Аннотация: This study investigates kinetic and production parameters of a glucose-utilizing bacterial strain, C. eutrophus B-10646, and its ability to synthesize PHA terpolymers. Optimization of a number of parameters of bacterial culture (cell concentration in the inoculum, physiological activity of the inoculum, determined by the initial intracellular polymer content, and glucose concentration in the culture medium during cultivation) provided cell concentrations and PHA yields reaching 110 g/L and 80%, respectively, under two-stage batch culture conditions. Addition of precursor substrates (valerate, hexanoate, propionate, ?-butyrolactone) to the culture medium enabled synthesis of PHA terpolymers, P(3HB/3HV/4HB) and P(3HB/ 3HV/3HHx), with different composition and different molar fractions of 3HB, 3HV, 4HB, and 3HHx. Different types of PHA terpolymers synthesized by C. eutrophus B-10646 were used to prepare films, whose physicochemical and physical-mechanical properties were investigated. The properties of PHA terpolymers were significantly different from those of the P3HB homopolymer: they had much lower degrees of crystallinity and lower melting points and thermal decomposition temperatures, with the difference between these temperatures remaining practically unchanged. Films prepared from all PHA terpolymers had higher mechanical strength and elasticity than P3HB films. In spite of dissimilar surface structures, all films prepared from PHA terpolymers facilitated attachment and proliferation of mouse fibroblast NIH 3T3 cells more effectively than polystyrene and the highly crystalline P3HB. Аннотация: The phenomenon of saturable absorption has been observed in aqueous suspensions of detonation nanodiamonds (DNDs) with 34, 50, and 110 nm sized clusters of nanoparticles under irradiation with 795-nm laser pulses of 120-fs duration. The saturable absorption intensity has been studied as a function of the DND concentration in suspension. At a concentration of 2 wt % of DNDs with 50-nm average cluster size, the saturable absorption intensity amounts to 950 GW/cm2. © 2015, Pleiades Publishing, Ltd.

Аннотация: Wind flows in meromictic saline lakes in which the water column is not mixed to the bottom for at least one year are studied. This leads to the formation of upper and depth layers with small density gradients, between which there is a water layer with a large density gradient. It has been shown that, depending on the density stratification and the wind speed, wind flows (in the vertical plane) of two types are possible: with one or two circulation zones. For a two-layer lake model, a criterion for the change in the wind flow regime is proposed.

Аннотация: The optical limiting (OL) of detonation nanodiamond (DND) suspensions in engine oil was studied at a temperature range of 20°C to 100°C. Oil suspensions were prepared on the basis of the DNDs with an average nanoparticle cluster size in hydrosols (Daver) of 50 and 110 nm. Raman spectroscopy was used to characterize the samples. The OL investigation was carried out by the z-scan technique. The fundamental (1064 nm) and second (532 nm) harmonic radiations of YAG:Nd3 laser with passive Q-switching as an excitation source were used. The OL thresholds for both suspensions at 532 and 1064 nm were determined. It is shown that a decrease in the average nanoparticle cluster size as well as an increase of the wavelength of the incident radiation leads to the OL threshold increase. It is established that the OL performance is not influenced by increasing the temperature from 20°C to 100°C. The results obtained show the possibility of using the DNDs suspensions in engine oil as an optical limiter in a wide temperature range. © 2017 Society of Photo-Optical Instrumentation Engineers (SPIE).

Аннотация: We report on a saturable absorption in aqueous dispersions of nanodiamonds with femtosecond laser pulse excitation at a wavelength of 795 nm. The open aperture Z-scan experiments reveal that in a wide range of nanodiamond particle sizes and concentrations, a lightinduced increase of transmittance occurs. The transmittance increase originates from the saturation of light absorption and is associated with a light absorption at 1.5 eV by graphite and dimer chains (Pandey dimer chains). The obtained key nonlinear parameters of nanodiamond dispersions are compared with those of graphene and carbon nanotubes, which are widely used for the mode-locking. © 2017 Society of Photo-Optical Instrumentation Engineers (SPIE).

Аннотация: Hybrid wound dressings have been constructed using two biomaterials: bacterial cellulose (BC) and copolymer of 3-hydroxybutyric and 4-hydroxybutyric acids [P(3HB/4HB)] – a biodegradable polymer of microbial origin. Some of the experimental membranes were loaded with drugs promoting wound healing and epidermal cells differentiated from multipotent adipose-derived mesenchymal stem cells. A study has been carried out to investigate the structure and physical/mechanical properties of the membranes. The in vitro study showed that the most effective scaffolds for growing fibroblasts were composite BC/P(3HB/4HB) films loaded with actovegin. Two types of the experimental biotechnological wound dressings – BC/P(3HB/4HB)/actovegin and BC/P(3HB/4HB)/fibroblasts – were tested in vivo, on laboratory animals with model third-degree skin burns. Wound planimetry, histological examination, and biochemical and molecular methods of detecting factors of angiogenesis, inflammation, type I collagen, and keratin 10 and 14 were used to monitor wound healing. Experimental wound dressings promoted healing more effectively than VoskoPran – a commercial wound dressing. © 2019

