The effect of radionuclide and heavy metal contamination of the Yenisei River on cytogenetics of aquatic plant Elodea canadensis / A. Bolsunovsky [et al.] // Radioprotection. - 2009. - Vol. 44, Is. 5. - P83-88, DOI 10.1051/radiopro/20095021 . - ISSN 0033-8451
Аннотация: The study was done to determine concentrations of radionuclides and heavy metals and to evaluate the frequency of chromosomal aberrations in samples of Elodea canadensis, a submerged plant, collected in different parts of the Yenisei River. Samples were collected in the area subjected to radioactive impact of theMining-and-Chemical Combine (MCC) at Zheleznogorsk and in the control areas, upstream of the MCC. The investigations showed that Elodea biomass in the area affected by MCC operation contained a long inventory of artificial radionuclides typical of the MCC discharges. Upstream of the MCC, in the control sampling areas, the sediments and the Elodea biomass contained only one artificial radionuclide - 137Cs. Thus, the exposure doses to Elodea shoots and roots upstream of the MCC are small (not more than 8 ?Gy/d) and the main contribution to them is made by natural radionuclides. At the MCC discharge site (the village of Atamanovo) and downstream of it, the total dose rate increases almost an order of magnitude, reaching its maximal values - 72 ?Gy/d for Elodea shoots and 58 ? Gy/d for roots. Cytogenetic investigations of Elodea roots showed that at the MCC discharge site (the village of Atamanovo) and downstream of it the occurrence of chromosomal aberrations in ana-telophase and metaphase cells of Elodea was considerably higher than in the control area. It is highly probable that this simultaneous dramatic increase in the total exposure rate and the occurrence of chromosomal aberrations in Elodea is associated with the radiation factor. It is suggested that Elodea is affected not only by the radiation factor but also by the chemical factor - toxicity of heavy metals. В© 2009 EDP Sciences.

Аннотация: A short survey and critical analysis of previously proposed criteria for growth control of populations of microorganisms in the chemostat are presented. Based on the analysis of a mathematical model of the steady-state of a microbial population in the chemostat, an adequate control criterion is suggested, along with a method to identify the corresponding regulating factors. The new control criterion is expressed as a product of the factor transformation coefficient and the biomass sensitivity coefficient (SC) with respect to the change of the factor at the chemostat inlet (referred to in the sequel as the biomass SC). The control criterion determines the strength of the control exerted by this or that factor. The method of determination of the regulating factors consists in experimental determination of the real SCs for factors and the biomass and in calculating on this basis the corresponding ideal SCs for constant factor transformation coefficients. The ideal SCs are shown to add up to an integer value, a constraint that we call "quantization" relationships. Such relationships are used to test the completeness of the drawn list of control factors. The proposed method was applied to our own and literature data.

Аннотация: The paper summarises the results of a three-year research study (European Union Grant: INTAS 97-0519) aimed at investigating the planktonic populations and trophic organization of the Lake Shira ecosystem - a saline lake in Khakasia, Siberia. The lake exhibits a stable summer-autumn stratification of the chemical-biological components. The mechanisms responsible for the 'blooming' of phytoplankton in the deeper layers were investigated in greater detail, using data from both field and laboratory experiments. The spectra of nutrition were examined to estimate the relationships between the specific growth rates of the hydrobionts and the influence of the limiting factors: light, nutrients. The observed heterotrophic capability of a metalimnetic phytoplankton population might help explain the development in the deeper waters of Lyngbya contorta. The scheme of trophic interactions was put up, based on the assessment of the carbon pools and carbon flows in the pelagic zone of the lake. A mathematical model of the vertical structure of the lake's plankton populations was constructed, using the ecosystem description and data of vertical turbulent diffusion. The role of light and nutrient limitations and grazing mortality in forming the vertical inhomogeneities, particularly in lowering the depth of the maximal cyanobacterial biomass, has been demonstrated. The theoretical curves for the stratification of chemical and biological parameters have been brought in conformity with the field observations, e.g. for the different patterns of the peaks, and for the biomass maxima of cyanobacteria, purple and green sulphur bacteria, oxygen, and hydrogen sulphide. The calculations revealed that for an adequate assessment of the parameters for the hydrogen sulphide zone it is necessary to introduce flows of allochthonous organic matter. Based on the form of the sulphur distribution curve, the allochthonous input of organic matter and the inflow of hydrogen sulphide from the bottom have been theoretically discriminated for the first time. It has also been ascertained that irrespective of the depth the allochthonous substances limiting bacterial growth, the bacteria are uniformly distributed over depth and can serve as an indicator of the presence of limitation (the effect of autostabilisation in space). Of indisputable interest to limnology are the specific methods developed for understanding the functioning of Lake Shira ecosystem. These include the autostabilisation of the limiting factors, the on-the-spot fluorescent method of determining the three classes of microalgae, the algal mixotrophy and the planktonic population interactions and feedbacks, and development of a more sensitive, bioluminescent method for mapping the nonhomogeneities. Owing to a balanced combination of classical approaches (field observations, in situ data on production-decomposition) and the more recent ones (satellite monitoring, biophysical methods of estimating interactions of populations, mathematical models based on the field and experimental data), many of the structural-function relationships in the ecosystem can now be explained, and the models can provide 'mutual control and mutual agreement' between the data collected using different approaches.

