Botryococcus -- Lipids -- Nitrogen limitation -- algae -- Botryococcus -- Botryococcus braunii -- Chlorophyta
Аннотация: The effect of nitrogen limitation in a medium on the composition of intracellular lipids in the alga Botryococcus braunii Kutz IPPAS H-252 in the course of culture development was investigated. Under the conditions of nitrogen limitation, the alga under investigation accumulated lipids as triacylglycerols, and this process was accompanied by substantial changes in the total fatty acid (FA) composition, which were manifested in a decrease in trienoic acids (from 52.8-57.2 to 19.5-24.7% of total FAs) and an increase in the content of oleic (from 1.1-1.2 to 17.1-24.4%) and saturated (from 23.7-26.0 to 32.9-46.1%) acids. In the control culture, the directionality of FA redistribution was less marked, and these changes were noticed at the later stages of culture development. Under nitrogen limitation, marked changes in the FA composition of polar lipids occurred by the 13th day, and they were characterized by an increase in the content of saturated acids (up to 76.8%) and a dramatic decrease in the content of all polyenoic acids (up to 6.8%). The changes in the FA composition of triacylglycerols were noticed as early as by the 7th day; these changes consisted in an increase in the content of oleic acid, and its high content (28.4-38.4%) was maintained up to the end of culturing. In the control culture, triacylglycerols with a high content of oleic acid were found by the 13th day, although, by this time, the content of total lipids and triacylglycerols did not change. Botryococcus -- Fatty acids -- Lipids -- Nitrogen deficiency -- biochemical composition -- green alga -- algae -- Botryococcus -- Botryococcus braunii -- Chlorophyta
Аннотация: A study has been carried out to investigate the influence of nitrogen deficiency on intracellular lipid composition, including total fatty acid composition of lipids, polar lipids, and triacylglycerols, of the alga Botryococcus braunii Kutz IPPAS H-252 in batch culture. Under nitrogen limitation, the alga accumulates lipids as triacylglycerols and the total fatty acid (FA) composition changes: trienoic acids decrease (from 52.8-57.2 to 19.5-24.7% of the total FAs) and the oleic acid increases (from 1.1-1.2 to 17.1-24.4%) as does the saturated acids (from 23.7-26 to 32.9-46.1%). A similar rearrangement in the FA spectrum occurs at later times in the control culture, but it is less pronounced. Under nitrogen limitation, considerable changes in the polar lipid FAs are registered at day 13: saturated acids increase (from 28.6-35.5 to 76.8%) and all polyenoic acids markedly decrease (from 56.9-64.1 to 6.8%). Changes in the triacylglycerol fatty acid spectrum are seen on day 7: the oleic acid increases (from 14.7 to 34.2%) and remains at a high level till the end of the culture. In the control, triacylglycerols with large contents of oleic acid are detected at day 13, the total lipids and triacylglycerols still remaining unchanged. В© Springer 2005.

Аннотация: Lipid composition and hydrocarbon structure of two colonial green algae of the genus Botryococcus, i.e., a mu seum strain and a field sample collected for the first time from Lake Shira (Khakasia, Siberia), have been compared. Polar lipids, diacylglycerols, alcohols, triacylglycerols, sterols, sterol esters, free fatty acids and hydrocarbons have been identified among lipids in the laboratory culture. The dominant fraction in the museum strain was formed by polar lipids (up to 50% of the lipids) made up of fatty acids from C12 to C24. Palmitic, oleic, C16 - C18 dienoic and trienoic acids were the main fatty acids of the museum strain. Aliphatic hydrocarbons were found in the lipid of the museum strain. However, these amounted maximally to about 1% of the dry biomass at the end of exponential growth phase. The qualitative and quantitative compositions of FAs and hydrocarbons of the museum strain of Botryococcus, (registered at the Cambridge collection as Botryococcus braunii Kutz No LB 807/1 Droop 1950 H-252) differed from those of the Botryococcus strain described in the literature as Botryococcus braunii. The Botryococcus sp. found in Lake Shira is characterized by a higher lipid content (< 40% of the dry weight). Polar lipids, sterols, triacylglycerols, free fatty acids and hydrocarbons have been identified among lipids in the field sample. The main lipids in this sample were dienes and trienes (hydrocarbons < 60% of total lipid). Monounsaturated and very long chain monounsaturated fatty acids, including C28:1 and C32:1 acids, were identified in the Botryococcus found in Lake Shira. The chemo-taxonomic criteria allow us to unequivocally characterize the organism collected from Lake Shira as Botryococcus braunii race A.

