Аннотация: The chemical composition of water conditioned by colonies of Moina macrocopa and involved in the regulation of their life cycle was analyzed in comparison with water samples from natural water bodies. Dechlorinated tap water was used as the control medium. The results showed that metabolism of crustaceans leads to an increase in the concentrations of phosphorus and nitrogen in all forms, the appearance of protein and carbohydrate fractions, and changes in the composition of lipid fractions in the water conditioned by their colonies. The contents of all organic components are very low, which makes their identification difficult. Concentrations of substances identified in the conditioned water do not exceed those occurring in nature. В© 2000 MAIK "Nauka/Interperiodica".

Аннотация: It has been demonstrated that the transgenic microorganism Escherichia coli Z905/pPHL7 (Ap'Lux+) can exist for a long time at an elevated concentration of mineral salts. The microorganism was introduced into microcosms with sterile brackish water (salinity variable from 21 to 22 g 1-1) taken from Lake Shira (Khakasia, Russia). The survivof the microorganism was estimated both by measuring the growth of the colonies on solid nutrient media and by the bioluminescence exhibited by the transgenic strain in samples from the microcosms and in the enrichment culture with the added selective factor - ampicillin (50 ?g/ml). In the enrichment culture, the bioluminescent signal was registered through the 160-day experiment. It has been shown that in the closed microcosms with brackish water the E. coli strain becomes heterogeneous in its ampicillin resistance. The populations of the transgenic strain were mainly represented by isolates able to persist in the medium containing 50 ?g/ml, but there were also the cells (about 10%) with the threshold of ampicillin resistance not more than 0.05 ?g/ml. Thus, it was shown that in the microcosms with brackish water and in the absence of the selective factor the transgenic strain survives and retains the recombinant plasmid. В© 2003 COSPAR. Аннотация: Manifestation of pleiotropic effects in the isogenic variants of the luminescent bacterium Photobacterium leiognathi 54 was investigated. The decrease or increase of the expression level of bioluminescence was caused by changes in lux operon regulation. The dynamics of the bioluminescence of dark and dim variants did not differ from the dynamics of the initial luminescent variant, but dependence of the level of luminescence intensity on the exogenous autoinducer of the lux operon was revealed. The investigated variants of P. leiognathi 54 inherited fairly stable morphological characteristics, colony architectonics, level of luminescence, and activity of some enzymes; variants with reduced bioluminescence formed colonies of the S type. Stable bright variants with S-and R-type colonies appeared both in the initial strain population and in the dark variant population, but with smaller frequency. Populations of the bright variant with R-type colonies were most heterogeneous; this can be determined by the lack of glucose repression of the bioluminescence in contrast to other investigated inherited variants of P. leiognathi. Аннотация: The foreign and Russian literature devoted to studying the effect of enhancing the growth of colonies of the cyanobacterium Microcystis aeruginosa Kutz em. Elenk. after their passage in a viable state through the digestive tract of filter-feeding aquatic animals (planktivorous fish, daphnia, and bivalves) has been analyzed. The main mechanisms of this effect are considered. Its role in the functioning of eutrophic reservoirs is discussed. The prospects and the need for further studies of the effect of enhancing Microcyctis growth after its viable passage through the digestive tracts of filter-feeding animals are shown for the development of a complete theory of the functioning of aquatic ecosystems.

Аннотация: Colonies produced by a consortium of nitrifying bacteria were studied using light and electron microscopy. The colonies were obtained by direct plating of inoculum from a two-stage nonsterile chemostat fermentor and by repeatedly passing the microbial community of the fermenter through selective media containing ammonium or nitrite. The colonies studied can be characterized by a specific combination of six types of cells differing in their ultrastructure and spatial location within the colony. The types of cells occurring within a given colony were found to depend on the nitrogen compound present in the medium. As a result of our study, morphological features of colonial bacterial communities were preliminarily identified. The proposed approach can be viewed as a method to describe microbial associations and communities.

Аннотация: Colonies of Flavobacterium sp. 56 and Flavobacterium sp. 22 were studied by electron microscopy without being washed off from solid nutrient medium and were shown to differ in the spatial arrangement and ultrastructure of their cells. Colonies of Flavobacterium sp. 56 contained a single type of cells, while those of Flavobacterium sp. 22 consisted of cells of two types differing in their ultrastructure. Within the colony volume, bacteria were oriented in a regular pattern characteristic of each strain studied. The evidence obtained relating to the ultrastructure, spatial orientation, and location of cells is sufficient for the unambiguous identification of bacterial strains and provides new information on colony architectonics.

