Аннотация: The study addresses the effect of humic substances on marine luminous bacteria Photobacterium phosphoreum exposed to Am-241 (3,000 Bq L -1, water solution). Luminescent intensity of the bacteria was applied as a marker of their physiological activity. Humic substances have been found to reduce the effect of Am-241 on luminescence, decrease damage to cells, and change distribution of Am-241 between bacterial cells and intercellular media. It was shown that water-soluble humic substances, being products of natural transformation of organic substances in soil and bottom sediments, can serve as protecting agents for water microorganisms exposed to alpha radionuclides. Аннотация: A submerged macrophyte of the Yenisei River, Elodea canadensis, was used to study the microdistribution of the artificial radionuclide 241Am among different components of the plant. The total amount of 241Am added to the experimental system was 1850 В± 31 Bq/L. The total amount of 241Am accumulated by the plants was 182 Bq per sample, or 758,333 В± 385 Bq/kg dry mass. It has been found that the major portion of 241Am accumulated by E. canadensis, up to 85%, was bound to solid components of the cells. It is observed that the microdistribution of 241Am within different components of the submerged plant E. canadensis was not uniform. 241Am distribution vary depending on the age of the leaf blades, the state of the cells and morphological features of the plant stem. Аннотация: In luciferase isolated from luminescing bacteria Vibrio harveyi the authors have studied the dynamics of the luminescence with aliphatic aldehydes C10, C12 and C14 taken in pairs in the reaction with photoreduced flavin mononucleotide (FMN) and in the conjugated system NAD В· H: :FMN-oxidoreductase-luciferase. The kinetic characteristics of endogenous aldehyde have been determined. It is shown that the process of switching of luciferase from one aldehyde substrate to another is dependent on chain length and the order of introducing the aldehydes into the reaction mixture. Analysis of the "matrix of successive perturbations" gave a numerical matrix of the probabilities of oxidation of the aldehydes in the luminescent reaction. An order of preference of the aldehydes on their binding to luciferase is constructed. В© 1989.

Аннотация: New results obtained in the detection of rhythms in the low frequency range in the light signal of a small number of photo bacteria are reported. The luminescence of a small number of bacteria was recorded with an apparatus the sensitivity of which was 12 quanta/pulse. To check the possible rhythms in the luminescence of the bacteria, correlation analysis was made of the interpulse intervals of the detection of the signal with a computer. This revealed a definite rhythm in the bacterial luminescence the fundamental frequency of which lies in the region 8 c/s. Profound modulation of a bioluminescent signal at this and multiple frequencies was demonstrated. The harmonic components found in the spectrum of the signal refute the existing view of the continuous character of bacterial luminescence.

Аннотация: 1. (1) The increase in the intensity of the luminescence of bacteria observed on addition of glucose follows a curve with a well-marked maximum after which attenuation sets in. 2. (2) For the relations studied by us between the amount of glucose and the amount of bacterial suspension (1 ml 0В·1 m solution of glucose per ml suspension) the time of attaining the maximum intensity of luminescence and the ratio of the maximum intensity of emission to its level before addition of glucose do not depend on the concentration of bacteria in the suspension. 3. (3) The highest uptake of glucose by bacteria occurs in the period from addition of glucose to the moment of maximum rise in luminescence. Change in the intensity of luminescence after addition of glucose follows a curve similar to that of glucose uptake. 4. (4) We have determined the radiant flux and all the radiant energy released by bacteria in the period from addition of glucose to the moment of attaining the maximum intensity. One bacterial cell on average emits 100 quanta/sec. 5. (5) Bioluminescence accounts for 10-6 parts of the entire energy released on oxidation of glucose. В© 1966.

Аннотация: The luminescence of obelin is initiated by NaOCl in a reaction mixture containing no calcium. The addition of Mn2+ enhances the light emission > 300-fold. Sodium azide and histidine, as singlet oxygen quenchers, inhibit NaOCl-activated obelin luminescence in the presence or absence of Mn2+. This suggests that the addition of NaOCl to the mixture causes singlet oxygen formation (stimulated by Mn2+ ions), and singlet oxygen initiates the light-emitting reaction.

Аннотация: Genes encoding luminescence of Photobacterium leiognathi have been cloned in Escherichia coli. The luminescent clones were readily apparent. Among them, a clone containing a recombinant plasmid with a 13.5-kb insertion was identified. This DNA fragment contained all of the luminescence-encoding genes. The luciferase-encoding genes (lux) in this DNA fragment were localized. We have sequenced a part of the cloned lux region and identified the luxA, luxB and luxG genes encoding the ? and ? subunits of luciferase and a ? protein with an Mr of 26 180, respectively. The analysis of deduced amino acid sequences and comparison with known luciferase sequences from Vibrio harveyi, indicate the common origin of these proteins. В© 1990.