Amphibiotic insects -- Essential fatty acids -- Metamorphosis -- Mosquitoes -- Short-chain polyunsaturated fatty acids
Аннотация: Emerging aquatic insects, including mosquitoes, are known to transfer to terrestrial ecosystems specific essential biochemicals, such as polyunsaturated fatty acids (PUFA). We studied fatty acid (FA) composition and contents of dominant mosquito populations (Diptera: Culicidae), that is, Anopheles messeae, Ochlerotatus caspius, Oc. flavescens, Oc. euedes, Oc. subdiversus, Oc. cataphylla, and Aedes cinereus, inhabited a steppe wetland of a temperate climate zone to fill up the gap in their lipid knowledge. The polar lipid and triacylglycerol fractions of larvae and adults were compared. In most studied mosquito species, we first found and identified a number of short-chain PUFA, for example, prominent 14:2n-6 and 14:3n-3, which were not earlier documented in living organisms. These PUFA, although occurred in low levels in adult mosquitoes, can be potentially used as markers of mosquito biomass in terrestrial food webs. We hypothesize that these acids might be synthesized (or retroconverted) by the mosquitoes. Using FA trophic markers accumulated in triacylglycerols, trophic relations of the mosquitoes were accessed. The larval diet comprised green algae, cryptophytes, and dinoflagellates and provided the mosquitoes with essential n-3 PUFA, linolenic, and eicosapentaenoic acids. As a result, both larvae and adults of the studied mosquitoes had comparatively high content of the essential PUFA. Comparison of FA proportions in polar lipids versus storage lipids shown that during mosquito metamorphosis transfer of essential eicosapentaenoic and arachidonic acids from the reserve in storage lipids of larvae to functional polar lipids in adults occurred. Аннотация: Food spectrum of microzooplancton in fresh-water ecological systems is lowly studied area. However, these organisms play very important role in transfer of substance and energy in water reservoirs. Traditional method of analysis of food in the microzooplancton does not makes it possible to determine real assimilation of organic substance. Therefore, biochemical methods have been developed which are based on analysis of compositions of fatty acids of organic substance. This leads to determining of the food assimilation. Fatty-acid compositions of lipids allow specialists to determine specific groups of organisms such as bacteria, microalgae etc. Data of the analysis are presented.

Frontal zones -- North Atlantic oscillation -- Sea surface temperature gradients -- South oscillation - El Ninjo -- Thermohaline circulation -- Variability of gradients
Аннотация: AVHRR MCSST data for the periods 1982-2000 (mean weekly data) were used to calculate mean gradient fields in the ocean for different periods of time. Three-month averaged sea surface temperature gradients (SSTG) and their mean seasonal variations have been studied for 25 points in the large-scale oceanic fronts zones. Major oceanic fronts in the Atlantic and Pacific have been identified and compared in literature. In the North Atlantic and Pacific, the areas under study were the North Polar Front and Subpolar Fronts. In the South Atlantic and Pacific we studied the region of the Antarctic Circumpolar Current (ACC) and the fronts formed by this current, known as the South Polar Front, and the Subantarctic Front. SSTG were also calculated for El Nino (Southeast Pacific) and Benguela Current (Southeast Atlantic). In warm periods seasonal SSTG in the North Atlantic markedly increased and exhibit some interannual cycles. The correlation between the North Atlantic Oscillation index and seasonal SSTG for a single point in the Gulf Stream zone can be the key point for evaluation of heat transfer by the currents to the coast of East Europe. In the Southern Atlantic, the SSTG values are low during the cold period (summer in the southern hemisphere) in the ACC zone and increase in the warm season (winter in the southern hemisphere). It also exhibits interannual cycles. In the Northwest Pacific for some points in the Subpolar Front the SSTG values are high in the cold period (winter). Here at seven points in the spring of 1993 and 2000 the calculations disclosed significant increase of the gradient. In these years, the anomalous SSTG in Subpolar Front and South Polar Front were found to vary synchronously in both hemispheres, with maximum intensity in spring (North Pacific) and in summer (South Pacific). Mean annual SSTG in the El Nino zone and south oscillation index have been found to exhibit some correlation. Major jet currents periodically form high-gradient temperature fields and from the temperature satellite data we can derive information about variation in the large-scale fronts in the Global Ocean. fatty acids -- triacylglycerols -- polar lipids -- gammarids -- aquatic larvae of insects
Аннотация: The composition and content of fatty acids (FAs) in total lipids, triacylglycerols (TAG) and polar lipids (PL) in dominant groups of benthic invertebrates: gammarids (Gammaridae, Amphipoda), chironomid larvae (Chironomidae, Diptera), caddisfly larvae (Trichoptera) and mayfly larvae (Ephemeroptera) were studied in the Yenisei river. For the first time data on the FA composition of species belonging to Trichoptera (Insecta) are presented. The groups of aquatic insect larvae and gammarids weakly differed in total content of essential polyunsaturated fatty acids (PUFAs). Hence, the strong invasion of gammarids which occurred in the last decades in the Yenisei river should not result in a decrease in potential yield of essential PUFA in the ecosystem and corresponding decrease in food resource quality for fish in respect to PUFA content. Significant differences in biomarker FAs in TAG were found which correlated to specific food sources. Different levels of long-chain PUFA in PL of the invertebrates are discussed in relation to the genetic ability of particular taxa to form these FAs. (C) 2002 Elsevier Science Inc. Аннотация: The surface properties of poly-3-hydroxybutyrate (P3HB) membranes were modified using oxygen and an ammonia radio-frequency (RF, 13.56 MHz) plasma. The plasma treatment procedures used in the study only affected the surface properties, including surface topography, without inducing any significant changes in the crystalline structure of the polymer, with the exception being a power level of 250 W. The wettability of the modified P3HB surfaces was significantly increased after the plasma treatment, irrespective of the treatment procedure used. It was revealed that both surface chemistry and surface roughness changes caused by the plasma treatment affected surface wettability. A treatment-induced surface aging effect was observed and resulted in an increase in the water contact angle and a decrease in the surface free energy. However, the difference in the water contact angle between the polymers that had been treated for 4 weeks and the untreated polymer surfaces was still significant. A dependence between cell adhesion and proliferation and the polar component of the surface energy was revealed. The increase in the polar component after the ammonia plasma modification significantly increased cell adhesion and proliferation on biodegradable polymer surfaces compared to the untreated P3HB and the P3HB modified using an oxygen plasma. © 2016 Elsevier B.V.

