Аннотация: Aptamers are short DNA and RNA fragments which bind their molecular targets with affinity and specificity comparable to those of antibodies. Here, we describe the selection of novel 2'-F-RNA aptamers against total human hemoglobin or its glycated form HbA1c. After SELEX and high-throughput sequencing of the enriched libraries, affinities and specificities of candidate aptamers and their truncated variants were examined by the solid-phase bioluminescent assay. As a result, we identified aptamers specific to both hemoglobins or only glycated HbA1c. The developed 2'-F-RNA aptamers have shown their applicability for detection of total and glycated hemoglobin in one sample.

Blood cell response -- Degradable polyhydroxyalkanoates (PHAs) -- Surface structure and properties
Аннотация: Four PHA types were synthesized in the culture of Cupriavidus eutrophus B-10646 under special conditions, poly(3?hydroxybutyrate) [P(3HB)] and of copolymers, which contained 3HB monomers and 4?hydroxybutyrate (4HB), 3?hydroxyvalerate (3HV), or 3?hydroxyhexanoate (3HHx). All copolymers had the Mw of about 550–670 kDa, and the homopolymer P(3HB) had a significantly higher Mw – 920 kDa. P(3HB?co?4HB) and P(3HB?co?3HHx) had the lowest Cx (42 and 49%) while P(3HB?co?3HV) and P(3HB) exhibited higher Cx values (76%). Polymer films were prepared from different PHAs. Electron microscopy showed differences in the surface microstructure of the films. Films prepared from the P(3HB) were more hydrophobic and the arithmetic mean surface roughness of 71–75 nm, than the copolymer films, which were hydrophilic (57–60°) and had considerably higher roughness (158–177 nm). Blood parameters (hemoglobin and hemolysis) and response of the cells (erythrocytes, platelets, and monocytes) were studied in experiments with blood directly contacting the surface of the films of PHAs with different compositions. Cultivation of blood cells on polymer films did not cause any adverse effects on adhesion and morphology of all cell types. Results of studying blood cell response suggested that the films made from low-crystallinity copolymers containing 4?hydroxybutyrate and 3?hydroxyhexanoate were the best for contact with blood. © 2019 Elsevier B.V.

Аннотация: We report a novel bioluminescent aptasensor, which consists of 2 '-F-RNA aptamer modules joined into a bi-specific aptamer construct. One aptamer module binds the analyte, then after structural rearrangement the second module recruits non-covalently Ca2+-dependent photoprotein obelin from the solution, thus providing a bioluminescent signal. This concept allows using free protein as a reporter, which brings such advantages as no need for aptamer-protein conjugation, a possibility of thermal re-folding of aptamer component with no harm to a protein, and simpler detection protocol. We developed the new 2 '-F-RNA aptamer for obelin, and proposed the strategy for engineering structure-switching bi-modular aptamer constructs which bind the analyte and the obelin in a sequential manner. With the use of hemoglobin as a model analyte, we showed the feasibility of utilizing the aptasensor in a fast and straightforward bioluminescent microplate assay. With a proper design of a secondary structure, this strategy of aptasensor engineering might be further extended to bi-specific aptamer-based bioluminescent sensors for other analytes of interest.

    Positive feedback between acidosis and hypoxia during the transition of COVID-19 to a severe form of the disease / D. Lagutkin, D. Semyonov, Y. E. Yegorov [et al.] // FEBS Open Bio. - 2021. - Vol. 11. - P450-450. - Cited References:0 . - ISSN 2211-5463

Аннотация: SARSCoV2 virus causes disease that has several distinctive characteristics compared to diseases caused by other viruses. We have put forward a hypothesis that relates COVID19 pathogenesis with acidosis, which frequently characterizes severe cases of this disease. It has been shown that hypoxia and acidosis affect the progression of severe COVID19 at various physiological levels such as organs, tissues, and cells. The physiological effects of acidosis and hypoxia range from the level of compensatory capabilities of the whole organism to the functioning of a single hemoglobin molecule. In our work, we consider several mechanisms that link the damaging factors of COVID19 with acidosis. These mechanisms reveal stepbystep processes with a pronounced positive feedback. In accordance with the well known Bohr effect, a decrease in blood pH leads to a drop in blood oxygen saturation. At the same time, this drop in saturation contributes to the further development of acidosis. This indicates a depletion of the body's compensatory capabilities to regulate acidosis and leads to deterioration of the patient's condition. In addition, a decrease in pH can cause conformational changes in the viral Sprotein, followed by changes in ability of some antibodies to recognize the virus. This might lead to the decrease in antibodies affinity and avidity, negatively affecting virus clearance. Low levels of pH and hypoxia in blood and tissues can induce a proinflammatory innate response even in the absence of antigen stimulation. Therefore, hypoxia and acidosis can lead to a restructuring of the immune system and multidirectional pro and antiinflammatory responses, which often, instead of recovery, lead to the disease aggravation.
