Аннотация: The behavior of Escherichia coli Z905, carrying a recombinant plasmid pPHL7 with genes determining ampicillin resistance and bacterial luminescence, and the efficiency of expression of cloned genes were studied after introduction of the strain into model aqueous ecosystems with different trophic chain lengths. The E. coli Z905 variants isolated from ecosystems after different periods of time were found to vary in their resistance to ampicillin (from 50 to 0.05 ?g/ml) and in the intensity of bioluminescence. An increase in the concentration of the selective factor (ampicillin) or in the extent of the aqueous microcosm blooming restored the expression of the recombinant plasmid genes in some clones.

    POPULATION-STRUCTURE DYNAMICS OF A RECOMBINANT STRAIN OF ESCHERICHIA-COLI IN CONTINUOUS CULTURE [Text] / L. Y. POPOVA [et al.] // Microbiology. - 1992. - Vol. 61, Is. 4. - P417-422. - Cited References: 10 . - ISSN 0026-2617

Аннотация: We studied the population structure of Escherichia coli strain Z905, carrying the recombinant plasmid of the Ap(r)Lux+ phenotype, in continuous culture in a chemostat. We found that the maintenance of a stable, defined ratio of plasmidless to plasmid-carrying cells in the population depended primarily on the presence in the medium of the selective factor (ampicillin), and also on the nature of the growth-limiting carbon and energy source.

bioluminescent assay -- MC1R receptor -- melanoma -- single nucleotide polymorphisms
Аннотация: Several polymorphisms in the melanocortin-1 receptor gene (MC1R) have been associated with melanoma risk. In particular, rs1805007, rs1805008, and rs1805009 mutations, which result in R151C, R160W, and D294H amino acid substitutions, respectively, and are associated with the phenotype of red-hair mutations, have also been connected with melanoma and nonmelanoma skin cancer risks. This work describes a method of detecting these polymorphisms using primer extension with subsequent dual bioluminescent assay. Model plasmids carrying polymorphic MC1R fragments, as well as several clinical DNA samples, were tested using the proposed technique. The results agreed well with those obtained by Sanger sequencing. © 2015, Pleiades Publishing, Inc.

Аннотация: As studies have shown, individuals from well morphologically distinct groups often represent the same species and may even belong to one population in fishes and lampreys from environments with unstable abiotic factors (Arctic, mountain, and desert regions). Phenotypic plasticity ensures broad variation ranges of morphological traits in unstable conditions, which require rapid transitions from one morphogenetic variant to another. The choice of a morphogenetic pathway can be influenced by the level of individual heterozygosity, changes in the copy numbers of certain DNA sequences, heteroplasmy, and the presence of several allelic variants in the genes that strongly affect the phenotype. A cyclic character is often observed for evolutionary processes driven by these mechanisms, and speciation usually does not take place in unstable environmental conditions. However, mobilization reserve accumulate in a species with a broad reaction norm, and particular morphogenetic pathways may be genetically fixed when its population finds its way into stable environmental conditions, facilitating fast allopatric speciation.
