Аннотация: Procedure for microencapsulation providing stable formation of high quality microspheres was designed. Conditions for deposition of the anthracycline antibiotic daunorubicin to polymer matrix were developed and microsheres loaded with various quantities of the drug were prepared. The kinetics of the in vitro and in vivo release of daunorubicin from the microsheres was studied. The rate of the rubomycin release to the medium in vitro (balanced phosphate buffer at 37.8В°C) in a 300-hour experiment directly depended on the quantity of the incorporated drug and averaged 0.81 В· 10 -4 to 2.3 В· 10 -4 mcg/mlВ·h. The experiment on laboratory animals with intraperitoneal administration of the rubomycin microsperes showed that the drug remained in the blood and abdominal liquid for a long time (up to 10 days). Possible control of the quantity of the rubomycin encapsulation to the polymer matrix, no sharp efflux of the drug at the early stages of the observation and low rate of the drug release to the medium allowed to conclude that the use of the biodegradable polymer microsperes as carriers of the high toxic antibiotic providing its prolonged action was prospective.

Аннотация: The effect of the preparation technique (chemical composition of a polymer, type and method of emulsion mixing, and molecular mass of a drug) on the yield, structure, and size of microparticles obtained from resorbable polyesters of microbiological origin, polyhydroxyalkanoates, is studied. It is found that the concentration of the polymer solution and the method of emulsion mixing are the most significant factors affecting the diameter of microparticles based on polyhydroxyalkanoates; the surface structure of particles depends to a higher extent on the chemical composition of the polymer. The family of microparticles from 100-200 nm to 50-70 mu m in diameter is synthesized. It is shown that the rate of drug release from microparticles in vitro into the medium is higher in the case of 3-hydroxybutyrate copolymers with 3-hydroxyvalerate than in the case of the homopolymer of 3-hydroxybutyrate. This parameter increases with the content of 3-hydroxyvalerate units in the copolymer and the porosity and mass fraction of the drug in particles with a decrease in their sizes. For in vitro systems containing a phosphate buffer, variation in the preparation parameters makes it possible to obtain microparticles with various characteristics suitable for deposition of drugs. For microparticles obtained from polyhydroxyalkanoates and having different diameters, the mathematical description of the kinetics of drug release from the polymer matrix is provided.

Аннотация: Crystals of recombinant obelin, the Ca2+-regulated photoprotein from the marine hydroid Obelia longissima, have been grown from a solution containing PEG 8000 and potassium phosphate. Hexamine-cobalt trichloride was used as an additive to increase the chance of crystallization. The crystals grow in a light yellow cubic form (0.5 x 0.5 x 0.45 mm) which diffracts to beyond 1.1 Angstrom resolution. The crystals belong to the space group C2, with unit-cell parameters a=83.43, b=54.92, c=52.99 Angstrom, beta = 112.00 degrees. The asymmetric unit contains one molecule. Crystals exposed to calcium ion before and after X-ray irradiation emit light, confirming that the crystals consist of an active photoprotein.

Аннотация: The study addresses the effect produced by different inorganic salts and detergents (SDS, Triton X-100, the Tween series) on the ATP-dependent bioluminescent reaction catalyzed by the luciferase of the new earthworm species Fridericia heliota (Annelida: Clitellata: Oligochaeta: Enchytraeidae). It has been shown that the effect of divalent metal salts on luminescence is determined by the action of cations. Three of them - Mg2+, Mn2+ and Ca2+ - can stimulate luciferase activity at concentrations varying within a wide range, and Mn2+ can act as a 100%-effective substitute for Mg2+ in F. heliota luminescence reaction in vitro. The inhibitory effect of monovalent metal salts on luminescence is largely determined by the action of the anion part of the molecule. The effectiveness of the inhibitory effect of anions increases in the following order: {Mathematical expression}. Of the sodium salts, dodecyl sulfate, which is an anionic detergent, produces the strongest inhibitory effect on luciferase. On the contrary, nonionic detergents produce a stimulatory effect on the F. heliota luciferase. The action of the most effective of them - Triton X-100 - is determined by its ability to reduce the actual concentration of lipid inhibitors in the reaction mixture. В© 2006 Elsevier B.V. Аннотация: Properties of non-canonical GC base pairs and their relations with mechanochemical cleavage of DNA are analyzed. A hypothesis of the involvement of the transient GC wobble base pairs both in the mechanisms of the mechanochemical cleavage of DNA and epigenetic mechanisms involving of 5-methylcytosine, is proposed. The hypothesis explains the increase in the frequency of the breaks of the sugar-phosphate backbone of DNA after cytosines, the asymmetric character of these breaks, and an increase in break frequency in CpG after cytosine methylation. As an alternative hypothesis, probable implication of GC(+)Hoogsteen base pairs is considered, which now exemplify the best-studied non-canonical GC base pairs in the DNA double helix. Аннотация: The application of detonation nanodiamonds (DNDs) in delivery systems has been demonstrated many times for a variety of substances, including anti-inflammatory drugs, antibiotics, antineoplastic agents, vitamins etc. The applicability of detonation nanodiamonds as a carrier for delivery of corticosteroids is investigated in this study. Commercial DNDs were modified to obtain particles with high colloidal stability in aqueous suspensions. Physical adsorption was then used to conjugate the DNDs with methylprednisolone sodium succinate and dexamethasone sodium phosphate. The experimental adsorption isotherms were fit with Langmuir and Freundlich adsorption models. The results of in vitro release study demonstrated an explicit prolonged effect of DNDs, due to which a sustained release of the adsorbed drugs was provided. © 2021 Elsevier B.V.