Аннотация: The processes resulting from the introduction of the transgenic microorganism (TM) E. coli Z905/pPHL7 into aquatic microcosms have been modeled experimentally. It has been shown that the TM E. coli is able to adapt to a long co-existence with indigenous heterotrophic microflora in variously structured microcosms. In more complex microcosms the numerical dynamics of the introduced E. coli Z905/pPHL7 population is more stable. In the TM populations staying in the microcosms for a prolonged time, changes are recorded in the phenotypic expression of plasmid genes (ampicillin resistance and the luminescence level) and chromosome genes (morphological and physiological traits). However, in our study microcosms, the recombinant plasmid persisted in the TM cells for 6 years after the introduction, and as the population adapts to the conditions of the microcosms, the efficiency of the cloned gene expression in the cells is restored. In the microcosms with high microalgal counts (107 cells/ml), cells with a high threshold of sensitivity to ampicillin dominate in the population of the TM E. coli Z905/pPHL7. В© 2003 COSPAR. Аннотация: The role of key environmental factors in adaptation of spore-forming and non-spore-forming transgenic microorganisms (TM) have been studied in model ecosystems. Model TM Escherichia coli Z905 (bearing plasmid genes of bacterial luminescence Ap1Lux+) has been found to have a higher adaptation potential than TM Bacillus subtilis 2335/105 (bearing genes of human ? 2-interferon Km1Inf+), planned for employment as a living vaccine under varying environmental conditions. Effects of abiotic factors on migration of natural and recombinant plasmids between microorganisms under model ecosystem conditions has been estimated. The transgenic microorganisms with low copy number survived better under introduction conditions in the microcosms studied. Аннотация: A structural approach to studying the regularities of the population dynamics of unstable recombinant bacterial strains in a chemostat was elaborated. The approach is based on the mathematical modeling of cell distribution in a population with different numbers of plasmid copies. The effect of decreased selective preference of plasmidless variants of the recombinant strain in the chemostat, which is related to a decrease in the number of plasmid copies in cells upon long-term incubation was analyzed. It is shown that the time of half-elimination of plasmids from the bacterial population in the steady state in the chemostat T1/2 does not depend on the maximum number of plasmid copies in cells N but is determined only by the mean time of generation g and the probability of the loss of one plasmid copy tau. The dependence of the preference of bacterial plasmidless variants on the efficiency of expression of genes cloned into plasmids in chemostat was analyzed using the recombinant strain E. coli Z905, whose plasmids pPHL-7 contain cloned genes for the luminescence system of marine luminescing bacteria Photobacterium leiognathi.

Аннотация: The behavior of Escherichia coli Z905, carrying a recombinant plasmid pPHL7 with genes determining ampicillin resistance and bacterial luminescence, and the efficiency of expression of cloned genes were studied after introduction of the strain into model aqueous ecosystems with different trophic chain lengths. The E. coli Z905 variants isolated from ecosystems after different periods of time were found to vary in their resistance to ampicillin (from 50 to 0.05 ?g/ml) and in the intensity of bioluminescence. An increase in the concentration of the selective factor (ampicillin) or in the extent of the aqueous microcosm blooming restored the expression of the recombinant plasmid genes in some clones.

Аннотация: An experimental approach for investigation of genetically modified microorganisms (GMMO) introduced into model ecosystems to evaluate potential risk of propagation of recombinant plasmids in surrounding medium has been developed. The object of modeling was Escherichia coli Z905 strain with a recombinant plasmid with bacterial luminescence genes, which was introduced into water microcosms of different structure. The approach involves comprehensive investigation of GMMO at four hierarchical levels: molecular (retaining the structure of the plasmid and expression of cloned genes); cellular (variation of metabolic activity); population (competitive power and metabolic interactions of GMMO with indigenous microflora, migration of recombinant and natural plasmids); ecosystem (effect of GMMO and cloned genes on ecosystem parameters). The experimental evidence and theoretical estimates are intended to form grounds to develop a basic version of an ecological certificate for different GMMO variants.

Аннотация: Исследована люминесценция экстрактов из полихет Arotonoe vittata. Но гомогенатах, полученных из элитр животных, показано, что биолюминесцентная система полихет данного вида относится к фотопротеиновому типу и стимулируется in vitro как О[2]{-} при использовании реагентов Фентона, так и при {-}ОС1 анионом при использовании гипохлорита натрия. Определены величины K[m] для реагентов Фентона в люминесцентной реакции, которые составили 0,37 мМ для FeSO[4] и 8,20 мМ для Н[2]О[2]. Аскорбат (1 и 10 мМ) ингибировал люминесценцию, инициированную реагентами Фентона, тогда как диэтилдитиокарбамат (0,1; 1,0 и 10 мМ) - увеличивал. Показано, что люминесценция экстрактов протекает с существенно большим квантовым выходом при стимуляции ее {-}OCl анионом по сравнению с О[2]{-}. Константы псевдопервого порядка спада люминесцентной реакции составили 0,34 с{-1} для NaOCl и 5,63 с{-1} для реагентов Фентона. Ни аскорбат, ни диэтилдитиокарбамат практически не влияли на люминесценцию, инициированную гипохлоритом натрия

Аннотация: The possibility of the development of the solid phase bioluminescent biotest using aerial mycelium of luminous fungi was investigated. Effect of organic and inorganic toxic compounds (TC) at concentrations from 10 -6 to 1 mg/ml on luminescence of aerial mycelia of four species of luminous fungi-Armillaria borealis (Culture Collection of the Institute of Forest, Siberian Branch, Russian Academy of Sciences), A. mellea, A. gallica, and Lampteromyces japonicus (Fungi Collection of the Botanical Institute, Russian Academy of Sciences)-has been studied. Culture of A. mellea was shown to be most sensitive to solutions of the model TC. It was demonstrated that the sensitivity of the luminous fungi is comparable with the sensitivity of the bacteria that are used for environmental monitoring. Use of the aerial mycelium of luminous fungi on the solid support as a test object is a promising approach in biotesting for the development of continuous biosensors for air monitoring.
