Аннотация: Methods of atomic absorption, flame photometry, and emission spectral analysis were used to study the concentrations of metals in water and major ecosystem components of the Yenisei River upstream of Krasnoyarsk City (conventionally background area). The mean bulk concentrations of Al and Cu in water exceeded the MAC for water bodies used for fishery. Cu concentration in freshwater shrimp was found to be reliably higher than that in the link of primary producers (periphyton), and Cd concentration in caddis fly larvae was found to exceed that in water moss. The maximal concentrations of metals among the examined aquatic organisms were recorded in periphyton. Cr concentration in the muscles of Arctic grayling was found to exceed some international standards. В© 2009 Pleiades Publishing, Ltd.

Contents of most parts of metals in soils, water, and macrophytes significantly correlated with each other. As concluded, their contents were determined by natural, general, geochemical peculiarities of the region. Heavy metals, contents of which were higher than federal upper limits of concentration, were revealed. In muscles of fish with different feeding spectra - crucian and perch - concentrations of some metals differed significantly; correlation graphs for metals also had different structures. Comparison of our data with those on diverse aquatic ecosystems of Siberia, Europe, North America, and China published in the last decade was carried out. It was concluded that a distribution of heavy metals in the compartments of an aquatic ecosystem presently have to be determined for each particular water body until general regularities are discovered.

Аннотация: During three field seasons (June-September) of 1997-99 contents of Na, K, Ca, Mg, Fe, Mn, Zn, Cu, Al, Cr, Ni, Cd, and Pb were determined in compartments of ecosystem (surrounding soils, bottom sediments, water, zoobenthos, macrophytes, and fish) of a fish and recreation pond situated at the edge of Krasnoyarsk City (Siberia, Russia). Аннотация: The study addresses the effect of abiotic (medium salinity and copper ions) and biotic (interactions between populations) factors on the formation of structured communities by binary associations consisting of halotolerant bacteria (Alcaligenes sp. 1-1 or Acinetobacter sp. 1-19) and a wild-type B. subtilis 2335 strain or a transgenic strain. The results showed that 250 mg l(-1) of copper ions inhibit formation of biofilms by monocultures of the tested strains. Binary associations of the strains were more resistant to high concentrations (250 mg l(-1)) of copper ions. At the lowest NaCl concentration (0.05% and 2.5%) and in the presence of copper ions, bacilli seemed to help halotolerant bacteria survive. Under increased salinity and in the presence of copper ions, structured communities developed due to halotolerant bacteria. Coexistence under stressful conditions was beneficial for the both groups of bacteria.

Аннотация: A new method for assessing biotoxicity of nanomaterials, based on the use of soluble bioluminescent coupled enzyme system NAD(P)ai...H:FMN oxidoreductase and luciferase, is proposed. The results of this study indicate a significant adverse biological effect exerted by nanoparticles at the molecular level. It was found that the most toxic nanoparticles the nanoparticles are based on copper and copper oxide, as well as single-walled carbon nanotubes and multi-walled carbon nanofibers, which are referred to hazard class II.

Аннотация: It was found that the catalytic effect of modified nanodiamonds (MND) in the H2O2–4-aminoantipyrine–phenol oxidative azo coupling reaction is due to microimpurities of iron and copper ions on the surface of nanoparticles. The efficiency of MND as a catalyst is determined by the amount of surface impurities of these ions and can be doubled by their additional adsorption on nanoparticles. Using MND for phenol indication ensures a linear yield of the colored product of the azo coupling reaction over an analyte concentration range of 0.05–10 ?g/mL. The possibility of reusing MND for phenol testing in aqueous samples was demonstrated. © 2017, Pleiades Publishing, Ltd.

Аннотация: This paper demonstrates the effectiveness of using detonation nanodiamonds (DNDs) for detecting phenol in aqueous medium. The study has shown that the catalytic effect of DNDs in the oxidative azo coupling reaction (phenol-4-aminoantipyrine-hydrogen peroxide) is produced by trace amounts of iron and copper ions adsorbed on the surface of nanoparticles. The effectiveness of DNDs as a catalyst is determined by the amounts of these adsorbates and can be enhanced by a factor of two by additional adsorption of these ions onto the nanoparticles. A rise in the temperature of the ONO-catalyzed azo coupling reaction leads to a considerable (4.5-fold) increase in the reaction product yield. DNDs used to detect phenol in aqueous medium enable a linear increase in the yield of the product of the azo coupling reaction at concentrations of the analyte of between 0.05 and 10 mu g/mlThe study demonstrates that DNDs can be reused to detect phenol in water samples.

Аннотация: A number of approaches have been proposed and tested to transfer enzymatic reactions into the functional elements of microfluidic chips on the example of the bienzyme bioluminescent reaction involving NAD(P)H:FMN-oxidoreductase and luciferase. Measurement of the catalytic activity of these enzymes (under the influence of pollutants) is the basis of enzymatic bioassay of various liquids. It was found that all of the components of the reaction must be placed in the same cell of the chip to improve the reproducibility of the measurements. The use of starch gel as a carrier for immobilization and gelatin as a scaffold in the reactor of the chip enables the preservation of enzyme activity in the course of sealing the chip at room temperature. It is shown that the components of the reaction should be vigorously stirred in a microfluidic chip reactor to improve the efficiency of the analysis. As a result of the studies, a prototype of microfluidic chip based on the enzymatic bioluminescent reaction is proposed. It is characterized by a detection limit of copper sulfate of 3 mu M that corresponds to the sensitivity of traditional lux-biosensors based on living cells. The analysis time is reduced to 1 min, and the analysis can be performed by individuals without special laboratory skills.