Аннотация: This chapter reports the experimental investigation of the effects of nonlinear light scattering on optical limiting and saturable absorption in nanodiamond suspensions. The results are obtained by using the nonlinear transmittance and Z-scan techniques, which were combined with the detecting of light pulses scattered in aqueous nanodiamond suspensions. The third-order nonlinear optical response of this material was studied in the nanosecond and femtosecond timescales at the wavelengths of 532, 800, and 1064 nm and within the optical communication window. The obtained results demonstrate that aqueous suspensions of detonation nanodiamonds possess a high ray stability and are attractive for optical limiting applications. The femtosecond Z-scan measurement at the wavelength of 800 nm also revealed a saturable absorption in this nanocarbon material. © 2019 Elsevier Ltd. All rights reserved.

Аннотация: Calcium-regulated photoproteins are found in at least five phyla of organisms. The light emitted by those photoproteins can be tuned by mutating the photoprotein and/or by modifying the substrate coelenterazine (CTZ). Thirty years ago, Shimomura observed that the luminescence activity of aequorin was dramatically reduced when the substrate CTZ was replaced by its analogv-CTZ. The latter is formed by adding a phenyl ring to the pi-conjugated moiety of CTZ. The decrease in luminescence activity has not been understood until now. In this paper, through combined quantum mechanics and molecular mechanics calculations as well as molecular dynamics simulations, we discovered the reason for this observation. Modification of the substrate changes the conformation of nearby aromatic residues and enhances the pi-pi stacking interactions between the conjugated moiety ofv-CTZ and the residues, which weakens the charge transfer to form light emitter and leads to a lower luminescence activity. The microenvironments of CTZ in obelin and in aequorin are very similar, so we predicted that the luminescence activity of obelin will also dramatically decrease when CTZ is replaced byv-CTZ. This prediction has received strong evidence from currently theoretical calculations and has been verified by experiments.

Аннотация: Bioluminescence of a number of marine organisms is conditioned by Ca2+-regulated photoprotein (CaRP) with coelenterazine as the reaction substrate. The reaction product, coelenteramide, at the first singlet excited state (S1) is the emitter of CaRP. The S1-state coelenteramide is produced via the decomposition of coelenterazine dioxetanone. Experiments suggested that the neutral S1-coelenteramide is the primary emitter species. This supposition contradicts with theoretical calculations showing that the anionic S1-coelenteramide is a primary product of the decomposition of coelenterazine dioxetanone. In this study, applying molecular dynamic (MD) simulations and the hybrid quantum mechanics/molecular mechanics (QM/MM) method, we investigated a proton-transfer (PT) process taking place in CaRP obelin from Obelia longissima for emitter formation. Our calculations demonstrate a concerted PT process with a water molecule as a bridge between anionic S1-coelenteramide and the nearest histidine residue. The low activation barrier as well as the strong hydrogen-bond network between the proton donor and the proton acceptor suggests a fast PT process comparable with that of the lifetime of excited anionic S1-coelenteramide. The existence of the PT process eliminates the discrepancy between experimental and theoretical studies. The fast PT process at emitter formation can also take place in other CaRPs. © 2021 American Chemical Society.

Аннотация: Bioluminescence of a number of marine organisms is conditioned by Ca2+-regulated photoprotein (CaRP) with coelenterazine as the reaction substrate. The reaction product, coelenteramide, at the first singlet excited state (S-1) is the emitter of CaRP. The S-1-state coelenteramide is produced via the decomposition of coelenterazine dioxetanone. Experiments suggested that the neutral S-1-coelenteramide is the primary emitter species. This supposition contradicts with theoretical calculations showing that the anionic S-1-coelenteramide is a primary product of the decomposition of coelenterazine dioxetanone. In this study, applying molecular dynamic (MD) simulations and the hybrid quantum mechanics/molecular mechanics (QM/MM) method, we investigated a proton-transfer (PT) process taking place in CaRP obelin from Obelia longissima for emitter formation. Our calculations demonstrate a concerted PT process with a water molecule as a bridge between anionic S-1-coelenteramide and the nearest histidine residue. The low activation barrier as well as the strong hydrogen-bond network between the proton donor and the proton acceptor suggests a fast PT process comparable with that of the lifetime of excited anionic S-1-coelenteramide. The existence of the PT process eliminates the discrepancy between experimental and theoretical studies. The fast PT process at emitter formation can also take place in other CaRPs.
