Аннотация: Sustainability is one of the most important criteria and evaluation of human life support systems intended for use during long space flights. The common feature of biological and physicochemical life support systems is that basically they are both catalytic. But there are two fundamental properties distinguishing biological systems: 1) they are auto-catalytic: their catalysts - enzymes of protein nature - are continuously reproduced when the system functions; 2) the program of every process performed by enzymes and the program of their reproduction are inherent in the biological system itself - in the totality of genomes of the species involved in the functioning of the ecosystem. Actually, one cell with the genome capable of the phenotypic realization is enough for the self-restoration of the function performed by the cells of this species in the ecosystem. The continuous microalgal culture of Chlorella vulgaris was taken to investigate quantitatively the process of self-restoration in unicellular algae population. Based on the data obtained, we proposed a mathematical model of the restoration process in a cell population that has suffered an acute radiation damage. В© 2003 COSPAR. Аннотация: The study was conducted to determine the biological value of proteins synthesized by hydrogen-oxidizing microorganisms-the hydrogen bacteria Alcaligenes eutrophus Z1 and Ralstonia eutropha B5786 and the CO-resistant strain of carboxydobacterium Seliberia carboxydohydrogena Z1062. Based on a number of significant parameters characterizing the biological value of a product, the proteins of hydrogen-oxidizing microorganisms have been found to occupy an intermediate position between traditional animal and plant proteins. The high total protein in biomass of these microorganisms, their complete amino acid content, and availability to proteolytic enzymes allow for us to consider these microorganisms as potential protein producers. Аннотация: The dynamics of accumulation of polyhydroxybutyrate (PHB) and the activities of key enzymes of PHB metabolism (?-ketothiolase, acetoacetyl-CoA reductase, PHB synthase, D-hydroxybutyrate dehydrogenase, and PHB depolymerase) in the hydrogen bacterium Ralstonia eutropha B5786 were studied under various conditions of carbon nutrition and substrate availability. The highest activities of ?-ketothiolase, acetoacetyl-CoA reductase, and PHB synthase were recorded during acceleration of PHB synthesis. The activities of enzymes catalyzing PHB depolymerization (PHB depolymerase and D-hydroxybutyrate dehydrogenase) were low, being expressed only upon stimulated endogenous PHB degradation. The change of carbon source (CO2 or fructose) did not affect the time course of the enzyme activity significantly.

Аннотация: Accumulation of americium ( 241Am) and plutonium ( 238,242Pu) and their distribution in cell compartments and biochemical components of the biomass of freshwater aquatic plants Elodea canadensis, Ceratophyllum demersum and Myrioplyllum spicatum and aquatic moss Fontinalis antipyretica have been investigated in laboratory experiments. Americium and plutonium taken up from water by Elodea canadensis apical shoots were mainly absorbed by structural components of plant cells (90% for 241Am; 89% for 238Pu and 82-87% for 242Pu). About 10-18% of isotope activity was recorded in the cytosol fraction. The major concentration (76-92%) of americium was bound to cell wall cellulose-like polysaccharides of Elodea canadensis, Myriophyllum spicatum, Ceratophyllum demersum and Fontinalis antipyretica, 8-24% of americium activity was registered in the fraction of proteins and carbohydrates, and just a minor concentration (<1%) in the lipid fraction. The distribution of plutonium in the biomass fractions of Elodea was similar to that of americium. Аннотация: The chemical composition of water conditioned by colonies of Moina macrocopa and involved in the regulation of their life cycle was analyzed in comparison with water samples from natural water bodies. Dechlorinated tap water was used as the control medium. The results showed that metabolism of crustaceans leads to an increase in the concentrations of phosphorus and nitrogen in all forms, the appearance of protein and carbohydrate fractions, and changes in the composition of lipid fractions in the water conditioned by their colonies. The contents of all organic components are very low, which makes their identification difficult. Аннотация: The method of atom-atom potentials has been used to study the glutamyl dipeptide in ionized (glu-) and non-ionized (glu) states. A calculation has been made with a computer of the conformational maps and the statistical sums of glu and glu-. It was found that on transition of glu > glu- fall in the energy exceeds the free energy of initiation of the helix with an increased probability of the helical state of the molecule of glu-. Аннотация: One of the ways of solving the problem of the human liquid wastes utilization in bioregenerative life support systems (BLSS) can be the use of halophytic vegetable plant Salicornia europaea capable of accumulating sodium chloride in rather high concentrations. Since the most specific higher plant function in BLSS, which at present cannot be substituted by physicochemical processes, appears to be the biosynthesis of a wide spectrum of nutritive substances necessary for a human, the object of the given work was the investigation of the S. europaea productivity, biochemical and mineral composition when grown under close to optimal BLSS vegetative component conditions. As the use of human urine after its preliminary physicochemical processing is supposed to be the mineral solution basis for the S. europaea cultivation, it is necessary to clear up the effect of reduced nitrogen on plants growth. Ground research was carried out. Biochemical composition of the S. europaea edible part showed that crude protein was contained in the highest degree. At that the content of crude protein (24% per dry weight) and cellulose (4.7% per dry weight) was higher in the plants grown on solutions containing amide nitrogen in comparison with the plants grown on solutions with nitrate nitrogen (15.4%-3.1% correspondingly). The water-soluble sugar contents were not high in the S. europaea edible part and depending on the nitrogen nutrition form they amounted to 1.1% (amide nitrogen) and 1.5% (nitrate nitrogen). The polysaccharide number (except cellulose) was rather higher and varied from 7.7% to 8.2%. Although the lipid content in the S. europaea plants was relatively low (7% per dry weight), it was shown that the plant lipids are characterized by a high nonsaturation degree mainly due to alpha linolenic and linoleic acids. Nitrogen nutrition form did not significantly affect the S. europaea productivity, and dry edible biomass of one plant was 8.6 g. Sodium and its concentrations predominated in the plant mineral composition and amounted in average to 9% per dry weight. Thus the S. europaea being the vegetable plant it can be the source of several biochemical substances and essential fatty acids. Аннотация: Most scenarios for a manned lunar base include a combination of physical-chemical and bioregenerative life support systems. Especially on the lunar surface, however, there is a series of special environmental factors which seriously affect the organisms suitable for food production and biological regeneration of the habitat atmosphere and water. So, e.g. the lunar day/night period creates difficult problems for higher plant culture. The paper presents the current scientific approaches to bioregenerative life support systems of a lunar base and discusses critically the possibilities of their realization. Moreover, a scientific strategy is developed with the biologist's point of view to implement in a stepwise manner bioregenerative life support modules into a lunar base covering the possibilities of the untilization of chemolytotrophic bacteria, microalgae and higher plants as well as those of animal breeding and protein production in intensive aquaculture systems. Аннотация: Genes encoding luminescence of Photobacterium leiognathi have been cloned in Escherichia coli. The luminescent clones were readily apparent. Among them, a clone containing a recombinant plasmid with a 13.5-kb insertion was identified. This DNA fragment contained all of the luminescence-encoding genes. The luciferase-encoding genes (lux) in this DNA fragment were localized. We have sequenced a part of the cloned lux region and identified the luxA, luxB and luxG genes encoding the ? and ? subunits of luciferase and a ? protein with an Mr of 26 180, respectively. The analysis of deduced amino acid sequences and comparison with known luciferase sequences from Vibrio harveyi, indicate the common origin of these proteins. В© 1990.

