four amino acid positions in and around EF?hand Ca2+?binding domains resulted in the isolation of six aequorin single or double mutants (AequorinXS) in EF2, EF3, and C?terminal tail, which exhibited markedly higher Ca2+ sensitivity than wild?type aequorin in vitro. The corresponding Redquorin mutants all showed higher Ca2+ sensitivity than wild?type Redquorin, and four of them (RedquorinXS) matched the Ca2+ sensitivity of GA in vitro. RedquorinXS mutants exhibited unaltered thermostability and peak emission wavelengths. Upon stable expression in mammalian cell line, all RedquorinXS mutants reported the activation of the P2Y2 receptor by ATP with higher sensitivity and assay robustness than wt?Redquorin, and one, RedquorinXS?Q159T, outperformed GA. Finally, wide?field bioluminescence imaging in mouse neocortical slices showed that RedquorinXS?Q159T and GA similarly reported neuronal network activities elicited by the removal of extracellular Mg2+. Our results indicate that RedquorinXS?Q159T is a red light?emitting Ca2+ sensor suitable for the monitoring of intracellular signaling in a variety of applications in cells and tissues, and is a promising candidate for the transcranial monitoring of brain activities in living mice. © 2020 by the authors. Аннотация: Long-chain polyunsaturated fatty acids of n-3 family (n-3 LC PUFAs) are physiologically essential compounds required for normal growth and development of animals, including humans. The ability of fish species to synthesize n-3 LC PUFAs varies significantly across different trophic levels. We have studied fatty acid (FA) content (mg/g of wet weight) and level (% of total FAs) in the brain, liver, heart, intestine, female and male gonads, muscle, and adipose tissues of commercially important wild freshwater Baikal grayling. Additionally, FA content and level of Baikal grayling juveniles have been studied. In all tissues of Baikal grayling, some LC PUFAs, namely, 24:5n-3 and 24:6n-3 (C24 PUFAs), have been found. These FAs are the intermediate products in the synthesis of docosahexaenoic acid (DHA, 22:6n-3) by the Sprecher pathway. The levels of C24 PUFAs in tissues differed significantly: the highest levels of C24 PUFAs were found in adipose tissue and the lowest values in the gonads of females, liver, brain, and head of juveniles. According to the dynamics of DHA and C24 PUFAs, the maximum rate of DHA synthesis is achieved in brain of Baikal grayling, while the lowest rate of DHA synthesis probably occurs in adipose tissue. Although all studied tissues had differences in the number of FAs and their levels, 16:0, 18:1n-9, 16:1n-7, 20:5n-3, and DHA dominated. Male gonads contained an extremely high level of furan FAs — presumably beneficial substances for human health. Аннотация: Present study investigated effect of dietary buckwheat in alleviating bisphenol A (BPA) mediated oxidative stress, concomitant sirtuin1 levels in serum, stomach, and liver of rats. Experimental group A and B ingested standard diet, C and D consumed buckwheat (30%); group A and C drank normal water, B and C had BPA contamination (10 mg L 1). Sirtuin1 mean B/A ratio nearing unity in all tissues reveals inertness of BPA towards sirtuin1. Dietary buckwheat improved sirtuin1 levels both in normal (mean C/A ratio of serum, 1.65; liver, 1.24; stomach, 1.78) and BPA fed state (mean D/B ratio of serum, 1.9; liver, 1.26; stomach, 1.75). Buckwheat augmented antioxidant status in BPA fed rats as seen in mean D/B ratio of serum (catalase, 2.4; glutathione reductase (GR), 1.33; Thiols, 1.2), liver (catalase, 2; GR, 2.5; Thiols, 1.36) and stomach (catalase, 1.31; GR, 1.5; Thiols, 1.33). Therefore, buckwheat counters BPA-led oxidative stress and modulates sirtuin1.