Аннотация: Closed ecological systems are of two-fold interest - as models of the Earth's biosphere explorable in experiments and as a facility for long-term autonomous human life support beyond the Earth. Theoretical analysis and experimental implementation of highly closed manned systems has been the subject of studies at the Institute of Biophysics (Russian Academy of Sciences, Siberian Branch) for many years. BIOS systems of increasing complexity with complete regeneration of atmosphere, water and partially food have been realized. In BIOS-3 experiments the system inhabited by 2-3 researchers for 4-6 months maintained its metabolic equilibrium without any negative physiological effect on the crew, which proves its sustainable condition. Specific for BIOS-3 is internal control by the people inhabiting the system. So, BIOS-3 is the first experimental implementation of V.l. Vernadsky's idea about the noosphere - habitable Biosphere controlled by human intelligence. Contrary to predictions of many environmentalists the closedness of the ecosystem is a factor that does not reduce, but increases its sustainability and makes its use for reliable life support outside the Earth realistic. The system is sustainable owing to permanent feedback between the monitoring of few key parameters of the system and automatic corrective actions on them. Main object of control is photo-biosynthesis regenerating parameters of human habitat disturbed by his vital activities. This principle has been realized in BIOS system and proved its reliability in long-term experiments. A new challenge is specified-optimal increase of trophic closedness of the system by reproduction within it essential proteins (peptides and amino acids), lipids, vitamins and other essential compounds. Alternative lines of attack on this problem by state-of-the-art biotechnological methods, GMO including, are under analysis. Reduced BIOS version - without complete closure - can be a breakthrough instrument to improve the quality of life of people living under extreme conditions on the Earth - in polar latitudes (Arctic, Antarctic), in deserts, in high mountains.

To sustain the LC-PUFA supply from natural populations, one needs to know how environmental and intrinsic factors affect fish fatty acid (FA) profiles and contents. We studied seven Salmoniformes species from two arctic lakes. We aimed to estimate differences in the FA composition of total lipids and two major lipid classes, polar lipids (PL) and triacylglycerols (TAG), among the species and to evaluate LC-PUFA contents corresponding to PL and TAG in muscles. Fatty acid profiles of PL and TAG in all species were characterized by the prevalence of omega-3 LC-PUFA and C16-C18 monoenoic FA, respectively. Fish with similar feeding spectra were identified similarly in multivariate analyses of total lipids, TAG and PL, due to differences in levels of mostly the same FA. Thus, the suitability of both TAG and total lipids for the identification of the feeding spectra of fish was confirmed. All species had similar content of LC-PUFA esterified as PL, 1.9-3.5 mg g(-1), while the content of the TAG form strongly varied, from 0.9 to 9.8 mg g(-1). The LC-PUFA-rich fish species accumulated these valuable compounds predominately in the TAG form.