Аннотация: Bioregenerative life support systems (BLSS) with different coefficients of closure are considered. The 66.2% coefficient of closure achieved in "BIOS-3" facility experiments has been taken as a base value. The increase in coefficient of closure up to 72.6-93.0% is planned due to use of soil-like substrate (SLS) and concentrating of urine. Food values were estimated both in a base variant ("BIOS-3"), and with increases in the coefficient of closure. It is shown that food requirements will be more fully satisfied by internal crop production with an increase in the coefficient of closure of the BLSS. Changes of massflow rates on an 'input-output' and inside BLSS are considered. Equations of synthesis and degradation of organic substances in BLSS were examined using a stoichiometric model. The paper shows that at incomplete closure of BLSS containing SLS there is a problem of nitrogen balancing. To compensate for the removal of nitrogen from the system in urine and feces, it is necessary to introduce food and a nitrogen-containing additive. В© 2005 COSPAR. Published by Elsevier Ltd. Аннотация: Ca{2+}-Регулируемый фотопротеин обелин определяет свечение морского гидроида Obelia longissima. Биолюминесценция инициируется кальцием и возникает вследствие окислительного декарбоксилирования связанного с белком субстрата целентеразина. Люцифераза светящегося морского коралла Renilla muelleri (RM) также использует целентеразин в качестве субстрата. Однако в биолюминесценцию этих животных in vivo вовлечено три белка: люцифераза, зеленый флуоресцентный белок и Ca{2+}-регулируемый целентеразин-связывающий белок (coelenterazine-binding protein (CBP)). Фактически, "субстратом" люциферазы RM в биолюминесцентной реакции in vivo является CBP, содержащий одну молекулу прочно связанного целентеразина. Целентеразин становится доступным для реакции с люциферазой и кислорода только после связывания с CBP ионов кальция. В отличие от Ca{2+}-регулируемых фотопротеинов в молекуле CBP не происходит активации кислородом молекулы целентеразина. В работе с помощью квантово-химических методов исследовано поведение субстратов в данных белках. Показано, что целентеразин может образовывать различные таутомерные формы - CLZ(2H) и CLZ(7H). Исследован механизм образования 2-гидропероксицелентеразина. Согласно полученным данным, эти белки используют для реакции различные формы субстратов: в обелине субстрат находится в форме CLZ(2H), из которой и происходит образование гидроперекиси. В RM целентеразин находится в форме CLZ(7H), и поэтому активации кислородом в CBP не происходит

Аннотация: Electron microscopical studies of the nucleoid structure of hydrogen bacteria using ultrahin sections and spread DNA from bacterial cell lysates revealed a different DNA packaging in the cell. A compact state of the major part of DNA at all growth stages and stability of nucleosome-like structures were shown. The use of antibodies to HU protein of E. coli labelled by protein A-colloidal gold demonstrated the immunological relationship between HU protein of E. coli and histone-like proteins of Alcaligenes eutrophus and their possible role in the nucleosome-like DNA packaging in procariotic genome.
