Аннотация: The age-related changes in the temperature dependence curves (TDC) of chlorophyll fluorescence were studied in leaf segments of wheat (Triticum aestivum L.), tomato (Lycopersicum esculentum Mill.), and cucumber (Cucumis sativum L.) plants grown under controlled photoculture conditions. Three major TDC patterns of chlorophyll fluorescence were identified within the temperature range of 25-70В°C, with each of the patterns corresponding to a certain phase of leaf development. The transition from one type of thermogram to another was a gradual and ordered process. The magnitude of the low-temperature TDC peak increased until leaves completely expanded and declined with leaf senescence. In the course of leaf senescence, the thermograms exhibited an additional shoulder, which further changed into a peak at 55-65В°C with increasing magnitude. Our data provide the basis for assessing leaf age from the type of chlorophyll fluorescence thermogram and the changes in the particular indices characteristic of TDC of chlorophyll fluorescence.

Аннотация: The usage of chlorophyll fluorescence induction (CFI) for estimating various types of plant resistance (primary, general, initial, adaptive) to stress factors is reviewed. The necessity of ontogenetic approach (considering the age-specific properties of the photosynthetic apparatus) in determining general and adaptive resistance of plants to prolonged action of stress factors by the CFI method is argued. In the plant Cucumbis sativus L., the possibility is shown of using age-specific qualitative and quantitative traits of leaf CFI (changes in the shape of chlorophyll fluorescence induction curves and in the dynamics of CFI parameters in the course of leaf ontogeny) for comparative study of differences between fully active and stressed plants. Possible criteria are suggested for estimating the effect of outer stress factors by the presence or absence of a steady-state phase in the dynamics of CFI parameters during leaf ontogeny. It is also suggested to use the duration of the steady-state phase following the termination of leaf growth (estimated by the dynamics of the slow phase of CFI as the ratio of fluorescence intensity at the peak P and the steady-state fluorescence intensity, FP/FS, or as the viability index Rfd) and the variability of CFI parameters during this period as qualitative estimates of plant resistance to prolonged action of stress factors.

Аннотация: The causes that limit the applicability of the chlorophyll fluorescence induction method to evaluating the state of the photosynthetic apparatus of leaves under long-term stress were analyzed. It was substantiated that it is necessary to use an ontogenetic approach, which consists in more accurately taking into account the age state of the plant leaves studied. Comparison of the calendar and ontogenetic methods for determining the leaf age by the example of cucumber plant under controlled conditions revealed significant differences in the results of evaluating the photosynthetic apparatus of plant leaves by the chlorophyll fluorescence induction method in two variants with contrasting illumination conditions (in white light at 400-700 nm and in stressful red light at 600-700 nm). It was shown that, under long-term stress, the unequivocal interpretation of the chlorophyll fluorescence induction parameters in evaluating the photosynthetic apparatus of plants depends significantly on the choice of the ontogeny period of the leaves of plants being compared and on the accuracy of determining their age. Аннотация: We studied soil-like substrate (SLS) as a potential candidate for plant cultivation in bioregenerative life support systems (BLSS). The SLS was obtained by successive conversion of wheat straw by oyster mushrooms and worms. Mature SLS contained 9.5% humic acids and 4.9% fulvic acids. First, it was shown that wheat, bean and cucumber yields as well as radish yields when cultivated on mature SLS were comparable to yields obtained on a neutral substrate (expanded clay aggregate) under hydroponics. Second, the possibility of increasing wheat and radish yields on the SLS was assessed at three levels of light intensity: 690, 920 and 1150 ?mol m-2 s-1 of photosynthetically active radiation (PAR). The highest wheat yield was obtained at 920 ?mol m-2 s-1, while radish yield increased steadily with increasing light intensity. Third, long-term SLS fertility was tested in a BLSS model with mineral and organic matter recycling. Eight cycles of wheat and 13 cycles of radish cultivation were carried out on the SLS in the experimental system. Correlation coefficients between SLS nitrogen content and total wheat biomass and grain yield were 0.92 and 0.97, respectively, and correlation coefficients between nitrogen content and total radish biomass and edible root yield were 0.88 and 0.87, respectively. Changes in hormone content (auxins, gibberellins, cytokinins and abscisic acid) in the SLS during matter recycling did not reduce plant productivity. Quantitative and species compositions of the SLS and irrigation water microflora were also investigated. Microbial community analysis of the SLS showed bacteria from Bacillus, Pseudomonas, Proteus, Nocardia, Mycobacterium, Arthrobacter and Enterobacter genera, and fungi from Trichoderma, Penicillium, Fusarium, Aspergillus, Mucor, Botrytis, and Cladosporium genera. Аннотация: Photosynthetic activity, the content of various photosynthetic pigments, and the chloroplast ultrastructure were examined in the leaves of cucumber (Cucumis sativus L.) and pea (Pisum sativum L.) plants of different ages grown under red light (600-700 nm, 100 W/m2). In pea leaves tolerant to red-light irradiation, chloroplast ultrastructure did not essentially change. In the first true leaves of cucumber plants susceptible to red-light irradiation, we observed a considerable increase in the number and size of plastoglobules, the appearance of chloroplasts lacking grana or containing only infrequent grana, and stromal thylakoids. In the upper leaves of 22-day-old cucumber plants, the chloroplast structure was essentially similar to that of the control chloroplasts in white light, and we therefore suppose that these plants have acclimated to red light.