Аннотация: Data on the seasonal changes in vertical heterogeneity of the physical-chemical and biological parameters of the thermally stratified Shira Lake ecosystem (Khakasia, Siberia) in 1996-2000 have been analyzed. The interaction mechanisms involving: (1) The plankton populations in aerobic and anaerobic zones, involving the cycling of carbon and sulphur, (2) the primary production limitation (by light and phosphorus) and inhibition (by light), and (3) the kinetic characteristics of plankton populations have been elucidated. A mathematical model of the vertical structure of the lake's plankton populations, based on the ecosystem description and on vertical turbulent diffusion of the matter, has been constructed. The green alga Dictyosphaerium tetrachotomum (Chlorophyta) and the cyanobacterium Lyngbya contorta (Cyanophyta), which dominated the phytoplankton biomass, were taken as oxygen producers. Arctodiaptomus salinus (a calanoid copepod) has been assumed as the main grazer in Shira Lake as it dominated the zooplankton biomass. Four groups of microorganisms involved in the sulphur cycle formation have been distinguished: sulphur, sulphur purple, sulphur green and SRB. H2S is oxidized to sulphate (only the green sulphur bacteria oxidize it to sulphur), and sulphate is reduced to H2S, forming neither sulphur nor its water-soluble compounds. The role of grazing, light and nutrient limitation, in forming the vertical inhomogeneities, particularly in lowering the depth of the maximal cyanobacterial biomass, has been demonstrated. When the model takes into account both light limitation and nutrient limitation of algal growth by P and consumption of algae by crustaceans: (a) in the scenario where the P is formed only by the cycling and decomposition of autochthonous organic matter, both the green algae and cyanobacteria are eliminated; (b) in the scenario involving an additional P flux in the deep water layers the peak of the cyanobacteria is at a depth of 10 m, and its amplitude is close to the one observed in the lake. The position of the peak remains stable owing to the 'double' limitation mechanism: light 'from above' and P 'from below'. Another mechanism responsible for the deep position of the peak of cyanobacteria was analyzed mathematically based on the model involving the experimentally proven assumption of the growth inhibition by light in the epilimnion and the light limitation in the hypolimnion. The main result is: the peak is positioned stable at its depth and does not change with time. The analytical and numerical calculations made for this positioning mechanism yielded the formulae relating the depth of the maximum of algal biomass, the 'width' of the peak base and the peak amplitude and a number of parameters (algae elimination, turbulent diffusion coefficient, sedimentation rate, light extinction coefficient and light intensity). The theoretical curves for the stratification of chemical and biological parameters have been brought in conformity with field observations, e.g. for the different patterns for the peaks, and the biomass maxima of cyanobacteria, purple and green sulphur bacteria, oxygen, and hydrogen sulphide. The calculations revealed that for an adequate assessment of the parameters for the hydrogen sulphide zone it is necessary to introduce flows of allochthonous organic matter. For the first time, theoretically, based on the form of the sulphur distribution curve, the allochthonous input of organic matter and the inflow of hydrogen sulphide from the bottom have been discriminated. The theoretical limit for the depth up to which the hydrogen-sulphide zone can ascend under the impact of allochthonous organic loading, has been determined.