Аннотация: Porous films have been prepared from degradable polymers—poly-3-hydroxybutyrate (PHB), poly-?-caprolactone (PCL) and a blend of these polymers (1:3)—by adding porogen (camphor) to the polymer solution at 10%, 30% or 50% of the total mass of the polymer and porogen, and leaching it out afterwards. After the rinse, camphor content in films decreased to about 0.025%. The structure, physical/mechanical and biological properties of the films were investigated as dependent on their composition and porosity, which varied depending on the amount of camphor added. The surface of PHB films was porous, the PCL films were relatively smooth, and the PHB/PCL films had an intermediate structure. The addition of camphor increased the thickness (from 35 to 45 µm, from 40 to 80 µm and from 20 to 65 µm for PHB, PCL and PHB/PCL, respectively) and porosity (from 4.2(±3.6)% to 50.0(±12.8)%, from 6.4(±5.5)% to 54.5(±6.0)% and from 4.9(±4.8)% to 51.5(±5.8)%, respectively) of the films. The introduction (and removal) of 10% camphor into the PHB and PHB/PCL films led to an approximately twofold increase in the polar component of the free surface energy (from 5.4 ± 0.38 to 11.8 ± 1.33 and from 2.7 ± 0.13 to 5.2 ± 0.09 mN/m, respectively) but in other cases, on the contrary, a decrease in this indicator was registered. The increase of camphor addition from 0% to 50% gradually impaired mechanical properties of the films: so, Young’s modulus decreased from 3.6 to 1.8 GPa, from 0.30 to 0.12 GPa and from 0.50 to 0.20 GPa for PHB, PCL and PHB/PCL, respectively. At the same time, the water vapor transmission rate considerably increased from 197.37 ± 23.62 to 934.03 ± 114.34 g/m2 /d for PHB films; from 1027.99 ± 154.10 to 7014.62 ± 280.81 g/m2 /d for PCL films; and from 715.47 ± 50.08 to 4239.09 ± 275.54 g/m2 /d for PHB/PCL films. Results of biocompatibility testing in the culture of NIH 3T3 mouse fibroblast cells showed that for the most of experimental samples cell adhesion and proliferation were comparable or superior to the corresponding parameters on the initial nonporous films. The best results were obtained for PHB films where at Day 3 of the experiment the registered cell density for experimental samples arrived at 2.66(±0.26) ? 105 cells/cm2 versus 1.29(±0.33) ? 105 cells/cm2 in the control. So, the proposed method can be used to construct highly porous cell scaffolds for cellular engineering. © 2020 by the authors. Licensee MDPI, Basel, Switzerland.

Аннотация: Films from biodegradable poly-3-hydroxybutyrate are treated with chemical reagents to improve their hydrophilicity and biocompatibility. Two approaches are tested: a single treatment with alkali, acids, oxidizing or reducing agents, and a step-by step treatment of the alkali pre-activated surface of polymer films with bromine water and amino-compounds (ammonia or triethylamine). The maximal level of hydrophilicity (the lowest water contact angle and the highest polar component of the surface free energy) is registered after a single treatment with NaOH and after the step-by-step treatment. These samples also showed the best adhesion of mouse fibroblasts of NIH 3T3 line on the film surface. So, the proposed methods can be used to enhance hydropilicity and biocompatibility of biopolymer surface. © 2021 Wiley-VCH GmbH

Аннотация: We present the presence/absence species list (Table 1) of rotifer, cladoceran, and copepod (Calanoida, Harpacticoida, and Cyclopoida) fauna from seven Arctic regions of Russia (the Kola Peninsula, the Pechora River Delta, the Bolshezemelskaya tundra, the Polar Ural, the Putorana Plateau, the Lena River Delta, and the Indigirka River Basin) based on our own and literature data. Our own records were obtained by analyzing samples of zooplankton, meiobenthos, and two cores of bottom sediments (from the Kola Peninsula and the Bolshezemelskaya tundra lakes) that we collected once in July or August in 1992, 1995–2017. To supplement the list, we used relevant literature with periods of research from the 1960s to the 2010s. The list is almost identical to “Dataset 2: Zooplankton and Meiofauna across Arctic Regions of Russia”, which was analyzed but not published in [1]. The detailed analysis of this list revealed the specific composition of the aquatic fauna associated with the climatic and geographical factors [1]. The data provide information on the current state of biodiversity and species richness in Arctic fresh waters and can serve as the basis for monitoring these environments and predicting how they are likely to change in the future. © 2021