Аннотация: Electron microscopy has shown that the colonies obtained as a result of joint growth of bacterial monocultures Flavobacterium sp. 56 and Bacillus cereus 60 consist of three main cell types. Two cell types of the mixed colonies preserve the fine structure specific for bacteria in the monocultures. At the same time, formation of a large amount of the third type of cells is stimulated, which occur as single cells in the monoculture Flavobacterium sp. 56. The cell types are not intermixed in the colony volume and have certain spatial positions.

Аннотация: Six-day-old colonies of Flavobacterium sp. 22 were studied by electron microscopy. Direct evidence was obtained of bacterial cell division across the entire colony volume, indicating that the colony growth of Flavobacterium sp. 22 is not purely peripheral. It is argued that the colony shape is determined not only by peripheral growth but also by physical forces acting upon a droplet of liquid on the surface. For bacterial colonies developing on solid nutrient media, the intercellular matrix plays the role of such a liquid.

Аннотация: The light- and electron-microscopic analysis of the colonies of a bacterial consortium capable of utilizing alkylsulfonates, which are the main ingredients of waste from the synthetic rubber industry, revealed the presence of eight types of cells. All types of cells were gram-negative and differed in shape, size, and the presence of capsule and cytoplasmic inclusions. Three types of cells were present in all the colonies studied. The presence of the other types of cells depended on the inoculum used and on the composition of the growth medium.

Аннотация: Colonies grown from vegetative Bacillus subtilis 2335 cells had a standard structure, with bacillar cells occupying the whole colony volume. At the same time, the colonies of this bacterium grown from germinated spores had an abnormal structure characterized by the location of cells in a surface layer 100-200 mum thick at the colony boundary with the air. The glycocalyx of the colonies grown from spores was characterized by a wetting angle theta(e) of 120degrees-160degrees, whereas that of the colonies grown from vegetative cells had an angle theta(c) as low as 5degrees-30degrees. It is suggested that spores and vegetative cells follow different strategies of substrate colonization and that the architectonics of bacterial colonies is determined by the physicochemical properties of the glycocalyx.

It has been shown that in the closed microcosms with brackish water the E. coli strain becomes heterogeneous in its ampicillin resistance. The wpopulations of the transgenic strain were mainly represented by isolates able to persist in the medium containing 50 mug/ml, but there were also the cells (about 10%) with the threshold of ampicillin resistance not more than 0.05 mug/ml. Thus, it was shown that in the microcosms with brackish water and in the absence of the selective factor the transgenic strain survives and retains the recombinant plasmid. (C) 2003 COSPAR. Аннотация: Diverse, multidirectional changes in biochemical composition of seston, plankton, and fish fry are detected in the coastal zone of a plain reservoir in the vicinity of a semiaquatic bird colony during vegetation season with extra high water level. Under these conditions, specific patterns of phyto- and zooplankton structure and quantitative characteristics, that, as was found out earlier, result from the vital activity of birds belonging to the family Ardeidae (namely, raising the share of mixotrophic phytoflagellates and copepods in plankton species composition) are leveled out. The main role in this process belongs to decomposition of immersed vegetation in the coastal zone, development of littoral macrophytes, and trophic relationships between algae and invertebrates. However, increase in atmospheric precipitation leads to increase in copepods abundance, which seems to be related to fast and intensive inflow of nitrogen-rich products of birds' vital activity. Due to this, during the whole period of observations, the following tendencies are revealed in biochemical composition of seston, plankton, and fish fry in the vicinity of the bird colony: 1) concentrations of carbon, nitrogen, and phosphorus and their ratios turn out to be auspicious for predominance of diatoms and mixotrophic phytoflagellates, rich in docosahexaenoic acid (DSA); 2) the abundance of phytoplankton, rich in polyunsaturated essential fatty acids (PEFA), favors propagation of those planktonic crustaceans which contain significantly more DSA and are more valuable food items for fish; 3) predominance of zooplankters, rich in PEFA, leads to higher concentration of PEFA in fry fish tissues, which, in certain periods, turns out to be advantageous for successful development and growth of dominant species of fish fry,such as roach, bream, white bream, and perch, by comparison with the test site.

Reliable inhibitors of colonies of all five test bacteria were nanoparticles FeP@Ag. CoP nanoparticles are reliable inhibitors of growth of 4 test bacteria (except for test bacteria Escherichia oli). NiP nanoparticles are reliable inhibitors of growth of 2 test bacteria: Escherichia oli and Klebsiella pneumoniae. Bacteria Escherichia oli were most sensitive to the effect of magnetic nanoparticles; and bacteria Pseudomonas aeruginosa and Staphylococcus aureus were most resistant to the effect of magnetic nanoparticles. The prospects of the method are in the possibility of multiple reuse of the magnetic particles with antimicrobial properties for bacterial decontamination of the studied sources of water and removal of magnetic nanoparticles from the treated liquids by electromagnet.