Аннотация: The effect of nitrogen limitation in a medium on the composition of intracellular lipids in the alga Botryococcus braunii Kutz IPPAS H-252 in the course of culture development was investigated. Under the conditions of nitrogen limitation, the alga under investigation accumulated lipids as triacylglycerols, and this process was accompanied by substantial changes in the total fatty acid (FA) composition, which were manifested in a decrease in trienoic acids (from 52.8-57.2 to 19.5-24.7% of total FAs) and an increase in the content of oleic (from 1.1-1.2 to 17.1-24.4%) and saturated (from 23.7-26.0 to 32.9-46.1%) acids. In the control culture, the directionality of FA redistribution was less marked, and these changes were noticed at the later stages of culture development. Under nitrogen limitation, marked changes in the FA composition of polar lipids occurred by the 13th day, and they were characterized by an increase in the content of saturated acids (up to 76.8%) and a dramatic decrease in the content of all polyenoic acids (up to 6.8%). The changes in the FA composition of triacylglycerols were noticed as early as by the 7th day; these changes consisted in an increase in the content of oleic acid, and its high content (28.4-38.4%) was maintained up to the end of culturing. In the control culture, triacylglycerols with a high content of oleic acid were found by the 13th day, although, by this time, the content of total lipids and triacylglycerols did not change. Аннотация: A study has been carried out to investigate the influence of nitrogen deficiency on intracellular lipid composition, including total fatty acid composition of lipids, polar lipids, and triacylglycerols, of the alga Botryococcus braunii Kutz IPPAS H-252 in batch culture. Under nitrogen limitation, the alga accumulates lipids as triacylglycerols and the total fatty acid (FA) composition changes: trienoic acids decrease (from 52.8-57.2 to 19.5-24.7% of the total FAs) and the oleic acid increases (from 1.1-1.2 to 17.1-24.4%) as does the saturated acids (from 23.7-26 to 32.9-46.1%). A similar rearrangement in the FA spectrum occurs at later times in the control culture, but it is less pronounced. Under nitrogen limitation, considerable changes in the polar lipid FAs are registered at day 13: saturated acids increase (from 28.6-35.5 to 76.8%) and all polyenoic acids markedly decrease (from 56.9-64.1 to 6.8%). Changes in the triacylglycerol fatty acid spectrum are seen on day 7: the oleic acid increases (from 14.7 to 34.2%) and remains at a high level till the end of the culture. In the control, triacylglycerols with large contents of oleic acid are detected at day 13, the total lipids and triacylglycerols still remaining unchanged. В© Springer 2005.

Аннотация: Seasonal dynamics of contents of essential polyunsaturated fatty acids (PUFA) in dominant groups of benthic invertebrates: gammarids (Gammaridae, Amphipoda), oligochaetes (Oligochaeta), chironomid larvae (Chironomidae, Diptera) and caddisfly larvae (Trichoptera), and dominant benthivorous fish, Siberian grayling Thymallus arcticus, have been studied in ecosystem of the large Siberian river. During the year of the study most benthic invertebrate taxa showed significant variations in the contents of both C-18 and long chain C-20-22 PUFAs. In contrast, the fish, which consumed the zoobenthos, had no significant seasonal variations in long chain PUFAs' contents. Thereby, the fish, as organisms of relatively higher evolution level than invertebrates, was supposed to have more strict metabolic control of long chain PUFAs' contents. Evidence was obtained that the studied fish species, grayling, may be capable to convert dietary EPA into DHA. В© 2006 Elsevier Inc. All rights reserved.

Аннотация: Contents of fatty acids in filets of unfrozen (control), boiled, fried, roasted and boiled in a small amount of water humpback salmon, collected from a wholesale market in Krasnoyarsk city (Siberia, Russia) were analyzed. Special attention was paid to essential polyunsaturated fatty acids of ?3 family: eicosapentaenoic, 20:5?3 (EPA) and docosahexaenoic, 22:6?3 (DHA). Heat treatment in general did not decrease content of EPA and DHA in humpback, except a modest reduction during frying. Cooked humpback appeared to be the valuable source of essential ?3 PUFAs, namely EPA and DHA. It was hypothesized that the absence of significant reduction of PUFAs' contents in red flesh of fishes of Salmonidae family during heat treatment may be due to a high level of natural antioxidants which formed in the course of evolution as adaptation to their ecological niche. Аннотация: A set of bioluminescent tests was developed to monitor water quality in natural and laboratory ecosystems. It consisted of four bioluminescent systems: luminous bacteria, coupled enzyme system NADH:FMN-oxidoreductase-luciferase and triplet enzyme systems with alcohol dehydrogenase and trypsin. The set of biotests was applied for a small forest pond (Siberia, Russia), laboratory microecosystems polluted with benzoquinone and a batch culture of blue-green algae. Thereby effects of natural water compared to those of models of heavy pollution and "bloom" of blue-greens on the bioluminescent tests were revealed. The set of biotests was not affected by a natural seasonal variability of water quality in the unpolluted pond, but responded to the heavy pollution and the "bloom" of blue-greens. The set of biotests could be recommended as the alarm test to control the acute toxicity of natural water bodies. В© 2001 Elsevier Science Ltd.