Аннотация: Tomato (Lycopersicon esculentum Mill), sunflower (Helianthus annuus L.), and cucumber (Cucumis sativus L.) plants were grown under red or white light at a radiance of 115 W/m2 under controlled conditions. The red spectra were characterized by either two well-distinguished spectral lines (RL1) or only one (RL2). Apparent photosynthesis (Pa), gross photosynthesis (Pg), dark (Rd), and light (R1) respiration, the content of pigments, and the biochemical composition of plants were estimated. Enhancing the destructive red light influence led to a rise in the Pa/R1 and Pa/Pg ratios and to a decrease in chlorophyll a, nitrate, and reduced nitrogen. RL1 was more inhibitory for plants than RL2. It is concluded that declined energy efficiency of respiration contributes to the destructive effect of RL in sunflower and cucumber plants. The rearrangement of metabolic processes, at which energy efficiency of respiration is maintained, is responsible for adaptation of tomato plants to red light. The obtained results allow one to outline the ways to study the species-specific mechanisms of plant responses to prolonged RL action.

Аннотация: The effect of light sources with discrete or continuous spectra of red light (50 W/m2, 600-700 nm) on the structural and functional characteristics of chloroplasts were studied in leaves of cucumber (Cucumis sativum L.) plants, which die under strong red light, and pea (Pisum sativum L.) plants, which tolerate red light under the same conditions. Leaf condition was assessed by measuring thermo-and photoinduced changes in the chlorophyll fluorescence yield and the photochemical and photophosphorylating activities of the chloroplasts. Different responses of the pigment apparatus of pea and cucumber plants to red light with continuous or line spectra were revealed. Pea plants responded to discrete-spectrum light by changing P700 content, the ratio of antenna chlorophyll to P700, and the position of the low-temperature peak of the temperature-induced chlorophyll fluorescence yield. In cucumber plants, disturbances in the energy transfer from the short-wavelength pigments to chlorophyll a were observed. In both plants, the effects of line spectrum light on the pigment apparatus were reversible; the ratio of cyclic to noncyclic photophosphorylation, photosynthetic control, and the extent of the coupling of photosynthetic electron transport to photophosphorylation did not change compared to the control light. The need for examining the line spectra of light sources for growing plants under moderate intensities of artificial light (about 50 W/m2 of photosynthetically active radiation) is discussed.

Аннотация: Cross sections and macerates were used to study leaf mesostructure in cucumber, sunflower, and tomato plants formed under controlled environmental conditions at an irradiance of 100 W.m-2 in the red and blue regions of the spectrum. White light served as the control. Three types of response to red light were isolated: the first type is a response not associated with harmful action of the light factor (tomato); the second a response associated with reversible changes of mesostructure (sunflower); and the third a response associated with irreversible changes of leaf mesostructure (cucumber). It is concluded that destructive changes during prolonged action of red radiation on the leaf are mainly associated with degradation of palisade parenchyma, whereas spongy parenchyma suffers less and takes upon itself the main functional role in certain species (sunflower). Species specificity was not clarified in the response of leaf mesostructure to radiation in the blue region of the spectrum.

Аннотация: Additional integral and kinetic parameters of induction curves have been introduced on the basis of analysis of currently employed parameters of slow induction of fluorescence (SIF) for a more complete quantitative description of SIF changes during ontogenesis of higher plant leaves. High sensitivity of the additional parameters to the age state of leaf tissue as compared with previously employed parameters is demonstrated on the example of cucumber leaves (Cucumis sativus L.). Analysis of the relationship between the photosynthetic rate and SIF parameters of separate cucumber leaf disks revealed the presence of a high positive correlation between the photosynthetic rate and the greatest possible average decline in fluorescence intensity during the inductive period for mature regions of the leaf and regions starting to senesce (the correlation coeffcient attained values of 0.95), which indicates the possibility of using the given SIF parameter in further elaboration of methods for estimation of chloroplast photosynthetic activity at the level of the higher plant leaf.

Аннотация: The slow fluorescence induction produced iii cucumber leaves by light in the range of wavelengths 380-540 nm and intensity of 180 W/m(2) was studied. The ratio of fluorescence maxima in the red region (F(734)/F(682)) in young and mature leaves was approximately 2. It is assumed that this value depends on an increase in the contribution of the long-wavelength fluorescence due to the spillover effect. In plants grown under natural conditions, the paramster F(734)/F(682) correlated with the concentrations of chlorophyll and carotenoids. In plants grown hi the light of red and blue regions no such correlation was observed and the F(734)/F(682) remained unchanged. It is concluded that the F(534)/F(682) is affected by the intensity and spectral composition of exciting light used during the growing.

Three major TDC patterns of chlorophyll fluorescence were identified within the temperature range of 25-70 degreesC, with each of the patterns corresponding to a certain phase of leaf development. The transition from one type of thermogram to another was a gradual and ordered process. The magnitude of the low-temperature TDC peak increased until leaves completely expanded and declined with leaf senescence. In the course of leaf senescence, the thermograms exhibited an additional shoulder, which further changed into a peak at 55-65 degreesC with increasing magnitude. Our data provide the basis for assessing leaf age from the type of chlorophyll fluorescence thermogram and the changes in the particular indices characteristic of TDC of chlorophyll fluorescence.
