Аннотация: Microparticles made from degradable polyhydroxyalkanoates of different chemical compositions a homopolymer of 3-hydroxybutyric acid, copolymers of 3-hydroxybutyric and 4-hydroxybutyric acids (P3HB/4HB), 3-hydroxybutyric and 3-hydroxyvaleric acids (P3HB/3HV), 3-hydroxybutyric and 3-hydroxyhexanoic acids (P3HB/3HHx) were prepared using the solvent evaporation technique, from double emulsions. The study addresses the influence of the chemical compositions on the size and ?-potential of microparticles. P3HB microparticles loaded with doxorubicin have been prepared and investigated. Their average diameter and ?-potential have been found to be dependent upon the level of loading (1, 5, and 10 % of the polymer mass). Investigation of the in vitro drug release behavior showed that the total drug released from the microparticle into the medium increased with mass concentration of the drug. In this study mouse fibroblast NIH 3T3 cells were cultivated on PHA microparticles, and results of using fluorescent DAPI DNA stain, and MTT assay showed that microparticles prepared from PHAs of different chemical compositions did not exhibit cytotoxicity to cells cultured on them and proved to be highly biocompatible. Cell attachment and proliferation on PHA microparticles were similar to those on polystyrene. The cytostatic drug encapsulated in P3HB/3HV microparticles has been proven to be effective against HeLa tumor cells. В© 2013 Springer Science+Business Media New York.

Аннотация: Microspheres have been prepared from the resorbable linear polyester of ?-hydroxybutyric acid (polyhydroxybutyrate, PHB) by the solvent evaporation technique and investigated in vitro and in vivo. Biocompatibility of the microspheres has been proved in tests in the culture of mouse fibroblast cell line NIH 3T3 and in experiments on intramuscular implantation of the microspheres to Wistar rats for 3 months. Tissue response to the implantation of polymeric microspheres has been found to consist in a mild inflammatory reaction, pronounced macrophage infiltration that increases over time, involving mono- and poly-nuclear foreign body giant cells that resorb the polymeric matrix. No fibrous capsules were formed around polymeric microparticles; neither necrosis nor any other adverse morphological changes and tissue transformation in response to the implantation of the PHB microparticles were recorded. The results of the study suggest that polyhydroxybutyrate is a good candidate for fabricating prolonged-action drugs in the form of microparticles intended for intramuscular injection. Аннотация: Along with the atmosphere, water and food regeneration processes in biological life support systems it is important to provide units and links responsible for utilization of unused plant biomass, human wastes and returning, if possible, the most of wastes into the intrasystem material cycle. The experience on construction of biological life support systems (BLSS) gained by the Institute of Biophysics SB RAS (Krasnoyarsk, Russia) allows us to suggest constructing an integrated biological-physical-chemical life support system with the biological unit predominating. It is possibly to partially mineralize urine and solid wastes by "wet incineration" by hydrogen peroxide in electric field. We suggest decomposing urea by a urease-enzymatic method using soybean or canavalia flour containing sufficient amount of urease. Consumption of 1.5 g of flour for decomposition of urea in daily urine and the possibility of producing flour from soybeans and canavalia grown inside the system make this method of urea decomposition rather prospective. Further ammonia distillation using the nitrification unit and evaporation of solution would make possible to return nitrogen and water back into the intrasystem cycle. Probably, in long-duration space expeditions the utilization of urine would be confined only by extraction of nitrogen and water from urine with further removal of dry residue to the stock, as the problem of returning sodium chloride into the intrasystem cycling has not been solved yet. As all biogenic elements contained in urine (except nitrogen) get lost at that, the solution of the problem with introducing NaCl and mineral elements into the cycle with the help of halophyte plants Salicornia europaea are of sufficient interest. This work presents the experimental results of growing Salicornia europaea on model solutions containing biogenic elements in the amounts equivalent to their content in urine and on urine, which undergone physically-chemically treatment by peroxide and ammonia distillation after urease-enzymatic decomposition. Taking into consideration that the mineral elements content in urine can vary, 2 variants of model solutions were used. In the first variant the content of P was 8-fold, S - 7-fold, K - 8-fold higher than in Knop's solution; the content of Ca and Mg almost complied with that in Knop's solution. In the variant P was 12-fold, S - 17-fold, K - 17-fold, Ca - 6-fold and Mg was 8-fold higher than in Knop's solution. The content of N and NaCl in both variants was the same and constituted 0.18 g/l and 10 g/l respectively. The results of carried experiments showed that growing plants on urine treated in the above-mentioned way is possible; though the productivity of plants would be less than on model solutions. The reasons of plant productivity drop and the possible ways of their removal have been discussed.

Аннотация: A procedure for preparation of microspheres from biodegradable linear polyether of microbiological origin (polyhydroxybutirate, PHB) with using the technology of solvent evaporation was developed considering a specific example of two- and three-component emulsions. The procedure provided permanent preparation of the microspheres of high quality. The influence of the procedure (emulsion type, dispersion process and medium temperature) on the yield of the microspheres, their structure and size was shown. The temperature had a significant impact on incorporation of the antitumor anthracycline antibiotic daunorubicin (rubomycin) to the polymer matrix. The microspheres with various levels of the drug load (29 and 90% of the initial content in the emulsion) were prepared and the kinetics of the in vitro rubomycin release was studied. The dynamics of the highly toxic rubomycin release from the microspheres was on the whole even with the curve profile reaching the plateau in 20-22 hours of the observation period. The rate of the rubomycin release to the medium depended on the value of the antibiotic incorporation and was maximum within the first two hours (3.3 and 13.0 mcg/mlВ·h) that corresponded to the release of 0.97 and 3.89 of the incorporated antibiotic. The average rate of rubomycin release during 300 hours was 0.81В·10-4 and 2.3В·10-4 mcg/mlВ·h. The release constituted respectively 3.9 and 13.11% of the antibiotic incorporated to the microspheres.

Аннотация: The morphotypes of human macrophages (MPh) were studied in the culture on nano-structured biopolymer substrates, made from polyhydroxyalcanoates (PHAs) of five various monomer compositions, followed by the solvent evaporation. Its surface relief, which was further in direct contact with human cells in vitro, was analyzed by atomic force microscopy (AFM) and scanning electron microscopy (SEM). It was shown, that the features of the micro/nano relief depend on the monomeric composition of the polymer substrates. Monocytes (MN) of patients with atherosclerosis and cardiac ischemia, undergoing stenting and conventional anti-atherosclerotic therapy, were harvested prior and after stenting. MN were isolated and cultured, with the transformation into MPh in direct contact with biopolymer culture substrates with different monomer composition and nano-reliefs, and transformed into MPh, in comparison with the same process on standard culture plastic. Sub-populations of cells with characteristic morphology in each phenotypic class were described, and their quantitative ratios for each sample of polymers were counted as an intermediate result in the development of “smart” material for cardiovascular devices. The results obtained allow us to assume, that the processes of MPh differentiation and polarization in vitro depend not only on the features of the micro/nano relief of biopolymer substrates, but also on the initial state of MN in vivo and general response of patients. © 2018 The Authors