Аннотация: The chemical composition of water conditioned by colonies of Moina macrocopa and involved in the regulation of their life cycle was analyzed in comparison with water samples from natural water bodies. Dechlorinated tap water was used as the control medium. The results showed that metabolism of crustaceans leads to an increase in the concentrations of phosphorus and nitrogen in all forms, the appearance of protein and carbohydrate fractions, and changes in the composition of lipid fractions in the water conditioned by their colonies. The contents of all organic components are very low, which makes their identification difficult. Concentrations of substances identified in the conditioned water do not exceed those occurring in nature. В© 2000 MAIK "Nauka/Interperiodica".

    The influence of food concentration and "water of aggregation" on the reproductive parameters of Moina macrocopa (Crustacea: Cladocera) females / E. S. Zadereev, V. G. Gubanov, I. V. Egorov // Zhurnal Obshchei Biologii. - 1998. - Vol. 59, Is. 1. - С. 55-57 . - ISSN 0044-4596
Аннотация: Combined influence of "water of aggregation" and food concentration on the reproductive parameters of Moina macrocopa females was investigated. In all experiments the influence of population density was associated with the effect of "water of aggregation" prepared earlier by keeping dense population (200 ?g/ml) of M. macrocopa. It was shown that life-span of M. macrocopa females in "water of aggregation" was not dependent on food concentration; under conditions of all tested food concentration it was less than in case of control clean water. There was no difference in proportion of males produced in "water of aggregation" and in control though more males were born under conditions of higher food concentration. In general "water of aggregation" causes a specific acceleration of female life history: the life-span and the period between clutches decrease, the amount of neonates in last clutches increases while the chance of male production at the beginning of reproduction period becames greater. The "water of aggregation" could also causes the formation of ephipial eggs instead of the first partenogenetic clutch. It seems that life strategy under conditions of very high mortality associated with "water of aggregation" is aimed to maximum realization of individual capacities in short period of time.

Аннотация: The change in reproductive mode from parthenogenesis to gametogenesis in Cladocera is controlled by multiple environmental cues. Maternal effects are involved in the control of reproductive switching. In this study, we estimated the readiness of Moina macrocopa females to change reproduction mode under the effect of conspecific chemicals on maternal and offspring generations. The results demonstrated that information about the chemical environment was not transmitted between generations (none of the females produced diapausing eggs in the control medium irrespective of their mothers' environment). Differences in maternal energetic investments were not significant, hence the maternal environment did not affect the fecundity. However, tested animals adapted to the effect of the diapause inducing factor. When offspring of mothers cultured in crowded water were also cultured in crowded water (the constant effect of conspecific chemicals), they switched less readily to gametogenesis than offsprings of mothers cultured in fresh medium.

Аннотация: The purpose of this work was to study the full-scale potential use of human mineralized waste (feces and urine) as a source of mineral elements for plant cultivation in a biological life support system (BLSS). Plants that are potential candidates for a photosynthesizing link were grown on a neutral solution containing human mineralized waste. Spring wheat Triticum aestivum L., peas Pisum sativum L. Ambrosia cultivar and leaf lettuce Lactuca sativa L., Vitaminny variety, were used. The plants were grown hydroponically on expanded clay aggregates in a vegetation chamber in constant environmental conditions. During plant growth, a determined amount of human mineralized waste was added daily to the nutrient solution. The nutrient solution remained unchanged throughout the vegetation period. Estimated plant requirements for macro-elements were based on a total biological productivity of 0.04 kg day-1 m-2. As the plant requirements for potassium exceeded the potassium content of human waste, a water extract of wheat straw containing the required amount of potassium was added to the nutrient solution. The Knop's solution was used in the control experiments. The experimental and control plants showed no significant differences in state or productivity of their photosynthetic apparatus. A small decrease in total productivity of the experimental plants was observed, which might result in some reduction of O{cyrillic}2 production in a BLSS. В© 2009 COSPAR.

