Аннотация: Contents of fatty acids in filets of unfrozen (control), boiled, fried, roasted and boiled in a small amount of water humpback salmon, collected from a wholesale market in Krasnoyarsk city (Siberia, Russia) were analyzed. Special attention was paid to essential polyunsaturated fatty acids of ?3 family: eicosapentaenoic, 20:5?3 (EPA) and docosahexaenoic, 22:6?3 (DHA). Heat treatment in general did not decrease content of EPA and DHA in humpback, except a modest reduction during frying. Cooked humpback appeared to be the valuable source of essential ?3 PUFAs, namely EPA and DHA. It was hypothesized that the absence of significant reduction of PUFAs' contents in red flesh of fishes of Salmonidae family during heat treatment may be due to a high level of natural antioxidants which formed in the course of evolution as adaptation to their ecological niche. Аннотация: The temperature dependence of bioluminescent activity of the Ca2+-activated photoprotein obelin from the hydroid polyp Obelia longissima and thermoinactivation of this protein at different concentrations of (NH4)2SO4 have been studied. The maximal intensity of luminescence of obelin was observed at 4-15-degrees-C. The activity of the photoprotein is completely stable to storage for 3 days at room temperature. Increasing the temperature to 40-degrees-C resulted in a 25-30% loss of enzyme activity in 1 h. The presence of ammonium sulfate during heating stabilizes the activity of obelin. Two breaks, at 11 +/- 3-degrees-C and 47 +/- 3-degrees-C, are observed in the Arrhenius plot of the first-order rate constant of the luminescence decay. The bioluminescent curves of obelin are biphasic in the temperature range 10-40-degrees-C. It is assumed that obelin may exist in two kinetically distinct conformers (active and inactive) whose ratio is temperature dependent.

Аннотация: The recombinant coelenterazine-dependent luciferases (isoforms MLuc 164 and MLuc39) from the marine copepod Metridia longa were expressed as inclusion bodies in E. coli cells, dissolved in 6 M guanidinium chloride and folded in conditions developed for proteins containing intramolecular disulfide bonds. One of them (MLuc09) was obtained in an active monomeric form with a high yield. The luciferase bioluminescence is found to be initiated not only by free coelenterazine, but also by Ca2+-dependent coelenterazine-binding protein (CBP) of Renilla muelleri on Ca2+ addition. The use of CBP as a "substrate" provides higher light emission and simultaneously the lower level of background. The high purity MLuc39 can be detected down to attomol with a linear range extending over 5 orders of magnitude. The MLuc39 reveals also a high stability towards heating and chemical modification; the chemically synthesized biotinylated derivatives of the luciferase preserve 35-40% of the initial activity The luciferase applicability as an in vitro bioluminescent reporter is demonstrated in model tandem bioluminescent solid-phase microassay combining the Ca2+-regulated photoprotein obelin and the Metridia luciferase.

Аннотация: The recombinant Ca2+-triggered coelenterazine-binding protein (CBP) from Renilla muelleri was investigated as a biospecifically labeled molecule for in vitro assay applications. The protein was shown to be stable in solutions in the frozen state, as well as stable under heating and to chemical modifications. Conjugates with biotin, oligonucleotide, and proteins were obtained and applied as biospecific molecules in a solid-phase microassay. CBP detection was performed with intact (no modifications were made) Renilla luciferase in the presence of calcium, and the detection limit was found to be 75 amol. Model experiments indicate that this approach shows much promise, especially with regard to the development of multianalytical systems.

Аннотация: The volatility and thermal stability of chemicals that are contained in the metabolic products of Moina macrocopa females that stimulate the production of diapausing eggs in these females have been studied in experiments with separately cultivated individuals. It has been shown that the gametogenesis-stimulating chemicals in this species are nonvolatile compounds: the effect of the medium that contains the metabolic products of the organism was not decreased after many hours of blowing with argon. Prolonged heating up to 80 degrees C also did not deactivate the ability of these products to change the reproductive mode. The effect of the medium that contains these products decreased after boiling, but if the possibilities of their oxidation or binding with salts of metals were excluded, they retained the ability to change the reproductive mode of the animals even after boiling.

Аннотация: The problem of correlation between rheological properties in macro- and micro- scales of media with biopolymers of polypeptide (gelatin) and polysaccharide (starch) nature is investigated. The viscosity of the biopolymer solutions with concentrations 0.5-5 wt% was estimated by standard rotational rheometry technique and with fluorescent molecular rotor at 15-50 A degrees C. Opposite trends were observed for relationship between microviscosity eta (m) and macroviscosity eta for two biopolymers: eta (m) << eta for gelatin and eta (m) >> eta for starch solutions. The temperature dependence of eta (m) followed the monoexponential decay law in all samples over the whole temperature range indicating insensitivity of microviscosity to gel mesh melting under heating. The dissimilarity of macro- and micro-rheological properties of gelatin and starch-containing media is discussed in terms of difference in architecture of the gels.

Аннотация: We studied structure, dynamics and functional (biogeochemical) activity of microbial complexes of cryogenic soils in larch forests in Central Evenkia and polygonal tundra on Samoilovskii Island, Lena Delta. We found that daily flux of methane from soil surface is 3–5 times less in forest soil than in the center of polygon in tundra. Short-term heating to 18.5–22.5 °C of permafrost-affected soil in larch forest caused sweetening of soil solution, shrinkage of eco-trophic groups of microorganisms and microbial biomass, as well as increase in greenhouse gases (CO2 and CH4) emission to the air. Notably the permafrost-affected soil on sandy deposits in tundra had highest microbial diversity of methanogenic archaea including Methanobacteriaceae, Methanomicrobiaceae, Methanosarcinaceae, Methanosaetaceae families. On the other hand only Methanosarcinacea were found in cryosols of larch forest. Both type I and type II methanotrophs were found in the forest soil, while only type II methanotrophs occurred in tundra soil. © 2017, Izdatel’stvo Nauka. All rights reserved.

Аннотация: Laboratory experiments were conducted in a hermetically sealed growth chamber with two soil samples obtained from the arctic tundra zone with different levels of moisture. Samples were maintained at a growing season typical of the region from which they were taken, and for the sample with a high level of moisture it was made twice: with the temperature in accord with natural conditions and one increased by 2 degrees C. It has been shown that heating of the overmoistened tundra soil by 2 degrees C can increased the average carbon dioxide emissions by almost two times (from 75 to 100-150 mg m(-2) h(-1)). Upon the application of heat, no significant increase in methane emission was observed.