Аннотация: Novel approaches to description of ecosystem changes under the impact of contaminants, dependent on their local concentration and the impact duration, are considered. It is shown that, using quite simple assumptions about biotic effects of a contaminant, this process can be treated as an ecological analogue to phase transition of the second order. For the analysis and verification of the model proposed, the data on forest ecosystems in the vicinity of powerful point sources of contamination (copper-smelting plants) had been used.

Аннотация: In the present study, we demonstrate the use of a disposable luciferase-based microfluidic bioassay chip for environmental monitoring and methods for fabrication. The designed microfluidic system includes a chamber with immobilized enzymes of bioluminescent bacteria Photobacterium leiognathi and Vibrio fischeri and their substrates, which dissolve after the introduction of the water sample and thus activate bioluminescent reactions. Limits of detection for copper (II) sulfate, 1,3-dihydroxybenzene and 1,4-benzoquinone for the proposed microfluidic biosensor measured 3 ?M, 15 mM, and 2 ?M respectively, and these values are higher or close to the level of conventional environmental biosensors based on lyophilized bacteria. Approaches for entrapment of enzymes on poly(methyl methacrylate) (PMMA) plates using a gelatin scaffold and solvent bonding of PMMA chip plates under room temperature were suggested. The proposed microfluidic system may be used with some available luminometers and future portable luminescence readers. © 2018 John Wiley & Sons, Ltd.

Аннотация: The research aimed to determine critical concentrations of heavy metals at which survival of resting eggs of the cladoceran Moina macrocopa is negatively affected. Resting eggs’ viability was not affected over a 30-days exposure towards copper, cadmium, zinc or nickel at concentrations up to 60–70 g/L. When resting eggs were exposed to sediment contaminated with heavy metals for 8 months, the hatching success was affected at 30 g copper/kg. Thus, resting eggs of Cladocera can tolerate heavy metals at concentrations that far exceed lethal concentrations of heavy metals to active life stage and exceed low or moderate levels of environmental pollution. Follow up investigation of life table parameters of hatchlings from resting eggs exposed to heavy metals demonstrated that neither lifespan nor fecundity of hatchlings differ from control animals. These results demonstrate that zooplankton may rapidly recover from resting egg bank once aquatic habitat becomes unpolluted. © 2018, Springer Science+Business Media, LLC, part of Springer Nature.

Аннотация: Enzymatic luminescent systems are a promising tool for rapid detection of heavy metals ions for water quality assessment. Nevertheless, their widespread use is limited by the lack of test procedure automation and available sensitive handheld luminometers. Herein we describe integration of disposable microfluidic chips for bioluminescent enzyme-inhibition based assay with a handheld luminometer, which detection system is based on a thermally stabilized silicon photomultiplier (SiPM). Microfluidic chips were made of poly(methyl methacrylate) by micro-milling method and sealed using a solvent bonding technique. The composition of the bioluminescent system in microfluidic chip was optimized to achieve higher luminescence intensity and storage time. Results indicate that developed device provided comparable sensitivity with bench-scale PMT-based commercial luminometers. Limit of detection for copper (II) sulfate reached 2.5 mg/L for developed biosensor. Hereby we proved the concept of handheld enzymatic optical biosensors with disposable chips for bioassay. The proposed biosensor can be used as an early warning field-deployable system for rapid detection of heavy metals salts and other toxic chemicals, which affect bioluminescent signal of enzymatic reaction.

Аннотация: In this paper, we have investigated the possibilities of application of enzymatic systems with increasing chain length as a bioassay to evaluate the soil contamination status. The sensitivity of monoenzyme reaction as well as double- and triple-enzyme chains based on NAD(P)H:FMN-oxidoreductase and luciferase of luminous bacteria and lactate dehydrogenase to pesticides and copper ions in water and water extracts from soils were estimated. For this, the toxicological parameter IC 20 reflecting the sensitivity limit of the enzyme system to the to-xicant was used. It was revealed that elongation of the coupled enzyme chain (from mono- to triple-enzyme) increases the sensitivity of the bioassay, in some cases by several orders of magnitude. This pattern can be used as a tool to improve the properties of enzymic bioassays. The effect of extracts from uncontaminated soils of various types on enzymatic systems also differs, which makes possible to design the specialized enzymatic bioassays as well. © 2018 Sankt Peterburg.All rights reserved.

Аннотация: The harmful potential of bulk bottom sediments of the Yenisei River has been assessed by the Allium test. Sediment samples have been taken in ten sites on a plot of about 100 km in length in the middle reaches of the Yenisei River affected by urban activity of the city of Krasnoyarsk and industrial plants. The samples of sediments differ considerably in the content of potentially toxic substances (heavy metals, artificial radionuclides, and organic pollutants) and nutrients and in physical properties. We use the mean length of onion root as an indicator of general toxicity, the mitotic index as an indicator of cytotoxicity, and the percentage of anatelophase cells in the apical root meristem with abnormal chromosomes as an indicator of genotoxicity. Both the stimulation and inhibition of indicator endpoints are revealed in sediment samples of the Yenisei River relative to the control (artificial sediments). An increased genotoxicity is recorded in the sample of sediments contaminated by a mixture of organic and chemical toxicants. A significant positive correlation between the percentage of abnormal cells and the content of copper and petrochemicals in the samples is revealed. No extremely strong reactions of onion endpoints to the quality of sediment samples are revealed, which is probably related to relatively low contamination of the sediments studied. This is in agreement with the data of chemical analyses. Based on the testing results, we can conclude that the endpoints of the Allium test are sensitive enough to the quality of bottom sediments of the Yenisei River and can be used for their biotesting.