Аннотация: Wheat was cultivated on soil-like substrate (SLS) produced by the action of worms and microflora from the inedible biomass of wheat. After the growth of the wheat crop, the inedible biomass was restored in SLS and exposed to decomposition ("biological" combustion) and its mineral compounds were assimilated by plants. Grain was returned to the SLS in the amount equivalent to human solid waste produced by consumption of the grain. Human wastes (urine and feces) after physicochemical processing turned into mineralized form (mineralized urine and mineralized feces) and entered the plants' nutrient solution amounts equal to average daily production. Periodically (once every 60-70 days) the nutrient solution was partly (up to 50%) desalinated by electrodialysis. Due to this NaCl concentration in the nutrient solution was sustained at a fixed level of about 0.26%. The salt concentrate obtained could be used in the human nutrition through NaCl extraction and the residuary elements were returned through the mineralized human liquid wastes into matter turnover. The control wheat cultivation was carried out on peat with use of the Knop nutrient solution. Serial cultivation of several wheat vegetations within 280 days was conducted during the experiment. Grain output varied and yield/harvest depended, in large part, upon the amount of inedible biomass returned to SLS and the speed of its decomposition. After achieving a stationary regime, (when the quantity of wheat inedible biomass utilized during vegetation in SLS is equal to the quantity of biomass introduced into SLS before vegetation) grain harvest in comparison with the control was at most 30% less, and in some cases was comparable to the control harvest values. The investigations carried out on the wheat example demonstrated in principle the possibility of long-term functioning of the LSS photosynthesizing link based on optimizations of biological and physicochemical methods of utilization of the human and plants wastes. The possibilities for the use of these technologies for the creation integrated biological-physicochemical LSS with high closure degree of internal matter turnover are discussed in this paper. Аннотация: Use of halophytes (salt-tolerant vegetation), in a particular vegetable Salicornia europaea plants which are capable of utilizing NaCl in rather high concentrations, is one of possible means of NaCl incorporation into mass exchange of bioregenerative life support systems. In preliminary experiments it was shown that S. europaea plants, basically, could grow on urine pretreated with physicochemical processing and urease-enzyme decomposing of urea with the subsequent ammonia distillation. But at the same time inhibition of the growth process of the plants was observed. The purpose of the given work was to find out the influence of excessive quantities of some mineral elements contained in products of physicochemical processing of urine on the production process and NaCl accumulation by S. europaea plants. As the content of mineral salts in the human liquid wastes (urine) changed within certain limits, two variants of experimental solutions were examined. In the first variant, the concentration of mineral salts was equivalent to the minimum salt content in the urine and was: K - 1.5 g/l, P - 0.5 g/l, S - 0.5 g/l, Mg - 0.07 g/l, Ca - 0.2 g/l. In the second experimental variant, the content of mineral salts corresponded to the maximum salt content in urine and was the following: K - 3.0 g/l, P - 0.7 g/l, S - 1.2 g/l, Mg - 0.2 g/l, Ca - 0.97 g/l. As the control, the Tokarev nutrient solution containing nitrogen in the form of a urea, and the Knop nutrient solution with nitrogen in the nitrate form were used. N quantity in all four variants made up 177 mg/l. Air temperature was 24 В°C, illumination was continuous. Light intensity was 690 ?mol/m2s of photosynthetically active radiation. NaCl concentration in solutions was 1%. Our researches showed that the dry aboveground biomass of an average plant of the first variant practically did not differ from the control and totaled 11 g. In the second variant, S. europaea productivity decreased and the dry aboveground biomass of an average plant totaled 8 g. The increase of K quantity in the experimental solutions resulted in an elevated content of the element in the plants. The increase of K uptake in the second experimental variant was accompanied by a 30-50% decrease of Na content in comparison with the other variants. Comparative Na content in the other variants was practically identical. N, Mg and P content in the control and experimental variants was also practically identical. The increase of S quantity in the second experimental variant also increased S uptake by the plants. But Ca quantity, accumulated in aboveground plants biomass in the experimental variants was lower than in the control. NaCl uptake by plants, depending on the concentration of mineral salts in the experimental solutions, ranged from 8 g (maximum salt content) up to 15 g (minimum salt content) on a plant growth area that totaled 0.032 m2. Thus, high concentrations of mineral salts simulating the content of mineral salts contained in urine did not result in a significant decrease of S. europaea productivity. The present work also considers the influence of higher light intensity concentrations on productivity and NaCl accumulation by S. europaea plants grown on experimental solutions with high salt content. Artificial-light culture -- Photosynthetic apparatus -- Phytocenosis -- Productivity -- Embryophyta
Аннотация: The processes limiting the production in higher plant phytocenoses under an artificial-light culture are analyzed in relation to the multilevel organization of the photosynthetic apparatus (PA). The authors consider the feasibility of overcoming these limitations by optimizing the physical parameters of irradiation (the structure of the light spectrum, the rate, and the ratio of radiation fluxes in photosynthetically active radiation (PAR) and infrared (IR) regions) at the molecular, leaf, plant, and cenotic levels of PA organization. To illustrate this approach, the authors used a complex experiment in an artificial ecosystem to evaluate the efficiency of the light control of production processes in multispecies phytocenoses by alleviating or removing the factors that limit plant production at the various levels of PA organization. An artificial-light culture is seen as an instrument for solving several problems of theoretical and applied plant physiology and related disciplines in the future.
